Mechanistic insights into hydroxy(tosyloxy)iodobenzene-mediated ditosyloxylation of chalcones: a DFT study.

In this paper, the mechanism of the hydroxy(tosyloxy)iodobenzene (HTIB)-mediated conversion of chalcones (α,β-unsaturated carbonyl compounds) to ditosyloxy ketones is investigated. Here, at β-carbon of the chalcone, an aryl group with a para-substituent is present. Our study focuses on investigating the effect of different nature of para-substituents on the reaction mechanism. The substituents considered in the study include -OCH3, -SCH3, -Cl and -NO2 groups. For these chalcones, different possible pathways at various steps during the reaction are investigated leading to formation of α,β-ditos

GABAA and serotonergic receptors participation in anxiolytic effect of chalcones in adult zebrafish.

The prevalence of anxiety is a significant public health problem, being the 24th leading cause of disability in individuals affected by this disorder. In this context, chalcones, a flavonoid subclass obtained from natural or synthetic sources, interact with central nervous system (CNS) receptors at the same binding site as benzodiazepines, the primary drugs used in the treatment of anxiety. Thus, our study investigates the anxiolytic effect of synthetic chalcones derived from the natural product 2-hydroxy-3,4,6-trimethoxyacetophenone isolated from Croton anisodontus Müll.Arg. in modulating anx

Chalcones and Five-Membered Heterocyclic Isosteres Bind to Alpha Synuclein Fibrils in Vitro.

A series of chalcone and heterocyclic isosteres, in which the enone moiety was replaced with an isoxazole and pyrazole ring system, was synthesized and their affinities for alpha synuclein (Asyn), amyloid beta (Aβ), and tau fibrils were measured in vitro. The compounds were found to have a modest affinity and selectivity for Asyn versus Aβ fibrils and low affinity for tau fibrils. Insertion of a double bond to increase the extendable surface area resulted in an increase in affinity and improvement in selectivity for Asyn versus Aβ and tau fibrils. The results of this study indicate that compou

Nanocrystallization Effectively Improves the Oral Efficacy of an Antileishmanial Chalcone.

Background/Objectives: Cutaneous leishmaniasis (CL) is a vector-borne neglected disease that can cause permanent deformities. Current chemotherapy based on injections with toxic drugs or oral miltefosine poses many drawbacks, urging the need for new oral therapies. Here, we proposed to increase the bioavailability of NAT22, an intralesionally but not orally active antileishmanial chalcone, through nanocrystallization to promote its oral use in CL. Methods/Results: NAT22 nanocrystals were produced using a solvent-free green process of dry and wet milling that reduced NAT22 crystal sizes by arou

Chalcone Scaffolds, Bioprecursors of Flavonoids: Chemistry, Bioactivities, and Pharmacokinetics.

Chalcones are secondary metabolites belonging to the flavonoid (C6-C3-C6 system) family that are ubiquitous in edible and medicinal plants, and they are bioprecursors of plant flavonoids. Chalcones and their natural derivatives are important intermediates of the flavonoid biosynthetic pathway. Plants containing chalcones have been used in traditional medicines since antiquity. Chalcones are basically α,β-unsaturated ketones that exert great diversity in pharmacological activities such as antioxidant, anticancer, antimicrobial, antiviral, antitubercular, antiplasmodial, antileishmanial, immunos

Ultrastructural alterations in Plasmodium falciparum induced by chalcone derivatives.

OBJECTIVE: Chalcones (1, 3-diaryl-2-propen-1-ones) and their derivatives are widely explored from the past decade for its antimalarial activity. To elucidate their mechanism of action on the malaria parasite, the ultrastructural changes with the action of these derivatives in different organelles of the parasite were studied in vitro. Infected RBCs [CQ sensitive (MRC-2) and CQ resistant (RKL-9) Plasmodium strain] were treated with three chalcone derivatives 1, 2 and 3 and standard drugs, i.e., CQ and artemisinin at twice their respective IC50 values for 24 h and then harvested, washed, fixed,

Transition-Metal-Free One-Pot Synthesis of (Hetero)chalcones with Cysteine Protease Inhibitory Activity.

We report a new and efficient one-pot methodology for the synthesis of (hetero)-chalcones via direct C-H functionalization. The protocol employs directed organolithiation of aromatic and heteroaromatic substrates, followed by in situ formylation using inexpensive DMF, which not only serves as the formylating agent but also generates lithium dimethylamide as the base for the subsequent aldol condensation with (hetero)-aryl ketones. This transition-metal-free and additive-free approach enables the preparation of 23 chalcone derivatives with broad structural diversity, varying both aromatic and h

Antiparasitic activity of chalcones analogue against Trichomonas vaginalis: biochemical, molecular and in silico aspects.

Trichomonas vaginalis is the etiologic agent of trichomoniasis, a worldwide distributed sexually transmitted infection (STI) that affects the genitourinary tract. Even though this disease already has a treatment in the prescription of drugs of the 5-nitroimidazole class, described low treatments adhesion, adverse side effects and cases of resistant isolates demonstrate the need for new formulations. With this in mind, chalcones emerge as a potential alternative to be tested, being compounds widely distributed in nature, easy to chemically synthesize and presenting several biological activities

Novel Novolac Phenolic Polymeric Network of Chalcones: Synthesis, Characterization, and Thermal-Electrical Conductivity Investigation.

A series of novolac phenolic polymeric networks (NPPN) were prepared via an acid-catalyzed polycondensation reaction of formaldehyde with chalcones possessing a p-phenolic OH group. When p-hydroxybenzaldehyde was treated with formaldehyde under the same conditions, a phenolic polymer (PP) was obtained. The resulting polymers were isolated in excellent yields (83-98%). Isolated polymers (NPPN, PP) were characterized using FTIR, TGA, and XRD. The results obtained from the TGA revealed that all prepared phenolic polymers have high thermal stability at high temperatures and can act as thermosettin

In vitro inhibition effect of some chalcones on erythrocyte carbonic anhydrase I and II.

In this study, 4'-(phenylurenyl/thiourenyl)chalcones (14-25) were prepared from 4'-(phenylurenyl/thiourenyl)acetophenones and benzaldehyde derivatives by Claisen-Schmidt condensation. In vitro inhibition effects of chalcone derivatives on purified carbonic anhydrase I and carbonic anhydrase II were investigated by using the CO2 hydration method of Maren. The result showed that all the synthesized compounds inhibited the CA isoenzymes activity. 18 and 19 were found to be most active (IC50 = 25.41 μM and 23.06 μM) for hCA I, respectively. For hCA II, 24 is the most active compound (IC50 = 14.40

Solvent-controlled switch between aziridination and Beckmann-type amidation of chalcones with hydroxylammonium salts.

Herein, we disclose a solvent-controlled switch between aziridination and Beckmann-type amidation of chalcones using MsONH3OTf as a common hydroxylammonium reagent. Specifically, the reaction in TFE delivers NH-aziridine derivatives via direct aziridination, whereas switching the solvent to THF promotes a Beckmann rearrangement to afford α,β-unsaturated amides. The solvent-dependent selectivity is supported by control experiments, HRMS detection of a hydroxylamine-derived intermediate, and the isolation of a bicyclic O-sulfonyl oxime, which is interpreted as a trapped intermediate-like species

Computationally guided synthesis and biological profiling of chalcones as antioxidant and anti-inflammatory activities.

Inflammation and oxidative stress are involved in the physiological changes associated with many chronic diseases, which has led to sustained interest in small molecules with pleiotropic properties. As part of this effort, this study reports the synthesis and evaluation of six chalcones. These six synthesized derivatives were produced using acetophenone and benzaldehyde, and structures were characterised through nuclear magnetic resonance (NMR) spectral analysis and Fourier transform infrared (FTIR) spectroscopy. The compounds were tested for their in-vitro antioxidant properties using the DPP

The Role of Natural Chalcones and Their Derivatives in Targeting Prostate Cancer: Recent Updates.

Prostate cancer (PCa) is the second most prevalent cancer among men and a major cause of cancer-related mortality worldwide. Despite an initial favorable response to hormone-based therapies, many patients ultimately develop an advanced and lethal form of the disease, referred to as castration-resistant PCa (CRPC). CRPC is associated with poor prognosis and a lack of effective curative treatments. As a result, new alternatives or improved therapeutic strategies to combat this life-threatening condition are urgently needed. Chalcones, also referred to as 1,3-diphenyl-2-propen-1-ones, have attrac
