Nervous necrosis virus capsid protein and Protein A dynamically modulate the fish cGAS-mediated IFN signal pathway to facilitate viral evasion.

Nervous necrosis virus (NNV), an aquatic RNA virus belonging to Betanodavirus, infects a variety of marine and freshwater fishes, leading to massive mortality of cultured larvae and juveniles and substantial economic losses. The enzyme cyclic guanosine monophosphate-adenosine monophosphate synthase (cGAS) is widely recognized as a central component in the innate immune response to cytosolic DNA derived from different pathogens. However, little is known about the response of cGAS to aquatic RNA viruses. This study found that Epinephelus coioides cGAS (EccGAS) overexpression inhibited NNV replic

Short-term effects of fish oil treatment on urinary excretion of high- and low-molecular weight proteins in patients with IgA nephropathy.

AIMS: Recently, it was shown that fish oil treatment improved renal survival in patients with IgA nephropathy. The precise mechanisms of this protective effect remained unclear. Omega-3 polyunsaturated fatty acids (PUFAs), important active substances of fish oil, are able to attenuate inflammatory responses. Thus, the renoprotective effects of fish oil may be the result of mitigation of glomerular or tubulo-interstitial inflammation. We hypothesized that such a decrease in glomerular or tubulo-interstitial inflammation could result in an improvement of glomerular permselectivity as reflected b

Fish oil supplementation protects against protein undernutrition-induced testicular and ovarian biochemical alterations in rats.

Proteins are required for biological functions and their inadequacy might impair the growth and development of the reproductive system. The study investigated the effects of fish oil (FO) supplementation on low-protein diet-induced alterations in male and female reproductive organs. Male and female rats were assigned randomly to four groups respectively. The NPD rats had five rats per group and were given 16% casein diet while the LPD rats had eight rats per group and received 5% casein diet. After the 8th week, FO was administered for 3 weeks via oral gavage at a concentration of 400 mg Kg-1

Production of a viral surface protein in Nannochloropsis oceanica for fish vaccination against infectious pancreatic necrosis virus.

Nannochloropsis oceanica is a unicellular oleaginous microalga of emerging biotechnological interest with a sequenced, annotated genome, available transcriptomic and proteomic data, and well-established basic molecular tools for genetic engineering. To establish N. oceanica as a eukaryotic host for recombinant protein synthesis and develop molecular technology for vaccine production, we chose the viral surface protein 2 (VP2) of a pathogenic fish virus that causes infectious pancreatic necrosis as a model vaccine. Upon stable nuclear transformation of N. oceanica strain CCMP1779 with the codon

Agouti-related protein as the glucose signaling sensor in the central melanocortin circuits in regulating fish food intake.

Agouti-related protein (AgRP) is a neuropeptide synthesized by AgRP/NPY neurons and transcribed as 132 amino acids in humans and 142 amino acids (AgRP1) in Japanese seabass (Lateolabrax maculatus) fish. AgRP neurons are activated by hormonal signals of energy deficits and inhibited by signals of energy surpluses and have been demonstrated to have the ability to sense the dynamics of blood glucose concentrations as the "glucose sensor" in mammals. It is widely recognized that AgRP is an endogenous antagonist of the melanocortin-3 and -4 receptors (MC3R and MC4R) in the hypothalamus, exhibiting

Modulation of Neural and Muscular Adaptation Processes During Resistance Training by Fish Protein Ingestions in Older Adults.

Assessments of both neural and muscular adaptations during interventions would provide valuable information for developing countermeasures to age-related muscle dysfunctions. We investigated the effect of fish protein ingestion on training-induced neural and muscular adaptations in older adults. Twenty older adults participated 8 weeks of isometric knee extension training intervention. The participants were divided into two groups who took fish protein (n = 10, Alaska pollack protein, APP) or casein (n = 10, CAS). Maximal muscle strength during knee extension, lower extremity muscle mass (body

A Comparative Genomic and Transcriptional Survey Providing Novel Insights into Bone Morphogenetic Protein 2 (bmp2) in Fishes.

Intermuscular bones (IBs) are only found in the muscles of fish. Bone morphogenetic protein 2 (bmp2) is considered to be the most active single osteogenesis factor. It promotes cell proliferation and differentiation during bone repair, as well as inducing the formation of bones and cartilages in vivo. However, detailed investigations of this family in fish are incredibly limited. Here, we have used a variety of published and unpublished bmp2 sequences for teleosts and cartilage fish in order to explore and expand our understanding of bmp2 genes in fish. Our results confirmed that teleost genom

Immunohistochemical detection of MTAP and BAP1 protein loss for mesothelioma diagnosis: Comparison with 9p21 FISH and BAP1 immunohistochemistry.

OBJECTIVES: Differentiating malignant pleural mesothelioma (MPM) from reactive mesothelial hyperplasia (RMH) is still challenging. Detection of homozygous deletion (HD) of 9p21 region including p16INK4A (p16) by fluorescence in situ hybridization (FISH) and immunohistochemical detection of loss of BRCA1 associated protein 1 (BAP1), are reliable markers for MPM diagnosis. However, not all laboratories are equipped to perform 9p21 FISH; immunohistochemistry (IHC) is a more common and feasible technique. Thus, we sought to develop a IHC-based method that could predict the deletion of p16 in MPM i

Gap junction protein in weakly electric fish (Gymnotide): immunohistochemical localization with emphasis on structures of the electrosensory system.

Electrotonic transmission via gap junctions appears to be essential for both the relay and integration of information in nuclear groups involved in the electrolocation and electrocommunication systems of weakly electric fish. An affinity-purified antibody against the 27 kD gap-junctional polypeptide (GJP) from rat liver was used to determine immunohistochemically the distribution of GJP-immunoreactivity (GJP-IR) in electrosensory structures and some other brain regions of the gymnotiform fish, Apteronotus leptorhynchus. At the ultrastructural level, immunolabelling with this antibody was local

Effects of age-related gastrointestinal conditions on the digestion of myofibrillar protein-stabilized fish oil oleogel emulsions.

Fish oil has polyunsaturated fatty acids, and fish oil oleogels have potential as animal fat substitution to supplement EPA and DHA. However, age-related gastrointestinal conditions may affect disintegration and digestion of fish oil oleogel emulsion. The objective of this study was to determine how age-associated differences in gastric pH, pepsin activity, and intestinal bile salt concentration regulate the digestion-induced interfacial remodeling of fish oil oleogel-in-water emulsions stabilized by golden pompano (Trachinotus ovatus) myofibrillar proteins. The results showed that increasing

Competitive Binding of Gingerol Homologs to Fish Myofibrillar Protein Reduces the Retention of Aldehydic Off-Odor Compounds.

Persistent aldehydic off-notes in fish products are influenced not only by lipid oxidation but also by protein-associated retention of odor-active aldehydes. A remaining question is how gingerol side-chain length and aldehyde structure jointly alter this apparent retention. Mackerel myofibrillar protein (MP) was therefore combined with (E)-2-decenal (T2D), (E,E)-2,4-decadienal (DDE), or trans-4,5-epoxy-(E)-2-decenal (E2D), with or without 6-, 8-, or 10-gingerol. Fish oil modulates macrophage P44/P42 mitogen-activated protein kinase activity induced by lipopolysaccharide.

BACKGROUND: Mitogen-activated protein kinase (MAPK) cascades represent a major signal system to transduce extracellular signals into cellular responses. Overactivity of MAPK has been implicated in the development of many diseases, including cancer and sepsis. This study investigated the hypothesis that fish oil altered the membrane phospholipid composition and modulated MAPK activity. METHODS: RAW 264.7 cells, a mouse macrophage (Mphi) cell line, were grown in eicosapentaenoic acid (EPA)-rich media (114 micromol/L) for 48 hours. Mphi were washed and exposed to Escherichia coli lipopolysacchar

Magnesium status in freshwater fish, common carp (Cyprinus carpio, L.) and the dietary protein-magnesium interaction.

Common carp juveniles were fed for 9 weeks one of the eight semipurified diets containing graded levels of magnesium, 0.08, 0.6, 1.1, 3, 2 g Mg kg(-1) and 25 or 44% protein.Fish growth and feed utilization were significantly affected by both Mg and protein levels in the diets. Significant interaction between these two studied variables existed in relation to the fish performance as well as to mean deposition rate of several minerals in common carp body. The fish fed diets containing 0.08 g Mg kg(-1) had reduced growth and developed deficiency signs such as muscle flaccidity and skin hemorrhage

Tracing the molecular evolution of Foxp2 proteins in vertebrates from fish to tetrapods: Insights into poly-Q tract variation, structural changes, and interaction networks.

Foxp2 is a transcription factor containing poly-Q repeats, commonly found in brain proteins. It plays essential roles in speech, motor function, cognition, and emotion, and is expressed during embryonic development in the brain, lungs, heart, and intestines. Foxp2 is highly conserved among vertebrates. This study investigated the molecular evolution of Foxp2 by analyzing its sequence, structure, post-translational modifications (N-glycosylation and phosphorylation), positive selection signals, conserved motifs, domains, and interaction networks across five representative vertebrates: human, ca

Expression pattern and developmental behaviour of cellular nucleic acid-binding protein (CNBP) during folliculogenesis and oogenesis in fish.

In vertebrates, folliculogeneis establishes an intricate system for somatic cell-oocyte interaction, and ultimately leads to the acquisition of their respective competences. Although the formation process and corresponding interactions are strikingly similar in diverse organisms, knowledge of genes and signaling pathways involved in follicle formation is very incomplete and the underlying molecular mechanisms remain enigmatic. CNBP has been identified for more than ten years, and the highest level of CNBP transcripts has been observed in adult zebrafish ovary, but little is known about its fun

Distribution of the phosphorylated form of microtubule associated protein 1B in the fish visual system during optic nerve regeneration.

Microtubule associated proteins are a heterogeneous group of proteins that have been implicated in regulating microtubule stability. They play an important role in the organisation of the neuronal cytoskeleton during neurite outgrowth, plasticity and regeneration. The fish visual system presents a considerable degree of plasticity. Thus, the retina grows continually throughout life and the optic nerve regenerates after crush. In the present study, we compared the distribution of the microtubule associated protein 1B in its phosphorylated form (MAP1B-phos) in the normal adult fish visual system

Controlled thermal unfolding of soy protein isolate modulates transglutaminase-induced gelation and network structure of fish gelatin.

Mixed-protein gel systems have emerged as an effective strategy for tailoring textural and structural properties in sustainable food systems. In this study, the effects of varying ratios (1:1, 2:1, 4:1) and thermal pretreatment (75, 85, 95 °C) of soy protein isolate (SPI) on the gelation behavior of transglutaminase (TGase)-crosslinked fish gelatin (FG) were systematically investigated. The results showed that TGase-induced FG-SPI complexes exhibited strengthened intermolecular interactions. However, the globular structure of native SPI constrained the formation of a dense and continuous porou
