Single-cell sequencing reveals cellular heterogeneity of nucleus pulposus in intervertebral disc degeneration.

The nucleus pulposus (NP) plays a vital role in intervertebral disc degeneration (IVDD). Previous studies have revealed cellular heterogeneity in NP tissue during IVDD progression. However, the cellular and molecular alterations of diverse cell clusters during IVDD remain to be fully elucidated. NP tissues were isolated from patients with different grades of IVDD undergoing discectomy, and then subjected to single-cell RNA sequencing (scRNA-seq). Cell subsets were identified based on unbiased clustering of gene expression profiles. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes

Fly Cell Atlas: A single-nucleus transcriptomic atlas of the adult fruit fly.

For more than 100 years, the fruit fly Drosophila melanogaster has been one of the most studied model organisms. Here, we present a single-cell atlas of the adult fly, Tabula Drosophilae, that includes 580,000 nuclei from 15 individually dissected sexed tissues as well as the entire head and body, annotated to >250 distinct cell types. We provide an in-depth analysis of cell type-related gene signatures and transcription factor markers, as well as sexual dimorphism, across the whole animal. Analysis of common cell types between tissues, such as blood and muscle cells, reveals rare cell types a

Chemokine receptor CXCR7 non-cell-autonomously controls pontine neuronal migration and nucleus formation.

Long distance tangential migration transports neurons from their birth places to distant destinations to be incorporated into neuronal circuits. How neuronal migration is guided during these long journeys is still not fully understood. We address this issue by studying the migration of pontine nucleus (PN) neurons in the mouse hindbrain. PN neurons migrate from the lower rhombic lip first anteriorly and then turn ventrally near the trigeminal ganglion root towards the anterior ventral hindbrain. Previously we showed that in mouse depleted of chemokine receptor CXCR4 or its ligand CXCL12, PN ne

Cell type-specific alterations in the nucleus accumbens by repeated exposures to cocaine.

BACKGROUND: The nucleus accumbens (NAc) is a brain region critically involved in psychostimulant-induced neuroadaptations. A major proportion of NAc neurons consists of medium spiny neurons (MSNs), commonly divided into two major subsets on the basis of their expression of D1 dopamine receptors (D1R-MSNs) or D2 dopamine receptors (D2R-MSNs). Although NAc MSNs are known to undergo extensive alterations in their characteristics upon exposure to drugs of abuse, the functional and structural changes specific to each type of MSN have yet to be fully resolved. METHODS: We repeatedly injected cocain

Objective assessment of conjunctival squamous metaplasia by measures of cell and nucleus dimensions.

The main aim of this work is to present a special method for conjunctival impression cytology (CIC) and objective measures of measure cell and nucleus dimensions. CIC using Millicell® Biopore filters was obtained from 50 Caucasian adults from the nasal bulbar conjunctiva. Giemsa-stained images of cells in a monolayer were photographed with a scale bar and projected at 1,500× to generate an overlay. Measures were made of both the long (L) and short (S) dimensions of the cells and the longest dimension of the nucleus (LN). The selected images comprised sets of 10 from different individuals, whic

Connections and oculomotor projections of the superior vestibular nucleus and cell group 'y'.

Attempts were made to determine brainstem and cerebellar afferent and efferent projections of the superior vestibular nucleus (SVN) and cell group 'y' ('y') in the cat using axoplasmic tracers. Injections of HRP, WGA-HRP and [3H]amino acids were made into SVN and 'y' using two different infratentorial stereotaxic approaches. Controls were provided by unilateral HRP injections involving the oculomotor nuclear complex (OMC), the interstitial nucleus of Cajal (INC) and the deep cerebellar nuclei (DCN). Large injections of SVN almost invariably involved 'y' and dorsal parts of the lateral vestibul

Antidiabetic DPP4 inhibitor Attenuates Intervertebral Disc Degeneration via Macrophage-Nucleus Pulposus Cell Crosstalk.

BACKGROUND AND PURPOSE: Intervertebral disc degeneration (IVDD), a leading cause of low back pain, lacks effective disease-modifying therapies. Diabetes exacerbates IVDD risk, but the causal mechanisms and therapeutic potential of glucose-lowering agents remain underexplored. This study aimed to identify glucose-lowering drug targets strongly associated with IVDD using Mendelian randomisation (MR) and to validate the therapeutic efficacy and mechanisms of sitagliptin, an inhibitor of dipeptidyl peptidase 4 (DPP4), in alleviating IVDD progression. EXPERIMENTAL APPROACH: Two-sample MR analysis

Surviving the Nucleus Pulposus Desert: Next-Generation Strategies for Intervertebral Disc Cell Therapy.

BACKGROUND: Low back pain remains the leading cause of disability worldwide, with intervertebral disc degeneration representing a major biological contributor. Although cell-based therapies have shown promise in preclinical models, clinical translation has yielded modest and inconsistent outcomes. Accumulating evidence suggests that therapeutic failure reflects not only limitations in cell source or differentiation potential, but also the hostile biochemical and biomechanical microenvironment of the degenerative disc. Hypoxia, nutrient deprivation, acidity, lactate accumulation, fibrosis, sene

Cluster replicability in single-cell and single-nucleus atlases of the mouse brain.

Single-cell RNA sequencing has advanced our understanding of cellular heterogeneity. Ensuring the replicability of identified cell clusters across studies is essential for determining their biological robustness. We assess the replicability of cell clusters identified in two large mouse brain atlases, one generated using single-cell RNA sequencing and the other with single nuclei. Both profile over 4 million cells and group them into over 5,000 clusters. Using transcriptome-wide neighbor voting, we identify 2,009 reciprocally matched cluster pairs with consistent spatial localization and coord

Single-nucleus atlas of cell-type specific genetic regulation in the human brain.

Genetic risk variants for common diseases are predominantly located in non-coding regulatory regions and modulate gene expression. Although bulk tissue studies have elucidated shared mechanisms of regulatory and disease-associated genetics, the cellular specificity of these mechanisms remains largely unexplored. Here we present a comprehensive, single-nucleus multi-ancestry atlas of genetic regulation of gene expression in the human prefrontal cortex, comprising 5.6 million nuclei from 1,384 donors of diverse ancestries. Through multi-resolution analyses spanning eight major cell classes and 2

Modulating Macrophage-Nucleus Pulposus Cell Crosstalk via Sequential Drug Delivery Attenuates Disc Degeneration.

Intervertebral disc degeneration (IDD) is characterized by an inflammatory environment and dysregulation of the extracellular matrix metabolism (ECM). Improving nucleus pulposus cells (NPCs) condition first requires a supportive surrounding environment. However, how to provide an excellent microenvironment before regulating the function of the NPCs is not yet a complete strategy. In this study, a sequential drug delivery system is comprised of chitosan hydrogel (G-CS) loaded with Angiotensin (1-7) and mesoporous silica nanospheres (MSN) coating with α-mangostin to achieve synergistic effects o

Crosstalk between reactive oxygen species mediated programmed cell death of nucleus pulposus cells.

It is yet unknown what causes intervertebral disc degeneration (IVDD), a chronic inflammatory systemic illness. Lower back pain may result from the disease's primary pathogenic characteristic, which is the degeneration of nucleus pulposus cells (NPCs). While IVDD can currently be improved by medications, there are still serious toxic side effects that require immediate attention. Consequently, the identification of novel therapeutic targets depends on a thorough examination of the mechanisms underlying the onset and progression of IVDD. Particularly in NPCs, annulus fibrosus, and cartilage end

Hyaluronan composite bioink preserves nucleus pulposus cell phenotype in a stiffness-dependent manner.

Intervertebral disc degeneration is a major cause of neck and back pain, representing a significant global socioeconomic burden. The polysaccharide hyaluronan is key to maintaining disc physiology and mediating disc disease through its structural and biological roles in the nucleus pulposus, a component of the intervertebral disc highly susceptible to degeneration. In this study, we introduce a novel composite bioink designed for extrusion bioprinting of structures resembling the nucleus pulposus. Our bioink combines levels of hyaluronic acid and collagen that approach physiological concentrat

Cell type-specific epigenetic priming of gene expression in nucleus accumbens by cocaine.

A hallmark of addiction is the ability of drugs of abuse to trigger relapse after periods of prolonged abstinence. Here, we describe an epigenetic mechanism whereby chronic cocaine exposure causes lasting chromatin and downstream transcriptional modifications in the nucleus accumbens (NAc), a critical brain region controlling motivation. We link prolonged withdrawal from cocaine to the depletion of the histone variant H2A.Z, coupled with increased genome accessibility and latent priming of gene transcription, in D1 dopamine receptor-expressing medium spiny neurons (D1 MSNs) that relate to aber

Single nucleus RNA sequencing reveals glial cell type-specific responses to ischemic stroke.

Reactive neuroglia critically shape the braińs response to ischemic stroke. However, their phenotypic heterogeneity impedes a holistic understanding of the cellular composition and microenvironment of the early ischemic lesion. Here we generated a single cell resolution transcriptomics dataset of the injured brain during the acute recovery from permanent middle cerebral artery occlusion. This approach unveiled infarction and subtype specific molecular signatures in oligodendrocyte lineage cells and astrocytes, which ranged among the most transcriptionally perturbed cell types in our dataset. Cocaine-induced projection-specific and cell type-specific adaptations in the nucleus accumbens.

Cocaine craving, seeking, and relapse are mediated, in part, by cocaine-induced adaptive changes in the brain reward circuits. The nucleus accumbens (NAc) integrates and prioritizes different emotional and motivational inputs to the reward system by processing convergent glutamatergic projections from the medial prefrontal cortex, basolateral amygdala, ventral hippocampus, and other limbic and paralimbic brain regions. Medium spiny neurons (MSNs) are the principal projection neurons in the NAc, which can be divided into two major subpopulations, namely dopamine receptor D1- versus D2-expressin

Releasing dentate nucleus cells from Purkinje cell inhibition generates output from the cerebrocerebellum.

The cerebellum generates its vast amount of output to the cerebral cortex through the dentate nucleus (DN) that is essential for precise limb movements in primates. Nuclear cells in DN generate burst activity prior to limb movement, and inactivation of DN results in cerebellar ataxia. The question is how DN cells become active under intensive inhibitory drive from Purkinje cells (PCs). There are two excitatory inputs to DN, mossy fiber and climbing fiber collaterals, but neither of them appears to have sufficient strength for generation of burst activity in DN. Therefore, we can assume two pos

Dynamic interactions between Pit-1 and C/EBPalpha in the pituitary cell nucleus.

The homeodomain (HD) transcription factors are a structurally conserved family of proteins that, through networks of interactions with other nuclear proteins, control patterns of gene expression during development. For example, the network interactions of the pituitary-specific HD protein Pit-1 control the development of anterior pituitary cells and regulate the expression of the hormone products in the adult cells. Inactivating mutations in Pit-1 disrupt these processes, giving rise to the syndrome of combined pituitary hormone deficiency. Pit-1 interacts with CCAAT/enhancer-binding protein a

cAMP pathway alterations from the cell surface to the nucleus in adrenocortical tumors.

The cyclic AMP (cAMP) pathway plays a major role in the development of endocrine tissues and various molecular defects of key components of this pathway (G protein, receptors, PKA, ...) have been observed in endocrine tumors. Hypersecretion of adrenocorticotropin hormone (ACTH), the key activator of the cAMP pathway in adrenal cortex, is associated with adrenocortical hyperplasia and cortisol oversecretion (Cushing's syndrome). The best example of "illegitimate" membrane receptors expression reported is the abnormal expression of the adenylyl cyclase activating gastric inhibitory peptide recep

Emergent dynamics of adult stem cell lineages from single nucleus and single cell RNA-Seq of Drosophila testes.

BACKGROUND/AIMS: Iron (Fe) can cause tissue injury and oxidative stress by catalyzing hydroxyl radical production and lipid peroxidation. Intravenous (i.v.) Fe preparations are routinely administered to treat anemia in patients with chronic renal failure (CRF), a condition marked by oxidative stress and inflammation. In an earlier study, we showed that iron overload augments oxidative stress in the cardiovascular tissues of CRF rats. This study was designed to expand these observations to other major organs. METHODS: Rats were randomized into CRF (5/6 nephrectomized) and sham-operated control
