The Nipah virus (NiV) infection is a highly fatal zoonotic disease with pandemic potential which has led to recurrent outbreaks in Bangladesh and India. While transmission pathways, including contaminated date palm sap and human-to-human spread, are increasingly identified, significant uncertainties remain. With no approved therapeutics or vaccines, prevention depends on addressing ecological and behavioural drivers of transmission. This viewpoint draws on selected evidence from NiV outbreaks and response to other zoonotic disease epidemics, such as Ebola, rabies, and the Marburg virus disease

We identified and isolated a novel Hendra virus (HeV) variant not detected by routine testing from a horse in Queensland, Australia, that died from acute illness with signs consistent with HeV infection. Using whole-genome sequencing and phylogenetic analysis, we determined the variant had ≈83% nt identity with prototypic HeV. In silico and in vitro comparisons of the receptor-binding protein with prototypic HeV support that the human monoclonal antibody m102.4 used for postexposure prophylaxis and current equine vaccine will be effective against this variant. An updated quantitative PCR devel

Innate immunity is the first line of defence elicited by the host immune system to fight against invading pathogens such as viruses and bacteria. From this elementary immune response, the more complex antigen-specific adaptive responses are recruited to provide a long-lasting memory against the pathogens. Innate immunity gets activated when the host cell utilizes a diverse set of receptors known as pattern recognition receptors (PRR) to recognize the viruses that have penetrated the host and responds with cellular processes like complement system, phagocytosis, cytokine release and inflammatio

The nucleolus is a common target of viruses and viral proteins, but for many viruses the functional outcomes and significance of this targeting remains unresolved. Recently, the first intranucleolar function of a protein of a cytoplasmically-replicating negative-sense RNA virus (NSV) was identified, with the finding that the matrix (M) protein of Hendra virus (HeV) (genus Henipavirus, family Paramyxoviridae) interacts with Treacle protein within nucleolar subcompartments and mimics a cellular mechanism of the nucleolar DNA-damage response (DDR) to suppress ribosomal RNA (rRNA) synthesis. Nipah virus causes outbreaks in humans with high case-fatality rates. In this study, we confirmed the presence of Nipah virus in Sri Lanka in Pteropus medius fruit bats, one of the known natural reservoir species. Sequences we generated were genetically related to Nipah virus strains from outbreaks in southern India.

Nipah virus (NiV) and Hendra virus (HeV) are bat-borne zoonotic paramyxoviruses that cause severe and often fatal respiratory and neurological disease in humans, for which no small-molecule therapies are currently approved. While host-protease-mediated viral entry has been implicated previously in henipavirus infection, the translational feasibility of targeting this pathway with clinically relevant compounds remains poorly defined. Here, we report a systematic evaluation of repurposed clinical-stage molecules using an integrated computational, in vitro, and in vivo framework to prioritize can

Nipah virus causes highly lethal disease, with case-fatality rates ranging from 40% to 100% in recognised outbreaks. No treatments or licensed vaccines are currently available for the prevention and control of Nipah virus infection. In 2019, WHO published an advanced draft of a research and development roadmap for accelerating development of medical countermeasures, including diagnostics, therapeutics, and vaccines, to enable effective and timely emergency response to Nipah virus outbreaks. This Personal View provides an update to the WHO roadmap by defining current research priorities for dev

Paramyxoviruses are a family of single-stranded negative-sense RNA viruses, many of which are responsible for a range of respiratory and neurological diseases in humans and animals. Among the most notable are the henipaviruses, which include the deadly Nipah (NiV) and Hendra (HeV) viruses, the causative agents of outbreaks of severe disease and high case fatality rates in humans and animals. NiV and HeV are maintained in fruit bat reservoirs primarily in the family Pteropus and spillover into humans directly or by an intermediate amplifying host such as swine or horses. Recently, non-chiropter

Nipah virus (NiV), a highly pathogenic Henipavirus in humans, has been responsible for annual outbreaks in recent years. Experiments involving live NiV are highly restricted to biosafety level 4 (BSL-4) laboratories, which impedes NiV research. In this study, we developed transcription and replication-competent NiV-like particles (trVLP-NiV) lacking N, P, and L genes. This trVLP-NiV exhibited the ability to infect and continuously passage in cells ectopically expressing N, P, and L proteins while maintaining stable genetic characteristics. Moreover, the trVLP-NiV displayed a favourable safety

Nipah virus (NiV), despite being a highly deadly zoonotic paramyxovirus and a major new public health concern in South and Southeast Asia, is still not well discussed in discussions of infectious disease preparedness. Along with Hendra virus (HeV), NiV is a member of the Henipavirus genus and is linked to severe respiratory and encephalitic illness. Case fatality rates have been reported to range from 40 % to 75 %. Recurrent outbreaks, especially in Bangladesh, have shown person-to-person transmission and underscored the virus's potential for epidemics since its initial discovery during the Ma

Nipah virus continues to pose a recurrent zoonotic threat in Bangladesh. The sporadic spillover events are associated with high case fatality and limited opportunities for clinical intervention. Despite nearly two decades of surveillance response, early warning systems remain reactive, relying heavily on laboratory confirmation and case accumulation. This Perspective argues that early warning has been conceptually misframed as outbreak detection, rather than risk anticipation. Drawing on the epidemiological characteristics of Nipah virus and Bangladesh's current surveillance architecture, this

Hendra virus is an emerging zoonosis endemic to Australia with high fatality of infection in horses and humans. The role of infected domestic horses as amplifying bridge hosts for onward transmission to humans led to the creation of an equine vaccine as a One Health solution against spillover. Despite the risk this disease represents, previously reported equine vaccination rates were low. This study explored vaccination trends between 2012 and 2023 at fine spatial scales and identified demographic and ecological predictors of vaccination against emerging zoonoses. Over 1 million doses of Equiv

Nipah virus (NiV) is a highly pathogenic zoonotic paramyxovirus with significant public health concern. Since 2018, Kerala, India, has experienced NiV outbreaks with variable clinical manifestations ranging from acute encephalitis syndrome (AES) to acute respiratory distress syndrome (ARDS). Recently, we observed two NiV outbreaks in Malappuram, Kerala, India during 2024, with AES as the predominant clinical manifestation in July and September 2024. Among all close contacts screened no secondary transmission of NiV was observed. The data from NiV outbreaks of Kerala state indicated that AES-pr

UNLABELLED: New approach methodologies, such as high-complexity in vitro systems, are increasingly prioritized in biomedical research as potential alternatives to animal experimentation. We show that cerebral organoids derived from human induced pluripotent stem cells can be leveraged to (i) investigate isolate-specific replication dynamics of Nipah virus and (ii) model key histopathological lesions found in the brain tissue of infected human patients. Furthermore, we discuss the importance of organoid models for the study of Risk Group 4 viruses.

In 2014, an outbreak of zoonotic Nipah virus (NiV) occurred on Mindanao Island, the Philippines. We investigated the prevalence of NiV in Philippine bats. Because neutralizing antibodies were detected in insectivorous bats on Siargao Island, public health officials should consider that the distribution range of NiV is not limited to Mindanao Island.

Nipah virus (NiV) is a highly pathogenic zoonotic virus that causes severe encephalitis and respiratory diseases and has a high mortality rate in humans (>40%). Epidemiological studies on various fruit bat species, which are natural reservoirs of the virus, have shown that NiV is widely distributed throughout Southeast Asia. Therefore, there is an urgent need to develop effective NiV vaccines. In this study, we generated recombinant vaccinia viruses expressing the NiV glycoprotein (G) or fusion (F) protein using the LC16m8 strain, and examined their antigenicity and ability to induce immunity.

Nipah virus is a relatively newly discovered emerging virus on the WHO list of priority pathogens which has the potential to cause outbreaks with high fatality rates. Whilst progress is being made in the development of animal models for evaluating vaccines and therapies, some of the more fundamental data on Nipah virus are lacking. We performed studies to generate novel information on the aerosol survival of Nipah virus and to look at the efficacy of two common disinfectants. We also performed studies to evaluate the inactivation of Nipah virus by using neutral buffered formalin. Nipah virus w
