Salvianolic acid B alleviates red blood cell storage lesion with GPX4 preservation and reduced lipid peroxidation.

BACKGROUND: Red blood cell (RBC) storage lesion is a cascade of biochemical and physical alterations that impair transfusion efficacy and safety. This study explored the role and underlying mechanism of salvianolic acid B (Sal B) against RBC storage lesion, with a specific focus on a ferroptosis-like process. MATERIALS AND METHODS: RBC from healthy volunteers were stored in MAP additive solution for 35 days, and samples were analyzed at weekly intervals. Ferroptosis was determined by the levels of cytosolic reactive oxygen species (ROS), antioxidant enzymes superoxide dismutase and glutathion

Endoplasmic reticulum stress inhibition reduces hypertension through the preservation of resistance blood vessel structure and function.

OBJECTIVE: Our purpose was to determine if endoplasmic reticulum stress inhibition lowers blood pressure (BP) in hypertension by correcting vascular dysfunction. METHODS: The spontaneously hypertensive rat (SHR) was used as a model of human essential hypertension with its normotensive control, the Wistar Kyoto rat. Animals were subjected to endoplasmic reticulum stress inhibition with 4-phenylbutyric acid (4-PBA; 1 g/kg per day, orally) for 5 weeks from 12 weeks of age. BP was measured weekly noninvasively and at endpoint with carotid arterial cannulation. Small mesenteric arteries were remov

A whole blood-based perfusate provides superior preservation of myocardial function during ex vivo heart perfusion.

BACKGROUND: Ex vivo heart perfusion (EVHP) provides the opportunity to resuscitate unused donor organs and facilitates assessments of myocardial function that are required to demonstrate organ viability before transplantation. We sought to evaluate the effect of different oxygen carriers on the preservation of myocardial function during EVHP. METHODS: Twenty-seven pig hearts were perfused ex vivo in a normothermic beating state for 6 hours and transitioned into working mode for assessments after 1 (T1), 3 (T3), and 5 (T5) hours. Hearts were allocated to 4 groups according to the perfusate com

The outcomes of perineal urethrostomy with preservation of the dorsal urethral plate and urethral blood supply.

OBJECTIVES: To describe the surgical technique and outcomes for perineal urethrostomy. We sought to identify factors that predicted surgery failure within our patient cohort and to describe key aspects of the operation aimed at preservation of the dorsal urethral plate and longitudinal blood supply within the urethra. METHODS: We reviewed our prospectively collected database and identified 45 men who underwent definitive perineal urethrostomy from 1989 to 2009. Primary success was defined as no need for additional treatment. If 1 urethral dilation was performed, outcome was defined as a secon

A novel bronchial artery catheterization technique with preserved blood flow in an ovine model.

The authors devised a novel bronchial artery catheterization technique to deliver agents directly into bronchial circulation with preserved blood flow in an awake ovine model. A polyurethane catheter was inserted into bronchial artery via an incision into the 4th intercostal space. Regional blood flow of the airway was measured by fluorescent microspheres before cannulation, after cannulation, and 7 days after the operative procedure. The blood flows were also measured in a sham group (no cannulation, no ligation, n = 6), cannulation group (bronchial artery cannulation, n = 5), and ligation gr

Ethyleneglycol-bis-(beta-aminoethylether)tetraacetate as a blood anticoagulant: preservation of antigen-presenting cell function and antigen-specific proliferative response of peripheral blood mononuclear cells from stored blood.

We examined the progressive and irreversible loss of antigen-specific lymphoproliferative responses in peripheral blood mononuclear cells (PBMC) obtained from blood exposed for prolonged periods to EDTA as an anticoagulant. The responses of these lymphocytes to interleukin-2 or to concanavalin A were, however, unaffected. The observed loss was not due to depletion of metal ions by EDTA, since the addition of several divalent cations to whole blood during storage in EDTA or to lymphocytes from EDTA-stored blood during antigen stimulation in vitro did not alleviate the defect. Reconstitution of

Optimal management of blood glucose, blood pressure and atrial fibrillation to reduce the risk of heart failure with preserved ejection fraction.

BACKGROUND: Type 2 diabetes mellitus (T2DM), hypertension and atrial fibrillation (AF) are risk factors for heart failure with preserved ejection fraction (HFpEF). AIM: To examine the effects of the simultaneous control of all three conditions on new-onset HFpEF in this population. METHODS: This prospective cohort study enrolled 552 patients with T2DM, hypertension and AF, but without clinical signs or symptoms of heart failure. The participants were followed up for 5 years to examine the effects of glycaemic control (haemoglobin A1c: <7.0%, 7.0%-8.0% and >8.0%), blood pressure (BP) control

Glibenclamide Attenuates Brain Edema After Intracerebral Hemorrhage Through Preservation of Blood-Brain Barrier Integrity and Glymphatic Function.

Brain edema is a major contributor to secondary injury following intracerebral hemorrhage (ICH). Neurochemical disturbances, including disruption of blood-brain barrier (BBB) tight junctions and altered aquaporin-4 (AQP4) polarization, are central to post-hemorrhagic fluid imbalance. However, their association with glymphatic dysfunction and tissue-level alterations remains incompletely understood. A rat model of ICH was established and treated with glibenclamide. BBB integrity, glymphatic function, and AQP4 polarization were systematically evaluated using molecular, histological, and tracer-b

Stepwise neutrophil-to-neuron targeting nanotherapy for mitochondrial protection and blood-brain barrier preservation in ischemic stroke.

Ischemic stroke is a leading cause of mortality and long-term disability worldwide, yet the clinical efficacy of reperfusion therapies remains severely constrained by a narrow therapeutic time window and secondary ischemia-reperfusion injury. Preserving the viability of the ischemic penumbra through early, multi-target neuroprotection represents a promising strategy to extend this window; however, effective approaches that simultaneously address neuronal mitochondrial dysfunction and blood-brain barrier (BBB) disruption are lacking. Here, we report a dual-modal, stepwise-targeting lipid nanopa

MSCs act as biopatches for blood-retinal barrier preservation to enhance functional recovery after retinal I/R.

Retinal ischemia/reperfusion (I/R) is one of the most common pathologies of many vision-threatening diseases and is caused by blood-retinal barrier (BRB) breakdown and the resulting inflammatory infiltration. Targeting BRB is promising for retinal I/R treatment. Mesenchymal stromal cells (MSCs) are emerging as novel therapeutic strategies. Although intravitreal injection targets the retina, the restricted number of injected cells still requires the precise biodistribution of MSCs near the injury site. Here, we found that retinal I/R led to BRB breakdown, which induced protein and cell leakage

Natural and Assisted Conceptions in Male Patients With Blood Cancers Who Stored Their Semen for Fertility Preservation.

PURPOSE: The recent improvement in the prognosis of blood cancers has focused attention toward fertility preservation. We aimed to provide real-world insights into sperm quality and rate of conception, both naturally and through assisted reproductive technology (ART), in men who stored their semen before treatments for hematologic cancers. MATERIALS AND METHODS: Patients with hematologic cancer who cryopreserved semen between 1987 and 2018 had a postbanking follow-up and survived for at least 5 years after therapies were included. A total of N = 464 patients were selected. RESULTS: Overall,

Endovascular treatment of spontaneous isolated common and external iliac artery dissections with preservation of pelvic blood flow.

Spontaneous isolated dissection of iliac arteries is very rare, with few reports in the literature. Medical, surgical, and endovascular treatment modalities have all been used to manage iliac artery dissections. We report a case of symptomatic, isolated, spontaneous dissection of the common iliac and external iliac arteries. Both dissections were successfully treated by separate percutaneous stent-graft placement, preserving hypogastric artery flow. This technique is interesting because it provides adequate sealing of proximal and distal dissection sites while preserving hypogastric artery and

A generalized consideration of myocardial preservation with cold crystalloid versus warm blood cardioplegia in heart valve replacement.

Continuous warm blood cardioplegia was widely used, as an effective means of myocardial preservation, in open heart surgery. The comparisons of myocardial protective effects between traditional cold crystalloid and warm blood cardioplegia, however, have been based mainly on hemodynamics, cardiac function and myocardial metabolism, other than clinical outcome. The present study was designed to examine myocardial protective effects by assessing clinical outcome, enzyme levels and myocardial cytochemistry. Twenty patients undergoing heart valve replacement were divided randomly into two groups: G

Camptothecin: solubility, in-vitro drug release, and effect on human red blood cells and sperm cold preservation.

BACKGROUND: Camptothecin (CPT) is an anticancer drug, and is not employed in the clinic because of its high hydrophobicity and low active form stability. CPT may also have potential for use in cold preservation. OBJECTIVE: To overcome these drawbacks, CPT solubility variations in the presence of cyclodextrins (CDs) and polyethylene glycol (PEG) were evaluated by Higuchi solubility experiments. MATERIALS AND METHODS: CPT was encapsulated in different cyclodextrins and polyethylene glycol using a co-evaporation method. The CPT interactions with CDs and PEG 6000 were investigated by Fourier-tra

Human placental mesenchymal stem cells improve stroke outcomes via extracellular vesicles-mediated preservation of cerebral blood flow.

BACKGROUND: Besides long-term trans-differentiation into neural cells, benefits of stem cell therapy (SCT) in ischemic stroke may include secretion of protective factors, which partly reflects extracellular vesicle (EVs) released by stem cell. However, the mechanism(s) by which stem cells/EVs limit stroke injury have yet to be fully defined. METHODS: We evaluated the protection effect of human placenta mesenchymal stem cells (hPMSC) as a potential form of SCT in experimental ischemic stroke 'transient middle cerebral artery occusion (MCAO)/reperfusion' mice model. FINDINGS: We found for the

Choosing the uterine preservation surgery for placental polyp determined by blood flow evaluation: A retrospective cohort study.

BACKGROUND: A placental polyp is an intrauterine polypoid mass or pedunculated mass occurring from residual trophoblastic tissue following abortion, cesarean section or vaginal delivery. Recently uterine preservation surgery represented by transcervical resection (TCR) has been performed for placental polyps. However TCR without intravascular intervention, including uterine artery embolization (UAE) may cause profound bleeding which necessitate emergency laparotomy. METHODS: Seventeen cases of placental polyp were retrospectively examined. We divided cases into two groups: strong vascularity

Intensive Blood Pressure Management Preserves Functional Connectivity in Patients with Hypertension from the Systolic Blood Pressure Intervention Randomized Trial.

BACKGROUND AND PURPOSE: The Systolic Blood Pressure Intervention (SPRINT) randomized trial demonstrated that intensive blood pressure management resulted in slower progression of cerebral white matter hyperintensities, compared with standard therapy. We assessed longitudinal changes in brain functional connectivity to determine whether intensive treatment results in less decline in functional connectivity and how changes in brain functional connectivity relate to changes in brain structure. MATERIALS AND METHODS: Five hundred forty-eight participants completed longitudinal brain MR imaging, i

Didymin mitigates neuroinflammation and preserves blood-brain barrier integrity after subarachnoid hemorrhage.

OBJECTIVE: Subarachnoid hemorrhage (SAH) is a highly lethal and disabling type of stroke. The main causes of poor prognosis are neuroinflammation, blood-brain barrier (BBB) disruption and brain edema following hemorrhage. Didymin has shown neuroprotective effects in intracerebral hemorrhage; however, its regulatory role in SAH remains unclear. METHODS: The rat SAH model was established using the internal carotid artery puncture method, while an in vitro model was developed by stimulating human brain microvascular endothelial cells (HBMECs) with hemoglobin (Hb). Following didymin treatment, ne

Blood Glucose Homeostasis is Preserved in Patients with Mild-to-moderate Spinocerebellar Ataxia Type 2.

Spinocerebellar ataxia type 2 (SCA2) is a neurodegenerative disorder that shows cerebellar glucose hypometabolism, systemic hypermetabolism and weight loss. This study aimed to explore novel molecular mechanisms of disease for SCA2, based on the assessment of blood glucose homeostasis. A case-control and correlational study was conducted in 79 normoglycemic Cuban patients with SCA2 and 83 sex- and age-matched control subjects during a fasting state. A subset of 20 patients with SCA2 and 19 control individuals underwent an oral glucose tolerance test (OGTT). Several indices for assessing blood
