Retinal glial cells in glaucoma and age-related retinal diseases: Inflammatory responses, disease transitions, and translational perspectives.

Microglia, Müller cells, and astrocytes play a crucial role in maintaining retinal structure, homeostasis, and neuronal function. In disease, they undergo reprogramming that drives chronic inflammation and neurodegeneration. Unique to the retina, these glial cells occupy specialized niches and interact closely with the blood-retinal barrier, creating distinct vulnerabilities. We summarized the glial activation mechanisms, shared triggers, including oxidative stress, metabolic dysfunction, aging, and systemic inflammation, as well as key pathways, such as nuclear factor kappa-B, mitogen-activat

The Gut-Immune Axis in Treated HIV Infection: From Mucosal Damage to Chronic Inflammation and Therapeutic Opportunities-A Clinician-Oriented Narrative Review.

Combined antiretroviral therapy (cART) has transformed HIV into a manageable chronic disease. However, people living with HIV (PLWH) experience a 16-year reduction in comorbidity-free life expectancy compared to HIV-negative individuals, driven by persistent chronic immune activation despite virological suppression. Serious non-AIDS events (SNAEs)-including cardiovascular disease, metabolic disorders, and malignancies-now represent the predominant cause of morbidity. This narrative review provides a clinician-oriented synthesis of immunopathophysiological mechanisms driving chronic inflammatio

The immunometabolic network in obesity-associated insulin resistance: a systemic multi-organ and multi-cellular perspective.

Obesity has escalated into a global pandemic, encompassing chronic low-grade inflammation-termed "meta-inflammation"-as a central pathophysiological nexus linking nutrient excess to insulin resistance (IR), type 2 diabetes mellitus (T2DM), and cardiovascular complications. While adipose tissue was historically viewed as the primary origin of inflammation, accumulating evidence indicates that metabolic perturbation involves a broader and more intricate cross-organ immunometabolic network. This review provides a comprehensive synthesis of the mechanisms underlying obesity-induced IR from a syste

High-cholesterol-load-triggered pyroptosis of gingival fibroblasts promotes periodontitis.

BACKGROUND: Periodontitis is a common inflammatory disease known to be closely associated with metabolic disorders. Studies have shown a positive correlation between serum cholesterol levels and the severity of periodontitis in patients, yet how cholesterol aggravates periodontal tissue damage remains unclear. METHODS: Single-cell sequencing dataset of patients with periodontitis was analyzed to identify the potential role of cholesterol in periodontitis. A high-cholesterol diet (HCD) rat model was established, and gingival RNA sequencing was performed. Then, in vitro, gingival fibroblasts' (

FGF4 activates FGFR1 - PI3K/AKT signaling to enhance Clec10a-mediated intracellular myelin debris processing and promote spinal cord repair.

Myelin debris accumulation after spinal cord injury (SCI) drives persistent neuroinflammation, lysosomal dysfunction, and lipid overload in macrophages, ultimately impairing tissue repair. Here, we identify fibroblast growth factor 4 (FGF4) as a previously unrecognized regulator of macrophage-mediated myelin debris clearance. Endogenous FGF4 transiently increased in the early phase of SCI but rapidly declined. Using in vitro models, we demonstrate that exogenous FGF4 markedly enhances myelin debris phagocytosis through activation of the FGFR1-PI3K/AKT signaling pathway, leading to upregulation

Autophagy as a Redox Rheostat Linking Cigarette Smoke, Electronic Cigarettes, and Nicotine Exposure to Lung Development, Disease, and Interorgan Communication.

Autophagy is a central cellular quality-control pathway that maintains metabolic and proteostatic homeostasis by degrading damaged organelles and proteins. In the lung, autophagy contributes to normal development, epithelial integrity, mitochondrial quality control, and immune regulation. Emerging evidence indicates that environmental exposures such as cigarette smoke (CS), electronic cigarette (EC) aerosols, and nicotine profoundly disrupt these processes, contributing to both chronic lung disease and developmental programming of respiratory pathology. In this review, we propose a unifying fr

Modulating the Gut-Liver Axis: Anti-Inflammatory Mechanisms of Probiotics and Prebiotics in MASLD.

Non-alcoholic fatty liver disease (NAFLD), recently reclassified as metabolic dysfunction-associated steatotic liver disease (MASLD), is a prevalent metabolic disorder with significant inflammatory underpinnings. Emerging evidence underscores the gut-liver axis as a pivotal pathway in MASLD pathogenesis through which dysbiosis drives cytokine-mediated inflammation, fibrosis, and disease progression. This review synthesizes preclinical and clinical findings on how probiotics and prebiotics modulate key inflammatory cytokines-including TNF-α, IL-6, IL-1β, IL-10, IL-17, and TGF-β-to ameliorate MA

Uric acid alleviates the inflammatory response in LPS-induced BV2 cells and MPTP-induced PD mice by resisting ferroptosis through the Nrf2 signalling pathway.

BACKGROUND: Neuroinflammation, oxidative stress, and ferroptosis are implicated in Parkinson' s disease (PD) pathogenesis. Epidemiological studies suggest that elevated uric acid (UA) levels may reduce PD risk, but the precise molecular mechanisms involved remain unclear. In this study, we investigated the effects of UA in lipopolysaccharide (LPS) or MPP+-stimulated BV2 microglia and MPTP-induced PD mouse models. METHODS: BV2 cells are recognized as a standardized and reproducible neural inflammatory cell model for mechanism exploration. The anti-ferroptosis and anti-inflammatory effects of U

Selenium Status Is Associated with Inflammation in Epicardial Adipose Tissue in Elderly Patients with Coronary Artery Disease.

BACKGROUND: Inflammation in epicardial adipose tissue (EAT) contributes to cardiovascular disease through the local production of pro-inflammatory cytokines affecting the adjacent myocardium. Selenium (Se) is essential for selenoprotein-mediated antioxidant and anti-inflammatory functions. We investigated associations between Se status and inflammatory markers in EAT and in the circulation in patients with coronary artery disease (CAD). METHODS: Patients with CAD undergoing coronary artery bypass grafting (n = 52) and valve disease patients receiving valve replacement serving as controls (n =

Irritant contact dermatitis: beyond a diagnosis of exclusion.

Irritant contact dermatitis (ICD) is the most common form of contact dermatitis and a leading cause of occupational skin disease, but remains underrecognized relative to allergic contact dermatitis (ACD). Direct chemical or physical injury to the epidermal barrier causes ICD, which can develop in anyone when exposure intensity or duration exceeds the skin's repair capacity. ICD is increasingly understood to involve more than barrier disruption: it is an innate immune-mediated inflammatory disorder involving keratinocyte-derived cytokines, neutrophil recruitment, C-C motif chemokine ligand 2-C-

Resveratrol as a potential natural compound to ameliorate cognitive impairment in aluminum-exposed mice: impacts on behavior, purinergic system, and brain inflammation.

Aluminum (Al³⁺) accumulates in the brain and has been linked to neurodegeneration. When combined with citrate, its absorption increase, raising the risk of neurodegenerative disorders such as Alzheimer’s disease. Resveratrol (RSV), a polyphenol with anti-inflammatory, antioxidant, and neuroprotective properties, has been associated with aging prevention by reducing oxidative stress. This in vivo study aimed to determine whether RSV could protect male Swiss mice from Al³⁺-induced cognitive impairment, alterations in purinergic signaling, and brain inflammation. The study was conducted using 60

Subclinical inflammation and early pathogenesis of rheumatic diseases: mechanistic insights relevant to primary care.

Rheumatic diseases exhibit insidious onset and non-specific early manifestations, causing substantial diagnostic delays in primary care. This delay is closely linked to a prolonged subclinical inflammation phase, where pathogenic immune processes are active yet clinically inapparent. The mechanistic basis of this early disease concealment remains incompletely synthesized. Subclinical inflammation represents a state of persistent, low-grade immune activation in the absence of overt clinical signs, structural damage, or marked laboratory abnormalities. This phase is driven primarily by innate im

Nicotine self-administration suppresses both pro- and anti-inflammatory accumbens cytokines but does not induce apoptosis in female rats.

Nicotine can alter neuroimmune signaling within the brain reward pathway, and specifically, within the nucleus accumbens (NAc). Despite clinically meaningful sex differences in nicotine use as well as known sex differences in immune system function, no studies have evaluated NAc neuroimmune consequences following nicotine use in females. Thus, the present study evaluated cytokine expression and neuronal apoptosis in the ventral striatum (which encompasses the NAc) in ovary-intact female Long-Evans rats following nicotine or saline self-administration. Circulating cytokine levels were also eval

Research Progress on Downstream Mechanisms of Glucose Metabolic Reprogramming and Its Role in the Occurrence and Progression of Type 2 Diabetes Mellitus.

Type 2 diabetes mellitus (T2DM) is a highly prevalent and devastating chronic metabolic disease worldwide, with pathogenesis centrally characterized by insulin resistance and pancreatic β-cell dysfunction. Accumulating evidence has demonstrated that glucose metabolic reprogramming represents an adaptive metabolic shift from oxidative phosphorylation to aerobic glycolysis in cells in response to a hyperglycemic microenvironment. This shift acts as an upstream important event driving the initiation and progression of T2DM. This review summarizes the characteristics of glucose metabolic reprogram

Escitalopram and N-acetylcysteine alleviate major depressive disorder with cognitive impairment through YTHDC1/ATF4/CHOP-mediated pyroptosis.

BACKGROUND: Cognitive impairment (CI) is an important factor contributing to poor prognosis in major depressive disorder (MDD). Endoplasmic reticulum stress and microglial inflammatory response are closely associated with the occurrence and development of MDD with CI. Escitalopram (ESC) and N-acetylcysteine (NAC) are two drugs used to improve depression. However, the therapeutic efficacy and underlying molecular mechanisms of ESC and NAC in treating MDD with CI are still not fully understood. METHODS: The depression rat was established using the chronic unpredictable mild stress (CUMS) protoc
