The role of MicroRNAs in Alzheimer's disease: from pathogenesis to therapeutic potential.

Alzheimer's disease (AD) is a major neurodegenerative disorder that severely impacts the global elderly population. It is characterized by progressive memory loss, cognitive impairment, and neuropsychiatric disturbances. To date, AD lacks definitive curative treatments, making it a persistent clinical challenge. Consequently, there is an urgent need to develop cost-effective and highly specific biomarkers for the early detection of AD. MicroRNAs (miRNAs) are evolutionarily conserved, small non-coding RNAs. They are highly enriched in the central nervous system (CNS), where they orchestrate ess

KATANIN promotes cell elongation and division to generate proper cell numbers in maize organs.

Microtubule severing is essential for proper eukaryotic cell elongation and division. Here we show that the microtubule severing protein, KATANIN p60, is encoded by two genes in Zea mays L. (maize) called Discordia3a (Dcd3a) and Dcd3b. A semi-dominant mutant with short stature, poor fertility, and a clumped tassel was identified in Dcd3b called Clumped tassel1 (Clt1). Genetic enhancers that further reduced stature and fertility were identified in inbred lines and mapped to the Dcd3a locus, identifying several dcd3a alleles. Loss-of-function p60 allele combinations reduce microtubule severing,

Molecular genotype-phenotype correlation in ACTB- and ACTG1-related non-muscle actinopathies.

Recent advances in Mendelian genomics reveal the importance of variant-level characterization of allelic disorders. Non-muscle actin isoforms, encoded by the genes ACTB and ACTG1, are the most abundant intracellular proteins, but historically, they are often regarded as merely being "housekeeping" molecules. Here, we illuminate the extraordinary clinical heterogeneity and complex pathobiology of genetic non-muscle actinopathies. To do this, we combine human genomics studies with molecular biology. Strikingly, variants in ACTB and ACTG1 isoforms generate at least eight distinct clinical disorde

MPK3-mediated phosphorylation inactivates MYB4 to promote FIT-dependent iron acquisition.

Iron (Fe) is an essential micronutrient for plant growth, and its homeostasis is critically regulated by the transcription factor FIT in response to deficiency. Here, we identify the R2R3-MYB transcription factor MYB4 as a previously unknown negative regulator of Fe acquisition. MYB4 directly interacts with FIT, competing with bHLH38/39 to prevent the formation of transcriptionally active heterodimers, thereby repressing the expression of the key Fe-uptake genes IRT1 and FRO2. We establish that the mitogen-activated protein kinase MPK3 acts upstream in this pathway. Under Fe-deficient conditio

Microproteins at the Frontiers of Biology and Medicine.

Small proteins, known as microproteins, typically consisting of fewer than 150 amino acids, have recently emerged as a previously unrecognized class of functional genes in biology. Advances in ribosome profiling, proteogenomics, and genome-wide functional screens have revealed that thousands of small open reading frames are actively translated, producing microproteins, some of which regulate essential processes in metabolism, immunity, cancer, and neurodegeneration. These discoveries have challenged long-held assumptions that gene function is confined to large, conserved proteins and have unde

Diagnostic and Prognostic Biomarkers for Sepsis-Associated Liver Injury: Current Status and Future Perspectives.

Sepsis-associated liver injury (SALI) is a complication of sepsis that carries a notably poor prognosis in the intensive care unit (ICU). So far, there remains no specific consensus regarding the diagnostic criteria for SALI. The quest for biomarkers associated with SALI continues, as they are essential for both early diagnosis and prognostic assessment. Traditionally utilized biomarkers include alanine aminotransferase (ALT), aspartate aminotransferase (AST), and bilirubin; however, their lack of specificity and sensitivity has hindered the accurate diagnosis of SALI and the prediction of sub

The transcriptional cortical response to daily torpor in Phodopus sungorus.

Torpor is an essential evolutionary strategy that allows mammals to conserve resources under extreme environmental conditions. Although the hypothalamus is a key regulator of systemic acclimatisation to energy scarcity and reduced temperature, it remains unclear how the cortex, with its high metabolic demand, endures this state. Here, we performed RNA sequencing on neuronal and non-neuronal nuclei isolated from the cortex of Djungarian hamsters (Phodopus sungorus) across different seasonal and metabolic states. We found that non-neuronal cells primarily adjust during the seasonal transition fr

Iron Homeostasis and Reproduction: Unveiling the Microbiome-Gut-Brain Axis Connection in the Mosquito Anopheles culicifacies.

Our study investigated how adult female Anopheles culicifacies mosquitoes regulate systemic iron homeostasis after blood feeding, a process essential for reproduction, and revealed striking parallels to iron deficiency disorders in mammals. This study identifies that coordinated transcriptional regulation of ferritin and transferrin plays a crucial role in follicle development and egg maturation. Silencing of both genes using ribonucleic acid interference led to severe reproductive impairment, including ovarian arrest in 50% of females, a 40% reduction in oocyte number, and a decrease in first

FlhF interacts with TonB2 to defend against oxidative stress in Campylobacter jejuni.

Campylobacter jejuni is the leading cause of bacterial foodborne diarrheal disease worldwide. Despite its microaerophilic nature, C. jejuni is ubiquitous in aerobic environments and must possess specific adaptation mechanisms against oxidative stress. Here, we identified a novel role for FlhF, a GTPase essential for proper flagellar assembly, in promoting resistance to hydrogen peroxide (H2O2). Comparative transcriptomic analysis under H2O2 stress revealed that deletion of flhF leads to significant downregulation of oxidative stress-related genes. FlhF directly interacts with TonB2, an iron tr

Notch1 activation and inhibition in T-cell acute lymphoblastic leukemia subtypes.

T-cell acute lymphoblastic leukemia (T-ALL) is an aggressive hematologic malignancy caused by the accumulation of genomic lesions that affect the development of T cells. Notch1 signaling controls the expression of numerous T-lineage genes, thus playing essential parts in T-cell differentiation. T-ALL can be classified into two subtypes according to the immunophenotypic and genetic makeup: early T-cell precursor acute lymphoblastic leukemia (ETP-ALL) and non-ETP-ALL. The relationship between constitutive activation of Notch1 signaling and non-ETP-ALL has been thoroughly studied; however, how No

SETDB1 is critically required for uveal melanoma growth and represents a promising therapeutic target.

Metastatic uveal melanomas are highly resistant to all existing treatments. To identify actionable vulnerabilities, we conducted a CRISPR-Cas9 knockout screen using a library composed of chromatin regulators. We revealed that the lysine methyltransferase, SETDB1, plays a critical role in metastatic uveal melanoma cell proliferation and survival. Functionally, SETDB1 deficiency induces a DNA damage response, senescence-like state and growth arrest. Knockdown of SETDB1 is associated with a decreased expression of genes related to replication and cell cycle. Moreover, deficiency in CDC6, an essen

Single-Cell Transcriptomics in Inherited Retinal Dystrophies: Current Findings and Emerging Perspectives.

Inherited retinal dystrophies (IRDs) represent a diverse group of disorders caused by mutations in genes essential for retinal function and maintenance. Traditional bulk RNA sequencing techniques provide valuable information for deciphering disease pathogenesis but lack the resolution to capture variation among specific cell clusters during disease progression. In contrast, single-cell transcriptomics, including single-cell RNA sequencing (scRNA-seq), enables detailed examination of distinct retinal clusters in both healthy and diseased states, uncovering unique gene expression signatures and

[Novel mutations in the calcium-sensing receptor encoding genes as a cause of familial hypocalciuric hypercalcemia].

Hypercalcemia is a rare reason for consultation in clinical practice and is often an incidental finding in a routine biochemical evaluation. Its most frequent cause is primary hyperparathyroidism. Rarely, hypercalcemia is due to mutations in the calcium-sensing receptor (CaSR) signaling pathway that give rise to the different forms of familial hypocalciuric hypercalcemia (FHH). Two aspects are essential to suspect a probable FHH, the first is the existence of other cases in the family of elevated calcium and PTH associated with low urinary calcium excretion. In this context, a calcium clearanc

Rare manifestations of tuberous sclerosis complex: low-grade oncocytic tumour and diffuse lipomatosis.

Tuberous sclerosis complex (TSC) is a rare genetic disorder caused by variants in the TSC1 or TSC2 genes. This case report highlights two rare TSC manifestations: diffuse lipomatosis and low-grade oncocytic tumour (LOT), a newly recognised entity with low malignant potential.A man in his 20s presented with renal tumours and diffuse lipomatosis of his left leg since his teenage years. Over the years, numerous biopsies and scans were conducted. The renal tumours were initially classified as chromophobe renal cell carcinomas, and the patient was scheduled for a nephrectomy. After finally diagnosi

The paddy frog genome provides insight into the molecular adaptations and regulation of hibernation in ectotherms.

Amphibians, like the paddy frog (Fejervarya multistriata), have played a critical role in the transition from water to land. Hibernation is a vital survival adaptation in cold environments with limited food resources. We decoded the paddy frog genome to reveal the molecular adaptations linked to hibernation in ectotherms. The genome contained 13 chromosomes, with a significant proportion of repetitive sequences. We identified the key genes encoding the proteins of AANAT, TRPM8, EGLN1, and VEGFA essential for circadian rhythms, thermosensation, and hypoxia during hibernation by comparing the hi

Engineering the microenvironment of P450s to enhance the production of diterpenoids in Saccharomyces cerevisiae.

Cytochrome P450 enzymes play a crucial role as catalysts in the biosynthesis of numerous plant natural products (PNPs). Enhancing the catalytic activity of P450s in host microorganisms is essential for the efficient production of PNPs through synthetic biology. In this study, we engineered Saccharomyces cerevisiae to optimize the microenvironment for boosting the activities of P450s, including coexpression with the redox partner genes, enhancing NADPH supply, expanding the endoplasmic reticulum (ER), strengthening heme biosynthesis, and regulating iron uptake. This created a platform for the e

Electrophysiological and radiological diagnosis of hereditary motor and sensory polyneuropathy.

Hereditary motor and sensory neuropathy (HMSN), also known as Charcot-Marie-Tooth disease (CMT), is a member of the inherited neuropathy family with specific clinical and genetical manifestations. More than twenty genes have been linked to HMSN, and the number might increase. Regarding diagnosis, a healthcare provider should be suspicious if the patient is young with a family history. Integrative diagnosis, which includes electrophysiological, radiological, and genetic screening, is of great value to exclude metabolic, nutritive-toxic, infectious, and inflammatory or autoimmunological causes a

Hippo signaling pathway regulates Ebola virus transcription and egress.

Filovirus-host interactions play important roles in all stages of the virus lifecycle. Here, we identify LATS1/2 kinases and YAP, key components of the Hippo pathway, as critical regulators of EBOV transcription and egress. Specifically, we find that when YAP is phosphorylated by LATS1/2, it localizes to the cytoplasm (Hippo "ON") where it sequesters VP40 to prevent egress. In contrast, when the Hippo pathway is "OFF", unphosphorylated YAP translocates to the nucleus where it transcriptionally activates host genes and promotes viral egress. Our data reveal that LATS2 indirectly modulates filov
