Preclinical Nanoparticle Approaches Targeting Tumor-Associated Macrophages in Breast Cancer: From Mechanisms to Therapeutic Strategies.

Breast cancer is the most common malignancy among women worldwide, with high incidence and mortality rates. Tumor-associated macrophages (TAMs) are key mediators in the immunosuppressive tumor microenvironment (TME), contributing to poor prognosis and reducing immunotherapy efficacy. This review examines the dual roles of TAMs in breast cancer progression. TAMs are known to promote tumor development through angiogenesis, immune evasion, and metastasis, while M1-polarized TAMs conversely enhance antitumor immunity. Herein, the nanoparticle-based strategies targeting TAMs presented in preclinica

Emerging trends in implication of nanocarriers in repolarization of tumor associated macrophages for improved triple negative breast cancer therapy.

Triple negative breast cancer (TNBC) is the most aggressive breast cancer subtype with highly complex tumor immune microenvironment. So far, chemotherapy is the effective treatment for TNBC, which ameliorated the patients' outcomes. However, treatment of advanced stage disease remains challenging. In recent years, marked progress in the area of TNBC immunomodulation has been achieved and put forth as effective approach. Tumor associated macrophages (TAMs) play a major role in immunomodulation and possess dual roles as pro-tumorigenic and anti-tumorigenic. In this review, a comprehensive discus

Multi-omics evaluation of cell lines as models for metastatic prostate cancer.

Cell lines are indispensable tools in prostate cancer research yet their suitability as models for distance metastasis remains incompletely characterized. Here, we conduct a systematic evaluation study using large-scale public multi-omics data. We reveal substantial genomic differences between cell lines and metastatic patient samples, and meanwhile pinpoint cell lines which more closely resemble metastatic prostate cancer. Notably, hypermutation significantly influences the tumor microenvironment, underscoring the importance of considering mutational burden in model selection. Surprisingly, t

Living Microbe-Driven Bioorthogonal Catalysts Enables Cytotoxin Generation, Cuproptosis and Lactate Depletion for Synergistic Tumor Therapy.

Bioorthogonal chemistry provides a compelling strategy for in situ cytotoxin generation. However, the clinical translation of conventional bioorthogonal catalysts is impeded by poor tumor accumulation, insufficient targeting specificity, limited in vivo catalytic efficiency, and inadequate spatiotemporal control. Meanwhile, excessive lactate accumulation in the tumor microenvironment (TME) drives metabolic reprogramming, malignant progression, and therapeutic resistance. Here, we develop a living bacterial hybrid reactor by integratingShewanella oneidensis MR-1 (S.o) with Cu(II)-based zeolitic

ESM1 and protein lysine L-lactylation modification (Pan-kla) in ovarian cancer: coexpression, clinical significance, and prognostic value.

BACKGROUND: Ovarian cancer (OC) remains a highly lethal gynaecologic malignancy. Endothelial cell-specific molecule 1 (ESM1) and protein lysine L-lactylation modification (Pan-kla) are key players in tumour microenvironment regulation, which involves metabolic reprogramming, angiogenesis, and immune modulation. However, their coexpression patterns, clinical relevance, and synergistic prognostic impact in OC patients have not been fully elucidated. METHODS: In this study, immunohistochemistry (IHC) was used to analyse ESM1 and Pan-kla expression in 131 ovarian cancer tissue samples from patient

Development of a prognostic model for thyroid cancer based on mitochondrial metabolism-related genes and immune profiling.

BACKGROUND: The preliminary study found that mitochondrial metabolism and structure are abnormal in thyroid cancer (THCA) patients. Therefore, this study systematically investigates the relationship between mitochondrial metabolism-related genes (MMRGs) and the prognosis of THCA patients, while establishing a prognostic model for THCA. METHODS: This study utilized THCA transcriptome data from the UCSC Xena database, performed differential expression analysis using the “limma” package, and intersected differentially expressed genes (DEGs) with MMRG to identify differentially expressed MMRGs (DE

Alprazolam Reduces Inflammatory Cytokine Production in Pancreatic Cancer-Associated Fibroblasts.

UNLABELLED: Pancreatic ductal adenocarcinoma (PDAC) diagnoses are often accompanied by a number of physical and psychologic symptoms, including anxiety and depression. As a result, many patients are prescribed anxiolytics such as benzodiazepines (BZD) that have unintended effects on the tumor. Previous work from our lab has highlighted that structural differences between BZD compounds may be responsible for different clinical outcomes influenced by their effects on cancer-associated fibroblasts (CAF) within the PDAC tumor microenvironment (TME). In this study, we demonstrate that the commonly

Nanoparticles-mediated dual metabolic-immunological intervention reverses immunosuppression in muscle invasive bladder cancer.

Muscle-invasive bladder cancer (MIBC) is an aggressive urological malignancy characterized by a profoundly immunosuppressive tumor microenvironment (TME) driven by dysregulated oncogenic metabolism. The oxidoreductase NAD(P)H:quinone oxidoreductase 1 (NQO1) stabilizes the core metabolic regulator hypoxia-inducible factor 1α (HIF-1α), which further transcriptionally activates the homeobox transcription factor SIX1, forming a coordinated NQO1/HIF-1α/SIX1 signaling axis that drives metabolic reprogramming and immune evasion in MIBC, yet targeted therapies disrupting this metabolic-immune nexus re

Modeling osteosarcoma invasion with fibroblast and tumor spheroids in functionalized hydrogels beads.

Osteosarcoma (OS) is a highly aggressive bone tumor with a strong propensity for metastasis. To develop a physiologically relevant in vitro platform for tumor invasion studies and drug testing, we engineered a three-dimensional OS model that mimics the tumor microenvironment using hydrogel-based multicellular spheroid beads. Tumor spheroids (∼125 μm) were generated and co-cultured with WS1 fibroblasts in VitroGel® RGD, a hydrogel matrix that promotes cell adhesion via integrin binding. The hydrogel beads, approximately 2-3 mm in size, supported robust growth of both MG63 and highly metastatic

Matrix stress relaxation drives glioblastoma cell response in viscoelastic biomaterials.

The extracellular matrix (ECM) of glioblastoma (GBM) is known to modulate cell behavior, yet the specific contributions of matrix biochemical and biomechanical signaling remain poorly understood. To address this, we engineer a tunable hyaluronan-elastin-like protein (HELP) hydrogel to independently control ligand presentation and matrix viscoelasticity. Two peptides mimicking fibronectin (FBN) and tenascin-C (TNC) are incorporated into HELP along with hyaluronan, all of which are highly up-regulated in GBM. Using dynamic covalent chemistry, we develop hydrogels with matched stiffness but disti

Harnessing lipid-driven immunometabolic pathways in omental metastases to enhance immunotherapy in patients with ovarian cancer.

Immunotherapy with immune checkpoint blockade (ICB) in epithelial ovarian carcinoma (EOC) shows limited clinical benefit only for a small subset of patients. Overall response rates are low, so that overcoming immunotherapy resistance and improved stratification are key. In this study, we investigated the immunometabolic landscape of EOC with a focus on omental metastases, identifying lipid-laden macrophages as central elements for actionable therapeutic vulnerabilities and giving rise to biomarkers for improved patient stratification. Using patient-derived explants, we demonstrated a functiona

CD1d remodels the tumor-infiltrating myeloid populations controlling antitumor immunity.

Myeloid cells play crucial roles in cancer progression, influencing tumor growth, metastasis, and response to immunotherapy. The mechanisms shaping their diverse functions in the tumors remain poorly understood and may offer therapeutic opportunities. Here, we identify the lipid-presenting molecule CD1d as a regulator of tumor progression and myeloid heterogeneity in the tumor microenvironment. Using several mouse models of breast cancer, we demonstrate that genetic deletion or antibody-mediated targeting of CD1d leads to reduced tumor growth, altered immune infiltration, and improved efficacy

AARS1-mediated lactylation reprograms macrophage lipid metabolism to restrict antitumor immunity.

BACKGROUND: Tumor-derived lactate has long been regarded as a metabolic waste product. However, accumulating evidence indicates that lactate also functions as a signaling molecule that actively remodels the tumor immune microenvironment. How lactate-driven post-translational modifications in immune cells contribute to immune evasion in hepatocellular carcinoma (HCC) remains incompletely understood. This study aimed to identify the immune cell population responsible for lactylation-driven immunosuppression in HCC and to elucidate the molecular mechanism by which lactylation rewires macrophage m

Kidney renal clear cell carcinoma-derived migrasomes mediate immunosuppressive microenvironment by inducing M2 macrophage polarization.

Kidney renal clear cell carcinoma (KIRC) is characterized by a highly immunosuppressive tumor microenvironment (TME) that drives disease progression and limits therapeutic efficacy. Migrasomes, a recently identified class of extracellular vesicles, mediate intercellular communication; however, their specific role in KIRC immunity remains largely unexplored. Here, we show that TSPAN4, a key migrasome marker, is significantly upregulated in KIRC. KIRC-derived migrasomes effectively induce macrophage polarization toward the M2 phenotype, as evidenced by increased M2 marker expression and reduced

Senescence-directed nanotherapy ameliorates fibrosis and overcomes immune exclusion in cancer.

Fibrotic remodeling of tissues and tumors establishes immunosuppressive microenvironments that drive organ dysfunction and, in cancer, limit response to immunotherapy. Senescent-like cells are conserved drivers of fibrosis and therapeutic targets, yet their functional heterogeneity complicates therapeutic intervention. Here, we show that P-selectin is expressed by a subset of senescent-like cells in fibrotic tissues and tumors. Leveraging fucoidan-based nanoparticles that bind P-selectin, we developed senescence-modulating nanoparticles (SMNPs) to selectively target these disease-associated st

Development of a cuproptosis-related prognostic signature to reveal heterogeneity of the immune microenvironment and drug sensitivity in acute lymphoblastic leukemia.

BACKGROUND: Cuproptosis is a brand-new copper-dependent type of cell death that has been linked to various tumors. However, the relationship between cuproptosis and acute lymphoblastic leukemia (ALL) remains to be further elaborated. METHODS: In ALL, 12 cuproptosis-related genes (CRGs) were analyzed at genetic and single-cell levels. Two molecular clusters were identified using "ConsensusClusterPlus". With the least absolute shrinkage and selection operator, a prognostic signature was built based on cuproptosis. The prognosis, clinical parameters, biological function, immune cell infiltration

Exosomes and their role in gastric cancer caused by Helicobacter pylori.

Helicobacter pylori (H. pylori) is one of the main pathogens linked to the etiology of gastric cancer (GC) and the cause of a large portion of the world's disease burden. Exosomes, small extracellular vesicles, play a crucial role in the pathogenesis and progression of GC through their unique RNA cargo. Exosomes produced from H. pylori-infected cells carry bioactive chemicals such as MicroRNA (miRNAs), that rewire recipient cells, so modulating immune responses, promoting angiogenesis, facilitating metastasis, and contributing to treatment resistance. Here, the role of H. pylori in GC is discu

Current progress in the elucidation of Acute Myeloid Leukemia (AML) immune landscapes.

Immune evasion is a pivotal hallmark of cancer as mechanisms revolving around this subject remain to be fully elucidated. In acute myeloid leukemia (AML), immune evasion plays a critical role in masking the disease from the host immune system. The heterogenous nature of this disease further complicates treatment as this affects several crucial components involved in immune regulation which resulted in AML progression. The tumor microenvironment (TME) could be influenced by AML as the bone marrow components could be hijacked. Moreover, AML modulates surface marker expression, this complicates t

Metabolic and aging related gene risk score for therapeutic stratification and precision oncology in gastric cancer.

Gastric cancer (GC) exhibits marked heterogeneity, patients with identical stage receive divergent outcomes. Metabolic reprogramming and aging are pivotal in reshaping the tumor microenvironment. However, their interplay in GC prognosis remains unexplored. We analyzed RNA-seq and clinical data from The Cancer Genome Atlas Program and Gene Expression Omnibus databases. Using univariate Cox, LASSO, and multivariate Cox regression, we identified candidate genes and constructed a prognostic signature.
