Almost half of the world's population is at risk of acquiring dengue virus (DENV) each year. However, no specific licensed prophylactic or antiviral treatment for dengue currently exists. Mosnodenvir, a novel DENV inhibitor, has been shown to inhibit DENV replication in vitro and in animal studies. Here, we provide new insights into the in vivo prophylactic inhibitory effect of mosnodenvir exposure on primary DENV-2 infection by fitting mechanistic within-host models of DENV infection to virological and serological data observed from pre-clinical challenge studies in AG129 mice and rhesus maca

During the recent pandemic, a rise in COVID-19 cases was followed by a decline in influenza. In the absence of cross-immunity, a potential explanation for the observed pattern is behavioural: non-pharmaceutical interventions (NPIs) designed and promoted for one disease also reduce the spread of others. We study short-term and long-term dynamics of two pathogens where NPIs targeting one pathogen indirectly influence the spread of another - a phenomenon we term behavioural spillover. We examine how perceived risk of and response to one disease substantially alter the spread of other pathogens, r

Spotted-wing drosophila, Drosophila suzukii Matsumura (Diptera: Drosophilidae) (SWD), is an important pest of soft-skinned fruit around the globe because of its propensity to oviposit into ripe fruit, directly impacting the marketable portion of a crop. Temperature and host availability are two major factors that influence SWD population dynamics, but predictive tools are absent for blueberries in southern Georgia, USA, where multiple non-crop hosts occur in habitat adjacent to blueberry farms and where high summer temperatures present challenges for SWD population growth. We used temperature

BACKGROUND: Peri-implant health depends on the complex interactions between the dental implant, surrounding soft/hard tissues and the oral microbial environment. However, existing 2D and monoculture models fail to replicate this complexity, limiting their clinical relevance. Therefore, this study aimed to develop a clinically relevant 3D in vitro model that integrates oral soft tissue, hard tissue and a titanium implant in a 3D setup to accurately replicate the peri-implant environment. In addition, the model was designed to integrate bacterial biofilms, in order to mimic incipient peri-implan

Despite major advances in radiation delivery, clinical outcomes remain constrained by tumor biology rather than technology. Conventional radiobiology has relied on reductionist two-dimensional (2D) systems that fail to capture the spatial, mechanical, and multicellular organization of tumors. Three-dimensional (3D) tumor models now resolve radiation response as a mechanobiological process coordinated across the extracellular matrix (ECM), adhesion signaling, cytoskeleton, and nucleus. This review focuses on a specific and we argue, underappreciated intersection: how 3D models expose integrin-m

Cell competition in multicellular organisms has been shown to play a critical role during the development of organisms, cancer progression, and in the establishment and maintenance of tissue homeostasis. Various mechanisms of cell competition have been identified, including active elimination via mechanical forces or induced apoptosis, as well as competition for nutrients and other beneficial factors. A recent experiment demonstrated hallmarks of cell competition, associated with cell cycle dynamics, between liver progenitor cells and colorectal cancer cells [A. Krotenberg Garcia et al., iScie

The origin and mechanics of whiplash injury from motor vehicle collisions are poorly understood. Among the proposed injury mechanisms, the inertial loading of the head and neck during whiplash exposures is theorized to produce injurious cerebrospinal fluid pressure (CSFP) transients. To better understand the mechanics and modal behavior of CSFP transients during whiplash exposures, we quantified the time-frequency relationship between input head kinematics and cervical CSFP responses in an in vivo pig model. Wavelet coherence analysis was used to correlate seven head kinematic parameters (incl

Multi-nucleated cells exist in all domains of life, ranging from animals, plants, and fungi to single-celled organisms such as the slime mold Physarum polycephalum. The large cell size, in the case of Physarum reaching centimeters and more, challenges the coordination of nuclei activity as signals need to cross large distances. In search of a mechanism for fast long-ranged communication among nuclei, we quantify nuclei dynamics and cytoplasmic flows in Physarum's tubular network. We observe nuclei in two interchangeable, dynamic states: mobile, flowing within the cytoplasmic shuttle flow, or t

Evidence suggests that insects may utilize resonant mechanics during flight to optimize energetic efficiency, though whether this mechanism is universal across all insect species remains debated. Microinsects appear particularly intriguing in this context: they exhibit agility comparable to larger species despite experiencing higher aerodynamic damping forces on their wings. We investigated mechanical resonance dynamics focusing on the miniature waspTiphodytes gerriphagus-a remarkable species capable of both aerial flight and underwater locomotion, using wings in both cases. This dual-mode mob

Healthy uterine function requires coordinated and spatially organized contractions over the menstrual cycle (oestrus in animals) and at term in pregnancy. The underlying mechanisms triggering and coordinating uterine contractions, without a distinct pacemaking region, are poorly understood. Potentially, gap-junction coupling between excitable smooth muscle cells themselves or between electrically passive cells (telocytes or fibroblasts) and excitable cells may be key. Here, we present a lattice-tissue model of coupled excitable and passive cells to investigate a potential mechanism of coordina

Maximum tolerated dose (MTD) and low-dose metronomic (LDM) schedules are widely used clinical strategies in cancer chemotherapy despite the fact that both approaches have inherent limitations. MTD often leads to drug resistance in tumors, whereas LDM usually results in the predominance of drug-sensitive or drug-resistant cancer cells, depending on the specific dose applied. To circumvent these unfavorable outcomes, we propose in this work a promising adaptive therapy strategy that alternates MTD and LDM schemes across the treatment cycles. By studying a three-component tumor system with replic

Drug-induced liver injury (DILI) presents a major challenge not only in new drug development but also in post-marketing withdrawals and the safety of food, cosmetics, and chemicals. Experimental model organisms such as the rodents have been widely used for preclinical toxicological testing. However, the tension exists associated with the ethical and sustainable use of animals in part because animals do not necessarily inform the human-specific ADME (adsorption, dynamics, metabolism and elimination) profiling. To establish alternative models in humans, in vitro hepatic tissue models have been p

Hypovolemic shock and hemorrhage continue to shape healthcare delivery and incur a heavy burden on public health and wellbeing. Many healthcare organizations have specific transfusion protocols that are referenced when a patient meets certain physiological criteria. These protocols are shaped around best practices and nuanced understanding of the physiology of the patient. Despite their importance, these protocols are extremely difficult to change or challenge and, to date, there does not exist a sufficient mathematical model that may be employed towards investigating simulated patient hemorrh

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by progressive motor neuron degeneration, muscle wasting, and eventual paralysis. The clinical and genetic complexity along with rapid disease progression has hindered efforts to model the disease and develop effective treatments. Rodent models and human tissue studies point to dysfunction in oligodendrocyte lineage cells early in disease, although the underlying mechanisms remain unclear. Advances in stem cell research have introduced novel platforms to investigate cells in the oligodendrocyte lineage and t

Infectious diseases have caused significant economic and human losses worldwide. Growing concerns exist regarding climate change potentially exacerbating the spread of these diseases, particularly those transmitted by vectors such as ticks and mosquitoes. Tick-borne diseases, such as Severe Fever with Thrombocytopenia Syndrome (SFTS), can be particularly detrimental to elderly and immunocompromised individuals. This study utilizes a mathematical modeling approach to predict changes in tick populations under climate change scenarios, incorporating tick ecology and climate-sensitive parameters.

In order to evaluate the performance of different types of middle-ear prostheses, a model of human ear was developed. The model was created using finite element (FE) method with the ossicles modeled as rigid bodies. First, the middle-ear FE model was developed and validated using the middle-ear transfer function measurements available in literature including pathological cases. Then, the inner-ear FE model was developed and validated using tonotopy, impedance, and relative BM motion level curves from literature. Both models are based on preexisting research with some improvements and were comb

Species interactions structure ecological communities through direct and indirect pathways with ecosystem-wide implications. Despite mounting interest in the importance of indirect interactions, empirical evidence remains limited. Here, we demonstrate the critical role of parasite-mediated competition in driving community outcomes in a multi-species system of conservation and management concern. We leveraged 2 years of detection/non-detection data of moose (Alces alces) and white-tailed deer (Odocoileus virginianus) and parasite loads in faecal samples within a hierarchical abundance-mediated

Periodontal disease, a chronic inflammatory condition affecting the supporting structures of teeth, is driven by an imbalanced interaction between the periodontal microbiota and the host inflammatory response. Beyond its local impact, periodontal disease is associated with systemic conditions such as diabetes mellitus, cardiovascular disease, and inflammatory bowel disease, emphasizing the importance of understanding its mechanisms. Traditional pre-clinical models, such as monolayer cultures and animal studies, have provided foundational insights but are limited by their physiological relevanc

The most widely used measurement of transmission dynamics in real time is the effective reproduction number $ R\left(t\right) $. However, in the context of human immunodeficiency virus (HIV)/acquired immunodeficiency syndrome (AIDS), $ R\left(t\right) $ has not been used frequently, possibly because of the slowly progressing nature of HIV infection that limits the knowledge of recent infection events. Gaining deeper insights into the practically used epidemiological metrics of HIV/AIDS is therefore vital. Notably, in many high-income countries, including Japan, the rate of clinical AIDS on dia

The development and implementation of novel treatments for temporomandibular joint disorders (TMDs) are limited by the lack of preclinical models that closely mimic in vivo conditions. Specifically, models that support three-dimensional (3D) cell culture and mechanical stimulation are needed, as both factors significantly influence cell behavior and, consequently, the success of TMDs treatments. We designed a novel 3D in vitro model using cells subjected to dynamic moderate loading (simulating chewing) or constant excessive loading (simulating clenching) applicable as an in vivo-mimetic system
