Emergence of insulin resistance in juvenile baboon offspring of mothers exposed to moderate maternal nutrient reduction.

Developmental programming of postnatal pancreatic β-cell and peripheral insulin function by maternal nutrient reduction (MNR) has been extensively investigated in rodents and sheep, but no data exist from nonhuman primate offspring of MNR mothers. We hypothesized that moderate levels of MNR would result in developmental programming of postnatal β-cell function and peripheral insulin sensitivity that lead to emergence of a prediabetic state prior to puberty. Prepregnancy phenotype of 18 nonpregnant baboons was matched. During pregnancy and lactation 12 mothers ate chow ad libitum (controls), wh

Plasma serotonin levels in Italian Fresian dairy cows.

The aim of this work was to investigate the metabolism of plasma serotonin or 5-hydroxytryptamine (5-HT), an important neurotransmitter, in Fresian dairy cows, a breed of zootechnical interest, using high-performance liquid chromatography with electrochemical detection. The subjects under study were at the stage of early lactation (n = 10; mean body weight 375 +/- 50 kg; average age of 3 years; body condition score 2.5), bred in a farm at an altitude of 150 m a.s.l. To evaluate animal welfare on this farm, which is closely connected to an animal's physiological status, tryptophan and cortisol

2,4-Dinitrofluorobenzene-induced contact hypersensitivity response in NC/Nga mice fed fructo-oligosaccharide.

Strategies to manipulate gut microbiota in infancy have been considered to prevent the development of allergic diseases later in life. We previously demonstrated that maternal dietary supplementation with fructo-oligosaccharide (FOS) during pregnancy and lactation modulated the composition of gut microbiota and diminished the severity of spontaneously developing atopic dermatitis-like skin lesions in the offspring of NC/Nga mice. The present study tested whether dietary FOS affects contact hypersensitivity (CHS), another model for allergic skin disease, in NC/Nga mice. In experiment 1, 5-wk-ol

Prenatal and postnatal mercury exposure, breastfeeding and neurodevelopment during the first 5 years.

OBJECTIVE: We evaluated the association between infant hair-Hg and Gesell schedules (GS). BACKGROUND: Longitudinal assessment of prenatal and postnatal Hg exposure during the first 60 months. METHODS: We used hair-Hg as a marker of postnatal Hg exposure (inorganic and methyl-Hg from breast milk, and ethyl-Hg from thimerosal) and GS measured at 6, 36, and 60 months. RESULTS: Hair-Hg at 6 months responded to events related to Hg exposure and breastfeeding. However, most neurodevelopment delays observed at 6 months were overcome with infant growth; at 60 months 87% of children showed adequate

Effects of folate and vitamin B12 deficiencies during pregnancy on fetal, infant, and child development.

The importance of folate in reproduction can be appreciated by considering that the existence of the vitamin was first suspected from efforts to explain a potentially fatal megaloblastic anemia in young pregnant women in India. Today, low maternal folate status during pregnancy and lactation remains a significant cause of maternal morbidity in some communities. The folate status of the neonate tends to be protected at the expense of maternal stores; nevertheless, there is mounting evidence that inadequate maternal folate status during pregnancy may lead to low infant birthweight, thereby confe

Accuracy of a user-friendly centrifuge for measuring creamatocrits on mothers' milk in the clinical setting.

The creamatocrit (CRCT), a simple, accurate, and inexpensive technique for the estimation of lipid and caloric content in mothers' milk, has been used extensively in lactation research, but has not been integrated into the routine management of clinical lactation problems such as slow weight gain in mothers' milk-fed preterm and term infants. The Creamatocrit Plus is a lightweight, noiseless centrifuge with an embedded reader that automatically calculates lipid and calories from the CRCT value, making it ideal for use in the clinical setting. This study compared intra-user and inter-user relia

Alterations of tooth eruption and growth in pups suckling from diabetic dams.

Several studies have confirmed a decrease in the quality and quantity of milk of mothers with diabetes during lactation. However, it remains unclear how maternal diabetes affects the offspring specifically during lactation. The aim of this study was to evaluate body and mandibular growth and tooth eruption in pups suckling from diabetic dams. The study was performed on 13 Wistar rat pups that were born to dams that were subjected to experimental diabetes on the day of parturition. Body weight and body size were recorded regularly throughout the study. The experimental pups and a group of eight

Effect of reproductive status on uptake of latex microparticles in rat small intestine.

This study investigates whether pregnancy or lactation affects microparticle uptake across the small intestinal mucosal barrier, since aspects of gastrointestinal physiology such as motility may be altered in these conditions. It also reports on validation of the model by several methods and discusses the findings in relation to possible mechanisms. Anaesthetised, pregnant, lactating, virgin female or male adult rats were gavaged with fluorescent latex microparticles. The small intestine was removed and fixed either 5 or 30 min later and successive segments of equal length were examined with f

Assessment of the perinatal effects of maternal ingestion of Solanum malacoxylon in rats.

A perinatal study was performed to verify the toxic effects of Solanum malacoxylon, which contains a glycoside conjugated to Vitamin D(3). In the gestational study, female rats received S. malacoxylon leaves in the diet at 0, 0.1, 0.2, 0.5, and 1% from days 6 to 21 of pregnancy. At 21 days of gestation, blood samples were taken from the dams for evaluation of serum Ca and P. A laparotomy was performed and the rats were examined for standard parameters of reproductive performance. Fetuses were examined for skeletal changes and histopathologic evaluation. In the second trial, dams were fed diets

Lactylation in the ageing nervous system: Epi-metabolic regulation, disease mechanisms, and translational challenges.

Lactate was long considered a waste metabolic byproduct. Currently, lactate functions as an energy substrate, signaling molecule, and epigenetic regulator in the central nervous system (CNS). Lysine lactylation (Kla), a novel lactate-dependent PTM, acts as a core epi-metabolic mediator by directly linking cellular metabolism to epigenetic remodeling. This review summarizes lactate production, shuttling, and the regulatory mechanism of lactylation (donors, writers, erasers, readers). We outline the multifaceted functions of lactylation in neurodevelopment, synaptic plasticity, and neural energy

Histone H3K9 lactylation activates the TXNIP/NLRP3 pathway to drive macrophage inflammation after spinal cord injury.

Spinal cord injury (SCI) induces metabolic and immune disruptions that impede tissue repair. However, the underlying mechanisms are poorly understood. In this study, we identified lactate accumulation as a critical driver of macrophage-mediated inflammation through histone H3K9 lactylation (H3K9la). Targeted metabolomics revealed elevated serum lactate levels in SCI patients, which were linked to increased glycolysis and lactate dehydrogenase activity. In mice, lactate accumulation after SCI was found to drive histone H3K9la in lesion-infiltrating macrophages and circulating monocytes. Integra

Inhibited proliferation and immune evasion of lung cancer: The key role of miR-4483 in lncRNA linc00824/miR-4483/PD-L1 pathway.

Growing evidence indicates that lactate and long non-coding RNAs (lincRNAs) exert a crucial influence on tumor development. This study aimed to investigate lactate-induced histone lactylation promotes the roles of lincRNA in lung cancer progression by enhancing proliferation and PD-L1-mediated inhibition of T cell antitumor function. Results showed that both linc00824 and lactate levels were elevated in NSCLC tissues compared to adjacent controls. Lactate upregulated linc00824 expression by regulating histone H3K18 lactylation. Functioning as an oncogene, linc00824 promoted NSCLC cell prolifer

FSTL1 Orchestrates Metabolic-Epigenetic Crosstalk: Glycolysis-Dependent H3K18 Lactylation Drives Cartilage Fibrosis in Osteoarthritis.

The progression of osteoarthritis (OA) is fundamentally characterized by the aberrant transformation of chondrocytes into a fibrotic phenotype, although the precise molecular mechanisms involved remain inadequately understood. In this study, the interplay between epigenetic modifications and metabolic reprogramming during the activation of fibrocartilage cells in osteoarthritis was investigated. The findings demonstrate that FSTL1 markedly upregulates key glycolytic enzymes, including LDHA, HK2, and PKM, in chondrocytes, triggering a "glycolytic burst" that results in elevated intracellular la

The effects of diaspirin cross-linked hemoglobin on hemodynamics, metabolic acidosis, and survival in burned rats.

OBJECTIVE: Diaspirin cross-linked hemoglobin (DCLHb) is a vasoactive hemoglobin-based oxygen carrier or "blood substitute" that has been shown to improve base deficit in several experimental studies of hemorrhagic shock. Our objective was to determine if the addition of DCLHb to the resuscitation regimen would improve hemodynamic parameters, metabolic acidosis, and survival in our rat burn shock model compared with currently used resuscitation therapy. METHODS: This was a randomized, controlled, experimental rat study. Male Wistar rats, weighing 200 to 250 g, were surgically prepared for an a
