Lipid infusion impairs physiologic insulin-mediated capillary recruitment and muscle glucose uptake in vivo.

Infusion of triglycerides and heparin causes insulin resistance in muscle. Because the vascular actions of insulin, particularly capillary recruitment, may contribute to the increase in glucose uptake by skeletal muscle, we investigated the effects of Intralipid/heparin infusion on the hemodynamic actions of insulin during clamp conditions. Saline or 10% Intralipid/heparin (33 U/ml) was infused into anesthetized rats at 20 microl/min for 6 h. At 4 h into the saline infusion, a 2-h hyperinsulinemic (3 mU. min(-1).kg(-1))-euglycemic clamp was conducted (Ins group). At 4 h into the lipid infusion

Effect of fluid pressure on the hydraulic conductance of interstitium and fenestrated endothelium in the rabbit knee.

A synovial cavity is separated from plasma by synovial intima in series with capillary endothelium. Because 20% of the intimal surface is bare interstitium, the system is a convenient model for the study of passive transport through serial endothelial and interstitial layers. Here hydraulic flow across the composite barrier was investigated in forty-seven knees of isolated, blood-perfused rabbit hindquarters, at intra-articular pressures between 4 and 30 cmH2O. In order to measure barrier conductance at constant intra-articular pressure, pressure on the opposite side of the barrier was varied,

[Macro- and micro-hemocirculatory shifts in the small intestine during pressor reflexes].

In acute experiments on cats, effects of occlusion of common carotid arteries, stimulation of afferent fibers of the brachial plexus, application of acetylcholine to reflexogenous zones of peritoneum and mesentery of the small intestine on macro- and microcirculation of the isolated small intestine perfused under constant flow, were studied. Under effects of occlusion and afferent stimulation, arteries and veins of the small intestine were constricted, whereas in stimulation of abdominal reflexogenous zones their reactions were different. All the reflexes induced augmentation of the capillary

Capillary plugging by granulocytes and the no-reflow phenomenon in the microcirculation.

Granulocytes are large, stiff viscoelastic cells that adhere naturally to the vascular endothelium. On their passage through the capillary network they have to be deformed, and recent evidence indicates that they may impose a significant hemodynamic resistance. The entry time of granulocytes into capillaries is about three orders of magnitude longer than that for red cells. Inside the capillary the granulocytes move with a lower velocity than red cells. Under conditions when the capillary perfusion pressure is reduced and/or elevated levels of inflammatory products are present that increase th

Acute effects of furosemide and mannitol on central haemodynamics in the early postoperative period.

The effects of furosemide and mannitol on central haemodynamics in the early postoperative period were investigated in 16 patients, who had undergone upper abdominal surgery. Measurements were performed prior to, and then 10, 30, 50 and 90 min after postoperative drug administration. Furosemide administration resulted in reductions of cardiac output, mean pulmonary arterial, pulmonary capillary wedge, and mean systemic arterial pressures, while systemic vascular resistance increased. Mannitol administration on the other hand, caused increases of cardiac output, mean pulmonary arterial and pulm

Definition and classification of pulmonary hypertension.

Pulmonary hypertension is defined as an increase of mean pulmonary arterial pressure ≥25 mmHg at rest as assessed by right heart catheterization. According to different combinations of values of pulmonary wedge pressure, pulmonary vascular resistance and cardiac output, a hemodynamic classification of pulmonary hypertension has been proposed. Of major importance is the pulmonary wedge pressure which allows to distinguish pre-capillary (pulmonary wedge pressure ≤15 mmHg) and post-capillary (pulmonary wedge pressure >15 mmHg) pulmonary hypertension. Pre-capillary pulmonary hypertension includes

Noninvasive determination of small vessel distensibility in man.

The present study describes and illustrates a noninvasive method for computing extensibility of the 'resistance vessels', from an experimentally obtained resistance vs transmural pressure plot, in a maximally dilated vascular bed of man. Limb blood flow was measured plethysmographically following 10 min of arterial occlusion, and blood pressure was measured by the arm cuff method. A ratio of two resistance values taken 30 mm Hg apart was used to compute percent change in radius and the extensibility, E. The computed values of E were 0.71 mm Hg-1 for normal hand and 0.13, 0.22 and 0.69 for the

Combining hyperpolarized 129Xe MR imaging and spectroscopy to noninvasively estimate pulmonary vascular resistance.

Hyperpolarized 129Xe MRI/MRS enables quantitative mapping of function in lung airspaces, membrane tissue, and red blood cells (RBCs) within the pulmonary capillaries. The RBC signal also exhibits cardiogenic oscillations that are reduced in precapillary pulmonary hypertension (PH). This effect is obscured in patients with concomitant defects in transfer from airspaces to RBCs, which increase RBC oscillation amplitudes. Here, we provide a framework for interpreting RBC oscillations and show their relationship to pulsatile blood flow, capillary blood volume, capillary compliance, and impedance o

[Pulmonary capillary pressure and the exercise test].

In the present work with radiocardiography the change of pulmonary capillary pressure as well as cross correlations of stroke index, cardiac index, pulmonary blood volume, pulmonary arterial pressure and pulmonary dispersion volume which reflects pulmonary capillary volume have been studied during muscular exercise. With increase in cardiac output the mathematical estimation showed decrease in pulmonary capillary pressure while stroke index, pulmonary blood volume and pulmonary dispersion volume increased. The pulmonary arterial pressure also rose but this increase was insignificant. The estim

Vascular resistance of carotid and vertebral arteries is associated with retinal microcirculation measured by laser speckle flowgraphy in patients with type 2 diabetes mellitus.

AIMS: Evaluation of the retinal microcirculation is key to understanding retinal vasculopathies, such as diabetic retinopathy. Laser speckle flowgraphy (LSFG) has recently enabled us to directly evaluate the vascular resistance in both retinal vessels and capillaries, non-invasively. We therefore assessed whether retinal vessel blood flow and/or the capillary microcirculation are associated with blood flow in the cervical arteries in diabetic patients without severe retinopathy. METHODS: We enrolled 110 type 2 diabetes patients, with no or mild non-proliferative diabetic retinopathy, in this

A thromboxane analog increases pulmonary capillary pressure but not permeability in the perfused rabbit lung.

Thromboxane has been implicated as a mediator of pulmonary hypertension and pulmonary edema in acute respiratory failure. Pulmonary edema may result from increased pulmonary capillary hydrostatic pressure or from increased pulmonary vascular permeability. We therefore studied the effects of a stable thromboxane analog, U46619, on these two parameters in the perfused rabbit lung. Pulmonary capillary pressure was measured by the double vascular occlusion method, and pulmonary vascular permeability was estimated by measurement of the pulmonary fluid filtration coefficient (Kf). U46619 infusion pr

[Preparation of chiral monolithic column with covalently bonded cellulose and their application to rapid enantioseparation].

A chiral monolithic capillary column for rapid enantioseparation was prepared by covalently bonding of cellulose tris(4-methylbenzoate) (CTMB) on N-acryloxysuccinimide-based monolith. The preparation and derivatization conditions of the monolithic column were optimized. The successful grafting of CTMB was confirmed on the characterizations of the infrared spectrum and the cathodic electroosmotic flow (EOF). The effects of acetic acid concentration and methanol content on the enantioseparation were studied. The solvent resistance, reproducibility and stability of the monolithic column have also

Haemostatic effect of prednisolone in thrombocytopenia.

In a short-time study, 8 patients with thrombocytopenia and clinical signs of bleeding were experimentally treated with prednisolone as the only therapeutic measure to control the bleeding tendency. 2 patients had idiopathic thrombocytopenic purpura and 6 acute non-lymphocytic leukaemia. 1 of the leukaemic patients was thrombocytopenic on admission. 5 had thrombocytopenia due to antileukaemic chemotherapy. The capillary fragility test, evaluated blindly, and bleeding time determination were performed just before treatment and repeated once after 2-4 d of treatment. Platelet counts were carried

Effects of prostaglandin E1 on microvascular haemodynamics in progressive systemic sclerosis.

The effects of prostaglandin E1 infusion on nailfold capillary haemodynamics were studied in eight patients with Raynaud's phenomenon secondary to progressive systemic sclerosis. Using a modified Landis microinjection technique the mean (+/- SEM) transcapillary pressure gradient was increased during and six weeks after infusion by 13.9 +/- 3.2 cm H2O (p less than 0.05) and 5.5 +/- 2.5 cm H2O (p less than 0.05) respectively. Capillary red cell velocity measured in two patients by video television microscopy also increased during and after infusion with prostaglandin E1. Six patients claimed sub

Acute impairment of insulin-mediated capillary recruitment and glucose uptake in rat skeletal muscle in vivo by TNF-alpha.

The vascular actions of insulin may contribute to the increase in glucose uptake by skeletal muscle. We have recently shown that when capillary recruitment by insulin is blocked in vivo, an acute state of insulin resistance is induced. Another agent that may have vascular effects is the inflammatory cytokine tumor necrosis factor-alpha (TNF-alpha), which has been reported to play an important role in the insulin resistance of obesity, type 2 diabetes, and sepsis in both animals and humans. Thus, in the present study, we have investigated the effect of an intravenous 3-h TNF treatment (0.5 micr

Microchip capillary electrophoresis dairy device using fluorescence spectroscopy for detection of ciprofloxacin in milk samples.

Detecting antibiotics in the milk supply chain is crucial to protect humans from allergic reactions, as well as preventing the build-up of antibiotic resistance. The dairy industry has controls in place at processing facilities, but controls on dairy farms are limited to manual devices. Errors in the use of these manual devices can result in severe financial harm to the farms. This illustrates an urgent need for automated methods of detecting antibiotics on a dairy farm, to prevent the shipment of milk containing antibiotics. This work introduces the microchip capillary electrophoresis dairy d

A Case Report of Pulmonary Arterial Hypertension Associated With Hereditary Hemorrhagic Telangiectasia Successfully Treated With Riociguat.

Hereditary hemorrhagic telangiectasia (HHT) is a rare vascular disorder that is occasionally complicated by pulmonary hypertension (PH). In most cases where PH is associated with HHT, it is typically of a post-capillary type, while pre-capillary PH is relatively rare. Herein, we report a rare case of pre-capillary PH in a 44-year-old woman. She presented with recurrent epistaxis, mucocutaneous telangiectasias, a family history of hemorrhagic events, and hepatic arteriovenous malformations. She met the four Curaçao criteria and was diagnosed with HHT. Right heart catheterization demonstrated se

Configuration of the microcirculation in pre-eclampsia: possible role of the venular system.

OBJECTIVE: To test the hypothesis that during pre-eclampsia microvascular function and structure are disturbed, which contributes to raised venular resistance. METHODS: The microcirculation of the skin and bulbar conjunctiva was studied in 11 women with preeclampsia and nine parous controls, both in the third trimester and 3 months postpartum. Using intravital videomicroscopy, arteriolar and venular diameters were determined in the conjunctiva. In addition, skin capillary densities and morphology were determined. RESULTS: Conjunctival venular diameters were 30% smaller in pre-eclampsia compa

Post-Capillary Pulmonary Hypertension: Clinical Review.

Pulmonary hypertension (PH) caused by left heart disease, also known as post-capillary PH, is the most common etiology of PH. Left heart disease due to systolic dysfunction or heart failure with preserved ejection fraction, valvular heart disease, and left atrial myopathy due to atrial fibrillation are causes of post-capillary PH. Elevated left-sided filling pressures cause pulmonary venous congestion due to backward transmission of pressures and post-capillary PH. In advanced left-sided heart disease or valvular heart disease, chronic uncontrolled venous congestion may lead to remodeling of t
