Solution structure of gamma 1-H and gamma 1-P thionins from barley and wheat endosperm determined by 1H-NMR: a structural motif common to toxic arthropod proteins.

The complete assignment of the proton NMR spectra of the homologous gamma 1-hordothionin and gamma 1-purothionin (47 amino acids, 4 disulfide bridges) from barley and wheat, respectively, has been performed by two-dimensional sequence-specific methods. A total of 299 proton-proton distance constraints for gamma 1-H and 285 for gamma 1-P derived from NOESY spectra have been used to calculate the three-dimensional solution structures. Initial structures have been generated by distance geometry methods and further refined by dynamical simulated annealing calculations. Both proteins show identical

Rice Endosperm Protein Improves the Anti-Inflammatory Effects of High-Density Lipoprotein and Produces Lower Atherosclerotic Lesion Accelerated by the Renal Mass Reduction than Casein in a Mouse Model.

Chronic kidney disease (CKD) impairs the anti-inflammatory effects of high-density lipoprotein (HDL) and increases cardiovascular mortality. Though the potential role of dietary interventions to manage HDL is well studied, the clinical trials aimed to increase HDL levels have failed to reduce cardiovascular risk, rendering HDL function to be explored as a more relevant clinical parameter. This study investigates the effects of rice endosperm protein (REP), a plant-based protein, on the anti-inflammatory properties of HDL and renal injury-driven atherosclerosis in comparison with casein, an ani

Complete amino acid sequence of puroindoline, a new basic and cystine-rich protein with a unique tryptophan-rich domain, isolated from wheat endosperm by Triton X-114 phase partitioning.

A new basic protein has been isolated from wheat endosperm by Triton X-114 phase partitioning. It contains five disulfide bridges and is composed of equal amounts of a polypeptide chain of 115 amino acid residues and of the same chain with a C-terminus dipeptide extension. The most striking sequence feature is the presence of a unique tryptophan-rich domain so that this protein isolated from wheat seeds has been named puroindoline. The similar phase partitioning behavior in Triton X-114 of this basic cystine-rich protein and of purothionins suggests that puroindoline may also be a membranotoxi

Characterization of barley (Horduem vulgare) lys3 mutants identifies genes under the regulation of the prolamin-box binding transcription factor and elucidates its role in endosperm promoter methylation during grain development.

Barley ranks fourth in global cereal production and is primarily grown for animal feed and malt. Hordeins, the principal barley seed storage proteins, are homologous to wheat gluten and when ingested elicit an immune response in people with Coeliac disease. Risø 1508 is a chemically induced barley mutant with low hordein levels imparted by the lys3.a locus that is reported to be caused by an SNP in the barley prolamin-box binding factor gene (BPBF). Reports suggest the lys3.a locus prevents CG DNA demethylation at the Hor2 (B-hordein) promoter during grain development subsequently causing hype

Parental genome imbalance in Brassica oleracea causes asymmetric triploid block.

Interploidy crosses fail in many plant species due to abnormalities in endosperm development. In the inbreeding species Arabidopsis thaliana, both paternal and maternal excess interploidy crosses usually result in viable seed that exhibit parent-of-origin effects on endosperm development and final seed size. Paternal excess crosses result in extended proliferation of the endosperm and larger seeds, while conversely maternal excess crosses result in early endosperm cellularisation and smaller seeds. Investigations into the effect of parental gene dosage on seed development have revealed that MA

CrAGL62 and potential paternally expressed gene CrbZIP contribute to paternal-excess postzygotic barriers in citrus.

Interploidy hybridization in plants is constrained by a postzygotic barrier. Triploid production via 2x × 4x interploidy hybridization is an important strategy for obtaining seedless cultivar in some fruit tree crops including citrus. However, as the molecular mechanisms of postzygotic barrier in citrus remain unclear, triploid breeding efficiency is limited. Here, we compared seed development in citrus 2x × 4x and 2x × 2x crosses. Embryo and endosperm developed normally in 2x × 2x crosses, while endosperm cellularization completely failed in 2x × 4x crosses, resulting in embryo arrest at the

Characterization of a near isogenic barley line with high grain β-amylase activity reveals a separation in the tight co-regulation of B-hordeins (Hor2) with endosperm-specific β-amylase (Bmy1).

GSHO 2096 is a near isogenic barley line with extremely high grain β-amylase activity, a desirable trait in the malting and brewing industry. High levels of grain β-amylase activity are caused by a surge in endosperm-specific β-amylase (Bmy1) gene expression during the early stages of grain development with high expression levels persisting throughout development. Origins of the high β-amylase activity trait are perplexing considering GSHO 2096 is not supposed to have grain β-amylase activity. GSHO 2096 is reported to be derived from a Bowman x Risø 1508 cross followed by recurrent backcrossin

Dosage-sensitive maternal siRNAs determine hybridization success in Capsella.

Hybrid seed failure arising from wide crosses between plant species is a recurring obstacle in plant breeding, impeding the transfer of desirable traits. This postzygotic reproductive barrier primarily occurs in the endosperm, a tissue that nourishes the embryo and functions similarly to the placenta in mammals. We found that incompatible seeds show a loss of DNA methylation and chromatin condensation in the endosperm, similar to seeds lacking maternal RNA polymerase IV activity. This similarity is linked to a decrease in small interfering RNAs in the endosperm (sirenRNAs), maternal RNA polyme

A single-nucleus transcriptomic atlas reveals distinct cell identities and key regulators of cellular differentiation during early rice seed development.

The early morphogenesis of the embryo and endosperm sets the upper limit of rice grain yield; yet, the cellular dynamics and regulatory mechanisms underlying this critical stage remain largely elusive. Here, we present a single-cell atlas of developing rice seeds based on single-nucleus RNA sequencing of 67,922 high-quality nuclei from rice caryopses. Integration with bulk RNA-seq, in situ hybridization, and promoter-GUS staining enabled systematic cell-type annotation and revealed a distinct population of embryo-endosperm interface (EEI) cells occupying the boundary between the developing emb

[Free and bound amino acids during germination of maize seeds with implemented foreign prolamines].

Wheat prolamines were preincorporated into the endosperm of maize seed, and concentrations of free and bound amino acids were measured in the endosperm and seedlings during seed germination. Incorporation of foreign prolamines lowered the decrease rate of free proline concentration in both the endosperm (sprouting and shoots) and the embryo (sprouting). The seedlings at the stage of shoots were characterized by higher concentrations of aspartic acid, glutamic acid, and tyrosine. At the stage of transition to autotrophic nutrition, the pool of amino acids that are physiologically important for

Effects of corn hardness and drying temperature on digestibility of energy and nutrients in diets fed to growing pigs.

Two experiments were conducted to test the hypothesis that corn kernel hardness and drying temperature influence the ileal digestibility of starch and amino acids (AA), as well as apparent total tract digestibility (ATTD) of gross energy (GE) and total dietary fiber (TDF) in diets for growing pigs. Two corn varieties with average or hard endosperm were grown and harvested under similar conditions, and after harvest, each variety was divided into 2 batches that were dried at 35 and 120 °C, respectively. Therefore, four batches of corn were used. In experiment 1, 10 pigs (67.00 ± 2.98 kg) with a

A survey of triploid block inconsistency: potential mechanisms and evolutionary implications on polyploid speciation.

The triploid block, primarily caused by endosperm developmental issues, is known as a significant barrier to interploidy hybridization among flowering plants and, thereby, polyploid speciation. However, its strength varies across taxa, with some instances of leakiness, questioning its universal role as a barrier. We conducted a literature survey to explore the causes of the variation in the strength of the triploid block across 11 angiosperm families. We assessed the impact of interploidy cross direction, types of endosperm development, endosperm persistence at seed maturity, and divergence be

Integrated metabolomic and transcriptomic analyses reveal that starch and sucrose metabolism regulate maize kernel hardness.

Maize kernel hardness, largely determined by the structural and compositional characteristics of the endosperm, is a key trait affecting grain quality, milling performance, and storage stability. Although vitreous and starchy endosperms exhibit markedly different physical properties, the underlying metabolic and transcriptional mechanisms-particularly the crosstalk between primary and specialized metabolic pathways-remain insufficiently understood. In this study, we conducted integrated metabolomic and transcriptomic analyses, along with cytological observations, to investigate these mechanism

The ratio of phytosiderophores nicotianamine to deoxymugenic acid controls metal homeostasis in rice.

The ratio of nicotianamine to deoxymugenic acid controls tissue-specific metal homeostasis in rice and regulates metal delivery to the endosperm. The metal-chelating phytosiderophores nicotianamine (NA) and 2'deoxymugenic acid (DMA) are significant factors for the control of metal homeostasis in graminaceous plants. These compounds are thought to influence metal homeostasis, but their individual roles and the effect of altering the NA:DMA ratio are unknown. We purposely generated rice lines with high and low NA:DMA ratios (HND and LND lines, respectively). The HND lines accumulated more iron (

Functional Analysis of the Autophagy-Related Gene OsATG4b in Rice Grain Chalkiness Regulation.

Grain chalkiness is an undesirable trait that significantly compromises rice quality, attracting considerable attention from both consumers and breeders. In this study, we characterized the role of the autophagy-related gene OsATG4b in rice grain development. OsATG4b was predominantly expressed in the endosperm. Compared with wild-type plants, OsATG4b-overexpressing lines exhibited significantly reduced grain chalkiness, whereas OsATG4b knockout mutants displayed a marked increase in chalkiness. Importantly, OsATG4b had no significant effect on other major agronomic traits. Ultrastructure anal

Developing seeds of Arabidopsis store different minerals in two types of vacuoles and in the endoplasmic reticulum.

Mineral-accumulating compartments in developing seeds of Arabidopsis were studied using high-pressure-frozen/freeze-substituted samples. Developing seeds store minerals in three locations: in the protein storage vacuoles of the embryo, and transiently in the endoplasmic reticulum (ER) and vacuolar compartments of the chalazal endosperm. Energy dispersive x-ray spectroscopy and enzyme treatments suggest that the minerals are stored as phytic acid (myo-inositol-1,2,3,4,5,6-hexakisphosphate) salts in all three compartments, although they differ in cation composition. Whereas embryo globoids conta

Analysis of the physical and chemical characteristics of stored and fresh Leucaena leucocephala seeds.

The long-term behavior of plant dietary fibers affects their usefulness in creating stable and sustainable functional ingredients. This study compared the structural and chemical features of Leucaena leucocephala seeds stored for 11 years under environmental conditions with freshly harvested seeds from the same plant. The anatomical parts, including the seed coat, cotyledons, and endosperm, were examined using physicochemical methods, as specified by the Association of Official Analytical Chemists (AOAC), as well as scanning electron microscopy (SEM) and energy-dispersive spectroscopy (EDS). Metabolic Engineering of Wheat Provitamin A by Simultaneously Overexpressing CrtB and Silencing Carotenoid Hydroxylase (TaHYD).

Increasing the provitamin A content in staple crops via carotenoid metabolic engineering is one way to address vitamin A deficiency. In this work a combination of methods was applied to specifically increase β-carotene content in wheat by metabolic engineering. Endosperm-specific silencing of the carotenoid hydroxylase gene (TaHYD) increased β-carotene content 10.5-fold to 1.76 μg g(-1) in wheat endosperm. Overexpression of CrtB introduced an additional flux into wheat, accompanied by a β-carotene increase of 14.6-fold to 2.45 μg g(-1). When the "push strategy" (overexpressing CrtB) and "block

Parental-genome dosage effects on the transcriptome of F1 hybrid triploid embryos of Arabidopsis thaliana.

Genomic imprinting in the seed endosperm could be due to unequal parental-genome contribution effects in triploid endosperm tissue that trigger parent-of-origin specific activation and/or silencing of loci prone to genomic imprinting. To determine whether genomic imprinting is triggered by unequal parental-genome contribution effects, we generated a whole-genome transcriptome dataset of F1 hybrid triploid embryos (as mimics of F1 hybrid triploid endosperm). For the vast majority of genes, the parental contributions to their expression levels in the F1 triploid hybrid embryos follow a biallelic
