NF-κB p65 iso5 Δ6/7 and p65 iso5 Δ10 isoforms drive immune-inflammatory remodeling in Fabry disease and GLA variants of uncertain significance.

INTRODUCTION: Fabry disease (FD) is an X-linked lysosomal storage disorder in which chronic inflammation contributes to disease progression beyond substrate accumulation. Although nuclear factor kappa B (NF-κB) signaling has been implicated in FD pathogenesis, the contribution of alternative NF-κB splicing to immune-inflammatory remodeling remains unknown. Here, we investigated the recently identified NF-κB p65 iso5 splice variants p65 iso5 Δ6/7 and p65 iso5 Δ10 in FD and in individuals carrying GLA variants of uncertain significance (GVUS). METHODS: We characterized p65 iso5 Δ6/7 and p65 iso

Impact of naturally occurring target-site polymorphisms on an autonomous Cas9/guide RNA-based gene drive system for population modification of Anopheles gambiae s.l.

Malaria remains a significant global health challenge with an estimated 282 million cases reported in 2024. CRISPR/Cas9-based gene-drive systems have emerged as promising tools to block Plasmodium transmission by mosquito vectors. The TP13 drive system targets the Anopheles gambiae cardinal (Agcd) gene and carries 2 engineered monoclonal antibodies to achieve rapid population modification to prevent parasite transmission. Previous cage trials demonstrated complete drive introduction in 3 to 6 generations and supported modeling predicting a potential >90% reduction in malaria incidence under op

Path planning, obstacle avoidance, and trajectory control of a differential-drive mobile robot based on improved RRT∗-APF and nonsingular quasi-sliding mode techniques.

This study presents a framework for path planning, trajectory generation, and trajectory control of a differential-drive mobile robot in obstacle-populated environments. In the path-planning phase, an improved RRT∗-APF (rapidly-exploring random tree star and artificial potential field) method is developed by integrating RRT∗ with APF, safety inflation, and a steering-aware smoothness cost. Furthermore, parent-node selection and connection in RRT∗ are determined by a indicator designed to consider path length, obstacle risk, and smoothness. The generated path is optimized by greedy pruning and

Chemotherapy-induced activation of caspase-1 and IL-1α release by cancer cells remotely skews myelopoiesis to drive pro tumorigenic systemic neutrophil-dominant inflammation.

While chemotherapy-induced tumor cell death is known to modulate the local immune landscape, its systemic impact on distant bone marrow-a site essential for immune cell maturation-remains underexplored. Here, we show that gemcitabine chemotherapy induces inflammatory caspase-1-dependent pyroptosis in epithelial cancer cells (epiCaspase-1). Despite its inflammatory nature, epiCaspase-1-mediated cell death is non-immunogenic. Clinically, elevated expression of an epiCaspase-1 gene signature correlates with worse patient outcomes. Mechanistically, epiCaspase-1 triggers the noncanonical release of

Adaptive Evolution of Serotype O Foot-and-Mouth Disease Virus Under Vaccine Pressure: Combined VP1 T142/Q153 Mutations Drive Antigenic Alteration and Immune Evasion.

Foot-and-mouth disease virus (FMDV) escapes host immune surveillance via adaptive evolution driven by vaccine-mediated selective pressure, leading to persistent breakthrough infections in immunized animals. In this study, the dominant neutralizing epitope VP1 G-H loop (141-160 aa) was analyzed among 46 serotype O FMDV strains belonging to Southeast Asian (SEA) and Middle East-South Asian (ME-SA) topotypes isolated during 1980-2019. The key amino acid residues at VP1 positions 142 and 153 underwent sequential stepwise evolution across three phases under natural selection. Based on the reverse g

Perspectives of Indian Gastroenterologists and Hepatologists on Nonalcoholic Fatty Liver Disease Diagnosis and Management: Insights From the Nationwide Web-Based Cross-Sectional DRIVE Survey.

BACKGROUND: Nonalcoholic fatty liver disease (NAFLD) and its progressive form, nonalcoholic steatohepatitis (NASH), represent an increasing clinical and public health burden in India. Despite their high prevalence, there are limited data on their diagnostic and management approaches among Indian health care providers. Real-world evidence on how Indian gastroenterologists and hepatologists diagnose and manage these conditions remains limited. OBJECTIVE: This study aimed to understand the current disease perspectives, diagnostic modalities, and management practices for NAFLD and NASH among Indi

Fibroblast Growth Factor 2-Immobilized Biointerfaces Drive Exogenous Transforming Growth Factor β1-Independent Chondrogenesis of Human Mesenchymal Stem Cell and Ectopic Cartilage Tissue Formation.

Articular cartilage regeneration remains challenging due to limited intrinsic repair capacity and the tendency of current human mesenchymal stem cell (hMSC)-based strategies to form fibrocartilage or undergo hypertrophy. Here, we present a defined biomaterial platform in which maltose-binding protein-fibroblast growth factor 2 (MBP-FGF2) is surface-immobilized to create a bioinstructive interface that directs in vitro hMSC chondrogenesis without exogenous transforming growth factor-β1 (TGF-β1). The immobilized FGF2 interface promotes spontaneous, scaffold-free spheroid assembly via FGFR1-hepar

The effect of different training intensities on Orexin-1 receptor protein expression in the nucleus accumbens and physical activity drive in male wistar rats.

AIM: An active lifestyle promotes both physical and mental health due to the release of endorphins, dopamine, and other neuropeptides. The orexinergic pathway might regulate physical activity. This study examined how continuous training (CT) and high-intensity interval training (HIIT) of equal volume influence the orexin-1 receptor expression in the nucleus accumbens and voluntary physical activity in male Wistar rats. METHODS: Thirty-three eight-week-old male Wistar rats were randomly divided into three groups: C (n = 11), CT (n = 11), and HIIT (n = 11). After an 8-week training period, each

Oxidative stress and inflammation distinctly drive molecular mechanisms of diastolic dysfunction and remodeling in female and male heart failure with preserved ejection fraction rats.

Heart failure with preserved ejection fraction (HFpEF) is a complex cardiovascular insufficiency syndrome presenting with an ejection fraction (EF) of greater than 50% along with different proinflammatory and metabolic co-morbidities. Despite previous work provided key insights into our understanding of HFpEF, effective treatments are still limited. In the current study we attempted to unravel the molecular basis of sex-dependent differences in HFpEF pathology. We analyzed left ventricular samples from 1-year-old female and male transgenic (TG) rats homozygous for the rat Ren-2 renin gene (mRe

Parents-children co-regulation as therapeutic variable and target in autism spectrum disorders. From observation of drive to need of cooperative parent-mediated therapy.

Autism spectrum disorders (ASDs) belong to the category of neurodevelopmental disorders. ASD emerges in early childhood and involves deficits in communication, language, behavioural inflexibility and fixity, and sensorial neurodivergent perception. ASDs have a biological pathogenesis related to genetic and epigenetic factors. Additionally, research has shown that starting from childhood, autistic persons could find emotional regulation challenging during communication with caregivers. The importance of emotional co-regulation has always been under-lined in psychology, starting with Freud who i

Arbuscular Mycorrhizal Fungi Drive Organo-Mineral Association in Iron Ore Tailings: Unravelling Microstructure at the Submicron Scale by Synchrotron-Based FTIR and STXM-NEXAFS.

Arbuscular mycorrhizal (AM) fungi play an important role in organic matter (OM) stabilization in Fe ore tailings for eco-engineered soil formation. However, little has been understood about the AM fungi-derived organic signature and organo-mineral interactions in situ at the submicron scale. In this study, a compartmentalized cultivation system was used to investigate the role of AM fungi in OM formation and stabilization in tailings. Particularly, microspectroscopic analyses including synchrotron-based transmission Fourier transform infrared (FTIR) and scanning transmission X-ray microspectro

IBD Subtype-Regulators IFNG and GBP5 Identified by Causal Inference Drive More Intense Innate Immunity and Inflammatory Responses in CD Than Those in UC.

Background: The pathological differences between Crohn's disease (CD) and ulcerative colitis (UC) are substantial and unexplained yet. Here, we aimed to identify potential regulators that drive different pathogenesis of CD and UC by causal inference analysis of transcriptome data. Methods: Kruskal-Wallis and Dunnett's tests were performed to identify differentially expressed genes (DEGs) among CD patients, UC patients, and controls. Subsequently, differentially expressed pathways (DEPs) between CD and UC were identified and used to construct the interaction network of DEPs. Causal inference wa

Prophylactic vaccines are potent activators of monocyte-derived dendritic cells and drive effective anti-tumor responses in melanoma patients at the cost of toxicity.

Dendritic cell (DC)-based immunotherapy is explored worldwide in cancer patients, predominantly with DC matured with pro-inflammatory cytokines and prostaglandin E2. We studied the safety and efficacy of vaccination with monocyte-derived DC matured with a cocktail of prophylactic vaccines that contain clinical-grade Toll-like receptor ligands (BCG, Typhim, Act-HIB) and prostaglandin E2 (VAC-DC). Stage III and IV melanoma patients were vaccinated via intranodal injection (12 patients) or combined intradermal/intravenous injection (16 patients) with VAC-DC loaded with keyhole limpet hemocyanin (

Usefulness of subtraction of 3D T2WI-DRIVE from contrast-enhanced 3D T1WI: preoperative evaluations of the neurovascular anatomy of patients with neurovascular compression syndrome.

BACKGROUND AND PURPOSE: High-resolution 3D MR cisternography techniques such as 3D T2WI-driven equilibrium radiofrequency reset pulse (DRIVE) are used preoperatively to assess neurovascular anatomy in patients with neurovascular compression syndrome, but contrast between vessels and cranial nerves at the point of neurovascular contact is limited. The postprocessing technique subtraction of 3D T2WI-driven equilibrium radiofrequency reset pulse from contrast-enhanced 3D T1WI (sDRICE) provides both high spatial resolution and excellent contrast in depicting the neurovascular contact. Gut bacteria drive Kupffer cell expansion via MAMP-mediated ICAM-1 induction on sinusoidal endothelium and influence preservation-reperfusion injury after orthotopic liver transplantation.

Bacteria in the gut microbiome shed microbial-associated molecule patterns (MAMPs) into the portal venous circulation, where they augment various aspects of systemic immunity via low-level stimulation. Because the liver is immediately downstream of the intestines, we proposed that gut-derived MAMPs shape liver immunity and affect Kupffer cell (KC) phenotype. Germ-free (GF), antibiotic-treated (AVMN), and conventional (CL) mice were used to study KC development, function, and response to the significant stress of cold storage, reperfusion, and orthotopic transplantation. We found that a cocktai
