Pulmonary artery pressure via endobronchial ultrasound: A feasibility and safety study.

OBJECTIVES: Pulmonary hypertension significantly influences perioperative outcomes in thoracic surgery. Right heart catheterization, the standard method to assess pulmonary artery pressure, is invasive and time-consuming. We aimed to assess the feasibility and safety of direct pulmonary artery pressure measurement through endobronchial ultrasound-guided puncture of the right pulmonary artery. METHODS: Twenty patients underwent ultrasound-guided puncture of the right pulmonary artery using a 22-gauge needle under real-time ultrasound guidance. Systolic, diastolic, and mean pulmonary pressures

TCR signal-enhancing mutation alters lipid metabolism of thymocytes and impairs antitumor immunity of mature T cells.

The T cell receptor (TCR), a master regulator of adaptive immunity, serves as a molecular transducer that converts antigen recognition into precisely modulated intracellular signals, orchestrating both T cell development and effector functions. In this study, we leveraged a germline CD3εI173A mutation, a previously characterized alteration that amplifies TCR signaling through the disruption of inhibitory lipid interactions, to dissect how thymocyte-intrinsic TCR signaling amplification influences the fate of mature T cells. Remarkably, thymic double-positive cells in CD3εI173A mice with altere

Kaempferide enhances type I interferon signaling as a novel broad-spectrum antiviral agent.

Broad-spectrum antivirals (BSAs) possess unique advantages of being effective against a wide range of both existing and unpredictable emerging viral infections. The host type I interferon (IFN) response serves as a universal defense against diverse viral infections nonspecifically, providing attractive targets to develop novel BSAs. In this study, we identified the flavonoid kaempferide as an enhancer of the type I IFN activated Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway, promoting the expression of IFN stimulated genes (ISGs) and the establishme

The role of JAK signaling in SpA pathogenesis and its inhibition.

INTRODUCTION: Spondyloarthritis (SpA) includes a group of chronic inflammatory disorders affecting the axial skeleton and/or peripheral joints. The Janus Kinase-Signal Transducer and Activator of Transcription (JAK-STAT) pathway is pivotal in cytokine-mediated signaling, contributing to SpA pathogenesis. AREAS COVERED: Cytokine inhibitors were the first biological therapies used in SpA. However, not all patients responded to this treatment. In this setting, Janus Kinase inhibitors (JAKi) have emerged as a promising option for SpA treatment. They act on JAK family members regulating many cytok

Long non-coding RNAs (lncRNAs) in cancer development: new insight from STAT3 signaling pathway to immune evasion.

Overcoming cancer and enhancing patient survival are becoming increasingly challenging due to the uncontrolled growth and metastasis of colorectal cancer cells. In order to provide effective cancer treatment and minimize the malignancy of cancer cells, it is necessary to understand how complex signaling networks contribute to their invasion and proliferation. The signal transducer and activator of transcription 3 (STAT3) is a promising target due to its involvement in various cellular functions, including apoptosis, immunosuppression, cell invasion, migration, and proliferation. Dysregulation

Chimeric antigen receptor modified hematopoietic stem cells (CAR-HSCs) arm all immune forces for anti-tumor in mice.

Chimeric antigen receptor T cell (CAR-T) is the main salvage therapy for relapsed or refractory large B-cell lymphoma (r/r LBCL). However, over 50% of patients relapse after CAR-T therapy. In this work, we transduced the CAR gene into hematopoietic stem cells (HSCs) using a lentiviral vector. Chimeric antigen receptor-modified HSC (CAR-HSCs) were transplanted into mice after lethal irradiation. CAR gene transduction did not compromise the ability of HSCs to expand, self-renew, or reconstitute. CAR was expressed on T cells, natural killer cells, B cells, monocytes, and neutrophils in the periph

[Research progress on intervention of "inflammation-cancer transformation" of chronic gastritis through regulation of NF-κB-related signaling pathways by traditional Chinese medicine].

Chronic gastritis, a chronic and insidious inflammatory condition of the gastric mucosa, is characterized by nonspecific early symptoms and a high risk of progression to gastric cancer. Persistent inflammatory stimulation accelerates the "inflammation-cancer transformation" by remodeling the microenvironment, and suppressing this process is critical for gastric cancer prevention and treatment. Nuclear factor-κB(NF-κB) serves not only as a central hub for inflammatory regulation but also as a key node connecting pathways such as Toll-like receptor 4(TLR4), NOD-like receptor protein 3(

Low-Intensity Focused Ultrasound Neuromodulation for Tic Disorder Therapy: Preclinical Validation With Clinically Relevant Parameters.

Tourette syndrome (TS) is typically treated with pharmacological therapies, which often induce tolerance. Recently, neuromodulatory therapies, such as deep brain stimulation (DBS) and transcranial magnetic stimulation (TMS), have emerged as alternatives for tic disorders. However, these modalities are constrained by invasiveness and insufficient spatial resolution. To address these limitations, we employed low-intensity focused ultrasound (LIFU), an emerging noninvasive therapeutic modality previously applied in various neurological disorders, and established effective stimulation parameters t

Sex-Specific Immune Responses Mediate Host Specificity in Hookworm Infections.

Hookworm infections affect 500-700 million people worldwide and can lead to chronic conditions, such as malnutrition and anemia. The challenge of managing these infections is heightened by the absence of effective vaccines and the potential for anthelmintic resistance to develop. A comprehensive understanding of the molecular interactions between the parasite and host is vital for unraveling the complexities of infection dynamics. This study aimed to identify the immune system components responsible for host specificity in hookworms by infecting immunodeficient mouse models. Findings herein in

Sphingolipid Metabolism in the Pathogenesis of Hashimoto's Thyroiditis.

Hashimoto's thyroiditis (HT) is the most common autoimmune thyroid disorder, characterized by progressive lymphocytic infiltration, follicular destruction, tissue fibrosis, and an elevated risk of thyroid carcinoma. While the precise mechanisms underlying HT remain incompletely defined, emerging evidence implicates dysregulated sphingolipid (SPL) metabolism, particularly the sphingosine-1-phosphate (S1P) signaling axis, as a central contributor to disease pathogenesis. S1P, a bioactive lipid mediator, integrates metabolic and immunological cues to regulate immune cell trafficking, cytokine pro

pSTAT3 activation of Foxl2 initiates the female pathway underlying temperature-dependent sex determination.

Ovarian development was traditionally recognized as a "default" sexual outcome and therefore received much less scientific attention than testis development. In turtles with temperature-dependent sex determination (TSD), how the female pathway is initiated to induce ovary development remains unknown. In this study, we have found that phosphorylation of the signal transducer and activator of transcription 3 (pSTAT3) and Foxl2 exhibit temperature-dependent sexually dimorphic patterns and tempo-spatial coexpression in early embryos of the red-eared slider turtle (Trachemys scripta elegans). Game changer: How Janus kinase inhibitors are reshaping the landscape of ulcerative colitis management.

Recent advancements in the treatment landscape of ulcerative colitis (UC) have ushered in a new era of possibilities, particularly with the introduction of Janus kinase (JAK)-signal transducer and activator of transcription inhibitors. These novel agents offer a paradigm shift in UC management by targeting key signaling pathways involved in inflammatory processes. With approved JAK inhibitors (JAKis), such as tofacitinib, filgotinib, and upadacitinib, clinicians now have powerful tools to modulate immune responses and gene expression, potentially revolutionizing the treatment algorithm for UC.

Delivery of Synthetic Interleukin-22 mRNA to Hepatocytes via Lipid Nanoparticles Alleviates Liver Injury.

Hepatocellular injury, a pivotal contributor to liver diseases, particularly hepatitis, lacks effective pharmacological treatments. Interleukin-22 (IL-22), crucial for liver cell survival, shows potential in treating liver diseases by regulating repair and regeneration through signal transducer and activator of transcription 3 (STAT3) activation. However, the short half-life and off-target effects limit its clinical applications. To address these issues, lipid nanoparticles are employed to deliver synthetic IL-22 mRNA (IL-22/NP) for in situ IL-22 expression in hepatocytes. The study reveals th

Uridine phosphorylase-1 promotes cell viability and cell-cycle progression in human epidermal keratinocytes via the glycolytic pathway.

Glycolysis is vital for the excessive proliferation of keratinocytes in psoriasis, and uridine phosphorylase-1 (UPP1) functions as an enhancer of cancer cell proliferation. However, little is known about whether UPP1 promotes keratinocyte proliferation and accelerates psoriasis development. This study revealed that UPP1 facilitates cell viability and cell-cycle progression in human epidermal keratinocytes (HEKs) by modulating the glycolytic pathway. Bioinformatics analysis of UPP1 gene expression and its correlation with the Reactome revealed that UPP1 mRNA expression, cell-cycle progression,

Targeting the long non-coding RNA MIAT for the treatment of fibroids in an animal model.

Our previous studies indicated that there is overexpression of MIAT in fibroids and MIAT is a sponge for the miR-29 family in these tumors. The objective of the present study was to determine if the knockdown of MIAT in fibroid xenografts will increase miR-29 levels and reduce the expression of genes targeted by this miRNA such as collagen and cell cycle regulatory proteins in a mouse model for fibroids. Ovariectomized CB-17 SCID/Beige mice bearing estrogen/progesterone pellets were implanted subcutaneously in the flank with equal weight of fibroid explants which had been transduced by lentivi
