The unusual phenotype of CD3+ T lymphocyte expressing B220, a marker originally attributed to B lymphocytes, was first observed in the liver of Fas/Fas-L-deficient mice as a marker of apoptotic T lymphocytes. However, other CD3+B220+ T lymphocyte populations were later described in the periphery as functional cytotoxic or regulatory cells, for example. Then, in this work, we studied whether hepatic CD3+B220+ T lymphocytes could play a role in experimental Trypanosoma cruzi infection. In control and infected mice, we observed two subpopulations that could be discerned based on CD117 expression,

At least two membrane receptors have been defined through which human T lymphocytes can be induced to proliferate and differentiate, namely the CD3-Ti antigen receptor complex and the CD2 molecule. Monoclonal antibodies directed at either CD2 or CD3 induce intracellular second messenger production and subsequent protein phosphorylation. On most human non-B lymphocytes, CD3-Ti and CD2 are coexpressed and seem to be functionally interrelated. But there are minor subpopulations in which these receptor systems can transduce signals despite a mutually exclusive expression, indicating that CD3-Ti an

We have derived 33 independent T cell clones from the cerebrospinal fluid (CSF) of a patient with subacute sclerosing panencephalitis using a single T cell cloning method. 6% (2 of 33) of these clones express the T cell receptor gamma (TCR-gamma) protein and are called CSF TCR-gamma clones. Phenotypic analyses of the CSF TCR-gamma clones indicate that they are WT-31-, CD3+, CD4-, and CD8-. The TCR-gamma protein exists on the cell surface as part of an 85-kD disulphide-linked dimer noncovalently associated with the CD3 polypeptides. The CSF TCR-gamma clones have NK-like activity that can be inh

Flow cytometry can establish T cell clonality by detecting a restricted expression pattern of the T cell receptor (TCR) β constant region (TRBC), expressed in association with CD3. However, T cell neoplasms frequently lose surface expression of the CD3/TCR complex, posing a challenge to demonstrating T cell lineage and clonality. To address this challenge, here we present a 12-color flow cytometry panel, called cytoTCR, to characterize cytoplasmic expression of CD3/TCR complex components. We apply cytoTCR to 38 patient specimens with immunophenotypically abnormal T cell populations, demonstrat

PROBLEM: The natural killer (NK) cell activity is depressed in the decidua of early normal pregnancy. Recently Morii et al. (Am J Reprod Immunol 1993;29:1-4) found that all early intradecidual CD3+ T cells expressed either T cell receptor (TCR) alpha/beta or gamma/delta but that the expression of the CD3+/TCR complex was down-regulated.
METHOD: To test whether these changes in decidual cellular immunity are different among normal pregnancy, anembryonic pregnancy and recurrent spontaneous abortion, we examined the immune cell subpopulations in the decidua from these three types of pregnancy us

T cells of autoimmune-prone mice homozygous for the lpr mutation respond poorly to mitogens in terms of proliferation and of IL-2 production. In a previous study, we have correlated this deficient activation with the inability of mitogens to stimulate hydrolysis of phosphatidylinositol 4,5-bisphosphate in lpr T cells, although these cells bind mitogen and express the TCR/CD3 complex. In order to determine whether activation-deficient lpr T cells contain functional GTP-binding (G) protein(s) and phospholipase C, we examined the effects of the G protein activating agent sodium fluoride plus Al+3

T cells infiltrating (T-TIL) B cell non-Hodgkin's lymphomas (NHL) are thought to represent a local host response to the tumor. However, tumor progression in the presence of this T cell infiltrate suggests that the T-TIL may be functionally impaired. To address this issue we determined whether response to stimulation of T-TIL from 25 patients with NHL through the T cell receptor (TCR/CD3) and the interleukin-2 (IL-2) receptor (IL-2R) was intact, since activation of these receptors is important for proliferation and cytokine production. Our results demonstrate defects in response to stimulation

AIMS: Normal human pregnancy is a complex process of many immunoregulatory mechanisms which protect fetus from the activation of the maternal immune system. The aim of the study was to investigate the apoptosis of lymphocytes in peripheral blood of normal pregnant patients and healthy nonpregnant women.
METHODS: Sixty pregnant women and 17 nonpregnant women were included in the study. Lymphocytes were isolated and labeled with anti-CD3, anti-CD4, and anti-CD8 monoclonal antibodies. Apoptosis was detected by CMXRos staining and analyzed using the flow cytometric method.
RESULTS: We found sign

BACKGROUND: Myeloid cells can express lymphoid markers, including components of the CD3-TCR complex. CD3+ macrophages have been identified in both infectious and non-infectious pathologies; however, their origin and signaling mechanisms remain poorly studied, primarily due to their low prevalence in human samples. Utilizing the RAW murine macrophage cell line as a model, this study aimed to ascertain whether nutrient deprivation induces CD3 expression and to identify the signaling molecules involved in the activation of a CD3-dependent proinflammatory profile.
METHODS: Firstly, the impact of

Immune mobilizing monoclonal T-cell receptors Against Cancer (ImmTAC®) molecules are innovative immunotherapies designed to harness the body's immune system to fight cancer. They consist of a soluble affinity-enhanced T-cell receptor (TCR) as the targeting arm, covalently linked to an antibody fragment specific to CD3 as the effector arm, enabling T-cell redirection and cancer cell killing. TCRs can target the intracellular antigenic landscape via recognition of peptides presented by surface localized Human Leukocyte Antigen (HLA) proteins, unlocking a choice of targets that are specific to th

T-cell engagers redirect cytotoxic lymphocytes toward malignant cells, but their efficacy in solid tumors is limited by uneven antigen expression and inefficient effector-target contact. Here, we report a modular dual-tumor-recognition trispecific T-cell nano-engager (Tri-NanoTCE) generated by Fc-mediated assembly of anti-PD-L1, anti-EGFR, and anti-CD3 IgG1 antibodies on an FcγR1-HSA/PLLA nano-adaptor. PD-L1 and EGFR provide complementary tumor recognition and anchoring, whereas CD3 recruits T cells. Tri-NanoTCE retained the binding activity of all antibodies, showed efficient antibody loading

Classically, B and T cells are differentiated by their mutually exclusive expression of surface markers such as CD3 and CD20, but it is now established that a few percent of T cells in human blood display also CD20. We analyzed CD3+CD20+ cells in detail by combining single-cell proteogenomics, FACS, and ImageStream. Thereby, we identified CD3+ B cells and found that about 40% of plasmablasts displayed CD3 in both blood and tonsils. In tonsils, where T-B interactions occur, about 80% of follicular helper T (Tfh) cells displayed CD20. In vitro, coactivation of T and B cells induced marker exchan

OBJECTIVE: CD3γ deficiency is an ultrarare autosomal recessive inborn error of immunity characterized by immune dysregulation and variable immunodeficiency. To date, only 16 predominantly pediatric cases have been reported. Here, we describe two additional adult patients with CD3γ deficiency and provide a comprehensive reevaluation of all previously published cases.
METHODS: Clinical, immunological, and genetic investigations were performed in two adult patients presenting with immune dysregulation and hypogammaglobulinemia. Whole-genome sequencing was used to identify pathogenic CD3G variant

CD3ε is an essential subunit of the T-cell receptor complex. It is required for T cell activation and immune modulation. Dysregulated CD3ε signaling results in abnormal T cell activation, contributing to the pathogenesis of various immune-related disorders, including autoimmune diseases. Although anti-CD3ε antibodies, such as muromonab-CD3, can effectively modulate T cell responses, their use is often limited by severe adverse effects caused by excessive T cell activation. In this study, we present K108.5, a fully human monoclonal antibody (mAb) specific to human CD3ε (hCD3ε), designed to modu

Bispecific antibodies, a novel immunotherapy with promising efficacy against multiple myeloma, form immune synapses between T-cell surface marker CD3 and malignant cell markers, including B-cell maturation antigen (BCMA), FcRH5, and G protein-coupled receptor GPRC5D. These bispecific antibodies so effectively deplete plasma cells (and to some extent T-cells) that patients are at increased risk of developing infections. A systematic review and meta-analysis of infections in published studies of patients with myeloma treated with bispecific antibodies was conducted to better characterize the inf

High rates of relapse and poor prognosis confer an urgent need for novel therapeutic agents for B cell non-Hodgkin lymphomas (B-NHLs). Herein, we describe a human IgG-like anti-CD79b/CD3 bispecific antibody (IBI38D9-L) that selectively depletes antigen-positive malignant B cells as an alternative treatment option for relapsed or refractory NHL patients. The antitumor activity and mechanism of action of IBI38D9-L were investigated in vitro using B-NHL cell lines and human primary effector cells and in vivo using xenograft models reconstituted with human PBMCs (peripheral blood mononuclear cells

INTRODUCTION: Severe brain injuries pose one of the most important problems on our health care because of their high morbidity and mortality.
MATERIAL AND METHODS: A group of 89 patients after severe brain injury (Glasgow Coma Scale< or =8) was included into our research of detecting the changes of immune system parameters and their relation to the application of mild hypothermia during the early period after the insult.
RESULTS: In both of the groups CD3+ and CD4+ lymphocytic levels decreased significantly after the insult and gradually got to normal (p<0.01). The NK cells levels have chang

The modulation of the cytokine response to coccidioidal antigen by lymphocytes from donors with coccidioidomycosis was examined. In initial experiments, samples from 13 healthy immune donors and seven donors with active coccidioidomycosis anergic to the coccidioidal antigen T27K were assessed for CD3 lymphocyte expression of intracellular IFN-gamma using whole blood analysis. Addition of 10 ng/ml of recombinant IL-12 significantly increased response to T27K among immune and anergic subjects (p<0.05), but the percent of cells expressing IFN-gamma was still significantly greater for immune subje
