Multi-pathway copper metabolisms regulation based on an engineered copper/ferrous nanoplatform for enhanced tumor cuproptosis therapy.

Cuproptosis is a currently discovered programmed cell death modality driven by copper (Cu) ions, which shows potential application prospects in overcoming apoptotic resistance in cancer therapy due to its unique mechanism. Nevertheless, the efficiency of cuproptosis is restricted by strict Cu metabolism regulation. Herein, elesclomol (ES) and glucose oxidase (GOx) co-loaded CuFe2O4 (CF) nanoplatform (termed as CFEG) was elaborately engineered to boost cuproptosis through multi-pathway copper metabolisms regulation. After triggered by tumor-overexpressed glutathione (GSH), the released ES conti

Nebivolol Mitigates the Pro-Oxidative and Pro-Inflammatory Effects of Cyclophosphamide in the Heart.

Cyclophosphamide (CP) is an alkylating agent commonly used to treat malignant conditions. However, CP is associated with various adverse effects, one of which is cardiotoxicity. To minimize this toxicity, a common clinical strategy involves combining CP with a cytoprotective agent. One of the main causes of cardiac toxicity from CP is redox imbalance. Nebivolol, an antihypertensive medication that selectively targets β1-adrenoceptors, has demonstrated cardioprotective effects in certain situations, partly due to its antioxidant properties. In this study, we evaluated whether nebivolol could re

Mitigating Cd stress in alfalfa: the role of melatonin and nano-calcium oxide in enhancing photosynthesis and antioxidant defense.

BACKGROUND: Heavy metal contamination, particularly cadmium (Cd), is a significant threat to agricultural productivity and food security. Melatonin, as a key biostimulant, plant growth regulator and stress resistance hormone, along with nano-calcium oxide (nCaO), can enhance plant resilience to stress. However, few studies have investigated the combined effects of melatonin and nCaO in mitigating Cd stress in plants. This study examined the effects of the combined application of melatonin (100 µM) and nCaO (100 mg kg-1) on alfalfa (Medicago sativa L.) seedlings. RESULTS: Under Cd stress, alfa

A nuclear-staining, water-soluble, polycationic two-photon DNA probe for identifying dead neuronal cells and monitoring traumatic brain injury.

Neuronal cells are in most parts exempt from the daily flux of cell birth and death. The identification of dead neuronal cells is essential for facilitating our understanding of the types, mechanisms, and roles of neuronal cell death in physiology and pathology and for the investigation and treatment of neurodegenerative diseases. It is highly desirable to fabricate fluorescent probes with strong binding affinity, high brightness, and long-wavelength excitation to identify dead neuronal cells. In this study, we developed a water-soluble, polycationic two-photon fluorescent probe (BTD-V) for id

Oxidative stress promotes intervertebral disc degeneration through NAT10-mediated IKKβ N4-acetylcytidine modification.

Oxidative stress is a significant risk factor in inducing intervertebral disc degeneration (IDD), yet its precise regulatory mechanism remains unknown. This study aimed to elucidate the role of NAT10-mediated N4-acetylcytidine modification in oxidative stress-induced IDD. Initially, human nucleus pulposus cells (hNPCs) subjected to hydrogen peroxide (H2O2) were examined to clarify how oxidative stress influences cell degeneration via NAT10. Subsequently, animal models were employed to validate the ameliorative impact of modulating NAT10 expression in vivo on IDD. Ultimately, a suite of techniq

Retinal Ischemia: Therapeutic Effects and Mechanisms of Paeoniflorin.

Retinal ischemia is a key factor in the progression of vision-threatening ocular diseases, including central retinal artery/vein occlusion, exudative age-related macular degeneration (eAMD), and proliferative diabetic retinopathy. This study investigates the effects of paeoniflorin along with its related neuroprotective molecular pathways in the treatment of retinal ischemia. Free radical or ischemic-like damage was induced by incubating retinal pigment epithelium (RPE) cells for 24 h with 1 mM hydrogen peroxide (H2O2) or by subjecting retinal neuronal cells to 8 h of oxygen-glucose deprivatio

A self-accelerating 'copper bomb' strategy activated innate and adaptive immune response against triple-negative breast cancer.

Triple-negative breast cancer (TNBC) presents therapeutic challenges due to its aggressive, drug-resistance, and low immunological reactivity. Cuproptosis, an emerging therapeutic modality, is a promising strategic intervention for treating TNBC. Nonetheless, the effectiveness of cuproptosis is compromised by tumor adaptations, including the Warburg effect, increased intracellular glutathione (GSH), and copper efflux, thus breaking the barrier of cuproptosis is the basis for developing cuproptosis-based clinical therapies. Herein, a self-accelerating strategy utilizing a pH-responsive copper f

Mechanisms of 10-hydroxyoctadecanoic acid resistance in Streptococcus pneumoniae.

Profiles of human nasal colonization consistently demonstrate that Staphylococcus aureus and Streptococcus pneumoniae can co-exist in the nasopharynx. Several studies have demonstrated the antagonist relationship between the two organisms via several molecular mechanisms, including competition for nutrients as well as via direct killing by hydrogen peroxide. During nasal colonization, the pneumococcus is in direct contact with the fatty acid h18:0, which is released into the extracellular environment by S. aureus. We report that h18:0 is specifically toxic to the pneumococcus among the pathoge

Glycosomal ABC transporter 3 (GAT3) deletion enhances the oxidative stress responses and reduces the infectivity of Trypanosoma cruzi.

Glycosomes, peroxisome-like organelles in Trypanosoma cruzi, contain enzymes involved in various metabolic processes, including glycolysis. Glycosomal ABC transporters (GATs) play a vital role in maintaining metabolic homeostasis by facilitating metabolite exchange between glycosomes and the cytoplasm. GAT3 is a member of the GAT family, which also includes GAT1 and GAT2. GAT3 transcript levels are downregulated in benznidazole-resistant T. cruzi populations; however, its specific functions remain unknown. Therefore, in this study, we generated GAT3 single-knockout and null mutant lines of the

Celastrol nanomedicine eye drops restore redox homeostasis and prevents keratoconus progression via PI3K/AKT/AP-1 signaling.

Keratoconus (KC) is a progressive corneal disorder primarily driven by oxidative stress, though its precise molecular mechanisms remain incompletely understood, and effective pharmacological treatments are currently lacking. Our proteomic analysis of human KC tissues identified significant oxidative stress signatures and potential PI3K pathway in disease pathogenesis. Subsequent immunohistochemical and Western blot analyses confirmed a pronounced Nox/Nrf-2 redox imbalance - characterized by elevated Nox-4 and Nox-2 and suppressed Nrf-2 - along with activation of the PI3K/AKT/AP-1 signaling axi

Accelerating chronic wound healing by breaking the pH limitations of biomolecule-loaded Nanozymes and remodeling the microenvironment in Cascade reactions.

The microenvironment around the wounds of chronic disease patients often exhibits weakly alkaline conditions, primarily due to defects in the patients' blood glucose regulation, tissue hypoxia, accumulation of metabolic products, and bacterial infection and growth. To regulate the wound microenvironment across a broad pH range, a Fe-doped ZIF-8 nanozyme loaded with glucose oxidase (Fe-ZIF-8@GOx) was synthesized via a one-step in situ growth method. This nanozyme, with ZIF-8 serving both as a protective matrix and immobilization scaffold, enhances the enzyme's operational microenvironment and m

Tobramycin enhances Mycobacterium abscessus fitness through whiB7 induction.

Nontuberculous mycobacteria (NTM) are opportunistic pathogens that cause pulmonary disease (PD) in people with bronchiectasis and other chronic airways diseases. Difficulty treating and eradicating NTM-PD highlights the need for improved understanding of bacterial mechanisms to establish chronic infections. People with the genetic disorder cystic fibrosis (CF) develop bronchiectasis and are the population at highest risk of NTM-PD, caused mainly by Mycobacterium avium or Mycobacterium abscessus (Mabsc). The majority of people with CF (pwCF) and bronchiectasis develop chronic Pseudomonas aerugi

Effect of levetiracetam on DNA oxidation and glutathione content in a temporal lobe epilepsy model.

Levetiracetam (LEV) is a drug commonly used as an anticonvulsant. However, recent evidence points to a possible role as an antioxidant. We previously demonstrated the antioxidant properties of LEV by significantly increasing catalase and superoxide dismutase activities and decreasing the hydrogen peroxide (H2O2) levels in the hippocampus of rats with temporal lobe epilepsy (TLE) showing scavenging properties against the hydroxyl radical. The aim of the present work was to evaluate, the effect of LEV on DNA oxidation, by determining 8‑hydroxy‑2‑deoxyguanosine (8‑OHdG) levels, and glutathione co

Dual action electrochemical bandage operated by a programmable multimodal wearable potentiostat.

We have developed electrochemical bandage (e-bandage) prototypes that generate the reactive oxygen species hypochlorous acid (HOCl) or hydrogen peroxide (H2O2) for potential use to treat biofilm-infected wounds in humans. We have shown that both e-bandage-generated HOCl and H2O2 kill biofilms in vitro and in infected wounds on mice, with the former being more active in vitro. The H2O2-generating e-bandage, more so than the HOCl-generating e-bandage, was associated with improved healing of infected wounds. Here, a strategy in which H2O2 and HOCl are alternately generated-for dual action-was exp

Ncf1 knockout in smooth muscle cells exacerbates angiotensin II-induced aortic aneurysm and dissection by activating the STING pathway.

AIMS: Aortic aneurysm and dissection (AAD) is caused by the progressive loss of aortic smooth muscle cells (SMCs) and is associated with a high mortality rate. Identifying the mechanisms underlying SMC apoptosis is crucial for preventing AAD. Neutrophil cytoplasmic factor 1 (Ncf1) is essential in reactive oxygen species production and SMC apoptosis; Ncf1 absence leads to autoimmune diseases and chronic inflammation. Here, the role of Ncf1 in angiotensin II (Ang II)-induced AAD was investigated. METHODS AND RESULTS: Ncf1 expression increased in injured SMCs. Bioinformatic analysis identified N
