A Ketogenic & Low-Protein Diet Slows Retinal Degeneration in rd10 Mice.

PURPOSE: Treatments that delay retinal cell death regardless of genetic causation are needed for inherited retinal degeneration (IRD) patients. The ketogenic diet is a high-fat, low-carbohydrate diet, used to treat epilepsy, and has beneficial effects for neurodegenerative diseases. This study aimed to determine whether the ketogenic diet could slow retinal degeneration. METHODS: Early weaned, rd10 and wild-type (WT) mice were placed on either standard chow, a ketogenic diet, or a ketogenic & low-protein diet. From postnatal day (PD) 23 to PD50, weight and blood β-hydroxybutyrate levels were

Maternal folic acid supplementation with vitamin B12 deficiency during pregnancy and lactation affects the metabolic health of adult female offspring but is dependent on offspring diet.

Epidemiologic studies have reported relationships between maternal high folate and/or low B12 status during pregnancy and greater adiposity and insulin resistance in children. The goal of this study was to determine the effects of maternal folic acid supplementation (10 mg/kg diet), with (50 μg/kg diet) and without B12, on adult female offspring adiposity and glucose homeostasis. Female C57BL/6J mice were fed 1 of 3 diets from weaning and throughout breeding, pregnancy, and lactation: control (2 mg/kg diet folic acid, 50 μg/kg diet B12), supplemental folic acid with no B12 (SFA-B12), or supple

The Influence of Metabolic Factors and Diet on Fertility.

Infertility is a disease globally affecting 20-30% of the reproductive age female population. However, in up to 50% on recorded cases, problems with infertility are ascribed to men; therefore, it is important to popularize healthy eating also in this group. During the last decade, it has been observed that society's lifestyle changed drastically: reduced energy expenditure in physical activity per day, increased consumption of hypercaloric and high-glycemic-index foods with high content of trans fats, and reduced consumption of dietary fiber, which negatively affects fertility. Increasing evid

Antiketogenic effects of monensin in early lactation.

Holstein cows, second lactation or later, were gradually introduced to monensin-containing concentrate 1 wk prepartum and fed complete diets containing 15 and 30 g monensin/ton of DM for 3 wk postpartum. The addition of 30 g monensin/ton of feed decreased the incidence of subclinical ketosis from 6 out of 12 to 1 out of 12. The concentration of beta-hydroxybutyrate in blood from cows in the high monensin group was decreased during the 3-wk postpartum experimental period. Acetate: propionate ratios decreased from 2.32 in the control group to 1.44 in the high monensin group. Feed intake in the l

n-3 Polyunsaturated fatty acids protect against pancreatic β-cell damage due to ER stress and prevent diabetes development.

SCOPE: In this study, we focus on the effects of n-3 polyunsaturated fatty acids (PUFAs) on tunicamycin-, streptozotocin-, or high fat diet (HFD)-induced β-cell damage and dysfunction. MATERIALS AND METHODS: Pretreatment with n-3 PUFAs protected RINm5F cells and mouse islets against tunicamycin-induced β-cell damage through suppression of ER stress and apoptosis induction. This protective effect of n-3 PUFAs on β-cells was further demonstrated by the normalization of insulin secretion in response to glucose in tunicamycin-treated islets. In multiple low-dose streptozotocin-induced diabetes mo

Impact of weight loss and reduction of ultra-processed foods on liver fat content in MASLD: A randomized controlled trial.

BACKGROUND & AIMS: Dietary interventions are a cornerstone of management for metabolic dysfunction-associated steatotic liver disease (MASLD). However, it remains unclear whether specific dietary patterns confer benefits beyond weight loss. We aimed to compare the effects of different dietary strategies on liver fat and metabolic health in patients with MASLD. METHODS: We conducted a 6-month, three-arm, parallel-group randomized controlled trial including patients with MASLD and overweight or obesity who were naïve to structured nutritional counselling. Participants were randomized to Mediter

A Metagenomics Investigation of Intergenerational Effects of Non-nutritive Sweeteners on Gut Microbiome.

To identify possible mechanisms by which maternal consumption of non-nutritive sweeteners increases obesity risk in offspring, we reconstructed the major alterations in the cecal microbiome of 3-week-old offspring of obese dams consuming high fat/sucrose (HFS) diet with or without aspartame (5-7 mg/kg/day) or stevia (2-3 mg/kg/day) by shotgun metagenomic sequencing (n = 36). High throughput 16S rRNA gene sequencing (n = 105) was performed for dams, 3- and 18-week-old offspring. Maternal consumption of sweeteners altered cecal microbial composition and metabolism of propionate/lactate in their

African American men and hereditary/familial prostate cancer: Intermediate-risk populations for chemoprevention trials.

The risk of prostate cancer diagnosis among African Americans is 66% greater than among European American men. For African Americans with a family history of hereditary prostate cancer the increased risk of diagnosis is even greater. Thus, this population should be a prime target for chemoprevention strategies. In addition to the higher incidence of prostate cancer among African Americans compared with other populations, the mortality of prostate cancer among this high-risk population is significantly greater than 100% compared with other populations, thus further demonstrating the need for ch

Dietary methionine restriction improves the impairment of cardiac function in middle-aged obese mice.

Dietary methionine restriction (MR) has been reported to extend lifespan, reduce obesity and decrease oxidative damage to mtDNA in the heart of rats, and increase endogenous hydrogen sulfide (H2S) production in the liver and blood. H2S has many potential benefits in the pathophysiology of the cardiovascular system. MR also increases the level of homocysteine (Hcy) in the liver and plasma, but elevated plasma Hcy is a risk factor for cardiovascular disease. Therefore, this study aimed to determine the effect of MR on cardiac function and metabolic status in obese middle-aged mice and its possib

Bioaccessibility of Polycyclic Aromatic Hydrocarbons (PAHs) in Grilled Meat: The Effects of Meat Doneness and Fat Content.

Exposure to polycyclic aromatic hydrocarbons (PAHs) through diet is gaining concern due to the risk it poses to human health. This study evaluated the bioaccessibility of PAHs contained in charcoal-grilled beef and chicken in different segments of the gastrointestinal tract (GIT) with regard to the degree of doneness and fat content of the meats. The levels of 15 PAHs in the grilled meat samples and bioaccessible fractions were determined using high-performance liquid chromatography (HPLC) equipped with PAH column, and UV and fluorescence detectors. Total PAHs were found in beef (30.73 ng/g) a

Validation of targets linked to weight gain in C57BL/6 mice and translational aspects in humans.

Obesity is linked to complex interactions between an organism's genetic background and its environment. Mouse models have been invaluable research tools to study obesity, with C57BL/6N being widely studied, given its propensity for weight gain on a high-fat diet (HFD). Despite this, the genetic basis of its susceptibility is poorly understood. In a partner paper we have identified SNP variants in 11 genes causing susceptibility to, or protection against, HFD-induced obesity in male C57BL/6N mice explaining 87% of the individual variation in fat gain. Surprisingly, most of them were maximally e

Investigating the effect of diet-induced prediabetes on the HPA-axis, sympathetic nervous system and endogenous opioids in male Sprague-Dawley rats.

BACKGROUND: Prediabetes is associated with metabolic dysfunction that may disrupt neuroendocrine regulation before the onset of overt type 2 diabetes mellitus. Although previous studies have independently examined alterations in the hypothalamic-pituitary-adrenal (HPA) axis, sympathetic nervous system (SNS), and endogenous opioid system during prediabetes, the temporal interactions among these systems during disease progression remain poorly understood. This study aimed to investigate changes in these interconnected neuroendocrine pathways in a diet-induced prediabetic rats. METHODS: Male Spr

Exercise attenuates atherosclerosis by inhibiting eWAT aging via targeting SIRT1/pyruvate carboxylase pathway.

Aging is the primary risk factor for atherosclerosis (AS). Epididymal white adipose tissue (eWAT) is a key driver of organismal aging and age-related diseases. Exercise is known to protect against AS, but the role of aging eWAT in this process remains unclear. To this end, we investigated whether exercise inhibits AS by regulating aging eWAT and elucidated the involved mechanisms. ApoE-/- and C57BL/6 mice were fed a high-fat diet (HFD) to mimic poor lifestyle habits and explore eWAT senescence evolution. We found eWAT is a senescence-susceptible organ, exhibiting time-dependent deterioration u

Nobiletin ameliorates hepatic insulin resistance by modulating the gut-liver axis.

Insulin resistance (IR) is a core pathological feature of type 2 diabetes mellitus (T2DM), with hepatic IR serving as a hallmark of systemic IR. Nobiletin (NOB) shows great potential in exerting hypoglycemic effects and improving IR; however, its molecular mechanisms remain incompletely elucidated. This study aims to investigate the molecular mechanisms by which nobiletin (NOB) ameliorates hepatic IR. Our results demonstrated that NOB effectively ameliorated IR in both high-fat diet/streptozotocin (HFD/STZ)-induced mice and palmitic acid (PA)-treated HepG2 cells. NOB administration improved dy

Inflammatory Factors Derived From Metabolic Dysfunction-Alcoholic Fatty Liver Disease: Inducers of Anxiety and Spatial Memory Impairment.

Metabolic dysfunction-associated steatotic liver disease (MASLD) has emerged as a global epidemic, with growing evidence suggesting its adverse impact on brain function. However, the underlying mechanisms linking hepatic metabolic dysfunction to neurodegeneration remain unclear. In this study, we systematically investigated the liver-brain axis by integrating genetic epidemiology, experimental neuroscience, and transcriptomics techniques. Two-sample Mendelian randomization (MR) analysis revealed a potential causal relationship between MASLD and cognitive decline. These findings were validated

Maternal overnutrition during pregnancy: impacts on pancreatic proteome of the offspring.

Maternal obesity, resulting from high caloric intake during pregnancy, can impair pancreatic development in the offspring and lead to an increased risk of diabetes. We hypothesized that maternal obesity during pregnancy results in reduced expression of proteins associated with pancreatic β cell function and increased expression of proteins associated with apoptosis. C57BL/6J (N = 6) female mice were fed either a control (CON; n = 3) or a high-fat (HFD; n = 3) diet for fourteen weeks prior to pregnancy, diets were continued throughout gestation and lactation. Offspring were weaned at four weeks

Study on Anti-Obesity Activities of Three Individual Black Soybean Meal-Derived Insoluble Dietary Fibers via Modulating Intestinal Microbiota and Their Metabolites.

Obesity, a global health crisis, is often accompanied by metabolic disorders such as dyslipidemia and insulin resistance. This study investigates the anti-obesity effects of black soybean meal insoluble dietary fiber modified by ultra-high pressure and its impact on gut microbiota and metabolites in high-fat diet-induced obese mice. UHP modification significantly enhanced the oil-holding capacity of IDF (9.47 ± 0.47 g/g) with minimal impact on purity (0.54% reduction). Animal experiments revealed that high-dose HPIDF and UP-IDF significantly inhibited weight gain, improved lipid profiles (redu

A fructo-oligosaccharide and sea buckthorn complex ameliorates glucose and lipid metabolism in type 2 diabetic (T2DM) rats via metabolic and anti-inflammatory pathways.

This study investigated the effects and underlying mechanisms of a fructo-oligosaccharide (FOS) and sea buckthorn complex (FS) on blood glucose and lipid metabolism in type 2 diabetic mellitus (T2DM) rats. The T2DM model was induced by a high-fat diet (HFD) combined with alloxan administration. Prior to rat experiments, the synergistic hypoglycemic and hypolipidemic effects of FOS and sea buckthorn have been verified by zebrafish experiments. FS intervention significantly reduced fasting blood glucose, triglycerides (TG), total cholesterol (TC), and low-density lipoprotein cholesterol (LDL-C)
