Feline calicivirus (FCV) infection causes nasal discharge, oral mucosa inflammation, ulcerations, gingivitis, and conjunctivitis, often progressing to chronic gingivostomatitis, severe pneumonia, and fatal systemic infections. With no antivirals currently available, poly(sodium 4-styrene sulfonate) (PSSNa) was identified in 2019 as a safe inhibitor in vitro. In this preliminary single-center, randomized, double-blind, placebo-controlled field study, we further characterized the PSSNa's safety profile and tested its efficacy in cats after topical oral application. Twenty-eight cats were enrolle

Developing novel antiviral drugs has always been a significant forefront in biological medicine research. Antiviral drugs can be extracted, purified, and synthesized from various biological sources and by different methods. However, they are less explored in veterinary medicine for animal viruses. This research primarily selected feline calicivirus (FCV) to screen the novel antiviral drug against animal viruses. A preliminary screening from a natural product library was conducted, with subsequent assessments to ascertain their toxicity levels and antiviral capabilities. The results showed that

Feline vaccination guidelines recommend less frequent boosters for the core vaccines (rhinotracheitis, calicivirosis and infectious panleucopenia). Most guidelines recommend boosters at 3-yearly intervals after a basic vaccination including primary vaccination and revaccination one year later. The objective of this study was to assess the duration of immunity induced by PUREVAX(®) RCPCh FeLV, a non-adjuvanted vaccine against feline rhinotracheitis, calicivirosis, infectious panleucopenia, chlamydiosis and leukemia. After primary vaccination followed by revaccination one year later with a vacci

High-resolution melting (HRM) analysis, a post-polymerase chain reaction (PCR) application in a single closed tube, is the straightforward method for simultaneous detection, genotyping, and mutation scanning, enabling more significant dynamic detection and sequencing-free turnaround time. This study aimed to establish a combined reverse-transcription quantitative PCR and HRM (RT-qPCR-HRM) assay for diagnosing and genotyping feline calicivirus (FCV). This developed method was validated with constructed FCV plasmids, clinical swab samples from living cats, fresh-frozen lung tissues from necropsi

Feline calicivirus (FCV) belongs to the Caliciviridae, which comprises small RNA viruses of both medical and veterinary importance. Once infection has occurred, FCV can persist in the cat population, but the molecular mechanism of how it escapes the innate immune response is still unknown. In this study, we found FCV strain 2280 to be relatively resistant to treatment with IFN-β. FCV 2280 infection inhibited IFN-induced activation of the ISRE (Interferon-stimulated response element) promoter and transcription of ISGs (Interferon-stimulated genes). The mechanistic analysis showed that the expre

The aim of this study was to perform the molecular characterization of conserved and variable regions of feline calicivirus capsid genome in order to investigate the molecular diversity of variants in Brazilian cat population. Twenty-six conjunctival samples from cats living in five public short-term animal shelters and three multicat life-long households were analyzed. Fifteen cats had conjunctivitis, three had oral ulceration, eight had respiratory signs (cough, sneeze and nasal discharge) and nine were asymptomatic. Feline calicivirus were isolated in CRFK cells and characterized by reverse

Feline calicivirus (FCV) is a highly infectious respiratory pathogen of domestic cats. The prevalence of FCV in the general cat population is high, particularly in multi-cat households, largely because many clinically recovered cats remain persistently infected carriers. In order to assess how FCV circulates in such groups and to assess the contribution that each individual animal makes to the epidemiology of the disease, we have carried out the first detailed analysis of long-term shedding patterns of FCV in individual cats within naturally infected colonies. The prevalence of FCV in each of

BACKGROUND: Feline calicivirus (FCV) is a significant causative pathogen of upper respiratory tract disease and virulent systemic infection in feline populations, yet specific targeted antiviral agents for FCV infection remain unavailable.
METHODS: A feline-derived single-chain variable fragment (scFv) phage display library targeting the VP1-CDE neutralizing region of FCV was constructed using peripheral blood mononuclear cells isolated from cats immunized with recombinant VP1-CDE protein. Positive scFv clones were screened via three rounds of affinity panning and phage ELISA. The scFv-Fc fus

In this study, nine monoclonal antibodies (mAbs) against the FCV VP1 protein were successfully prepared using traditional hybridoma technology, and all of them could simultaneously recognize linear and conformational epitopes. Their subtypes mainly included IgG1/kappa, IgG2a/kappa, IgG1/lambda, IgG2b/kappa, IgG2b/lambda, and IgM/kappa. Based on the mAb 8F9 with the highest blocking activity, a blocking enzyme-linked immunosorbent assay (ELISA) method for detecting cat calicivirus antibodies was established. After systematic optimization, the antigen coating concentration was determined to be 1

We investigated primitively the molecular basis of the neural spread of a feline calcivirus isolate (FCV-S) from the spinal cord of a cat that died after manifesting excitation. Experimental infections of cats with three clones from parent virus isolate FCV-S, isolated based on plaque size, were performed, and virus recovery from the spinal cord and the nucleotide and predicted amino acid sequences of the viral capsid protein region (ORF2) were compared. In the experimental infection with the one-time cloned virus (C1L1) isolated from a large plaque, the C1L1 was recovered from the spinal cord

Norovirus (NoV) causes serious gastrointestinal disease worldwide and is regarded as an important foodborne pathogen. Due the difficulties of in vitro cultivation for human NoV, alternative caliciviruses (i.e., feline calicivirus, FCV, or murine NoV) have long been used as surrogates for in vitro assessment of the efficacy of antivirals. Essential oils (EOs) are natural compounds that have displayed antimicrobial and antioxidant properties. We report in vitro the virucidal efficacy of four EOs, Melissa officinalis L. EO (MEO), Thymus vulgaris L. EO (TEO), Rosmarinus officinalis L. EO (REO), an

BACKGROUND: The feline viral rhinotracheitis, calicivirus, and panleukopenia (FVRCP) vaccine, prepared from viruses grown in the Crandell-Rees feline kidney cell line, can induce antibodies to cross-react with feline kidney tissues.
OBJECTIVES: This study surveyed the prevalence of autoantibodies to feline kidney tissues and their association with the frequency of FVRCP vaccination.
METHODS: Serum samples and kidneys were collected from 156 live and 26 cadaveric cats. Antibodies that bind to kidney tissues and antibodies to the FVRCP antigen were determined by enzyme-linked immunosorbent ass

Hand disinfection is an important measure to prevent transmission of norovirus (formerly called Norwalk-like viruses) from hands or environmental surfaces to other objects. Therefore, three types of alcohol (ethanol, 1- and 2-propanol) were examined for their virus-inactivating properties against feline calicivirus (FCV) as a surrogate for norovirus. Tests were performed as quantitative suspension assays or as in vivo experiments with artificially contaminated fingertips. The in vitro experiments showed that 1-propanol was more effective than ethanol and 2-propanol for the inactivation of FCV:

Reverse transcription-polymerase chain reaction (RT-PCR) was used to direct the amplification of a 670 to 680 base pair segment that included the hypervariable regions of the capsid protein gene of feline calicivirus (FCV). The segment was amplified from 13/13 cultivated FCV strains including 12 field isolates collected over 18 years. The sensitivities of culture and this RT-PCR for the detection of FCV in conjunctival swabs over the course of infection were then compared and correlated with clinical signs in 5 vaccinated and 3 unvaccinated experimentally-infected cats. Conjunctival swabs were

Feline calicivirus (FCV) is a highly contagious pathogen prevalent in domestic cats, often leading to upper respiratory tract infections and oral diseases. In clinical settings, particularly veterinary hospitals and shelters, nosocomial outbreaks of FCV can occur, sometimes involving virulent systemic disease (VSD) strains that result in severe systemic illness with high mortality rates. This retrospective study reviews the clinical and diagnostic features of 15 cats that developed postoperative ulcerative tongue lesions, ptyalism and skin lesions after elective ovariohysterectomy (OVH) perfor

Sanitation of environmental surfaces with chlorine based-disinfectants is a principal measure to control outbreaks of norovirus or Clostridium difficile. The microbicidal activity of chlorine-based disinfectants depends on the free available chlorine (FAC), but their oxidative potential is rapidly eliminated by organic matter. In this study, the microbicidal activities of weakly acidified chlorous acid water (WACAW) and sodium hypochlorite solution (NaClO) against feline calcivirus (FCV) and C. difficile spores were compared in protein-rich conditions. WACAW inactivated FCV and C. difficile sp

Feline calicivirus (FCV) is a significant pathogen responsible for upper respiratory tract disease and oral ulceration in felids. Genetic and antigenic variation in circulating FCV strains limits the utility of current diagnostic assays, and no validated genotype II-specific antibodies exist for rapid test development. In this study, we targeted the C, D, E, and partial F regions of the major capsid protein VP1 (termed VP1-CDEF'). The recombinant VP1-CDEF' protein was expressed in a prokaryotic system and used to immunize SPF chickens. A chicken-derived phage display single-chain variable frag

The protective efficacy of intranasal (IN) administration of inactivated feline calicivirus (FCV) vaccine against homologous or heterologous FCV infection was investigated. Groups of cats immunized with the experimental inactivated, non-adjuvanted FCV vaccine via either the IN or subcutaneous (SC) route were exposed to homologous or highly heterologous FCV. Both the IN and SC immunization protocols established robust protection against homologous FCV infection. Although neither immunization regimen conferred protection against the heterologous strain, clinical scores and virus titres of oral s

BACKGROUND: Feline calicivirus (FCV) is a common virus, found worldwide, mainly responsible for chronic ulceroproliferative faucitis and periodontitis. This virus has a high mutation rate, leading to the presence of numerous FCV strains in the field. The objectives of this study was to evaluate and compare the efficacy of two vaccines (Leucofeligen™ FeLV/RCP and Purevax™ RCP FeLV), which differ by their nature (live vs. inactivated) and the vaccinal strains, against circulating FCV strains. Thirty 9-week-old specific pathogen free (SPF) kittens were thus randomised into 3 groups and were eithe
