We have investigated whether granzyme B (GzmB) is required for effective cytotoxic T lymphocyte (CTL) mediated control of lymphocytic choriomeningitis virus (LCMV) infection. Clearance of LCMV from tissues of GzmB-deficient (GzmB-) mice following intraperitoneal infection with LCMV was impaired compared with control mice; however, the virus was ultimately eliminated. The impaired clearance of LCMV in GzmB- mice was not due to a deficiency in the generation of LCMV-specific T cells. In addition, CTL from LCMV-infected GzmB- mice efficiently lysed virus-infected cells in vitro, but were deficien

We previously showed that T cells expressing granzyme (gzm) A are more frequent in skin lesions of susceptible mice than in those of resistant mice infected with the intracellular parasite Leishmania major. To determine the in vivo role of gzm in cutaneous leishmaniasis, we examined the course of L. major infection in gzmA-deficient mice. Despite a delay in host colonization of susceptible mice, the lack of gzmA did not influence the course of lesion development or result in a discernible alteration of the interferon-gamma and interleukin-4 production. Moreover, no differences in these paramet

OBJECTIVES: Behçet's disease (BD) is a chronic inflammatory condition with immune system dysregulation. Recent evidence suggests proinflammatory roles of extracellular granzyme K (GzmK), particularly in aged immune cells. This study investigated GzmK-expressing CD8+ T cells and their senescent subset in BD pathology.
METHODS: We analysed lymphocyte subsets in blood samples from 19 active BD (aBD), 19 inactive BD (iaBD), and 19 healthy controls (HC) using flow cytometry and granzymes, as well as IL-6 using enzyme-linked immunosorbent assay.
RESULTS: Active BD patients showed significantly hig

Granzyme B (GzmB), beyond its established role in corneal and conjunctival disease, is increasingly recognized in the retinal and choroidal distribution of several pathologies. GzmB targets tight-junction proteins (e.g., ZO-1, JAM-A) and extracellular proteins such as fibronectin and laminin within the retinal pigment epithelium (RPE) and Bruch's membrane, thereby disrupting the outer blood-retinal barrier and contributing to the pathogenesis of age-related macular degeneration (AMD) and Macular neovascularization (MNV). This mini review highlights recent mechanistic insights into how GzmB dri

Background/Objectives: Parkinson's disease (PD) is a neurodegenerative disease with a heterogeneous nature. Many molecular pathways are engaged in PD pathogenesis. Granzyme B (GrB) is an enzyme released by CTLs and has roles in neuroinflammation, axonal degeneration, demyelination, and neuronal ischemic death. However, little is reported about its role in PD pathogenesis and deterioration. To evaluate the role of GrB in PD. Method: A total of 94 participants were recruited from outpatient neurology clinics (47 PD patients and 47 healthy controls). Serum GrB levels were measured using ELISA. Re

The cornea and conjunctiva are critical structures of the ocular surface that are prone to a range of inflammatory diseases impacting the vision of millions globally. Moreover, due to their complex etiology and pathophysiology, these conditions remain a significant clinical challenge. Granzyme B (GrB), a serine protease traditionally recognized for its intracellular role in targeting tumor or pathogen-infected cells, has been implicated in numerous pathogenic mechanisms observed in inflammatory corneal and conjunctival diseases, such as dysregulated inflammation and tissue remodeling, barrier

Psoriasis is an inflammatory disease with systemic manifestations that most commonly presents as itchy, erythematous, scaly plaques on extensor surfaces. Activation of the IL-23/IL-17 pro-inflammatory signaling pathway is a hallmark of psoriasis and its inhibition is key to clinical management. Granzyme K (GzmK) is an immune cell-secreted serine protease elevated in inflammatory and proliferative skin conditions. In the present study, human psoriasis lesions exhibited elevated GzmK levels compared to non-lesional psoriasis and healthy control skin. In an established murine model of imiquimod (

Age-related macular degeneration (AMD), a prevalent and progressive degenerative disease of the macula, is the leading cause of blindness in elderly individuals in developed countries. The advanced stages include neovascular AMD (nAMD), characterized by choroidal neovascularization (CNV), leading to subretinal fibrosis and permanent vision loss. Despite the efficacy of anti-vascular endothelial growth factor (VEGF) therapy in stabilizing or improving vision in nAMD, the development of subretinal fibrosis following CNV remains a significant concern. In this review, we explore multifaceted aspec

BACKGROUND AND AIMS: Primary sclerosing cholangitis (PSC) is a chronic cholestatic liver disease characterized by biliary inflammation and fibrosis. We showed an elevated interferon γ response in patients with primary sclerosing cholangitis and in multidrug resistance protein 2-deficient ( Mdr2-/- ) mice developing sclerosing cholangitis. Interferon γ induced expression of the cytotoxic molecules granzyme B (GzmB) and TRAIL in hepatic lymphocytes and mediated liver fibrosis in sclerosing cholangitis.
APPROACH AND RESULTS: In patient samples and Mdr2-/- mice, we identified lymphocyte clusters

BACKGROUND: Granzyme K (GzmK) is a serine protease with minimal presence in healthy tissues while abundant in inflamed tissues. Initially thought to play an exclusive role in immune-mediated cell death, extracellular GzmK can also promote inflammation.
OBJECTIVES: To evaluate the role of GzmK in the pathogenesis of atopic dermatitis (AD), the most common inflammatory skin disease.
METHODS: A panel of human AD and control samples was analysed to determine if GzmK is elevated. Next, to determine a pathological role for GzmK in AD-like skin inflammation, oxazolone-induced dermatitis was induced

BACKGROUND: Low-dose computed tomography (LDCT) has improved the early detection of lung cancer. However, LDCT scans present several disadvantages, including the abundance of false-positive results, which lead to a high socioeconomic cost, psychological burden, and repeated exposure to radiation. Therefore, the identification of complementary biomarkers is needed to select high-risk individuals for LDCT. Here, we showed that granzyme B testing with the novel immunosensor has diagnostic value for identifying patients with lung cancer.
METHODS: We enrolled 44 patients with lung cancer and 51 he

BACKGROUND: Lung inflammation plays a vital role in the pathogenesis of chronic obstructive pulmonary disease (COPD), but the characteristics of the inflammatory process remain unclear. There is growing interest in the role of granzyme B (GzmB) because CD8+ T cells can induce apoptosis of target cells by releasing GzmB, which in turn may cause tissue injury and remodeling. However, GzmB is also expressed by regulatory cells, which are able to suppress CD8+ T cell. The role of GzmB+ cells needs to be defined in COPD.
METHODS: GzmB+ and CD8+ cells on alveolar wall of surgically resected lungs o

A novel conception of CD4(+) T cells with cytolytic potential (CD4(+) CTL) is emerging. These cells appear to have a part in controlling malignancies and chronic infections. Human parvovirus B19 can cause a persistent infection, yet no data exist on the presence of B19-specific CD4(+) CTLs. Such cells could have a role in the pathogenesis of some autoimmune disorders reported to be associated with B19. We explored the cytolytic potential of human parvovirus B19-specific T cells by stimulating peripheral blood mononuclear cell (PBMC) with recombinant B19-VP2 virus-like particles. The cytolytic

In the livers of susceptible C57BL/6 (B6) mice infected with Leishmania donovani, CD8(+) T cell mechanisms are required for granuloma assembly, macrophage activation, intracellular parasite killing, and self-cure. Since gene expression of perforin and granzymes A and B (GzmA and GzmB), cytolytic proteins linked to CD8(+) cell effector function, was enhanced in infected liver tissue, B6 mice deficient in these granular proteins were used to gauge host defense roles. Neither perforin nor GzmA was required; however, mice deficient in GzmB (GzmB(-/-), GzmB cluster(-/-), and GzmA×B cluster double k

Cytotoxic lymphocytes (CLs) contain lysosome-related organelles (LROs) that perform the normal degradative functions of the lysosome, in addition to storage and release of powerful cytotoxins employed to kill virally infected or abnormal cells. Among these cytotoxins is granzyme B (GrB), a protease that has also been implicated in activation (restimulation)-induced cell death of natural killer (NK) and T cells, but the underlying mechanism and its regulation are unclear. Here we show that restimulation of previously activated human or mouse lymphocytes induces lysosomal membrane permeabilisati

Thyroid carcinoma (THCA), the most prevalent endocrine system cancer, is rising worldwide. Granzyme family member GZMM causes tumor cell inflammation and programmed cell death. However, the role of GZMM in THCA has not been investigated. Bioinformatics analysis and assays showed that THCA GZMM protein levels were down-regulated and associated with certain clinicopathological features. Additionally, univariate and multivariate Cox analysis and ROC curve analysis showed that low GZMM expression was related with poor overall survival and might be employed as a prognostic and diagnostic factor. We

Neurodegenerative diseases activate innate and adaptive immune responses that can either slow or accelerate disease progression. Here, we sought to define beneficial immune pressures that emerge during tauopathy development in mice and humans. Using mice that express mutant human tau in neurons, we observed that microglia slowed tauopathy development by controlling the spread of phosphorylated tau (pTau) in the central nervous system and blood. However, over time microglia converted into distressed antigen-presenting cells, acquired neuronal transcripts and were targeted by resident, clonally

Chronic obstructive pulmonary disease (COPD) is caused by smoking, but only a small proportion of smokers have disease severe enough to develop COPD. COPD is not always progressive. The question then arises as to what explains the different trajectories of COPD. The role of autoimmunity and regulatory T (Treg) cells in the pathogenesis of COPD is increasingly being recognized. Nine published studies on Treg cells in the lung tissue or bronchoalveolar lavage fluid have shown that smokers with COPD have fewer Treg cells than smokers without COPD or nonsmokers. Three studies showed a positive cor
