Unravelling lumbar disc herniation severity beyond MRI : integrated transcriptomic and metabolomic analyses highlight glycerophospholipid metabolism and inform a machine-learning diagnostic model: a pilot study.

AIMS: While MRI serves as a tool for assessing the severity of lumbar disc herniation (LDH), it has been observed that imaging diagnoses do not always align with clinical symptoms in nearly half of patients. The absence of dependable prognostic biomarkers impedes the early and accurate diagnosis of LDH, which is critical for the development of further treatment approaches. Thus, the aim of this study was to elucidate the molecular mechanisms that determine pain and LDH severity. METHODS: We conducted a pilot study with 55 patients, employing transcriptomic and metabolomic analyses on blood sa

Bottom-up structural analysis of amides by identifying collision-induced dissociation fragment ions: An application in bile acid-amino acid conjugates-targeted sub-metabolome profiling.

BACKGROUND: Amide serves as the key functional group for several chemical families, such as proteins, peptides, amides, and amino acid-conjugated bile acids (AA-BAs), and the featured fragment ions (e.g., a+/-, b+/-, c+/-, x+/-, y+/-, and z+/-) are usually the determinant clues for MS/MS-dependent structural analysis. Here, efforts were made to identify the fragment ions through matching full exciting energy ramp (FEER)-MS3 spectra with authentic amines and/or known substructures, and the justified strategy was thereafter applied to profile AA-BAs-targeted sub-metabolome in biological matrices

Farnesol ameliorates DSS-induced IBD by regulating inflammatory cytokines, repairing the intestinal barrier, reversing the gut microbiota imbalance, and influencing fecal metabolome in C57BL/6 mice.

The incidence of inflammatory bowel disease (IBD) is rising globally, increasing interest in food ingredients for its prevention and control. This study evaluated the effect of farnesol (FAR), a key component of pomelo flower volatile oil, on dextran sodium sulfate (DSS)-induced colitis in C57BL/6 mice. FAR significantly alleviated DSS-induced colitis and secondary liver injury, as shown by improved body weight, DAI, colon length, and pathology, as well as liver function and blood lipid indices. The mechanism involves FAR-mediated regulation of inflammatory cytokines, increased expression of t

Short- and medium-term exposures of diazepam induce metabolomic alterations associated with the serotonergic, dopaminergic, adrenergic and aspartic acid neurotransmitter systems in zebrafish (Danio rerio) embryos/larvae.

INTRODUCTION: Diazepam is a well-known psychoactive drug widely used worldwide for the treatment of anxiety, seizures, alcohol withdrawal syndrome, muscle spasms, sleeplessness, agitation, and pre/post-operative sedation. It is part of the benzodiazepine family, substances known to primarily act by binding and enhancing gamma-aminobutyric acid (GABAA) receptors. The objective of the present work was to investigate the influence of short and medium-term diazepam exposures on neurotransmitters measured through targeted metabolomics using a zebrafish embryo model. METHODS: Short-term (2.5 h) and

Comprehensive Metabolomic Analysis in Blood, Urine, Fat, and Muscle in Men with Metabolic Syndrome: A Randomized, Placebo-Controlled Clinical Trial on the Effects of Resveratrol after Four Months' Treatment.

Resveratrol possesses several beneficial metabolic effects in rodents, while the effects of resveratrol in humans remain unclear. Therefore, we performed a non-targeted comprehensive metabolomic analysis on blood, urine, adipose tissue, and skeletal muscle tissue in middle-aged men with metabolic syndrome randomized to either resveratrol or placebo treatment for four months. Changes in steroid hormones across all four matrices were the most pronounced changes observed. Resveratrol treatment reduced sulfated androgen precursors in blood, adipose tissue, and muscle tissue, and increased these me

Metabolically active extracellular vesicles released from hepatocytes under drug-induced liver-damaging conditions modify serum metabolome and might affect different pathophysiological processes.

Hepatocytes are involved in the endogenous and drug metabolism; many of the enzymes involved in those processes are incorporated into extracellular vesicles and secreted into the bloodstream. Liver-damaging conditions modify the molecular cargo of those vesicles significantly. However, no information about the effect of these hepatic vesicles on the extracellular environment is available. Drug-induced liver damage increases the number of circulating extracellular vesicles and affects the release and content of hepatocyte-derived vesicles. In this work, we evaluated the metabolic effect of thes

Co-expression analysis, proteomic and metabolomic study on the impact of a Deg/HtrA protease triple mutant in Synechocystis sp. PCC 6803 exposed to temperature and high light stress.

Members of the DegP/HtrA protease family are widespread in nature and play an important role in proteolysis of misfolded and damaged proteins. The cyanobacterium Synechocystis sp. PCC 6803 contains three Deg proteases, HhoA (Sll1679), HhoB (Sll1427) and HtrA (Slr1204). Using the proteomic or metabolomic approach we investigated a triple deletion mutant (Δdeg) exposed to light or temperature stress. To cope with the stress conditions the triple mutant reduces its energy metabolism and stress-related proteins are induced to protect the cells. Additionally the co-expression of the genes encoding

Evaluation of an Infertile, All-Male ZZ Line Exhibiting Female-like Growth in Chinese Tongue Sole (Cynoglossus semilaevis): Growth Performance, Flesh Quality, and Muscle Metabolome.

Chinese tongue sole (Cynoglossus semilaevis) is an important marine aquaculture species that exhibits pronounced female-biased sexual size dimorphism, which limits the economic value of male stocks. The F4 generation of genome-edited dmrt1-/- ZZ males carried a stable 8 bp deletion in dmrt1, developed ovarian lamella-like gonads and were completely sterile, as in vitro fertilization with their gonads produced no viable embryos. From 12 to 15 months post-hatch (mph), their growth rate was significantly higher than that of wild-type males and ultimately comparable to that of females. They weighe

Subacute exposure of male adolescent rats to 2,2',5,5'-tetrachlorobiphenyl-4-ol via a polymeric implant causes gene expression changes in the brain and metabolomic disruption in serum.

Polychlorinated biphenyls (PCBs) remain an environmental health concern due to their persistence and ongoing release from legacy and emerging sources. 2,2',5,5'-Tetrachlorobiphenyl (PCB52), a PCB congener frequently detected in the environment and human blood, is oxidized to 2,2',5,5'-tetrachlorobiphenyl-4-ol (4-52). The neurotoxicity of this hydroxylated (OH-PCB) metabolite remains poorly characterized. In this study, we exposed 4-week-old male Sprague Dawley rats to 4-52 via a polymeric implant drug delivery system grafted in the subcutaneous cavity at 4-52 concentrations of 0 %, 1 %, 5 %, a
