Base-Promoted Conversion of Propargylic Alcohols to 1,3-Enynes.

Under acidic conditions, propargylic alcohols undergo Meyer-Schuster or Rupe rearrangements to afford two isomeric α,β-unsaturated ketones. Herein, we disclose a mechanistically distinct base-mediated regioselective conversion of TMS ethers of propargylic alcohols to 1,3-enynes in high yields with broad functional-group compatibility. Alternatively, trapping of the in situ-generated lithium acetylide with electrophiles enables access to functionalized internal 1,3-enynes. Owing to the ready accessibility of propargylic alcohols, this method provides a practical and attractive entry to syntheti

Diversity of ThDP-Dependent Enzymes Forming Chiral Tertiary Alcohols.

Thiamine diphosphate (ThDP)-dependent enzymes are well known biocatalysts for CC bond-forming reactions. While this enzyme class is mainly investigated for the formation of acyloins of secondary alcohols, recent studies have expanded its scope to utilize ketones as electrophiles in asymmetric carboligation reactions for the formation of tertiary alcohols. Chiral tertiary alcohols are ubiquitous motifs in natural products and important building blocks for the synthesis of bioactive compounds. ThDP-dependent enzymes are emerging as one of the most promising classes of biocatalysts for synthesiz

Photoredox-catalyzed deoxygenative radical transformation of alcohols to sulfinamides.

Sulfinamides play a crucial role in organic synthesis and pharmaceuticals. In this study, we introduce a highly effective method for the deoxygenative radical addition to N-tritylsulfinylamine, which produces sulfinamides via photoredox catalysis. This method is compatible with a diverse array of functional groups and the resulting sulfonamides were achieved in moderate to high yields. Furthermore, the synthetic applications to access various sulfur(vi)-centered functional groups highlight the practicality of this approach.

Sugar Alcohols as Crosslinking Delay Additives for Fracturing Fluids.

The development of thermally stable fracturing fluids is essential for the effective stimulation of deep and low-permeability reservoirs. The stabilizing additives used in these fluids typically fall into three categories: crosslinking delay molecules, oxygen scavengers, and pH buffers. However, many conventional additives raise toxicity and environmental concerns, prompting the search for safer alternatives. This study investigates the use of sugar alcohols, commonly used as low-calorie sweeteners, as environmentally responsible additives for high-temperature fracturing fluids. A guar-based f

Dehydrogenative Coupling of Alcohols with Hydrazines under Nickel Catalysis.

The development of efficient and robust catalytic systems based on earth-abundant transition metals for fundamentally new transformations is crucial for sustainable chemical synthesis. Herein, an effective and selective Ni-catalyzed dehydrogenative coupling of alcohols with hydrazines with the liberation of ammonia gas is reported. Although several methods were documented for the N-alkylation reaction, the present strategy is conceptually novel, and the reaction proceeds through a pathway involving N-N bond cleavage of phenylhydrazine followed by hydrogen autotransfer. This unprecedented metho

Trivalent Nickel-Catalyzing Electroconversion of Alcohols to Carboxylic Acids.

The comprehension of activity and selectivity origins of the electrooxidation of organics is a crucial knot for the development of a highly efficient energy conversion system that can produce value-added chemicals on both the anode and cathode. Here, we find that the potential-retaining trivalent nickel in NiOOH (Fermi level, -7.4 eV) is capable of selectively oxidizing various primary alcohols to carboxylic acids through a nucleophilic attack and nonredox electron transfer process. This nonredox trivalent nickel is highly efficient in oxidizing primary alcohols (methanol, ethanol, propanol, b

Antidotes for poisoning by alcohols that form toxic metabolites.

The alcohols methanol, ethylene glycol and diethylene glycol share many characteristics. The most important is that the compounds themselves are relatively nontoxic but are metabolized, initially by alcohol dehydrogenase, to various toxic intermediates. These compounds are readily available worldwide in commercial products as well as in homemade alcoholic beverages, both of which lead to most of the poisonings, from either unintentional or intentional ingestion. Although relatively infrequent, toxic alcohol poisonings do unfortunately occur in outbreaks and can result in severe morbidity and m

Pincer Nickel-Catalyzed Olefination of Alcohols with Benzylphosphine Oxides.

N,N,P-Pincer nickel complexes effectively catalyze reaction of alcohols with benzylphosphine oxides to form alkenes in good yields. The protocol suits for a wide scope of substrates and generates only E-configurated alkenes. The method also shows good compatibility of functional groups. Methoxy, methylthio, trifluoromethyl, ketal, fluoro, chloro, bromo, thienyl, and furyl groups are tolerated. The mechanism studies support that the reaction proceeds through catalytic dehydrogenation of alcohols to aldehydes or ketones followed by condensation with benzyldiphenylphosphine oxides in the presence

Cobalt-Catalyzed Photo-Semipinacol Rearrangement of Unactivated Allylic Alcohols.

We report a photochemical method for the semipinacol rearrangement of unactivated allylic alcohols. Aliphatic as well as aromatic groups participate as migrating groups, yielding a variety of α,α-disubstituted ketones. The reaction proceeds under mild conditions and is compatible with ethers, esters, halides, nitriles, carbamates, and substituted arenes. The operationally simple and fully catalytic conditions prescribe 1 mol % benzothiazinoquinoxaline as organophotocatalyst, 0.5 mol % Co-salen, and 10 mol % lutidinium triflate and, importantly, display reactivity complementary to procedures em

Enable biomass-derived alcohols mediated alkylation and transfer hydrogenation.

A single-atom catalyst with generally regarded inert Zn-N4 motifs derived from ZIF-8 is unexpectedly efficient for the activation of alcohols, enabling alcohol-mediated alkylation and transfer hydrogenation. C-alkylation of nitriles, ketones, alcohols, N-heterocycles, amides, keto acids, and esters, and N-alkylation of amines and amides all go smoothly with the developed method. Taking the α-alkylation of nitriles with alcohols as an example, the α-alkylation starts from the (1) nitrogen-doped carbon support catalyzed dehydrogenation of alcohols into aldehydes, which further condensed with nit

Deracemization of racemic alcohols combining photooxidation and biocatalytic reduction.

We described a cascade reaction for deracemization of racemic alcohols combining photooxidation and enzymatic reduction under mild conditions without the isolation of intermediate ketones. Using different ketoreductases, a variety of racemic alcohols can be successfully converted into (R)- or (S)-enantiomers in high yields (up to 95%) and stereoselectivity (up to 99%).

Ruthenium-Catalyzed 1,3-Aryl Redox Isomerization of Allylic Alcohols.

The isomerization of allylic alcohols to the corresponding carbonyl compounds is a well-established reaction in organic synthesis. However, 1,3-carbon migration is a quite challenging field, and thus transformation has remained elusive until now. Herein, we present the ruthenium-catalyzed intramolecular 1,3-aryl migrative isomerization, which provides a mild and environmentally friendly method to synthesize various ketones with high step- and atom-economy and broad substrate scope. Meanwhile, the Ru(III)-catalyzed C(sp3)-C(aryl) bond cleavage of unactivated allylic alcohols may serve as a heur

Iron-Catalyzed C-C Single-Bond Cleavage of Alcohols.

An iron-catalyzed deconstruction/hydrogenation reaction of alcohols through C-C bond cleavage is developed through photocatalysis, to produce ketones or aldehydes as the products. Tertiary, secondary, and primary alcohols bearing a wide range of substituents are suitable substrates. Complex natural alcohols can also perform the transformation selectively. A investigation of the mechanism reveals a procedure that involves chlorine radical improved O-H homolysis, with the assistance of 2,4,6-collidine.

Direct Trifluoromethylation of Alcohols Using a Hypervalent Iodosulfoximine Reagent.

The direct trifluoromethylation of a variety of aliphatic alcohols using a hypervalent iodosulfoximine reagent afforded the corresponding ethers in moderate to good yields (14-72 %). Primary, secondary, and even tertiary alcohols, including examples derived from natural products, underwent this transformation in the presence of catalytic amounts of zinc bis(triflimide). Typical reaction conditions involved a neat mixture of 6.0 equivalents of the alcohol with 1.0 equivalent of the reagent, with the majority of reactions complete within 2 h with 2.5 mol % of the Lewis acid catalyst. Enzymatic synthesis of enantiopure alcohols: current state and perspectives.

Enantiomerically pure alcohols, as key intermediates, play an essential role in the pharmaceutical, agrochemical and chemical industries. Among the methods used for their production, biotechnological approaches are generally considered a green and effective alternative due to their mild reaction conditions and remarkable enantioselectivity. An increasing number of enzymatic strategies for the synthesis of these compounds has been developed over the years, among which seven primary methodologies can be distinguished as follows: (1) enantioselective water addition to alkenes, (2) enantioselectiv

In Vitro Effects of Amino Alcohols on Echinococcus granulosus.

Cystic echinococcosis is a globally distributed zoonotic disease, which is caused by the larval stage of Echinococcosus granulosus sensu lato. The chemotherapy of the disease is limited to the use of benzimidazoles. Recently, mefloquine and its analogues, aminoalcohol-carbazole, and some amino alcohol derivatives were reported to display inhibitory effects on parasites. Here, the activities of 130 amino alcohol compounds against E. granulosus were tested on protoscoleces and germinal cells at a concentration of 20 μg/ml over a period of three days. As a result, sixteen compounds totally were e

Stereoselective Chiral Recognition of Amino Alcohols with 2,2'-Dihydroxybenzil.

2,2'-Dihydroxybenzil is demonstrated to be a highly diastereoselective stereodynamic receptor for the chiral recognition of amino alcohols. The receptor by forming diimine compounds with amino alcohols showed good (11:1) to excellent (>50:1) diastereoselectivity in chloroform. The existence of intramolecular hydrogen bonding with amino alcohols only in an axial conformer is demonstrated by 1H NMR and CD spectroscopy, X-ray crystallography, and DFT computations. The exciton chirality method can be used with diazo-attached 2,2'-dihydroxybenzil.

Copper-catalyzed trifluoromethylation of trisubstituted allylic and homoallylic alcohols.

An efficient copper-catalyzed trifluoromethylation of trisubstituted allylic and homoallylic alcohols with Togni's reagent has been developed. This strategy, accompanied by a double-bond migration, leads to various branched CF3-substituted alcohols by using readily available trisubstituted cyclic/acyclic alcohols as substrates. Moreover, for alcohols in which β-H elimination is prohibited, CF3-containing oxetanes are isolated as the sole product.
