Interleukin-4 alleviate polycystic ovary syndrome through promoting the polarization of M2 type macrophages via targeting NF-κB/IκB pathway.

Polycystic ovary syndrome (PCOS) represents a prevalent endocrine disorder that constitutes a significant threat to female reproductive health. Immune dysregulation and chronic low-grade inflammation are increasingly recognized as pivotal contributors to ovarian dysfunction in this condition. In the present study, we enrolled PCOS patients clinically and established a dehydroepiandrosterone (DHEA)-induced PCOS mouse model. Our findings revealed elevated levels of pro-inflammatory M1 macrophages, diminished anti-inflammatory M2 macrophages, and aberrant distribution of associated inflammatory c

Integrated network toxicology suggests potential mechanisms involving environmental flame retardant TDCPP in ovarian cancer progression via "liver-to-ovary crosstalk.".

BACKGROUND: The environmental flame retardant tris(1,3-dichloro-2-propyl) phosphate (TDCPP) is epidemiologically associated with several core risk factors for ovarian cancer (such as hormonal dysregulation and infertility); however, the molecular mechanisms linking these adverse health outcomes remain unclear. This knowledge gap limits the precision and specificity of risk assessment. This study aims to utilize bioinformatics methods to suggest potential molecular pathways through which TDCPP influences the development of ovarian cancer. METHODS: We employed an integrative analysis strategy co

Myo-inositol versus D-chiro-inositol in murine in vitro follicular development: An experimental study relevant to polycystic ovary syndrome.

Inositol plays a crucial role in follicular development by regulating insulin signaling and ovarian function. However, its precise mechanism of action remains unclear. This study investigated the effects of myo-inositol (MI) and D-chiro-inositol (DCI) on the development of murine ovarian follicles in vitro. Follicles treated with DCI exhibited larger diameters than controls on Day 6 (275.20 ± 12.54 μm; p = 0.037) and Day 8 (277.47 ± 11.47 μm; p = 0.048), indicating a modest, marginally significant effect that was not maintained by Day 10. The rate of follicular antrum formation was significant

Correlation Between Baseline Serum Testosterone Levels and Cognitive Gain Following Combined Oral Contraceptive Treatment in Patients With Polycystic Ovary Syndrome.

Background Combined oral contraceptives (COCs) are the cornerstone therapy for polycystic ovary syndrome (PCOS). While COCs have been shown to improve cognitive performance, the relationship between these gains and hormonal profiles, especially testosterone, remains unclear. This study investigated whether baseline testosterone levels correlate with cognitive changes after COC treatment. Methods This quasi-experimental study evaluated 35 nulliparous women (aged 18-35 years) with PCOS. Baseline total serum testosterone levels were measured before starting a three-cycle course of COCs (levonorge

Dynamic changes of Kisspeptin during controlled ovarian hyperstimulation (COH) in polycystic ovary syndrome patients and its correlation with COH outcomes.

BACKGROUND: Polycystic ovary syndrome (PCOS) is a common cause of anovulatory infertility in women, and patients often exhibit ovarian hyperresponse during controlled ovarian hyperstimulation (COH) in assisted reproductive technology. Kisspeptin is a key upstream regulator of the hypothalamic-pituitary-ovarian axis (HPOA). This study aims to explore the dynamic changes in serum and follicular fluid (FF) Kisspeptin levels during COH in PCOS patients and analyze their correlation with COH outcomes and pregnancy outcomes, in order to evaluate the potential value of Kisspeptin as a predictive biom

Kaempferol supplementation at 150 mg/kg improves reproductive performance of aged laying hens via modulation of the gut-liver-ovary axis.

The decline of egg production in aged laying hens is primarily due to age-related declines in hepatic and ovarian functions as well as changes in reproductive hormones and reduced antioxidant capacity. Kaempferol (KAE), a flavonoid compound with potent antioxidant activity also affects ovarian functions in mammals, but its role in chickens remains poorly understood. We aimed to investigate the potential of KAE to mitigate reproductive decline in aged laying hens through modulation of the gut-liver-ovary axis. A total of 400 Hy-Line Brown aged laying hens (420 days old) were randomly assigned t

BRCA1 alleviates inflammation, oxidative stress, and ovarian granulosa cell apoptosis by inhibiting endoplasmic reticulum stress, thereby ameliorating polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS) is a prevalent endocrine disorder characterized by metabolic imbalance, oxidative stress, and granulosa cell (GC) dysfunction. Given the established role of BRCA1 in maintaining genomic stability and regulating stress responses, its potential involvement in PCOS pathogenesis warrants investigation. We examined the impact of BRCA1 on PCOS using a dehydroepiandrosterone (DHEA)-induced mouse model and testosterone-stimulated KGN cells. Lentiviral vectors overexpressing BRCA1 were administered by ovarian injection in PCOS mice or used to infect KGN cells. The IL-1β

An ovary-enriched piRNA controls ovarian development via Gadd45a-JNK-mediated nurse-cell apoptosis in the arbovirus vector Aedes albopictus.

The Asian tiger mosquito (Aedes albopictus) is a rapidly expanding invasive species and a global vector of arboviruses. Its transmission depends on the female's blood meal-driven reproductive cycle. Although mosquito PIWI-interacting RNAs (piRNAs) research has predominantly focused on antiviral immunity, their roles in reproduction remain poorly understood. Here we profiled sex-biased piRNA expression in Ae. albopictus and identified a female-enriched piRNA, AalpiRNA-18529. This piRNA accumulates in pre-vitellogenic ovaries and is rapidly downregulated after a blood meal. Gain-of-function expe

Increased versatility and convenience: Advances and strategy optimization of Receptor-Mediated Ovary Transduction of Cargo-mediated genetic modification in insects.

Genetic modification via gene editing has become a widely adopted and demonstrably effective method in functional gene research within entomology. However, the optimal efficiency and simplicity of delivering exogenous guide RNA-clustered regularly interspaced short palindromic repeats-associated protein 9 complexes into target tissues are crucial for successful gene editing. The Receptor-Mediated Ovary Transduction of Cargo (ReMOT) strategy, which simplifies the delivery process, target-site selection, technical requirements, and delivery cost compared with embryonic microinjection, enabling e
