Surface-enhanced Raman spectroscopy for breast cancer personalized management:From liquid biopsy and intraoperative navigation to therapy monitoring.

Surface-enhanced Raman spectroscopy (SERS) has emerged as a powerful analytical platform for addressing the critical challenges in breast cancer precision diagnostics. This review systematically examines the recent methodological advances that position SERS as a transformative tool across the entire clinical continuum of breast cancer management-spanning non-invasive liquid biopsy screening, intraoperative surgical margin navigation, and personalized therapeutic guidance-with an emphasis on analytical innovation rather than clinical outcomes alone. We critically assess the core analytical stra

A novel extracellular vesicle (EV)-based nanodrug for tongue squamous cell carcinoma: in vitro efficacy in HNO-97 cells.

BACKGROUND: Squamous cell carcinoma of the tongue (TSCC) is an aggressive malignancy with few therapeutic modalities available. Cisplatin, a commonly used chemotherapeutic drug, is often associated with significant toxicities and resistance. The use of an exosome-enriched fraction of extracellular vesicles (EVs) as a drug delivery system has become an attractive way to potentiate therapeutic effects and decrease toxicity. In particular, our focus is to study the therapeutic effect of cisplatin-loaded EVs obtained from umbilical cord mesenchymal stem cells (UC-MSCs) on TSCC. The goal is to asse

Emerging pathological mechanisms of Alzheimer's disease pathogenesis: from neuroimmune interactions to intercellular communication.

Alzheimer's disease (AD) research has transcended the traditional paradigm centered on amyloid-beta (Aβ) shifting toward a neuroimmune network perspective. This article systematically elucidates the evolving mechanisms underlying disease progression, from neuroimmune interactions to intercellular communication. Studies indicate that microglial and astrocytic dysfunctions are key contributors to disease progression, operating within a complex multifactorial framework. Upon transformation into disease-associated microglia (DAM), microglia exhibit a significant decline in Aβ clearance capacity an

Mechanistic analysis and clinical translation strategies of m6A erasers in the metabolism-immunity axis of hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is a malignant tumor with high heterogeneity and immunotherapeutic resistance worldwide, presenting a severe challenge in clinical prevention and treatment. N6-methyladenosine (m6A) is the most common reversible post-transcriptional modification on eukaryotic mRNA, whose dynamic balance is precisely regulated by "demethylases" (Erasers). Recent studies have confirmed that the two core m6A Erasers (FTO and ALKBH5) exhibit significant expression imbalance in HCC, which strongly drives malignant tumor progression. Erasers enhance the ferroptosis resistance of HCC ce

Cell competition within breast tumor microenvironment: emerging role of cancer stem cells and immune cells in tumor progression and therapeutic implications.

Breast cancer remains the most prevalent malignancy among women worldwide, with approximately 2.3 million new cases diagnosed annually, accounting for nearly 12% of all cancer diagnosis globally. Increasing evidence highlights the tumor microenvironment (TME) as a dynamic and interactive ecosystem that governs tumor behaviour. This review consolidates emerging evidence that positions cell competition as a critical regulator of clonal dynamics within the breast cancer TME. We examine how breast cancer stem cells (BCSCs), defined by CD44+/CD24- phenotype and elevated ALDH activity, acquire enhan

Intelligent-responsive hydrogel synergistically mediates immune remodel-antibacterial-angiogenesis cascade for diabetic foot ulcer repair.

The healing of diabetic foot ulcers (DFU) is severely hindered by dysregulated immune microenvironment, recurrent infections, and insufficient angiogenesis. The persistent and resistant nature, coupled with elevated morbidity and amputation rates, has driven the scientific community to pursue the development of novel therapies. Current wound dressings mostly target a singular aspect and fail to achieve systematic regulation of these dynamically related factors, highlighting the urgent need for dressings capable of accurate sense and response to the complex pathological process of wound and pre

Solution structure of mouse HBS1L/SKI7-specific UBA domain in complex with ubiquitin: Implications for stalled ribosome recognition.

Human HBS1L and SKI7 (HBS1LV3) are isoforms encoded by the same gene. HBS1L forms a complex with PELO to recognize ribosomes stalled on non-stop mRNAs and promotes ribosome splitting, whereas SKI7 acts as a bridge between the exosome and the SKI complex to mediate mRNA decay on stalled ribosomes. Despite substantial differences in the sequence and function of their C-terminal regions, the two isoforms share an identical N-terminal domain (termed UBAh) that resembles the ubiquitin binding UBA and CUE domains (collectively referred to as the three-helix bundle ubiquitin-binding [THB-Ub] group).

Integrated regulation of ferroptosis in prostate cancer covering mechanisms, resistance, and translational opportunities.

Ferroptosis, an iron dependent form of regulated cell death driven by lipid peroxidation, has emerged as a pivotal process in prostate cancer biology and therapy. This review summarizes the multifaceted regulation of ferroptosis in prostate cancer from molecular, metabolic, and microenvironmental perspectives. Core regulators such as GPX4, SLC7A11, and ACSL4 coordinate redox balance, glutathione metabolism, and lipid peroxidation, together determining ferroptotic sensitivity. Transcriptional, epigenetic, and post translational mechanisms including STAT3, the JMJD6 ATF4 axis, and TRIM family pr

Metabolic regulation of tumor angiogenesis in colorectal cancer: mechanisms and therapeutic opportunities.

Colorectal cancer (CRC) progression is closely linked to abnormal vascular remodeling. Tumor vessels are structurally disorganized and poorly perfused, creating spatially heterogeneous regions of hypoxia and nutrient deprivation. These conditions reshape tumor metabolism by increasing glycolysis, glutamine utilization, and lipid metabolic plasticity. Metabolic changes, in turn, alter endothelial function and the surrounding tumor microenvironment through lactate accumulation, lipid-derived mediators, amino acid metabolism, and extracellular vesicle-mediated signaling. This reciprocal interacti

Oral Colon-Targeted Lipid Nanoparticles Enhance Upadacitinib Delivery and Efficacy in a Murine Model of Ulcerative Colitis.

Ulcerative colitis (UC) is a chronic inflammatory disorder of the colon characterized by dysregulated mucosal immunity and progressive epithelial injury. Upadacitinib (UPA), a selective Janus kinase 1 (JAK1) inhibitor, has demonstrated clinical efficacy in UC, but its therapeutic application is often constrained by adverse effects arising from systemic drug exposure. This underscores the need for advanced, site-specific delivery systems that enhance local efficacy while minimizing systemic toxicity. Here, we developed a colon-targeted natural lipid nanoparticle formulation of UPA (UPA-nLNP) to

ADAM19: biological characteristics and implications in disease pathology and clinical applications.

PURPOSE OF REVIEW: This article critically examines ADAM19-mediated inflammatory remodeling within five distinct subtypes of solid tumors prone to inflammation, employing non-neoplastic inflammatory states strictly as a comparative reference. As a transmembrane protease belonging to the ADAM family, ADAM19 undergoes maturation through plasmin-dependent cleavage, with its expression tightly regulated by both transcriptional and epigenetic controls. Although dysregulated ADAM19 levels are evident in various inflammatory lesions and malignancies, its context-specific dual biological roles necessi

Multidimensional Regulatory Mechanisms and Targeted Therapeutic Strategies for Inhibited Keratinocyte Proliferation in Diabetic Wounds.

Diabetic wound healing impairment represents a pressing clinical challenge worldwide, with its high disability rate and recurrence rate imposing a heavy burden on patients and healthcare systems. Keratinocytes are the core effector cells that drive re-epithelialization during wound healing, and their impaired proliferative capacity is a core pathological mechanism underlying healing arrest and chronic wound development. The functional status of other repair cell populations, including fibroblasts, endothelial cells and immune cells, also exerts direct or indirect regulatory effects on the enti

Evaluation of non-sputum-based diagnostics for pediatric tuberculosis: the Pediatric TB Diagnostic (PDTBDx) cohort protocol.

Tuberculosis (TB) is a significant cause of morbidity and mortality in children and adolescents, causing 172,000 deaths in 2024 in children and adolescents worldwide. Diagnostic challenges are pronounced in pediatrics, in which collecting respiratory specimens is challenging and TB is often paucibacillary, leading to delayed diagnosis and increased mortality. We describe the protocol and methodology of the Pediatric TB Diagnostic (PDTBDx) cohort, a study with the primary aim of evaluating non-sputum-based TB diagnostics for diagnosis and treatment response in children. This is a prospective ob

The application of tissue engineering in cartilage regeneration: technological advances and future challenges.

Cartilage tissue, owing to its avascular, aneural, and alymphatic nature, possesses a highly limited capacity for self-repair. Damage caused by trauma, degenerative diseases, or congenital malformations seldom heals spontaneously, constituting a long-standing clinical challenge in orthopedic and plastic surgery. Tissue engineering offers a promising strategy for cartilage regeneration by combining seed cells, biomaterial scaffolds, and bioactive molecules to construct substitutes that recapitulate the structure and function of native cartilage. The evolution of biomaterials and scaffold design

Biological pathways related to mirna-125a-5p in behavioral variant of frontotemporal dementia.

BACKGROUND: Frontotemporal dementia (FTD) comprises a group of neurodegenerative disorders that lead to progressive changes in language, behavior, personality, and movement. Accounting for approximately 60% of cases, the behavioral variant (bvFTD) represents the most common clinical presentation of FTD. MicroRNAs (miRNAs) are small non-coding RNAs involved in gene regulation and are expressed in various biological fluids. Considering bvFTD as an inflammatory disorder, this study investigated the expression of inflammation-related miRNAs in individuals with bvFTD compared with age- and sex-matc

GFAP+ FOXF2+ ependymal cells promote blood-brain barrier repair via DLL4-NOTCH signaling after neural injury.

Ependymal cells in the adult ventricular-subventricular zone are increasingly recognized for functions extending beyond cerebrospinal fluid dynamics; however, their identity and functional specialization remain incompletely understood. While ependymal cells (EP) have been implicated in interactions with the stem cell niche and the vasculature, their role in repair processes following neural injury remains elusive. In this study, we employed region-specific single-cell transcriptomics of the subventricular zone (SVZ) and ipsilesional peri-infarct territory in mice to identify a distinct subpopu
