Beyond the 'second brain': the gut microbiota as a constitutive co-constructor of embodied cognitive network.

Traditional cognitive science has historically confined the mind within the cranium. While the "second brain" metaphor underscores the autonomy of the enteric nervous system, it remains entrenched in a neurocentric paradigm. Here, we propose a transformative framework: the gut microbiota may function as a constitutively relevant contributor to specific embodied cognitive architectures. We contend that cognition, emotion, and behavior are not fully understandable in brain-isolated terms. Instead, these processes emerge from a sustained, bidirectional dialogue between the host and its symbiotic

Diagnostic Accuracy of Deep Learning for Detection of Incipient and Advanced Radiographic Interproximal Caries in Mixed Dentition Periapical Radiographs.

OBJECTIVES: This study aimed to develop and compare deep-learning models for detecting proximal carious lesions of different radiographic severity on mixed-dentition periapical radiographs. METHODS: A retrospective diagnostic accuracy study included 1,838 digital periapical radiographs from pediatric patients in the mixed dentition stage. Two calibrated examiners independently annotated proximal radiographic lesions using bounding boxes (inter-examiner κ=0.89; intra-examiner κ=0.92), with disagreements resolved by consensus and specialist adjudication. Lesions were classified as incipient or

Use of patient-derived organoids for pleural mesothelioma 3-D modeling.

Defining preclinical models is of utmost importance for pleural mesothelioma (PM) to improve prognosis and predict therapeutic response. Using cells isolated from pleural fluid (PF) and diagnostic pleural biopsy (PB), we generated PM patient-derived organoids (PM-PDOs) and reactive-mesothelial (RM) patient-derived organoids (RM-PDOs) aiming at assessing the proportion of successful cultures both from PF and PB. We also compared the architectural and immune-histochemical features of PM-PDOs with those of parental tissues and evaluated the PM-PDOs response to chemoimmunotherapy. We obtained 11 s

Utilizing deep learning via the 3D U-net neural network for the delineation of brain stroke lesions in MRI image.

The segmentation of acute stroke lesions plays a vital role in healthcare by assisting doctors in making prompt and well-informed treatment choices. Although Magnetic Resonance Imaging (MRI) is a time-intensive procedure, it produces high-fidelity images widely regarded as the most reliable diagnostic tool available. Employing deep learning techniques for automated stroke lesion segmentation can offer valuable insights into the precise location and extent of affected tissue, enabling medical professionals to effectively evaluate treatment risks and make informed assessments. In this research,

Raman microspectroscopy as a diagnostic tool for the non-invasive analysis of fibrillin-1 deficiency in the skin and in the in vitro skin models.

UNLABELLED: Fibrillin microfibrils and elastic fibers are critical determinants of elastic tissues where they define as tissue-specific architectures vital mechanical properties such as pliability and elastic recoil. Fibrillin microfibrils also facilitate elastic fiber formation and support the association of epithelial cells with the interstitial matrix. Mutations in fibrillin-1 (FBN1) are causative for the Marfan syndrome, a congenital multisystem disorder characterized by progressive deterioration of the fibrillin microfibril/ elastic fiber architecture in the cardiovascular, musculoskeleta

Vertebral fractures are associated with increased cortical porosity in iliac crest bone biopsy of men with idiopathic osteoporosis.

In men, vertebral fractures are poorly associated with bone density, and both cortical and trabecular micro-architectural changes could contribute to bone fragility. Bone histomorphometry makes it possible to investigate both the thickness and porosity of cortical bone, which has been reported to have a major impact on the biomechanical properties of bone. We therefore conducted a cross sectional study using iliac crest biopsies to investigate the trabecular and cortical bone structure in men with or without vertebral fractures. We selected 93 bone biopsies from men with idiopathic osteoporosi

Ubiquitin family as novel protein partners for molecular glues.

Molecular glues are small molecules that induce ternary complexes between a protein of interest and a partner protein, creating therapeutic mechanisms distinct from traditional inhibitors. This minireview examines binding-site architectures of clinically tested molecular glues and extends these insights to ubiquitin-family partner proteins. Structural analysis suggests recurring recognition features, while emerging computational methods may accelerate discovery and optimization of ubiquitin-directed glues for therapeutic applications.

3D Ewing Sarcoma Modeling for Metabolic Studies.

Three-dimensional (3D) in vitro models provide a physiologically relevant platform that more accurately recapitulates tissue architecture, cell-cell and cell-extracellular matrix interactions, and the physical microenvironment than traditional two-dimensional (2D) cultures. In particular, these models reproduce metabolic gradients, including oxygen and nutrient availability, enabling more precise investigations of cellular metabolism. This chapter describes two methods to grow Ewing sarcoma cell lines as 3D spheroids using physiological media, as well as the collection of spheroids in bulk or

Role of 68Ga-DOTA-TOC PET/CT in the Evaluation of Meningiomas with Osseous Involvement.

Meningiomas with osseous involvement present unique diagnostic and therapeutic challenges due to their variable patterns of bone remodelling, hyperostosis, and direct bone infiltration. While MRI and CT remain the standard imaging modalities for evaluating soft-tissue extension and bony architecture respectively, they often fall short in accurately distinguishing true tumour invasion from reactive changes, particularly in complex skull-base and calvarial locations. 68Ga-DOTA-TOC PET/CT represents a pivotal advancement in the imaging of meningiomas with osseous involvement, enhancing diagnostic

A Water-Stable Ultramicroporous Metal-Organic Framework with Aromatic Pore Walls for Coal-Mine Methane Capture.

Selective separation of methane (CH4) from nitrogen-rich coal-mine methane remains challenging owing to their similar physicochemical properties. Here we report a water-stable ultramicroporous metal-organic framework, Ni(Hbzza)2, featuring densely packed aromatic pore walls that create a π-enriched confined environment for CH4 adsorption. The 3-fold interpenetrated architecture generates narrow channels that may strengthen London dispersion interactions with CH4, affording an uptake of 40 cm3 g-1 at 298 K and a CH4/N2 selectivity of 5.6 (IAST). Breakthrough experiments demonstrate efficient dy

Asymptomatic Pulmonary Involvement as the Initial Presentation of Vascular Ehlers-Danlos Syndrome with a Novel COL3A1 Frameshift Variant: A Case Report.

Vascular Ehlers-Danlos syndrome (vEDS), a rare connective tissue disorder caused by pathogenic COL3A1 variants, rarely presents with isolated pulmonary manifestations in asymptomatic cases. We herein report the case of a 36-year-old asymptomatic man with incidentally discovered fluctuating pulmonary nodules and cavitary lesions. A histopathological examination revealed nodular fibrosis and focal pulmonary ossification, indicating a disruption of the pulmonary architecture and pulmonary hemosiderosis. Genetic testing revealed a novel COL3A1 frameshift variant, confirming that the attenuated vED

Using the DIVA model to better understand speech motor disorders.

The Directions Into Velocities of Articulators (DIVA) model provides a comprehensive neurocomputational framework for understanding speech production and speech motor disorders. This theoretical model describes three main control processes: feedforward control, auditory feedback control, and somatosensory feedback control. By mapping these processes to specific brain regions and neural circuits, DIVA offers insights into the mechanistic basis of various speech disorders and provides a foundation for developing targeted therapeutic interventions. This paper reviews the DIVA model's architecture

Supra-hepatic gallbladder in cirrhosis presenting as a mimic of intra-abdominal fat necrosis: Diagnostic pitfalls and clinical implications.

UNLABELLED: Supra-hepatic gallbladder is an exceptionally rare form of ectopic gallbladder. Its identification can be particularly challenging in patients with cirrhosis, where distortion of hepatic architecture obscures expected anatomical relationships. This report describes a case of supra-hepatic gallbladder in a patient with chronic liver disease, in which surrounding inflammatory oedema mimicked intra-abdominal fat necrosis, creating a significant diagnostic dilemma on imaging. CONTRIBUTION: This case underscores the importance of recognizing unusual gallbladder locations, understanding

Pathology of the Human Temporal Bone in a Rare Case of Combined Usher Syndrome and Cystic Fibrosis.

Usher syndrome is characterized by congenital or early-onset sensorineural hearing loss and progressive retinitis pigmentosa. We present the temporal bone pathology of a 23-year-old woman with Usher syndrome and concurrent cystic fibrosis who had received long-term tobramycin therapy for recurrent pulmonary infections. Histopathologic evaluation revealed largely preserved cochlear architecture with degeneration of the organ of Corti in the basal turn, diffuse strial atrophy, and mild vestibular hair cell loss. These surprising findings indicate a largely preserved sensorineural elements within
