Nanotechnology-based reformulation of AUY922 mitigates retinal toxicity and retains potent anti-tumor activity.

AUY922, a heat-shock protein 90 inhibitor, shows broad antitumor activity but is limited by dose-limiting ocular toxicity. In this study, we engineered FLIM02, a PLGA/DSPE-PEG2000 (1,2-Distearoyl-sn-glycero-3-phosphoethanolamine-N-[poly(ethylene glycol)-2000])-based nanoparticle via single-emulsion, and evaluated its physicochemical profile, biodistribution, ocular safety, and efficacy. FLIM02 formed ∼100-nm particles with efficient encapsulation and retained stability after lyophilization. In tumor-bearing mice, LC-MS/MS biodistribution showed reduced ocular retention of AUY922 with preserved

A neutrophil-intrinsic CKLF1-PKM2 axis drives glycolytic flux for de novo DAG synthesis and pro-inflammatory ROS production.

Reactive oxygen species (ROS) burst and subsequent recruitment to the ischemic brain characterize neutrophil activation in the hyperacute phase of ischemic stroke, yet the underlying metabolic drivers remain elusive. Here, we report that neutrophil-intrinsic chemokine-like factor 1 (CKLF1) acts as a key immunometabolic regulator following stroke. CKLF1 was rapidly upregulated in neutrophils within 6 h post-ischemia in both humans and mice. Mechanistically, CKLF1 bound directly to the tetrameric form of pyruvate kinase M2 (PKM2), enhancing glycolytic flux and diverting intermediates toward de n

Chemotactic sensing of extracellular antibiotic resistance genes enables their efficient removal by Stutzerimonas stutzeri.

The dissemination of antibiotic resistance genes (ARGs) in wastewater environments poses a severe threat to public health. Extracellular ARGs (eARGs) persist as free DNA fragments that are refractory to efficient removal by conventional physicochemical treatment technologies. Here, we isolated Stutzerimonas stutzeri CHY07 from municipal sewage and demonstrated that extracellular DNA fragments, including eARGs, can serve as chemoattractants for environmental bacteria. Through genomic mining, molecular docking, surface plasmon resonance (SPR), isothermal titration calorimetry (ITC) and protein-l

A synergistic multiparametric MRI strategy for FAPα-targeted tumor fibrosis based on NaGdF4@PEG-FAPI nanoprobes.

Tumor-associated fibrosis, a key pathological feature of the tumor microenvironment (TME), is driven by cancer-associated fibroblasts (CAFs), promoting tumor progression and immune evasion, while fibroblast activation protein α (FAPα) overexpressed in over 90% of solid tumor stroma but absent in normal fibroblasts serves as an ideal target for early tumor diagnosis via fibrotic lesion detection. Herein, we developed a FAPα-targeted nanoprobe NaGdF4@PEG-FAPI for multiparametric MRI diagnosis of tumor fibrosis. This nanoprobe was engineered for dual-mode T1/T2 MRI, enabling comprehensive tumor c

Proteomic profiling of ANO10-deficient SH-SY5Y cells reveals candidate pathways relevant to SCAR10.

Autosomal recessive spinocerebellar ataxia type 10 (SCAR10) is a rare, slowly progressive neurodegenerative disorder characterised by ataxia, cerebellar atrophy, and oculomotor abnormalities, caused by variants in the ANO10 (anoctamin 10) gene. While ANO10-mediated calcium dysregulation in Purkinje cells has been proposed to cause SCAR10, the precise pathogenic mechanism remains unclear. To investigate cellular responses to ANO10 deficiency, four ANO10-mutant neuronal cell lines - a knockout and three lines harbouring recurrent SCAR10-associated pathogenic variants - were engineered using CRIS

A self-iterative orthogonal base-editing platform enables multiplex N-to-N diversification and genome-scale functional screening in Escherichia coli.

Base editing enables precise genome modification without double-strand breaks but remains limited by narrow editing windows, DNA repair pathway biases, and restricted nucleotide diversity. Here, we report MUTATOR, a MUlTiplexAble and self-iTerative ORthogonal base-editing platform that enables N-to-N diversification in Escherichia coli. MUTATOR combines CWBE and ABE with iterative editing on two complementary DNA strands, thereby overcoming endogenous DNA repair constraints and expanding A-to-N and C-to-N editing outcomes across both strands. This strategy substantially expands accessible nucl

A modular platform for measuring substrate oxidation by CO2 production in diverse cell culture systems.

Metabolism underpins cellular physiology, whereby the preference for specific substrates and catabolic pathways shapes the production of energy, anabolic substrates, and metabolite signals to address bioenergetic demands. Substrate catabolism can be directly examined by measuring metabolic endpoints. For instance, substrate oxidation can be quantified by the incorporation of carbon from labelled glucose or fatty acids into carbon dioxide, providing a sensitive and specific readout of metabolic flux. However, current platforms require relatively large culture volumes, lacking adaptability for s

The Multifaceted Role of Extracellular Vesicles in Triple Negative Breast Cancer.

Triple negative breast cancer (TNBC) is an aggressive and heterogeneous subtype of breast cancer characterized by the absence of the estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor 2 (HER2), resulting in limited options for targeted therapy and high rates of metastasis, recurrence and death. Extracellular vesicles (EVs) have emerged as central mediators of TNBC pathophysiology, functioning as key intercellular communication vehicles transporting oncogenic proteins, nucleic acids, lipids, and metabolites. These EV-mediated interactions promote tumo

Establishment of a novel brain cell line from grass carp (Ctenopharyngodon idella) with high transfection efficiency and CRISPR/Cas9-mediated genome editing capacity.

Fish cell lines serve as valuable tools in aquaculture research, particularly in immunology, pathology, and toxicology. In this study, we successfully established a novel cell line derived from brain tissue of grass carp, designated CIB. This cell line has been subcultured over 100 times and exhibits a fibroblast-like morphology. Chromosomal analysis revealed that the diploid chromosome number of CIB cells is 2n = 48, while sequencing of the 18S rRNA gene confirmed the cell line's origin. Notably, CIB cells demonstrated a transfection efficiency of 62.4% with pEGFP-N3, highlighting their poten

Extracellular Vesicles as Key SASP Carriers Driving Cellular Senescence, Inflammaging, and Therapeutic Opportunities in Aging and Age-Related Diseases.

The progressive decline in physiological processes with aging is a complex biological process that increases susceptibility to age-related diseases. Cellular senescence is a primary contributor to this process, a state of sustained cell cycle arrest accompanied by a distinctive secretory profile known as the Senescence-Associated Secretory Phenotype (SASP). In recent years, extracellular vesicles (EVs), including exosomes and microvesicles (MVs), have emerged as key mediators of intercellular communication by carrying bioactive molecules, such as proteins, lipids, and nucleic acids. MEK inhibitor mirdametinib promotes fracture healing in osteofibrous dysplasia RASopathy.

Osteofibrous dysplasia (OFD) is a skeletal RASopathy presenting with periosteal bone lesions that may progress to fracture and delayed healing (pseudarthrosis). MET gene mutations reducing ubiquitin-mediated protein degradation via loss of the juxtamembrane domain (METΔJMD) were previously identified in patients with OFD, resulting in ligand-dependent gain of function. The effect of METΔJMD expression on skeletal progenitor cell differentiation and the potential efficacy of targeted therapies remain unclear. We engineered MetΔJMD mice and showed that MetΔJMD expression inhibited osteogenic dif

DHA Targets RAB1B to Mediate the Driving Force Transmission of Copper Ion Vesicular Endocytosis Under Copper Overload for Promoting Cuproptosis and Ferroptosis in Hepatic Stellate Cells.

Liver fibrosis represents a central pathological process in chronic liver diseases and poses a severe threat to human health. Cuproptosis, a copper ion-dependent form of regulated cell death, offers a potential therapeutic strategy for liver fibrosis. This study demonstrated that dihydroartemisinin (DHA) transiently upregulates SLC31A1, thereby promoting copper influx. Under copper overload, SLC31A1 undergoes ubiquitin-mediated degradation, while DHA targets RAB1B to activate RAB10-dependent vesicular endocytosis as a compensatory mechanism for sustained copper accumulation. This endocytic pro

Tracking HIV-1 DNA fate from cell culture to humanized mice tissues.

Unambiguous identification of HIV reservoirs is essential for their characterization and for developing curative strategies. A major obstacle to curing HIV is the persistence of silent viral genomes in host cells that evade immune detection and resist therapy. Here, we present a fluorescence microscopy-based method for direct in vivo visualization of full-length double-stranded HIV-1 DNA, an achievement not previously realized. To achieve this, we developed an advanced imaging platform adapted for replication-competent HIV-1. This system leverages a bacterial-derived two-component tagging stra

Hybrid chitosan sponges biofunctionalized with decellularized porcine bladder extracellular matrix for in vivo dermal regeneration.

The limited availability of wound dressings necessitated the development of low-cost options for burn treatment. In this study, chitosan and porcine extracellular matrix (ECM) sponges were developed for potential use in dermal regeneration. These sponges are designed for wounds that require a material to promote fibroblast growth, which is essential for the formation of new, healthy tissue. Natural materials, such as chitosan and ECM, provide antimicrobial and regenerative properties, as well as structural proteins and bioactive signals essential for the repair of damaged tissue. Therefore, us

Cyclin-Dependent Kinase 5 Contributes to Bruton's Tyrosine Kinase Inhibitor Resistance via the IRE1α/XBP1 Axis in Mantle Cell Lymphoma.

The mechanisms of resistance to Bruton tyrosine kinase inhibitors (BTKi) in MCL are not well understood. We found that cyclin-dependent kinase 5 (CDK5), a serine/threonine kinase, was upregulated in ibrutinib-resistant MCL cell lines and in primary MCL cells obtained from patients who progressed on BTKi. Furthermore, primary MCL cells upregulated CDK5 in BAFF/CD40L stromal conditions, and CDK5 mRNA overexpression in primary MCL tumors was associated with inferior outcomes. Genetic and pharmacologic (GFB-12811) manipulation of CDK5 in MCL cell lines revealed that CDK5 contributed to proliferati
