From Lysosomal Storage to Neurodegeneration: Sphingolipid Signaling as a Driver of CNS Pathology and Biomarker Strategy in Neuronopathic Gaucher Disease.

Gaucher disease is a prototypical lysosomal sphingolipid storage disorder caused by pathogenic variants in GBA1, resulting in glucocerebrosidase deficiency and accumulation of bioactive lipids, including glucosylceramide and glucosylsphingosine (lyso-Gb1). While non-neuronopathic Gaucher disease is effectively managed with enzyme replacement and substrate reduction therapies, neuronopathic forms remain largely refractory to treatment due to progressive central nervous system (CNS) involvement and limited penetration of current therapies across the blood-brain barrier. Targeting the cytokine-redox axis: dose-dependent immunomodulation by methotrexate in a biomarker-guided ovine endotoxaemia model.

BACKGROUND: Oxidative stress and excessive inflammation are central to the pathophysiology of sepsis and its large-animal analogue, endotoxaemia. Current anti-inflammatory treatments in ruminants lack efficacy in modulating redox balance or acute-phase responses. Low-dose methotrexate (MTX), a known immunometabolic modulator, may offer targeted attenuation of these processes. OBJECTIVE: To evaluate the anti-inflammatory, antioxidant, and organ-sparing effects of two low-dose MTX regimens in a controlled ovine model of lipopolysaccharide (LPS)-induced endotoxaemia. METHODS: 20 clinically heal

Expressive writing to improve resilience to trauma: A clinical feasibility trial.

BACKGROUND AND PURPOSE: Trauma is highly prevalent, with estimates that up to 90% of the U.S. population have been exposed to a traumatic event. The adverse health consequences of trauma exposure are diverse and often long-lasting. While expressive writing has been shown to improve emotional and physical health in numerous populations, the feasibility and potential effectiveness of a novel expressive writing program provided in a clinical setting to improve resilience is unknown. Our objective was to determine the feasibility and potential effectiveness of a 6-week expressive writing course pr

Integrated Transcriptomic and Functional Analyses Identify SERPINE1 as a Ferroptosis-Associated Mediator of Glioblastoma Radioresistance.

BACKGROUND: Radiotherapy failure remains a major challenge in glioblastoma (GBM), yet the stress-adaptive programs that enable tumor cells to survive irradiation-induced lethal injury remain incompletely defined. This study investigated whether serpin family E member 1 (SERPINE1) contributes to GBM radioresistance by modulating ferroptosis-associated lipid peroxidation, a process linked to iron-dependent oxidative injury. METHODS: Radiotherapy-treated GBM cohorts from TCGA (n = 93; 56 radiosensitive and 37 radioresistant cases), the Chinese Glioma Genome Atlas (CGGA)-325 (n = 100), and CGGA-6

Transcranial direct current stimulation promotes functional recovery in rats with traumatic brain injury by inhibiting SLC7A11-mediated Disulfidptosis and Neuroinflammation.

BACKGROUND: Traumatic brain injury (TBI) lacks effective pharmaceutical treatments. While transcranial direct current stimulation (tDCS) shows promise in neurorehabilitation, its potential to mitigate disulfidptosis, a novel cell death mechanism driven by disulfide stress, remains unexplored. OBJECTIVE: This study aimed to investigate whether tDCS facilitates neurofunctional recovery in TBI by modulating SLC7A11-associated disulfidptosis. METHODS: A rat model of TBI was established using controlled cortical impact. Integrated transcriptomic and untargeted metabolomic analyses were performed

Comorbidity risk characteristics of rheumatoid arthritis in the context of depression-associated lipid metabolism.

BACKGROUND: Rheumatoid arthritis (RA) and major depressive disorder (MDD) exhibit significant comorbidity, with shared pathological mechanisms such as inflammation and abnormal lipid metabolism. However, the specific molecular features and functional pathways linking the two diseases remain unclear. METHODS: This study integrated public transcriptomic data from five MDD and five RA cohorts. Lipid metabolism-related module genes were screened in MDD cohorts and intersected with RA-upregulated differentially expressed genes to construct a candidate set. Placenta at single-cell resolution in early and late preeclampsia: insights and clinical implications.

Preeclampsia, one of the great obstetrical syndromes, manifests through diverse maternal and fetal complications and remains a leading contributor to adverse perinatal outcomes. In this review, we describe our work on single-cell and single-nuclei RNA sequencing to elucidate the molecular mechanisms that underlie early- and late-onset preeclampsia. Analysis of 46 cell types, encompassing approximately 90,000 cells from placental tissues collected after delivery, demonstrated cellular dysregulation in early-onset preeclampsia, whereas late-onset preeclampsia showed comparatively subtle changes.

Varying black soldier fly (Hermetia illucens) levels as a fish meal replacement in Nile tilapia (Oreochromis niloticus).

The quest for sustainable alternative protein sources in aquaculture has been driven by rising costs and limited supply of traditional fish meal; insect meals have emerged as viable options. Among these, the high nutritional value and environmental sustainability of black soldier fly larval (BSFL) diet have garnered attention. The purpose of this study was to assess how a dietary BSFL meal affected the Nile tilapia's (Oreochromis niloticus) growth performance, antioxidant capacity, immunological response, digestive enzyme activity, and histological features. For 60 days, fish were fed diets wi

Staphylococcus aureus Strain Newman Photoinactivation and Cellular Response to Sunlight Exposure.

Sunlight influences microbial water quality of surface waters. Previous studies have investigated photoinactivation mechanisms and cellular photostress responses of fecal indicator bacteria (FIB), including Escherichia coli and enterococci, but further work is needed to characterize photostress responses of bacterial pathogens. Here we investigate the photoinactivation of Staphylococcus aureus (strain Newman), a pigmented, waterborne pathogen of emerging concern. We measured photodecay using standard culture-based assays and cellular membrane integrity and investigated photostress response by

Variable role of carotid bodies in cardiovascular responses to exercise, hypoxia and hypercapnia in spontaneously hypertensive rats.

KEY POINTS: Carotid bodies play a critical role in maintaining arterial pressure during hypoxia and this has important implications when considering resection therapy of the carotid body in disease states such as hypertension. Curbing hypertension in patients whether resting or under stress remains a major global health challenge. We demonstrated previously the benefits of removing carotid body afferent input into the brain for both alleviating sympathetic overdrive and reducing blood pressure in neurogenic hypertension. We describe a new approach in rats for selective ablation of the carotid

[Systems of chemoperception in decapod crayfish].

The review presents data on some peripheral and central structures in the system of perception of chemical stimuli in crayfish and other Decapoda. The hair receptors on chelipeds, antennas, antennules are innervated by mechano-and chemoreceptor neurons. Antennules are the specialized crayfish chemoreceptor organs, on the surface of which groups of exteroceptors are located. There is an ordered disposition of exteroceptor receptive fields in the form of receptor hair bushes on claws of ambulatory feet (AF), antennas, antennules, and other movable appendages. Neuroendocrine response to CRF stimulation in veterans with and without PTSD in consideration of war zone era.

BACKGROUND: Alterations in hypothalamic-pituitary-adrenal (HPA) axis activity have been observed in Gulf War veterans with posttraumatic stress disorder (PTSD) which differ from those observed in other veteran groups, raising the possibility that there is a unique neuroendocrine profile in this group of veterans. This study seeks to further characterize the effects of PTSD, military cohort (Vietnam, 1991 Gulf War, Operations Enduring Freedom/Iraqi Freedom (OEF/OIF)), and their interaction on the neuroendocrine response to synthetic corticotrophin-releasing factor (CRF) stimulation. Endothelium-targeted and untargeted nanoparticle therapies in thromboinflammatory vascular diseases.

Endothelial cells (ECs) line the vasculature and integrate hemodynamic, inflammatory, and metabolic cues that are crucial in maintaining vascular health. Endothelial dysfunction is a driver of thromboinflammatory conditions including atherosclerosis, thrombosis, stroke, and sepsis-induced multiorgan dysfunction syndrome. Therapies for these conditions that are given systemically fail to target the site of disease and can be associated with off-target side effects. EC-targeted nanoparticles represent a novel method of concentrating the drug- or gene-based therapy in ECs at the site of disease.

Integrative Phosphoproteomic Characterization of ULK1 Phosphosites Reveals Co-Regulatory Networks in Autophagy and Metabolic Adaptation.

Unc-51-like autophagy-activating kinase 1 (ULK1), a serine/threonine kinase, is the central regulator of autophagy initiation and plays a crucial role in cellular homeostasis, metabolic adaptation, and stress responses. We performed a large-scale phosphoproteomic data analysis of ULK1 by systematically integrating and analyzing 891 profiling and 245 differential human phosphoproteomic datasets. S450, S623, and S638 emerged as the predominant phosphosites in ULK1, with the highest frequency of detection across multiple cellular phosphoproteomics datasets. Conservation analysis showed that these

Entomopathogenic nematode recent applications in precision agriculture: Methods, challenges, and future perspectives.

This review examines recent advances in the application technologies of entomopathogenic nematodes (EPNs) as sustainable biological control agents in agricultural systems. Although synthetic pesticides have long been used for pest management, their adverse effects on non-target organisms and the environment have raised significant concerns, leading to increasing restrictions and a growing demand for alternative strategies. In this context, EPNs have emerged as promising biocontrol agents due to their effectiveness against a wide range of insect pests, safety for humans and the environment, and

Integrated multi-omics analysis reveals shared genetic and immunological biomarkers linking arsenic-induced autophagy to carcinogenesis.

Autophagy is a long-standing biological mechanism that degrades and recycles damaged cellular components. It has become a significant mediator of arsenic poisoning, but its role is complicated and depends on the situation. Chronic inorganic arsenite exposure is a considerable global environmental health issue, strongly linked to multiple cancer types via processes that remain inadequately elucidated. This research employed an integrated multi-omics approach to investigate the molecular network linking inorganic arsenite exposure to cancer progression via disruption of autophagy, examining gene

Neuroimmune State Configurations at the Amyloid-Tau Interface in Alzheimer's Disease: A Testable Framework.

Neuroimmune mechanisms are central to Alzheimer's disease (AD), but a single continuum of "more inflammation" does not explain why similar amyloid-β (Aβ) and tau burdens are associated with different clinical trajectories, tissue responses, and treatment risks. We present a testable neuroimmune-state framework that integrates, rather than replaces, the AT(N) system, cellular-phase models, and transcriptional descriptions of disease-associated microglia and reactive astrocytes. The framework treats neuroimmune states as multidimensional, regionally heterogeneous configurations defined by pathol

Potential Relevance of Bidens pilosa L. to Atopic Dermatitis: Mechanistic Evidence, Drug Development Challenges, and Future Perspectives.

Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by epidermal barrier dysfunction, immune dysregulation, oxidative stress, microbial dysbiosis, and persistent inflammation. Although current therapies have improved disease management, long-term treatment remains challenging because of disease relapse, adverse effects, treatment costs, and variability in therapeutic response. These challenges have stimulated interest in botanical agents with diverse phytochemical profiles and potentially complementary biological activities. Bidens pilosa L. has attracted attention beca

Cerebral cavernous malformations: epidemiology, molecular pathogenesis, diagnosis and therapy.

Cerebral cavernous malformations (CCMs) are cerebrovascular diseases characterized by vascular lesions of the central nervous system (CNS) that can result in epileptic seizures, headaches, focal neurologic deficits (FNDs), and hemorrhagic stroke. Germline and somatic loss-of-function (LOF) mutations in one of the three established CCM-associated genes, namely CCM1 (or krev interaction trapped protein 1, KRIT1), CCM2, and CCM3 (or programmed cell death 10, PDCD10), which have been validated as core drivers of CCM pathogenesis and progression.
