Lumpy skin disease threat in Europe: Current situation, transmission dynamics and future prospects.

Lumpy skin disease (LSD), caused by Lumpy Skin Disease Virus (LSDV, genus Capripoxvirus), is an emerging transboundary disease of cattle and water buffalo. Although rarely fatal, it results in severe economic losses due to decreased productivity, hide damage, infertility, and trade limitations. This review summarizes the etiology, susceptible hosts, modes of transmission, and advances in control, vaccination, and treatment strategies, with a particular focus on Europe, where spread into previously unaffected areas has recently been reported. Since its first identification in Zambia in 1929, LS

Laser-Induced Graphene Enabled Highly Sensitive Lumpy Skin Disease Detection and Vaccine Validation.

Lumpy skin disease (LSD), caused by Capripoxvirus, poses significant threats to global livestock due to its high contagion and economic impact. Current diagnostic methods are limited by their dependence on laboratories, while vaccine efficacy monitoring relies on cumbersome virus neutralization tests (VNTs). To address these challenges, a laser-induced graphene (LIG)-based electrochemical immunoassay has been developed for the sensitive detection of LSD and the validation of vaccination. The platform employs recombinant F13L antigen and IgM antibody, configured for dual applications: F13L quan

Development and Validation of a New DIVA Real-Time PCR Allowing to Differentiate Wild-Type Lumpy Skin Disease Virus Strains, Including the Asian Recombinant Strains, from Neethling-Based Vaccine Strains.

The current epidemic in Asia, driven by LSDV recombinants, poses difficulties to existing DIVA PCR tests, as these do not differentiate between homologous vaccine strains and the recombinant strains. We, therefore, developed and validated a new duplex real-time PCR capable of differentiating Neethling-based vaccine strains from classical and recombinant wild-type strains that are currently circulating in Asia. The DIVA potential of this new assay, seen in the in silico evaluation, was confirmed on samples from LSDV infected and vaccinated animals and on isolates of LSDV recombinants (n = 12),

Development of recombinant goatpox virus expressing Echinococcus granulosus EG95 vaccine antigen.

Goatpox disease and cystic hydatidosis may be simultaneously endemic in a given area. Their pathogens are goatpox virus (GPV) and Echinococcus granulosus (E. granulosus), respectively. Both E. granulosus EG95 subunit vaccine and live-attenuated GPV AV41 vaccine have been widely used for prevention of both diseases in China. However, it has been rarely reported that a bivalent vaccine is developed to prevent both diseases. The GPV is an ideal vector for developing recombinant multivalent vaccines to deliver immunogenic proteins in animals. In this study, we constructed an EG95 antigen-expressin

Biphasic phenotypic adaptation of sheeppox virus consequent to serial passaging in heterologous host cell type.

Sheeppox virus (SPPV) is a highly host-restricted capripoxvirus, and its biological behavior in heterologous cells remains poorly defined. Although heterologous cell lines such as Vero cells are widely used for SPPV propagation and vaccine production, the effects of prolonged adaptation to these systems on viral replication and release dynamics have not been systematically examined. In this study, the SP3-Nur strain of SPPV (PV167793.1) was serially passaged 45 times in Vero cells. The parent virus (P0) was assessed in lamb testicular (LT) cells, while Vero-adapted passage levels (P15, P30, an

Genetic Evolutionary Analysis of Lumpy Skin Disease Virus Strain Under Immune Pressure Exerted by Heterologous Goat Poxvirus Vaccines.

Lumpy skin disease (LSD) is an important infectious disease that threatens the global cattle industry. Recently, LSD has occurred frequently in Asia. The attenuated goat poxvirus (GTPV) vaccine is widely used to prevent LSD in cattle in China; however, sporadic cases of LSD still occur in immunized cattle. This study aimed to investigate the genetic evolution of isolated LSD virus (LSDV) strain under GTPV heterologous vaccine immune pressure. We isolated a new strain of LSDV, named LSDV/China/SX/2023, from a cattle herd immunized with a GTPV-attenuated vaccine in Shaanxi Province, Northwest Ch

Lessons Learned from Active Clinical and Laboratory Surveillance during the Sheep Pox Virus Outbreak in Spain, 2022-2023.

In September 2022, more than 50 years after its eradication from Spain, Sheep pox virus was confirmed by laboratory analysis in sheep showing characteristic lesions. This was the start of an outbreak that lasted 9 months and infected 30 farms dispersed over two different areas, Andalusia and Castilla-La Mancha. Early after the initial confirmation, an active surveillance based on clinical inspection with laboratory confirmation of sheep with clinical signs was started in restricted areas. This allowed the confirmation of Sheep pox in 22 out of 28 suspected farms, where limited numbers of sheep

Comparative innocuity and efficacy of live and inactivated sheeppox vaccines.

BACKGROUND: Sheeppox (SPP) is one of the priorities, high-impact animal diseases in many developing countries, where live attenuated vaccines are routinely used against sheeppox virus (SPPV). In an event of an SPP outbreak, historically disease-free countries would hesitate to use of live vaccines against SPPVdue to the safety and trade reasons. Currently no killed SPPV vaccines are commercially available. In this study, we developed an inactivated Romanian SPPVvaccine and assessed its efficacy and potency in comparison with a live attenuated Romanian SPPV vaccine. Four naïve sheep were vaccin

Multicolor quantum dot-encoded microspheres for the fluoroimmunoassays of chicken newcastle disease and goat pox virus.

Semiconductor nanocrystals (or quantum dots, QDs) have the potential to overcome some of the limitations encountered by traditional fluorophores in fluorescence labeling applications. The unique spectroscopic properties of QDs make them hold immense promise as versatile labels for biological applications. In this work, we employ the layer-by-layer (LbL) method for the construction of bio-functional multicolor QD-encoded microspheres. Polystyrene microspheres with diameter of 3 microm were used as templates for the deposition of different sized CdTe QDs/polyelectrolyte multilayers. Two differen

A review: Surveillance of lumpy skin disease (LSD) a growing problem in Asia.

Lumpy skin disease (LSD) is a viral disease caused by lumpy skin disease virus (LSDV), a member of Capripoxvirus, genus of Poxviridae family. It is a transboundary infection of monetary significance that primarily affects water buffaloes and cattle. LSD was known to be once endemic in Saharan regions of Africa but later on reported in central Asian and neighboring countries of Pakistan like, India, Iran and China. It is a vector borne disease and arthropods are believed to be the main perpetrators. It is discernible by its high morbidity and low mortality. Characteristic lumps on skin and high

Capripox disease in Ethiopia: Genetic differences between field isolates and vaccine strain, and implications for vaccination failure.

Sheeppox virus (SPPV), goatpox virus (GTPV) and lumpy skin disease virus (LSDV) of the genus Capripoxvirus (CaPV) cause capripox disease in sheep, goats and cattle, respectively. These viruses are not strictly host-specific and their geographical distribution is complex. In Ethiopia, where sheep, goats and cattle are all affected, a live attenuated vaccine strain (KS1-O180) is used for immunization of both small ruminants and cattle. Although occurrences of the disease in vaccinated cattle are frequently reported, information on the circulating isolates and their relation to the vaccine strain

Enhancing lumpy skin disease control: Effective competitive and indirect ELISAs for serological surveillance.

Lumpy skin disease (LSD), caused by the LSD virus (LSDV) from the Capripoxvirus genus, affects cattle, water buffalo, and wild bovines, leading to significant economic losses. Characterised by fever, skin nodules, and mucosal lesions, LSD raises global concerns due to vector-borne transmission. The World Organisation for Animal Health (WOAH) classifies LSD as a notifiable disease, emphasising the need for rapid diagnostic methods for timely disease confirmation and control. This study evaluates the performance of two previously developed ELISA tests - competitive and indirect. The validation i

Design of a multi-epitope vaccine against goatpox virus using an immunoinformatics approach.

INTRODUCTION: Goatpox, a severe infectious disease caused by goatpox virus (GTPV), leads to enormous economic losses in the livestock industry. Traditional live attenuated vaccines cause serious side effects and exist a risk of dispersal. Therefore, it is urgent to develop efficient and safer vaccines to prevent and control of GTPV. METHODS: In the present study, we are aimed to design a multi-epitope subunit vaccine against GTPV using an immunoinformatics approach. Various immunodominant cytotoxic T lymphocytes (CTL) epitopes, helper T lymphocytes (HTL) epitopes, and B-cell epitopes from P32

A real time high-resolution melting PCR assay for detection and differentiation among sheep pox virus, goat pox virus, field and vaccine strains of lumpy skin disease virus.

In this paper, we report on the development of a real time high-resolution melting (HRM) PCR assay for detection and differentiation among sheep pox virus (SPPV), goat pox virus (GTPV), field isolates and vaccine strains of lumpy skin disease virus (LSDV) based on high-resolution melting curve analysis of their target PCR amplicons. A 111 bp region of LSDV010 ORF, which harbors unique genetic differences for each of these viral species, was selected as the PCR target in this study. During the validation of this assay using DNA from clinical isolates originated from naturally infected animals f

Goatpox virus (G20-LKV) vaccine strain elicits a protective response in cattle against lumpy skin disease at challenge with lumpy skin disease virulent field strain in a comparative study.

In this comparative study, we examine the safety of the sheeppox (SPP) and goatpox (GTP) vaccines and the protective response of these vaccines in cattle against a virulent lumpy skin disease (LSD) field strain. The vaccine safety was tested in rabbits, mice and cattle using ten times recommended dose. In the safety trial, none of the vaccinated animals showed any deviation from physiological norms or fever, inappetence or local/ generalized skin reactions. In the challenge trial, both SPP and GTP vaccine groups developed virus-neutralizing antibodies with an average titre of 2.1 log2 at 21 da

Development of a Duplex TaqMan-MGB qPCR Assay for Differential Detection of Chinese Epidemic Lumpy Skin Disease Virus Strains and Goatpox Virus.

Lumpy skin disease (LSD) is a transboundary viral disease of cattle, including Asian water buffalo and yaks, and certain wild ruminants (e.g., African buffalo, giraffe, wildebeest, eland, and Arabian oryx). It is caused by lumpy skin disease virus (LSDV), a member of the genus Capripoxvirus (family Poxviridae) together with goatpox virus (GTPV) and sheeppox virus (SPPV). High nucleotide identity and serological cross-reactivity among these viruses hinder differential diagnosis. Elimination of toxicity and enhanced detection of lumpy skin disease virus on cell culture from experimentally infected bovine semen samples.

Lumpy skin disease virus (LSDV), a poxvirus of the genus Capripoxvirus, is shed in the semen of infected bulls. The screening of semen for infectious virus requires a sensitive diagnostic method. The isolation of the virus on cell cultures and/or the polymerase chain reaction (PCR) are sensitive diagnostic tests which may be used to screen semen for LSD viral DNA prior to artificial insemination. Although cell culture detects infectious virus and is a sensitive method, there are major difficulties in using this method due to the toxic effect of semen on the cells. The aim of this study was to
