Both elevated lipopolysaccharide and D-lactic acid concentrations in blood contribute to systemic inflammation in acute ruminal acidosis.

Acute ruminal acidosis is a major metabolic disorder in agriculturally important ruminants, such as cattle and sheep. A common symptom of acute ruminal acidosis is systemic inflammation. This study aimed to identify the underlying causes of systemic inflammation during acute ruminal acidosis using both in vivo and in vitro experiments. In the in vivo experiments, twelve 9-month-old sheep were randomly assigned to two groups, six sheep per group. Sheep in the acidotic group were fasted for 24 h and then provided ad libitum access to a 1:1 mix of whole corn and wheat flour to induce acute rumina

A randomized trial on the effects of heat abatement during the pre-weaning phase on growth and reproductive performance of heifers and health, reproductive and productive performances of cows.

Evaluation of heat stress abatement for pre-weaned dairy calves is a rare endeavor. We aimed to assess the impacts of cooling the environment of pre-weaned calves through ceiling fans on their performance after weaning and during their first lactation. We randomly assigned female Holstein calves to one of two treatment at birth (day 0): individual frame-wire hutches in a non-cooled barn ("SH", n = 125) and individual frame-wire hutches in a barn equipped with ceiling fans ("SHF", n = 101). Calves were housed under the same barn, with treatments applied in three alternating sections. Ceiling fa

Structured antenatal milk expression education for nulliparous pregnant people: results of a pilot, randomized controlled trial in the United States.

BACKGROUND: Hand-expression, collection, and storage of breast milk during pregnancy (i.e., antenatal milk expression or AME) is a safe, potentially effective practice to reduce early, undesired infant formula supplementation among women with diabetes. The feasibility and potential impact of AME on lactation outcomes in the United States (U.S.) and among non-diabetic birthing people is unknown. METHODS: The purpose of this study was to examine the feasibility of a structured AME intervention among nulliparous birthing people in the United States. We recruited 45 low-risk, nulliparous individu

Maintaining Breastfeeding During Severe Infant and Maternal HSV-1 Infection: A Case Report.

INTRODUCTION: Childbearing women with primary or secondary Herpes Simplex Virus type 1 infection are at risk of transmitting neonatal herpes infection to their infants; a medical emergency that is associated with high mortality and morbidity. Neonatal herpes infection has been commonly associated with Herpes Simplex Virus type 2, or genital herpes, but can be caused by either subtype and the presentations are indistinguishable. This case describes the course of diagnosis, treatment, and recovery for a mother and infant during a severe maternal and neonatal herpes infection, and how lactation w

Effects of oral administration of acidogenic boluses at dry-off on performance and behavior of dairy cattle.

The aim of this study was to evaluate the potential of oral acidogenic mineral boluses (196 g) containing anionic salts to facilitate the transition from lactation to the dry stage by inducing a mild and temporary metabolic acidosis at dry-off. In experiment 1, 84 lactating cows were randomly allocated to 1 of 3 treatment groups consisting of an oral administration of 0, 1, or 2 boluses 5 d before dry-off to evaluate the effects on milk production. In experiment 2, 16 lactating cows were involved in a crossover study to evaluate the effects of the administration of 2 boluses on milk production

Metabolic and energy status during the dry period is crucial for the resumption of ovarian activity postpartum in dairy cows.

It is well known that the degree of negative energy balance in high-producing dairy cows is the major cause of delayed resumption of the ovarian cyclicity that closely relates to fertility. Recent evidence suggests that the energetic situation during early lactation critically affects nutrient partitioning, metabolism, and the reproductive axis, whereas the effect of energy status during the dry period is widely unknown. The aim of the present study was to investigate the effect of energy status throughout the entire dry period until early lactation on the onset of the ovarian cyclicity. Blood

Genetic analysis of fertility in the Italian Brown Swiss population using different models and trait definitions.

The aim of this study was to estimate genetic parameters for fertility and production traits in the Brown Swiss population reared in the Alps (Bolzano-Bozen province, Italy). Fertility indicators were interval from parturition to first service, interval from first service to conception (iFC), and interval from parturition to conception, either expressed as days and as number of potential 21-d estrus cycles (cPF, cFC, and cPC, respectively); number of inseminations to conception; conception rate at first service; and non-return rate at 56 d post-first service. Production traits were peak milk y

Urinary excretion of collagen degradation markers by sows during postpartum uterine involution.

Incomplete uterine involution is the putative cause of the increased embryo mortality and reproductive failure often exhibited by sows that lactate for less than 21 days. Since such short lactation lengths are common in American swine production, an effective technique to monitor the postpartum involution process and test this hypothesis might be valuable. Rapid and extensive catabolism of uterine collagen is essential for normal postpartum involution. The objective of this study was to characterize postpartum excretion of two biochemical markers of collagen degradation. In experiment I, urine

Effect of short-term fasting on plasma concentrations of leptin and other hormones and metabolites in dairy cattle.

We determined the effects of short-term fasting and refeeding on temporal changes in plasma concentrations of leptin, insulin, insulin-like growth factor- 1 (IGF-1), growth hormone (GH), glucose, and nonesterified fatty acids (NEFA), in early lactating cows, non-lactating pregnant cows, and postpubertal heifers. In experiment 1, Holstein cows in early lactation were either fed ad libitum (Control, n=5) or feed deprived for 48 h (Fasted, n=6). Plasma leptin, insulin, and glucose concentrations rapidly declined (P<0.05) within 6h, and IGF-1 by 12h, but all these variables sharply returned to con

Rod photoreceptors have a dual dependency on both aerobic glycolysis and OXPHOS and diverge metabolically from other retinal neurons.

The retina metabolizes glucose into lactate, a hallmark of aerobic glycolysis known as the Warburg effect. Although evidence points to rod photoreceptors as the primary source of aerobic glycolysis, a comparison of the energy metabolism in different retinal neurons has yet to be performed. We combined two-photon fluorescence lifetime imaging of biosensors with pharmacological protocols to analyze metabolic dynamics in healthy and diseased rod photoreceptors and RBPMS-positive ganglion and calretinin-positive amacrine cells. Our data reveal distinct metabolic profiles among retinal neurons, ide

Practical strategies to incorporate maternal nutrition into neonatology practice to impact infant outcomes.

BACKGROUND: Maternal diet is an important clinical adjunct to care for optimal infant outcome. Clinicians should have a heightened awareness to the importance of the first 1000 days from conception to 2 years for key critical nutrients. The Registered Dietitian can be a key partner to ensure mothers are assessed nutritionally for food insecurity and dietary adequacy in pregnancy and lactation. METHODS: A clinical review of the PubMed literature was done to establish recent guidelines for pregnancy and lactation- highlighting key dietary patterns, calories and nutrients that have been associat

A feedback mechanism from prostate cancer cells to macrophages, reinforced by STAT1, regulates tumor progression and resistance to radiotherapy.

The resistance to radiotherapy of prostate cancer is driven by interactions within the tumor microenvironment, particularly between prostate cancer cells and tumor-associated macrophages, however the underlying mechanisms remain poorly understood. In this study, we found that STAT1 enhanced the transcription of critical glycolytic enzymes, leading to an increase in lactate secretion from prostate cancer cells. Then, the lactate was transported to macrophages via the MCT1 transporter, activating the NFκB1 pathway, which subsequently promoted macrophage polarization to the M2 phenotype and activ

LDH isozymes as targets for cancer therapy.

The Warburg effect, a hallmark of cancer, positions lactate dehydrogenase (LDH) as a key therapeutic target. Mammals possess three LDH isozymes (LDH-A, LDH-B, LDH-C) with distinct properties. This review critically re-evaluates the simplistic 'aerobic-anaerobic' paradigm, emphasizing that all isozymes catalyze reversible pyruvate-lactate conversion and contribute to tumor metabolism in a context-dependent manner. While LDH-A inhibition is a primary focus, challenges include metabolic plasticity and compensatory LDH-B upregulation. LDH-B plays a critical role in mitochondrial lactate oxidation.

Maternal vitamin B12 deprivation exacerbates offspring obesity by reducing early-life colonization with Bifidobacterium pseudolongum.

Vitamin B12 deficiency during pregnancy and lactation is common, yet its mechanistic impact on reproductive outcomes and offspring health remains poorly understood. Here, we show that maternal dietary vitamin B12 deprivation not only impairs maternal glucose metabolism and reproductive outcomes but also exacerbates high-fat-diet-induced obesity in offspring. These effects are mediated by gut microbiota and associated with a marked reduction of Bifidobacterium pseudolongum (B. pseudolongum) in both dams and their offspring. Maternal vitamin B12 deprivation limits early-life acquisition of B. ps

Long-term histone lactylation connects metabolic and epigenetic rewiring in innate immune memory.

Trained immunity, a de facto innate immune memory characterized by enhanced responsiveness to future challenges, is underpinned by epigenetic and metabolic rewiring. In individuals vaccinated with Bacille Calmette-Guérin (BCG), lactate release was associated with enhanced cytokine responsiveness upon restimulation. Trained monocytes/macrophages are characterized by lactylation of histone H3 at lysine residue 18(H3K18la), mainly at distal regulatory regions. Histone lactylation was positively associated with active chromatin and gene transcription, persisted after the elimination of the trainin

Pre-stroke exercise does not reduce atrophy in healthy young adult mice.

Stroke is the main cause of acquired disability in adults. Exercise reduces the risk for stroke and protects against functional loss after stroke. An exercise-induced reduction in key risk factors probably contributes to the protective effect, but direct effects on the brain may also contribute to stroke protection. We previously reported that exercise increases angiogenesis and neurogenesis through activation of the lactate receptor HCA1. Here we exposed young adult wild-type mice and HCA1 knockout mice to interval exercise at high or medium intensity, or to intraperitoneal injections of L-la

Protein Lactylation Critically Regulates Energy Metabolism in the Protozoan Parasite Trypanosoma brucei.

Lysine lactylation has been recognized as a novel post-translational modification occurring on histones. However, lactylation in non-histone proteins, especially in proteins of early branching organisms, is not well understood. Energy metabolism and the histone repertoire in the early diverging protozoan parasite Trypanosoma brucei, the causative agent of African trypanosomiasis, markedly diverge from those of conventional eukaryotes. Here, we present the first exhaustive proteome-wide investigation of lactylated sites in T. brucei. We identified 387 lysine-lactylated sites in 257 proteins of

The role of vitamin A and its pro-vitamin carotenoids in fetal and neonatal programming: gaps in knowledge and metabolic pathways.

Vitamin A (VA) and its pro-vitamin carotenoids are naturally occurring lipophilic compounds involved in several cellular processes and metabolic pathways. Despite their broad spectrum of activities in the general population, dietary deficiencies of these compounds can potentially affect pregnancy outcomes. Since maternal nutritional status and diet composition during pregnancy and lactation can have long-lasting effects in offspring until adulthood, this study presents an overview of VA and the role of pro-VA carotenoids during pregnancy and lactation - the nutrition, metabolism, and biologica

Can maternal exposure to paracetamol impair reproductive parameters of male rat offspring?

Paracetamol is a widely used medication during gestation and lactation periods for the treatment of pain and fever. Several studies have shown that exposure to paracetamol can increase the incidence of cryptorchidism and decrease testosterone production. Therefore, the present study aimed to evaluate if maternal treatment with paracetamol during gestation and gestation/lactation periods can alter reproductive and behavioral parameters in male offspring. Female Wistar rats were treated daily by gavage with water or paracetamol (350 mg/kg/day) during gestation (CTRG and PARG) or gestation/lactat
