Analysis of testicular migration during the fetal period in humans.

PURPOSE: We present an analysis concerning the testicular migration and its position correlated to body weight, crown-rump length and gestational age during the fetal period in humans without congenital abnormalities. MATERIALS AND METHODS: We studied bilaterally 142 testes taken from 71 fresh human fetuses between 10 and 35 weeks after conception. The fetuses were also evaluated in regard to crown-rump length and body weight. The position of the testes was correlated to the fetal parameters. RESULTS: In 37 fetuses (74 testes) at 10 to 23 weeks after conception only 7 testes (9.45%) had migr

Retinoic acid embryopathy: case report and review of literature.

Isotretinoin use as a treatment for acne has increased tremendously and, with it, the problems of associated birth defects. We feel that pathologists should be familiar with isotretinoin embryopathy and its pathogenesis in order to assist in differentiating this syndrome from other genetic syndromes that involve branchial arch defects, such as DiGeorge syndrome or velocardiofacial syndrome. Although selected autopsy findings have been presented in epidemiologic reports, to our knowledge a detailed autopsy report has not been published. We therefore wish to present a complete case study of isot

[Early embryonic formation of the human vomeronasal organ].

We studied the origin and development of the vomeronasal system in early human embryos of 10-18 mm crown-rump length under normal and pathological conditions. The formation of the vomeronasal organ from the vomeronasal groove and placode in a 10-mm embryo is described. We propose that the vomeronasal cavity originates in the schizocoel way. We studied the development of the vomeronasal and olfactory nerves. Attention is paid to the development of the nasolacrimal duct and the epithelial plug. The possible use of the vomeronasal system by embryos during the first seven weeks of development is d

The development of ependyma in the human fetal brain: an immunohistological and electron microscopic study.

The stratified inner layer of the embryonic fetal brain, the ventricular zone (VZ), contains glial fibrillary acidic protein (GFAP)-positive cell bodies of radial glia. The adult cerebral ventricle is lined by a single layer of cuboidal, ciliated common ependymal cells which are, immunohistologically, GFAP negative. In late gestation, the ventricular lining is formed by tanycytes, ependymal cells with short, intensely GFAP-positive basal fibres. The development of ependyma was examined, morphologically and immunohistologically, in human fetal brain from between 11 weeks gestation to 6 months p

Teratogenic and cytogenetic effects of ivermectin and its interaction with P-glycoprotein inhibitor.

Experiments in animals proved that P-glycoprotein (Pgp) forms a functional barrier between maternal and fetal blood circulation in the placenta, thus protecting the fetus from exposure to xenobiotics during pregnancy. In this study we aimed to demonstrate the effects of administration of ivermectin (anthelmentic drug, Pgp substrates), either alone or simultaneously with verapamil (Pgp inhibitor) in Wister rats on fetal development, maternal bone marrow for detection of micronuclei (MN), chromosomal aberrations and mitotic index (MI) and embryonic liver cells for cellular proliferation indicate

Effects of acute doses of ethanol administered at pre-implantation stages on fetal development in the mouse.

The objective of the present study was to investigate the effects of maternal administration of acute doses of alcohol at preimplantation stages of pregnancy on the fetus and fetal membranes. Single doses of 0.02 ml and 0.03 ml/g body wt. of 25% v/v of absolute alcohol in saline were administered to groups of MF1 mice on one of days 1-6 of gestation. Controls were either pair-fed and pair-watered or saline-treated. Fetuses and placentae were collected on day 15. Resorption and fetal death were increased in most of the alcohol treatment groups. The reduction in fetal body weight was significant

Gamma secretase inhibition: Effects on fertility and embryo-fetal development in rats.

Avagacestat inhibits γ-secretase, a protease that cleaves the amyloid precursor protein (APP) to produce amyloid beta (Aβ). Aβ plaques, a predominant lesion in Alzheimer's patient's brain, is considered a mechanism driving neurodegeneration. As part of the nonclinical reproductive safety assessment, avagacestat effects on fertility and early embryonic development and embryo-fetal development were evaluated in rats. In the embryo-fetal development study, avagacestat was a selective developmental toxicant with dose-related increased fetal mortality, decreased fetal growth, and increased fetal ma

Macrophage colony-stimulating factor receptor marks and regulates a fetal myeloid-primed B-cell progenitor in mice.

Although it is well established that unique B-cell lineages develop through distinct regulatory mechanisms during embryonic development, much less is understood about the differences between embryonic and adult B-cell progenitor cells, likely to underpin the genetics and biology of infant and childhood PreB acute lymphoblastic leukemia (PreB-ALL), initiated by distinct leukemia-initiating translocations during embryonic development. Herein, we establish that a distinct subset of the earliest CD19(+) B-cell progenitors emerging in the E13.5 mouse fetal liver express the colony-stimulating facto

Tissue-resident neutrophils serve homeostatic and immunological functions in embryos.

Development of neutrophils in the bone marrow and their crucial role in first-line defense are well understood in adults, but remarkably little is known about fetal neutrophils. Here, we analyze the production, distribution, and functions of neutrophils during embryonic development in the mouse. We discover that multiple non-hematopoietic steady-state organs harbor substantial numbers of immature and mature neutrophils, many of which are localized in the tissue parenchyma outside the blood vasculature. Using single-cell transcriptomic analyses, we reveal the presence of neutrophil progenitors

Cytogenetic and genetic studies of radiation-induced chromosome damage in mouse oocytes. I. Numerical and structural chromosome anomalies in metaphase II oocytes, pre- and post-implantation embryos.

The incidences of X-ray induced numerical and structural chromosome anomalies were screened in a range of developmental stages from metaphase II oocytes through to post-implantation embryos. Following 1 Gy of acute X-rays to immediately preovulatory stage oocytes, the rate of hyperploidy (chromosome gain) was found to be elevated over levels in unirradiated controls, at metaphase II, in 1-cell and 3.5 day pre-implantation embryos but not in 8.5 day post-implantation foetuses. In the latter, however, the frequency of mosiacism was significantly increased. A similar response of an increase in mo

Fetal cardiac surgery.

Elucidation of the mechanisms of placental dysfunction after bypass and the negative effects of fetal stress (Fig 9) has allowed these issues to be addressed effectively using indomethacin and appropriate fetal anesthesia. The effective management of these two major problems has made a dramatic difference in the ability of fetal animals to survive surgical intervention and extracorporeal circulation. Characterization of various aspects of placental vascular hemodynamics using the isolated placental preparation also has added new insights into the behavior of the placental vasculature during ex

Development and function of the fetal adrenal.

The adrenal cortex undergoes multiple structural and functional rearrangements to satisfy the systemic needs for steroids during fetal life, postnatal development, and adulthood. A fully functional adrenal cortex relies on the proper subdivision in regions or 'zones' with distinct but interconnected functions, which evolve from the early embryonic stages to adulthood, and rely on a fine-tuned gene network. In particular, the steroidogenic activity of the fetal adrenal is instrumental in maintaining normal fetal development and growth. Here, we review and discuss the most recent advances in our

Do placental weights have clinical significance?

This study attempted to determine if placental size has implications for fetal/neonatal health and for subsequent childhood growth and development. 38,351 placentas were trimmed and weighed in a standardized way. The following factors were found to be associated with low placental weight: low maternal pregravid body weight, low pregnancy weight gain, high maternal hemoglobin levels during pregnancy, gestational hypertension, paid employment outside the home during pregnancy, and low parity. Taking these factors into consideration, placentas that were underweight for birth weight were associate

Fetal stem cells from extra-embryonic tissues: do not discard.

Stem cells hold promise to treat diseases currently unapproachable, including Parkinson's disease, liver disease and diabetes. Seminal research has demonstrated the ability of embryonic and adult stem cells to differentiate into clinically useful cell types in vitro and in vivo. More recently, the potential of fetal stem cells derived from extra-embryonic tissues has been investigated. Fetal stem cells are particularly appealing for clinical applications. The cells are readily isolated from tissues normally discarded at birth, avoiding ethical concerns that plague the isolation embryonic stem

Fetal development after assisted reproduction--a review.

Despite the success of assisted reproduction technologies (ART) in allowing conception in couples with infertility problems, a growing body of evidence points to implication of ART on fetal birth weight alterations, fetal malformations, chromosomal aneuploidies and syndromes related to genomic imprinting modifications. Different causes can be accounted for to explain the increased risk of fetal defects. Pregnancies generated by ART differ from spontaneously achieved pregnancies in that gametes and embryos are cultured in vitro, more than one conceptus is transferred into the uterine cavity, an

[Fetal uropathies and pulmonary hypoplasia].

The author discusses the role of fetal uropathies in the development of pulmonary hypoplasia. The predisposing abnormalities may occur at various locations, including the kidneys and the efferent urinary ducts (ureters, urocyst, urethra). Following a brief general overview of the normal development of the urinary system, the developmental abnormalities are discussed. These anomalies interfere with the secretion of urine or, alternatively, may hinder or obstruct its elimination through the urinary ducts, thus leading to oligohydramnios. Inadequate production of amniotic fluid hinders the respir

Fetal growth and development.

The foetal growth is, for the greater part, determined genetically. There are, however, favouring and inhibiting factors, which influence this growth. In such a matter, the mother, the placenta and the foetus play an important part. The mother shows as well metabolic as vascular adaptations. The placenta must let through nutrients and oxygen; the uteroplacental circulation is characterized by a high flow, together with a low vascular resistance. The foetus itself plays an important part owing to its own growth factors. The principal growth factors are insulin and the growth factors analogous t
