Single-subject-based whole-brain MEG slow-wave imaging approach for detecting abnormality in patients with mild traumatic brain injury.

Traumatic brain injury (TBI) is a leading cause of sustained impairment in military and civilian populations. However, mild TBI (mTBI) can be difficult to detect using conventional MRI or CT. Injured brain tissues in mTBI patients generate abnormal slow-waves (1-4 Hz) that can be measured and localized by resting-state magnetoencephalography (MEG). In this study, we develop a voxel-based whole-brain MEG slow-wave imaging approach for detecting abnormality in patients with mTBI on a single-subject basis. A normative database of resting-state MEG source magnitude images (1-4 Hz) from 79 healthy

Modeling Pressure-Dependent Wave and Vessel Compliance in the Brain Following Middle Cerebral Artery Occlusion.

OBJECTIVE: This study demonstrates the impact of alterations in pressure, vascular compliance, arterial pulsatility, and autoregulation on tissue perfusion following middle cerebral artery (MCA) occlusion using mathematical modeling. METHODS: Our previous mathematical model of the cerebral circulation is expanded to include vessel compliance and pulsatility of blood flow. An experimentally-obtained pressure waveform is used as an incoming boundary condition to simulate the effects of vascular compliance and pulsatility of flow on perfusion following MCA occlusion. The waveform is adjusted to

Auditory Stimulation of Slow-Wave Sleep Promotes Recovery after Brain Injury in an Animal Model.

OBJECTIVE: Traumatic brain injury (TBI) significantly reduces the quality of life for millions of survivors worldwide, causing persistent brain tissue damage and cognitive impairments, with no established therapeutic interventions currently available. Slow-wave activity, a hallmark of deep sleep, has been implicated in recovery after TBI, but pharmacological approaches to enhance it lack specificity and scalability, complicating efforts to identify slow-wave activity as a direct mechanistic contributor and severely limiting clinical translation. METHODS: To overcome these limitations, we deve

Dynamics of the "Cognitive" Brain Wave P3b at Rest for Alzheimer Dementia Prediction in Mild Cognitive Impairment.

Alzheimer's disease (AD) is the most common cause of dementia that involves a progressive and irrevocable decline in cognitive abilities and social behavior, thus annihilating the patient's autonomy. The theoretical assumption that disease-modifying drugs are most effective in the early stages hopefully in the prodromal stage called mild cognitive impairment (MCI) urgently pushes toward the identification of robust and individualized markers of cognitive decline to establish an early pharmacological intervention. This requires the combination of well-established neural mechanisms and the devel

Riding the wave into wellbeing: A qualitative evaluation of surf therapy for individuals living with acquired brain injury.

Nature has long demonstrated the capacity to facilitate wellbeing. Interventions involving the natural environment such as surf therapy, are increasingly being used to facilitate aspects of wellbeing in clinical populations. However, explorations of how nature-based interventions such as surf therapy may be used to promote wellbeing in the context of neurorehabilitation are missing from the peer-reviewed literature. Here we characterize the experience of a five-week surfing intervention involving fifteen adults living with the psycho-social and cognitive sequelae of acquired brain injury. Insi

When brain networks crash: connectivity, slow-wave activity and neural replay in Alzheimer's disease.

Brain connectivity disruption is a hallmark of neurodegenerative disorders, including Alzheimer's disease (AD), yet how network breakdown gives rise to cognitive deficits remains unclear. Here, we review the evidence bearing on three interconnected processes that have been proposed to link molecular pathology to cognitive decline. First, we summarize evidence that misfolded proteins propagate along brain connectivity pathways, making the connectome the biological substrate for disease progression. Second, we discuss the divergent trajectory reported across the AD continuum, an initial compensa

A Pilot Study of the Feasibility of In-Ear EEG for Long-Term Temporal Lobe Brain Wave Monitoring.

Background: Traditional scalp electroencephalography (EEG), though clinically validated, presents significant limitations, including lengthy setup, patient discomfort, and limited mobility. These factors hinder its utility for long-term ambulatory monitoring. In-ear EEG offers a promising alternative by leveraging the ear canal's anatomical proximity to the temporal lobes and natural mechanical stability. To address this, we developed SeizEAR, a standardized, three-size in-ear EEG system using paste-based electrodes with cross-ear referencing. Methods: This pilot study evaluated SeizEAR device

A comparison of continuous-wave fNIRS with quantitative fMRI-derived indices of brain function in the visual cortex.

Functional near-infrared spectroscopy (fNIRS) and functional magnetic resonance imaging (fMRI) both rely on the phenomenon of neurovascular coupling (NVC) to probe brain activity through their sensitivity to cerebral blood oxygenation. However, the relationship between fNIRS chromophores (oxy- and deoxyhaemoglobin, HbO and HbR), and fMRI (Blood Oxygen Level Dependent and Arterial Spin Labeling, BOLD and ASL) measurements, and whether this relationship remains consistent across subjects and physiological conditions, has only been partially characterised. We acquired concurrent continuous-wave f

Role of Yoga and Meditation as Complimentary Therapeutic Regime for Stress-Related Neuropsychiatric Disorders: Utilization of Brain Waves Activity as Novel Tool.

During recent decades, stress-related neuropsychiatric disorders such as anxiety, depression, chronic tension headache, and migraine have established their stronghold in the lives of a vast number of people worldwide. In order to address this global phenomenon, intensive studies have been carried out leading to the advancement of drugs like anti-depressants, anxiolytics, and analgesics which although help in combating the symptoms of such disorders but also create long-term side effects. Thus, as an alternative to such clinical practices, various complementary therapies such as yoga and medita

Effects of gabapentin on brain hyperactivity related to pain and sleep disturbance under a neuropathic pain-like state using fMRI and brain wave analysis.

Neuropathic pain is the most difficult pain to manage in the pain clinic, and sleep problems are common among patients with chronic pain including neuropathic pain. In the present study, we tried to visualize the intensity of pain by assessing neuronal activity and investigated sleep disturbance under a neuropathic pain-like state in mice using functional magnetic resonance imaging (fMRI) and electroencephalogram (EEG)/electromyogram (EMG), respectively. Furthermore, we investigated the effect of gabapentin (GBP) on these phenomena. In a model of neuropathic pain, sciatic nerve ligation caused

The use of hyperventilation in the treatment of plateau waves in two patients with severe traumatic brain injury: contrasting effects on cerebral oxygenation.

We present the case reports of two patients with severe traumatic brain injury who, in the absence of external stimuli, developed episodes of acute elevation of intracranial pressure (plateau waves) associated with jugular bulb oxyhemoglobin (SjO2) desaturation, severe reduction of cerebral tissue PO2 (PbrO2), and deterioration of neurological status. In all of these episodes hyperventilation was successful in extinguishing plateau waves, but in one patient it was associated with an improvement of both the global (increased SjO2) and local (increased PbrO2) cerebral perfusion, while in the oth

Noninvasive ultrashort wave neuromodulation enhances glymphatic clearance to alleviate traumatic brain edema and Alzheimer's pathology.

The glymphatic system clears cerebral waste, yet strategies to enhance its function remain elusive. Here, we show that 40.68 MHz ultrashort wave (USW) rapidly enhances glymphatic bulk flow in healthy mice within 10 minutes. USW potentiates both glymphatic influx and efflux, concomitant with increased Delta wave neuronal synchronization, as confirmed by local field potentials and electroencephalograms. Chemogenetic silencing confirms that USW-driven glymphatic enhancement depends on neuronal activity, while pharmacological blockade of aquaporin-4 (AQP4) abolishes the effects, revealing dual rel

Mild Traumatic Brain Injury Alters Hippocampal Sharp-Wave Ripple Properties: Implications in Memory Impairments.

A large body of evidence has shown that sharp wave ripples (SWRs), highly synchronized oscillations of the local field potential (LFP) in the CA1 pyramidal layer of the hippocampus, play a prominent role in memory consolidation. SWRs typically occur in periods of non-REM sleep, but have also been observed during awake rest periods of animals engaged in cognitive tasks. These events have been observed in rodents, non-human primates, and humans, suggesting they are evolutionarily conserved and contribute to memory formation. Numerous neurological diseases and insults, including traumatic brain i

Multimodal assessment of the effects of radial pressure wave therapy on spastic muscles in patients with traumatic brain injury.

OBJECTIVES: Radial pressure wave therapy (rPWT) has shown promise in reducing spastic symptoms; however, its application in patients with spasticity following traumatic brain injury, particularly in those with prolonged disorders of consciousness (pDoC), remains underreported. The study aimed to investigate the immediate effects of rPWT on spastic muscles in patients with TBI-associated pDoC. METHODS: Ten patients with pDoC and upper limb spasticity and ten healthy controls received a single session of rPWT (2.5 Bar, 2000 shots) to the biceps brachii. Assessments were conducted at four time p

Triangular Wave tACS Improves Working Memory Performance by Enhancing Brain Activity in the Early Stage of Encoding.

Working memory is an executive memory process that includes encoding, maintenance, and retrieval. These processes can be modulated by transcranial alternating current stimulation (tACS) with sinusoidal waves. However, little is known about the impact of the rate of current change on working memory. In this study, we aimed to investigate the effects of two types of tACS with different rates of current change on working memory performance and brain activity. We applied a randomized, single-blind design and divided 81 young participants who received triangular wave tACS, sinusoidal wave tACS, or

Meningeal Damage and Interface Astroglial Scarring in the Rat Brain Exposed to a Laser-Induced Shock Wave(s).

In the past decade, signature clinical neuropathology of blast-induced traumatic brain injury has been under intense debate, but interface astroglial scarring (IAS) seems to be convincing. In this study, we examined whether IAS could be replicated in the rat brain exposed to a laser-induced shock wave(s) (LISW[s]), a tool that can produce a pure shock wave (primary mechanism) without dynamic pressure (tertiary mechanism). Under certain conditions, we observed astroglial scarring in the subpial glial plate (SGP), gray-white matter junctions (GM-WM), ventricular wall (VW), and regions surroundin
