Gene expression differences in nitric oxide and reactive oxygen species regulation point to an altered innate immune response in chronic rhinosinusitis.

BACKGROUND: The complex interplay between host, environment, and microbe in the etiopathogenesis of chronic rhinosinusitis (CRS) remains unclear. This study focuses on the host-microbe interaction, specifically the regulation of nitric oxide (NO) and reactive oxygen species (ROS) against the pathogenic organism Staphylococcus aureus (S. aureus). NO and ROS play crucial roles in innate immunity and in the first-line defense against microbial invasion. METHODS: Sinonasal tissue samples were harvested from CRS and control patients during surgery. CRS patients were classified S. aureus biofilm-po

Fimbrial profiles predict virulence of uropathogenic Escherichia coli strains: contribution of ygi and yad fimbriae.

Escherichia coli, a cause of ∼90% of urinary tract infections (UTI), utilizes fimbrial adhesins to colonize the uroepithelium. Pyelonephritis isolate E. coli CFT073 carries 12 fimbrial operons, 5 of which have never been studied. Using multiplex PCR, the prevalence of these 12 and 3 additional fimbrial types was determined for a collection of 303 E. coli isolates (57 human commensal, 32 animal commensal, 54 asymptomatic bacteriuria, 45 complicated UTI, 38 uncomplicated cystitis, and 77 pyelonephritis). The number of fimbrial types per E. coli isolate was distributed bimodally: those with low (

Combinatorial phenotypic signatures distinguish persistent from resolving methicillin-resistant Staphylococcus aureus bacteremia isolates.

Persistent methicillin-resistant Staphylococcus aureus (MRSA) bacteremia (PB) (positive blood cultures after ≥7 days of therapy) represents a clinically challenging subset of invasive MRSA infections. In this investigation, we examined the potential correlation of specific virulence signatures with PB versus resolving MRSA bacteremia (RB) (negative blood cultures within 2 to 4 days of therapy) strains. Thirty-six MRSA isolates from patients enrolled in a recent multinational clinical trial were studied for (i) susceptibility to host defense cationic peptides (HDPs) (i.e., thrombin-induced plat

Antifungal Activity, Antibiofilm and Association Studies with O-Alkylamidoximes against Cryptococcus spp.

This is the first study that describes the antifungal and anti-biofilm potential of O-alkylamidoximes against strains of Cryptococcus neoformans and Cryptococcus gattii. In vitro tests have shown that O-alkylamidoximes are capable of inhibiting fungal growth and biofilm formation of the C. neoformans and C. gattii strains, suggesting, from molecular docking, the potential for interaction with the Hsp90. The associations between O-alkylamidoximes and amphotericin B were beneficial. Therefore, O-alkylamidoximes can be a useful alternative to contribute to the limited arsenal of drugs, since they

Unveiling the impact of atmospheric temperature on antifungal resistance and virulence factors in Candida spp. isolated from forest ecosystem.

Antifungal resistance is an emerging global health concern, particularly in opportunistic fungal pathogens like Candida. While environmental factors such as atmospheric temperature fluctuations have been linked to antimicrobial resistance in bacteria, their impact on fungal resistance remains underexplored. This study investigates the relationship between atmospheric temperature variations and antifungal resistance as well as virulence factors in Candida isolated from a forest ecosystem. A total of 30 fecal samples were collected from the Margalla Hills region, and Candida isolates were identi

Voices of Eukaryotic Microbes: Chemical Communication Via Quorum Sensing.

Quorum sensing (QS) is a cell-cell communication mechanism mediated by secreted hormone-like signaling molecules that operates in both Gram-positive and Gram-negative bacteria, driving coordinated alterations in gene expression once a critical cell density is reached. In these prokaryotic systems, bacteria produce, release, detect, and respond to small autoinducers, such as acyl-homoserine lactones in Gram-negative bacteria, oligopeptides in Gram-positive bacteria, and the universal autoinducer-2, to regulate community behaviors including biofilm formation, virulence factor production, and str

Antibacterial mechanism of octyl gallate against methicillin-resistant staphylococcus aureus and its application in pork preservation: insights from multi-omics and computational simulation.

The widespread emergence of methicillin-resistant Staphylococcus aureus (MRSA) in the food chain, particularly in pork, has become an urgent public health and food safety challenge. However, safe and effective food-grade preservatives capable of both eliminating MRSA and elucidating clear antibacterial mechanisms remain limited. Herein, the food-grade additive octyl gallate (OG) exhibited remarkable activity against MRSA, with a minimum inhibitory concentration (MIC) of only 20 μg/mL and completely eradicating MRSA at 40 μg/mL within 2 h, demonstrating potent efficacy comparable to vancomycin.

Prevalence of quorum sensing luxS gene in multidrug resistant uropathogens.

BACKGROUND: The increasing prevalence of antimicrobial resistance (AMR) among urinary tract infections (UTIs) is making clinical management challenging. Biofilm formation is a key mechanism contributing to antibiotic resistance in uropathogens. Quorum sensing is known to regulate several bacterial processes like bioluminescence, virulence, and biofilm formation which is known to play a role in the development of drug resistance. Out of three major quorum sensing systems, luxS/AI-2 is a universal system used by both Gram-positive and Gram-negative bacteria. METHODS: Community-acquired AMR UTI

Targeting cyclic di-AMP signaling through diadenylate cyclase inhibition reduces methicillin resistance in clinical MRSA isolates.

BACKGROUND: Methicillin-resistant Staphylococcus aureus (MRSA) remains a major clinical challenge due to persistent β-lactam resistance and biofilm-associated tolerance. While canonical resistance mechanisms are well defined, non-canonical pathways such as cyclic di-adenosine monophosphate (c-di-AMP) signaling remain poorly understood, and no effective inhibitors have yet been shown to reduce MRSA resistance. METHODS: Clinical MRSA isolates were analyzed for c-di-AMP levels, gene expression, biofilm formation, and host immune responses under sub-inhibitory methicillin exposure. Structure-base

KpFhaB/FhaC is a virulence-associated TPS system of the globally-disseminated Klebsiella pneumoniae ST15 high-risk clone.

INTRODUCTION: The global dissemination of Klebsiella pneumoniae sequence type 15 (ST15), a Q9 high-risk clone frequently resistant to carbapenems and third-generation cephalosporins, represents a major public health concern. While chaperone-usher pili (CUP) systems (i.e. Kpi system) are known mediators of K. pneumoniae adhesion and biofilm formation, additional adhesin mechanisms remain poorly characterized in this pathogen. In this study, we identified and functionally characterized a two-partner secretion (TPS) system, KpFhaB/FhaC, in the outbreak strain K. pneumoniae Kp3380 (ST15). METHODS
