Optic nerve head: A gatekeeper for vitreous infectious insults?

The axons of retinal ganglion cells (RGCs) pass through the optic nerve head (ONH) and form the optic nerve (ON). The ONH serves as an anatomical interface between the vitreous cavity and subarachnoid space. After inducing acute neuroinflammation by intravitreal injection of lipopolysaccharides (LPS), we observed inflammatory activation in the retina, but detect no signs of inflammation in the posterior ON or infiltration of inflammatory cells in the ONH. Therefore, we hypothesized that the ONH functions as a barrier to vitreous inflammation. Using transmission electron microscopy, we identifi

Solute carrier transporters: the metabolic gatekeepers of immune cells.

Solute carrier (SLC) transporters meditate many essential physiological functions, including nutrient uptake, ion influx/efflux, and waste disposal. In its protective role against tumors and infections, the mammalian immune system coordinates complex signals to support the proliferation, differentiation, and effector function of individual cell subsets. Recent research in this area has yielded surprising findings on the roles of solute carrier transporters, which were discovered to regulate lymphocyte signaling and control their differentiation, function, and fate by modulating diverse metabol

Liver sinusoidal endothelial cells - gatekeepers of hepatic immunity.

Liver sinusoidal endothelial cells (LSECs) line the low shear, sinusoidal capillary channels of the liver and are the most abundant non-parenchymal hepatic cell population. LSECs do not simply form a barrier within the hepatic sinusoids but have vital physiological and immunological functions, including filtration, endocytosis, antigen presentation and leukocyte recruitment. Reflecting these multifunctional properties, LSECs display unique structural and phenotypic features that differentiate them from the capillary endothelium present within other organs. It is now clear that LSECs have a cri

Metabolic gatekeeper function of B-lymphoid transcription factors.

B-lymphoid transcription factors, such as PAX5 and IKZF1, are critical for early B-cell development, yet lesions of the genes encoding these transcription factors occur in over 80% of cases of pre-B-cell acute lymphoblastic leukaemia (ALL). The importance of these lesions in ALL has, until now, remained unclear. Here, by combining studies using chromatin immunoprecipitation with sequencing and RNA sequencing, we identify a novel B-lymphoid program for transcriptional repression of glucose and energy supply. Our metabolic analyses revealed that PAX5 and IKZF1 enforce a state of chronic energy d

The cellobiose sensor CebR is the gatekeeper of Streptomyces scabies pathogenicity.

UNLABELLED: A relatively small number of species in the large genus Streptomyces are pathogenic; the best characterized of these is Streptomyces scabies. The pathogenicity of S. scabies strains is dependent on the production of the nitrated diketopiperazine thaxtomin A, which is a potent plant cellulose synthesis inhibitor. Much is known about the genetic loci associated with plant virulence; however, the molecular mechanisms by which S. scabies triggers expression of thaxtomin biosynthetic genes, beyond the pathway-specific activator TxtR, are not well understood. In this study, we demonstrat

Hypertensive encephalopathy and the blood-brain barrier: is deltaPKC a gatekeeper?

Hypertensive encephalopathy is a life-threatening condition due to elevation of cerebral perfusion pressure beyond the limits of autoregulation. Breakdown of the blood-brain barrier (BBB) leads to cerebral edema and reduced blood flow. In this issue of the JCI, Mochly-Rosen and colleagues demonstrate a novel molecular strategy for preserving the BBB in a model of hypertension-induced encephalopathy (see the related article beginning on page 173). Using a rationally designed peptide inhibitor of deltaPKC, they stabilized the BBB and improved mortality in hypertensive rats. This study highlights

Tumor suppressor genes in breast cancer: the gatekeepers and the caretakers.

Tumor suppressor genes encode for proteins whose normal function is to inhibit cell transformation and whose inactivation is advantageous for tumor cell growth and survival. A variety of mechanisms result in the inactivation of tumor suppressor genes, including intragenic mutations, chromosomal deletions, and loss of expression by methylation-mediated transcriptional silencing or increased proteolysis. Tumor suppressor genes participate in a variety of critical and highly conserved cell functions, including regulation of the cell cycle and apoptosis, differentiation, surveillance of genomic in

Diagnostic management by gatekeepers is not cost effective for neuro-ophthalmology.

PURPOSE: To determine whether the "gatekeeper physician system" for evaluating neuro-ophthalmologic problems is cost effective. METHODS: The authors retrospectively reviewed the records of 588 patients referred for neuro-ophthalmologic evaluation between July and December 1989 to determine the frequency and cost of unnecessary diagnostic testing ordered by "gatekeeper physicians." Pre-referral diagnostic testing costs were compared with the cost of neurophthalmologic consultation for four common problems: (1) optic neuropathy; (2) diplopia; (3) ptosis; and (4) proptosis. RESULTS: Between 16%

Tryptophan metabolic gatekeeping in epithelial repair: GPR35-KLF5 circuitry decodes mucosal damage signals for repair programming.

The impaired repair of intestinal mucosal damage is an important pathological feature of ulcerative colitis (UC). The critical role of intestinal epithelial cells (IECs) proliferation and migration in the repair of damaged mucosal epithelium has been well established. However, the molecular circuitry that decodes IECs sense intestinal mucosal damage signals to initiate and drive repair program remains elusive. Here, we identify a tryptophan (Trp) metabolic gatekeeping mechanism wherein G protein-coupled receptor 35 (GPR35) senses intestinal mucosal damage through monitoring Trp-kynurenine (KYN

Metabolic gatekeepers shape immune cell function in rheumatoid arthritis.

Immune cell function is tightly coupled to cellular metabolism, with glycolysis, oxidative phosphorylation, and amino acid and lipid metabolism governing activation, differentiation, survival, and effector responses. In rheumatoid arthritis (RA), an autoimmune disease characterized by destructive joint inflammation, metabolic checkpoints have emerged as critical regulators of immune-cell behavior and drivers of tissue pathology. RA CD4+ T cells undergo profound metabolic reprogramming: glycolysis is suppressed, and glucose is redirected into the pentose phosphate pathway, resulting in NADPH ac

Beyond the biofilm: KLF2 as a molecular gatekeeper in periodontal health and disease.

Periodontal disease is a chronic inflammatory condition resulting from complex interactions between microbial dysbiosis and host immune responses, leading to progressive destruction of the periodontium. Kruppel-like factor 2 (KLF2), a zinc-finger transcription factor, has emerged as a key regulator of immune modulation and tissue homeostasis. KLF2 orchestrates anti-inflammatory pathways, controls immune cell activation, and influences periodontal tissue integrity. Dysregulation of KLF2 is implicated in enhanced inflammation and periodontal tissue breakdown. This review summarizes the molecular

Oral epithelial barrier: Gatekeeper of mucosal immunity and driver of disease.

The oral mucosal epithelium functions not only as a physical barrier but also as a dynamic immunomodulatory interface at the boundary between the oral environment and the host. It actively maintains host-microbial homeostasis and orchestrates mucosal immunity. Disruption of this intricate barrier is a hallmark of various oral inflammatory diseases, underscoring its pivotal role in pathogenesis. This review synthesizes recent advances demonstrating how the epithelium, as a central component of the mucosal immune community, contributes to health and disease through mechanisms including barrier i

Astrocytes and Their Role in the Development and Progression of Alzheimer's Disease: Gatekeepers of Neurodegeneration.

Astrocytes are increasingly recognized as central players in the pathogenesis of Alzheimer's disease (AD), exhibiting both neuroprotective and neurotoxic functions, which complicates their role in disease progression. Under physiological conditions, astrocytes support neuronal homeostasis, facilitate synaptic function, and promote the clearance of Amyloid-β (Aβ), thereby contributing to neuroprotection. In the context of AD, however, reactive astrocytes can adopt detrimental phenotypes, releasing pro-inflammatory cytokines, generating oxidative stress, and disrupting neuronal networks, thereby

Maternal small RNAs as gatekeepers of hybrid compatibility in plants and animals.

Hybridization between species can enable introgression of advantageous traits, yet hybrid offspring often show reduced fitness, sterility, or developmental arrest due to postzygotic hybrid incompatibility. Understanding the molecular bases of these barriers could help overcome reproductive limits and enable the creation of currently inaccessible hybrids. In this review, we compare two small RNA-associated routes through which parental genomes can become incompatible: maternally biased sirenRNAs in the endosperm of flowering plants and animal PIWI-interacting RNAs (piRNAs), with emphasis on Dro

A glycolytic intermediate gatekeeps global mRNA decay in Escherichia coli.

Polynucleotide phosphorylase (PNPase) is a highly conserved exoribonuclease crucial for RNA degradation across all domains of life. In bacteria, PNPase is found in multi-protein ribonucleolytic complexes that include glycolytic enzymes. While evidence supports a link between RNA degradation and glycolysis in Escherichia coli, the precise contribution of PNPase and the underlying regulatory mechanism remain unknown. Here we establish PNPase as a metabolic sensor that physically and functionally links glycolysis to RNA stability. We find that the intermediate of glycolysis, fructose 1,6-diphosph

Autophagy in endothelial cells: A gatekeeper of immune cell trafficking and function.

Endothelial cells (ECs) are increasingly recognised as dynamic regulators of immunity rather than passive vascular barriers. By integrating inflammatory, metabolic, and mechanical cues, ECs actively shape leukocyte trafficking and determine inflammatory outcomes. Autophagy, a conserved lysosomal degradation pathway essential for cellular homeostasis, has emerged as a key determinant of endothelial function in health and disease. Recent work identifies EC autophagy as a context-dependent determinant of leukocyte trafficking, vascular inflammation, antigen presentation, and endothelial phagocyti

Polyamines as Gatekeepers of Virus Replication and Central Nervous System Homeostasis.

Polyamines are small, positively charged molecules essential for fundamental cellular processes, including transcription, translation, and membrane fluidity. In the central nervous system (CNS), these molecules serve as homeostatic gatekeepers by modulating neuroreceptors like NMDA and supporting autophagic clearance. While basal polyamine levels are necessary for proper neuronal differentiation and memory formation, their dysregulation is a hallmark of neurodegenerative pathologies such as Alzheimer's and Parkinson's diseases. Retinal Gatekeepers: Molecular Mechanism and Therapeutic Role of Cysteine and Selenocysteine.

Oxidative stress is a key contributor to retinal degeneration, as the retina is highly metabolically active and exposed to constant light stimulation. This review explores the crucial roles of cysteine and selenocysteine in redox homeostasis and retinal protection. Cysteine, primarily synthesized via the transsulfuration pathway, is the rate-limiting precursor for glutathione (GSH), the most abundant intracellular antioxidant. Selenocysteine enables the enzymatic activity of selenoproteins, particularly glutathione peroxidases (GPXs), which counteract reactive oxygen species (ROS). Experimenta
