Attention-Deficit/Hyperactivity Disorder (ADHD) is often accompanied by other psychiatric and somatic conditions. Telomere shortening as an accelerated biological aging marker may underlie this aggregate burden. Telomere length (TL) has previously been associated with childhood inattention and hyperactivity, as well as other neuropsychiatric conditions. We hypothesize that TL and ADHD may share overlapping genetic influences, suggesting that some of the same genes could affect both cellular aging and ADHD traits. We investigated the pleiotropic and causal associations between these traits. Usi

BACKGROUND: As a key index of reproductive performance in pigs, gestation length (GL) exerts a direct influence on litter size and the survival rate of piglets. Phenotypic variability in GL has been observed across Large White pig populations. To elucidate the genetic mechanisms underlying GL, this study analyzed 22,783 reproductive records from 9,057 pigs across five Large White populations. We employed a combination of diverse analytical strategies, encompassing genome-wide association studies (GWAS), GWAS meta-analysis, Bayesian fine-mapping (BFM), transcriptome-wide association studies (TW

Despite considerable efforts investigating the genetic aetiology of rare diseases in the past decades, approximately 50% of cases remain without a genetic diagnosis. Many missing diagnoses can be attributed to the limitations of short-read sequencing (SRS), compounded by (mis)-alignment to incomplete and inaccurate reference genomes such as GRCh37/38. SRS cannot resolve many regions that are challenging to map, including large contiguous tandem repeats, segmental duplications (SDs), sites of complex structural variants (SV), or highly diverged population-specific loci. Long-read sequencing (LR

BACKGROUND AND AIMS: LMNA-related dilated cardiomyopathy (LMNA-DCM) is a progressive genetic disorder characterized by conduction disease, malignant arrhythmias, myocardial fibrosis, and heart failure. Although LMNA mutations have traditionally been associated with cardiomyocyte-intrinsic defects, the mechanisms driving fibrotic remodelling remain incompletely understood.
METHODS: Spatial transcriptomics and integrated single-nuclei multiomics were performed on explanted human LMNA-DCM hearts to define endothelial transcriptional and epigenomic states associated with fibrosis. Patient-specifi

Cognitive-behavioral therapy (CBT) is a widely used treatment for mental disorders, yet the biological mechanisms underlying its effects and the factors contributing to treatment response remain poorly understood. DNA methylation, an epigenetic mechanism shaped by both genetic and environmental influences, may provide insights into individual differences in psychotherapy outcomes. We performed an epigenome-wide association study of psychotherapy response in 889 adults with obsessive-compulsive disorder (OCD) receiving the Bergen 4-Day Treatment, an intensive, concentrated exposure-based interv

The emergence of carbapenem-resistant Klebsiella pneumoniae (CRKP) poses a critical threat to global public health due to limited therapeutic options. This situation is magnified by CRKP strains with elevated virulence. This study aimed to characterize the virulome, resistome, and blaKPC genetic context of CRKP strains exhibiting increased virulence from hospital-associated infections in southeastern Brazil, focusing on their molecular evolution and clinical impact. Despite being classified as classical variants, the strains displayed a dense virulome, averaging 14 ± 0.55 virulence genes, many

Less than half of the individuals with hereditary cerebellar ataxia receives a genetic diagnosis. Repeat expansions account for disproportionate number of hereditary cerebellar ataxia and have genetically heterogeneous causes. These genetic loci include ATXN1, ATXN2, ATXN3, CACNA1A, ATXN7, ATXN8OS, ATXN10, PPP2R2B, TBP, ATN1, FMR1, BEAN1, NOP56, GLS, THAP11, GAA-FGF14, ZFHX3, FXN and RFC1. This study aims to assess the yield of short-read whole genome sequencing in the molecular diagnosis of hereditary cerebellar ataxia. We recruited 380 patients (351 probands) from a national ataxia centre in

Early life, including the embryonic period, is particularly vulnerable to environmental exposures. Previous studies have largely focused on the relationship between individual environmental factors and population health. In reality, individuals are exposed to multiple environmental factors, which together form an "exposome" that reflects the environmental influences on human health. Exposome-based analyses utilizing biological samples have been conducted in various countries to assess the impacts of environmental endocrine disruptors and other pollutants on maternal and fetal health during pre

BACKGROUND: Suicide is a leading cause of death among younger veterans; however, the genetic basis of suicide remains largely unknown. While prior genome-wide association studies (GWAS) have identified common variants associated with suicidal ideation and attempts, a pressing need remains for unbiased assessment of rare and coding genetic variation with respect to suicide phenotypes.
METHODS: Here, we conducted an exome-wide association study (ExWAS), examining both single variant and gene-based tests in relation to suicidal ideation and attempts in a cohort of post-9/11 era veterans (N = 139

Porcine reproductive and respiratory syndrome (PRRS) severely impacts global swine production. While studies have revealed host genetic factors and molecular mechanisms of fetal response to PRRSV infection, the relationship between fetal genotype, thymic transcription and PRRS susceptibility remains unclear. This study integrates genomic and thymic transcriptomic analyses of fetal pigs from a maternal PRRSV-2 challenge model. Fetuses were categorized by susceptibility to PRRS, following maternal inoculation at gestation day 84-86 into four groups: complete resistance (CR), partial resistance (

BACKGROUND: Azathioprine (AZA) is a widely used immunosuppressant drug. Leukopenia is a serious adverse effect of the drug which often necessitates dose reduction or drug withdrawal. Predictors of leukopenia include genetic and nongenetic factors. Genetic polymorphism of AZA-metabolizing enzyme, thiopurine S-methyltransferase (TPMT) is well established. There is inconclusive evidence about the role of Nudix hydrolase (NUDT15) gene polymorphism. This case-control study assessed the association of genetic polymorphisms of NUDT15 and TPMT with leukopenia induced by AZA.

Exposure to adverse life events is linked to somatic disorders. The study aims to evaluate the association between adverse events at varying life stages and the risk of chronic kidney disease (CKD), a condition affecting about 10% population worldwide. This prospective cohort study included 140,997 participants from the UK Biobank. Using survey items related to childhood maltreatment, adulthood adversity and catastrophic trauma, we performed latent class analysis to summarize five distinct patterns of exposure to adverse life events, namely "low-level exposure", "childhood exposure", "adulthoo

Many Mendelian disorders, such as Huntington's disease (HD) and spinocerebellar ataxias, arise from expansions of CAG trinucleotide repeats. Despite the clear genetic causes, additional genetic factors may influence the rate of those monogenic disorders. Notably, genome-wide association studies discovered somewhat expected modifiers, particularly mismatch repair genes involved in the CAG repeat instability, impacting age at onset of HD. Strikingly, FAN1, previously unrelated to repeat instability, produced the strongest HD modification signals. Diverse FAN1 haplotypes independently modify HD,

BACKGROUND: Otosclerosis is a common cause of adult-onset progressive hearing loss, affecting 0.3%-0.4% of the population. It results from dysregulation of bone homeostasis in the otic capsule, most commonly leading to fixation of the stapes bone, impairing sound conduction through the middle ear. Otosclerosis has a well-known genetic predisposition including familial cases with apparent autosomal dominant mode of inheritance. While linkage analysis and genome-wide association studies suggested an association with several genomic loci and with genes encoding structural proteins involved in bon

Metabolic diseases are a host of complex conditions, including obesity, diabetes mellitus, and metabolic syndrome. Endocrine control systems (eg, adrenals, thyroid, gonads) are causally linked to metabolic health outcomes. N/NIH Heterogeneous Stock (HS) rats are a genetically heterogeneous outbred population developed for genetic studies of complex traits. Genetic mapping studies in adult HS rats identified loci associated with cardiometabolic risks, such as glucose intolerance, insulin resistance, and increased body mass index. This study determined underappreciated metabolic health traits an

Genomics can play important roles in biodiversity conservation, especially for Extinct-in-the-Wild species where genetic factors greatly influence risk of total extinction and probability of successful reintroductions. The Christmas Island blue-tailed skink (Cryptoblepharus egeriae) and Lister's gecko (Lepidodactylus listeri) are two endemic reptile species that went extinct in the wild shortly after the introduction of a predatory snake. After a decade of management, captive populations have expanded from 66 skinks and 43 geckos to several thousand individuals; however, little is known about
