Valproic acid as a therapeutic scaffold: Advances in structural modification and drug design.

Valproic acid (VPA), a short-chain branched fatty acid with a broad pharmacological profile, remains one of the most versatile molecules in neuropsychopharmacology. Beyond its established use in epilepsy and bipolar disorder (BD), VPA functions as a histone deacetylases (HDACs) inhibitor, revealing significant potential in oncology and other therapeutic areas. However, its clinical application is limited by serious adverse effects, notably hepatotoxicity and teratogenicity. Owing to its simple and modifiable chemical structure, VPA has served as a valuable scaffold for the development of new a

Design, Synthesis, and Study of Prenylated Carbazole Derivatives as Potential Antimicrobial Agents.

Crop diseases caused by agricultural pathogens are a constant threat to food security. New antimicrobial agents are urgently required to address the toxicity and resistance issues of existing commercial agents. Inspired by the active fragments of natural prenylated carbazole alkaloids, this study synthesized a series of prenylated carbazole derivatives using a skeleton hybridization and fragment splicing strategy. Compounds C1-4, C1-15, and C1-16 demonstrated broad-spectrum in vitro antimicrobial effects which were superior to commercial drugs thiodiazole copper and bismerthiazol. In particula

Exploration of 4'-fluoro fleximer nucleoside analogues as potential broad-spectrum antiviral agents.

A series of 4'-fluoro fleximer nucleoside analogues in addition to their corresponding prodrugs were designed, synthesized, and evaluated for their antiviral potential. The impetus for this series was based on the success of other 4'-modified nucleoside antiviral drugs such as adafosbuvir and balapiravir. One analogue, KAD-008, exhibited potent, low single-digit micromolar activity against DENV-2. Another analogue, KAD-025, had broad-spectrum antiviral activity across several viral families including flaviviruses, filoviruses, and coronaviruses. All of the analogues tested showed no toxicity u

N,N'-bridged heterocyclic bis-iminium salts as potent antimalarial agents against multi-resistant Plasmodium falciparum.

The emergence of Plasmodium resistance to past and newly introduced antimalarial drugs reinforces the need for new antimalarial agents with innovative mechanisms of action. In this regard, the search for new redox-cycling compounds might offer an interesting approach to treat malaria since P. falciparum is sensitive to oxidative stress. Based on our experience in the design of powerful organic reducers, we undertook here the synthesis and evaluation of different families of N-heterocyclic iminium salts that could provide more selective redox-cycling candidates against the K1 multi-resistant Pl

Rational design of heparin antagonist: Guanidine-based mimetics unveil key carbohydrate-carbohydrate interactions.

Small-molecule inhibitors targeting heparin (HP)-protein interactions represent a promising strategy for developing therapeutic agents against serious bleeding complications. Herein, we report a rational design and synthesis of a library of eight trisaccharide HP mimetics incorporating positively charged guanidinium residues aimed at disrupting the ionic interactions of HP and modulating HP-mediated biological activities. The introduction of guanidine residue in HP backbone significantly influenced the conformational plasticity of l-idose and l-iduronic acid, shifting the major 4C1-conformatio

New Structure Activity Relationship Insight into the Role of the C-3 Extension on Rifamycin Antimycobacterial Activity.

Herein, the antimycobacterial screening of a series of rifamycin analogues, modified at their C-3 extension, is reported. Overall, these compounds display potent activity against a wild-type Mtb strain assayed in three different growth media. Several promising C-3 extensions are identified through this screen, with compounds featuring rigid tertiary alicyclic hydrazones displaying superior activity to amino compounds. In addition, a general correlative trend between logP and biological activity is observed. This study adds to the growing literature surrounding structure activity relationship p

Balinatunfib: A Clinical Oral Small Molecule TNFα Inhibitor.

Most diseases are accompanied by an inflammatory response, making effective pharmacological control highly desirable. Tumor necrosis factor alpha (TNFα) is a key cytokine driving inflammatory and autoimmune diseases, such as rheumatoid arthritis and inflammatory bowel disease. Although biological TNFα inhibitors revolutionized treatment, they have drawbacks including lacking blood-brain barrier penetration, parenteral administration, and immunogenicity. Recent studies highlight the potential of small-molecule approaches to target TNFα by stabilizing an asymmetrical, receptor-incompetent trimer

Synthesis and structure-activity relationship of azithromycin derivatives against non-tuberculous mycobacteria.

Non-tuberculous mycobacteria (NTM), particularly Mycobacterium avium complex (MAC), have emerged as significant pulmonary pathogens. Current first-line treatments for pulmonary MAC disease typically involve the macrolide azithromycin (AZM). However, the increasing prevalence of drug-resistant MAC has created an urgent need for new therapeutics. Here, we present a novel series of Glc-AZM derivatives, KU15 to KU27, featuring C11 glucosyl moieties designed to anchor the macrolide to the ribosome and enhance antibacterial activity. These compounds were selectively synthesized using our boron-media

Advancing N,N'-disubstituted diamines in antimalarial drug development: Structure-activity relationships and mechanistic studies.

Previously we reported the synthesis of thirty N,N'-disubstituted diamines, several of which exhibited potent activity against apicomplexan parasites, including Plasmodium falciparum and Toxoplasma gondii. Building on this, we expanded the series with fifty-four new compounds, which were evaluated against the same parasites. These analogs were synthesized via a one-pot reductive amination of aliphatic diamines with various aromatic aldehydes. The new library showed great potential, with several compounds achieving a pIC50 greater than 6.0 against P. falciparum and with an acceptable selectivit

Structure-activity relationship studies of tanzawaic acid A, an antifungal agent against Rhizopus oryzae.

Through structure-activity relationship studies of tanzawaic acid A (TZAA), this research aims to contribute the development of potent antifungal drugs for treating mucormycosis infections. To this end, new TZAA analogs bearing chemical modifications in the side chain and/or C8-methyl group moieties were synthesized. These analogs are not accessible from natural TZAA and therefore required total synthesis. As a result, four of the newly synthesized TZAA analogs exhibited more potent and selective inhibitory activity against R. oryzae compared to TZAA.

Discovery of novel NLRP3 inhibitors enabled by a high-throughput screen.

NLRP3 is a key regulator of the innate immune system involved in sensing a variety of pathogen and danger signals. Priming and activation of NLRP3 leads to the release and maturation of pro-inflammatory cytokines, as well as gasdermin D-mediated cell death. Inhibition of dysregulated NLRP3 activity has been associated with promising therapeutic opportunities for a variety of systemic and neurological diseases including atherosclerosis and Parkinson's disease. Herein, we discuss how a high-throughput screen (HTS) allowed us to discover new chemical scaffolds that specifically bind to NLRP3 and

Fine-tuning 5-HT7 receptor selectivity, inverse agonism, and metabolic stability of new (aryloxy)ethyl-piperidines toward antidepressant and pro-cognitive properties.

Affective disorders, the leading causes of disability and premature death worldwide, require new and effective treatment strategies. Clinically used antidepressants and second-generation antipsychotic drugs, including vortioxetine and lurasidone, act as potent 5-HT7 receptor antagonists and improve cognitive functions in the patients with mood disorders. Additionally, 5-HT7 receptor-mediated activation of matrix metalloproteinase 9 (MMP-9) induces depressive-like behavior in mice. We designed and synthesized a series of 27 arylsulfonamide derivatives of 2-[(2-aryl/2-heteroaryl)phenoxy]ethyl-pi

Gallium Hematoporphyrin inhibits Mycobacteroides abscessus in vitro, including biofilm formation and stability.

The nontuberculous mycobacterial (NTM) pathogen, Mycobacteroides abscessus, can cause severe pulmonary infections. Emerging multidrug resistance to current antibiotics poses significant challenges for treatment of M. abscessus infections. Thus, new antibiotics are needed, preferably ones that target new microbial pathways. Earlier, we examined the in vitro inhibitory activities of several gallium compounds, Ga(NO3)3, GaCl3, gallium meso-tetraphenylporphyrin (GaTP) and gallium nanoparticles (GaNP), against intra- and extracellular M. abscessus. We have previously shown that these gallium compou

Targeting virulence to disarm the pathogen: Enzyme-activated-substrate inhibition of Anthrax edema factor.

Since the discovery by Fleming that a mold could inhibit the growth of bacteria, each new antibiotic developed to treat infections quickly lost its efficacy due to the emergence of resistant strains. As a result, the ongoing threat by antibiotic-resistant pathogens would benefit from new strategies to combat bacterial infections. An ideal drug is one which is efficacious, can limit selective pressure against the pathogen, and potentially augment the currently available antibiotics to restore their efficacy. Targeting virulence factors used by bacteria to establish infections has the potential

Amoebicidal thymol analogues against brain-eating amoeba, Naegleria fowleri.

Naegleria fowleri, known as the brain-eating amoeba, is the pathogen parasite that causes primary amoebic meningoencephalitis. None of the currently available therapies are fully effective, mainly due to the inefficacy of pharmacotherapy. In this regard, natural products and related compounds represent a promising strategy for amoebicidal drug discovery. Herein, a series of eight monoterpene phenol derivatives of thymol bearing ester, carbonate, or carbamate moieties were prepared, and screened as potential amoebicidal agents on N. fowleri. The cytotoxicity of these compounds on murine macroph

Accessing Promising Passerini Adducts in Anticancer Drug Design.

The 3-component Passerini reaction (3CPR), discovered little more than 100 years ago, has been demonstrated in the last few decades to be a valuable tool for accessing structural diversity and complexity, essential topics to consider in drug discovery programs. Focusing on accessing a fine-tuned family of α-acyloxyamide-oxindole hybrids, we underline herein our latest insights regarding the use of this mild reaction approach to obtain promising anticancer agents. Cheap and commercially available isatin was used as starting material. The library of α-acyloxyamide-oxindole hybrids was tested aga

Identification of 6,9-dihydro-5H-pyrrolo[3,2-h]quinazolines as a new class of F508del-CFTR correctors for the treatment of cystic fibrosis.

Although substantial advances have been obtained in the pharmacological treatment of cystic fibrosis (CF) with the approval of Kaftrio, a combination of two correctors (VX-661, VX-445) and one potentiator (VX-770), new modulators are still needed to rescue F508del and other CFTR mutants with trafficking defects. We have previously identified PP compounds based on a tricyclic core as correctors with high efficacy in the rescue of F508del-CFTR on native epithelial cells of CF patients, particularly in combination with class 1 correctors (VX-809, VX-661). Compound PP028 was found as a lead candid

Design, synthesis, and activity evaluation of water-soluble propofol derivatives as anesthetic drugs.

In this work, two series of water-soluble derivatives were designed and synthesized based on the structure of propofol as the lead compound. Furthermore, the anesthetic activities of the synthesized compounds were evaluated in vivo against mice, and the in vitro propofol release rate from five target compounds was determined. The findings of this study have shown that series II compounds which possess the structure feature of propofol + γ-hydroxybutyric acid + α-aminoacetate or γ-aminobutyrate have higher therapeutic index than that of series I compounds which possess the structure feature of
