Organoid-based models in cerebrovascular disorders: Mechanistic insights and precision medicine.

Cerebrovascular diseases are the leading cause of death and long-term disability worldwide, including ischemic stroke, intracerebral hemorrhage, intracranial aneurysms, and cerebrovascular malformations, imposing a heavy burden on society and families. Organoid technology, as a major breakthrough in the field of biomedicine, provides a new platform for the study of cerebrovascular diseases. By constructing organoid-based models of cerebrovascular diseases, researchers can simulate their pathogenesis in vitro and deeply explore key links such as angiogenesis and neurovascular unit interactions,

Improving diagnostic approaches to cerebrovascular disorders in patients with hypertensive encephalopathy.

BACKGROUND: Modern research today proves an increase in the incidence of ischemic stroke. Risk factors for vascular disease have become more common among young people. Clinical studies have shown that hypertension, dyslipidemia, and diabetes are traditional risk factors among the elderly. The aim of the study was to improve diagnostic approaches to cerebrovascular disorders in patients with hypertensive encephalopathy (НE) by clarifying the development of pathogenesis mechanisms, based on a comprehensive clinical study. MATERIALS AND METHODS: This qualitative study was carried by clinical and

Cerebrovascular disorders and Fahr's disease: Report of two cases and literature review.

Fahr's disease is a rare idiopathic nosological entity, characterized by calcification of the basal ganglia and dentate nuclei of the cerebellum. Sometimes it may be associated to other diseases like cerebrovascular disorders. However, this link remains unclear and it needs to be further validated. We report two cases of patients with cerebrovascular disorders and Fahr's disease. In the first case, a 69-years-old woman with right internal capsule-basal ganglia haemorrhage. In the second case, a 72-years-old woman with ischemic stroke and pericallosal artery aneurysm. The physiopathology is dis

Cerebrovascular and neurological disorders associated with antiphospholipid antibodies in CSF and serum.

Paired serum and cerebrospinal fluid (CSF) samples from 70 patients with inflammatory and non-inflammatory neurological diseases, as well as 10 sera from patients with primary antiphospholipid syndrome (PAS), six of which presented with cerebrovascular ischemic syndromes, were studied for the presence of anticardiolipin antibodies (ACA) of the G and M classes. PAS sera and some selected paired CSF and serum specimens, were also analyzed for the presence of anti-phosphatidylserine (PS) and anti-phosphatidylethanolamine (PE) antibodies. High levels of IgG and IgM ACA were synthesized intrathecal

Three-dimensional transcranial Doppler blood flow mapping in patients with cerebrovascular disorders.

We investigated 60 patients with cerebrovascular disorders using a three-dimensional transcranial Doppler blood flow mapping system. A composite display of the circle of Willis is created with computer assistance, allowing accurate vessel identification and optimal data documentation of blood flow velocity and direction in the basal cerebral arteries. The basilar artery was insonated in every patient; the middle cerebral artery and the most distal internal carotid artery were found in 95% of the patients, the anterior cerebral artery in 85%, and the posterior cerebral artery in 84%. Insonation

Impaired endothelium-mediated cerebrovascular reactivity promotes anxiety and respiration disorders in mice.

Carbon dioxide (CO2), the major product of metabolism, has a strong impact on cerebral blood vessels, a phenomenon known as cerebrovascular reactivity. Several vascular risk factors such as hypertension or diabetes dampen this response, making cerebrovascular reactivity a useful diagnostic marker for incipient vascular pathology, but its functional relevance, if any, is still unclear. Here, we found that GPR4, an endothelial H+ receptor, and endothelial Gαq/11 proteins mediate the CO2/H+ effect on cerebrovascular reactivity in mice. CO2/H+ leads to constriction of vessels in the brainstem area

Cognitive-communicative disorders of right cerebrovascular accident patients and reimbursement for treatment.

Speech-language pathologists frequently diagnose and treat individuals with cognitive-communicative disorders, but they sometimes encounter difficulty receiving reimbursement for those services from third-party payers. Often, reimbursement becomes an issue because the documentation provided fails to emphasize the linguistic and communication aspects of treatment that are precisely the components that make the speech-language pathologist's role with this population unique. By analyzing how the elements of language, cognition, and communication are interactively affected for each patient and by

[Cerebrovascular disorders associated with livedo (Sneddon's syndrome): its relation to lupus anticoagulant].

The clinical manifestations of Sneddon's syndrome (cerebrovascular disorder, livedo reticularis, peripheral venous thrombosis++, cardiac pathology, obstetric pathology--fetal loss and intrauterine fetal death) are characteristic of the antiphospholipid syndrome. A lupus anticoagulant (LA), one of the types of antiphospholipid antibodies, was detected in the blood plasma of 18 out of 30 patients with Sneddon's syndrome. The negative results of LA-examination in 12 patients don't exclude the presence of other antiphospholipid antibodies. So 16 of 30 patients had anticardiolipin antibodies, among

[Features of aphasic disorders in patients with acute cerebrovascular disorder in the posterior cerebral artery basin].

Aphasia is a systemic disorder of formed speech that develops as a result of local brain lesions. Most aphasias are characterized by damage to secondary cortical fields, which in turn are responsible for the performance of the functions of gnosis and praxis, which explains the variability in the manifestations of speech disorders in patients with acute cerebrovascular accidents. However, it is necessary in each case to diagnose the central pathological mechanism, which underlies the development of the entire syndrome and determines the entire clinical picture. The most important task of a spee

Insulin Resistance Across Cerebrovascular and Related Disorders: Mechanisms, Measurement, Genetics, and Clinical Implications.

Insulin resistance (IR) Insulin resistance (IR) is a central metabolic disturbance implicated in a broad range of cardiometabolic and neurological disorders. Increasing evidence suggests that IR plays a pivotal role in the initiation and progression of cerebrovascular diseases (CeVD) and contributes to neurodegeneration through shared molecular and vascular mechanisms. In the brain and its vasculature, impaired insulin signaling-particularly involving the IRS-PI3K-Akt-GSK-3β axis-interacts with inflammation, oxidative stress, endothelial dysfunction, and neurovascular unit impairment, thereby

Validity of Aspiration Prevention Surgery in Elderly Patients with Cerebrovascular or Neurodegenerative Disorders.

The risk of aspiration pneumonia has been reported to increase with age, especially in frail elderly patients with cerebrovascular and/or neurodegenerative disorders. To prevent this type of pneumonia, aspiration prevention surgery (APS) procedures have been demonstrated to be effective. However, the comprehensive efficacy of APS, not only in terms of survival rates, but also in terms of medical costs and caregiver burden, has not been fully evaluated in elderly patients requiring aspiration care. Thus, the aim of the current study was to demonstrate the validity of APS in elderly patients wit
