Potentiation of halothane hepatotoxicity by chronic ethanol administration in rat: an animal model of halothane hepatitis.

To determine if chronic ethanol administration modifies the effect of halothane on the liver, fourteen male Wistar rats were pair-fed nutritionally adequate liquid diets containing either ethanol (36% of calories) or isocaloric carbohydrate (controls) for 6 weeks. After halothane anesthesia of these animals under different oxygen concentration, the livers were examined light microscopically as well as biochemically. The livers from rats fed ethanol which received halothane at low oxygen concentration showed multifocal or patchy necrosis primarily in the centrilobular regions with parenchymal l

Detection and molecular characterization of hepatitis E virus in food-producing animals in Slovakia.

Hepatitis E virus (HEV) is an emerging public health concern in developed countries, with most infections linked to the foodborne transmission of the zoonotic genotype HEV-3. With increasing number of viral hepatitis E cases in human population in Slovakia, knowledge of the situation in the food-producing animal population is becoming increasingly important. Detection of HEV RNA was performed in 290 samples (collected during period of March 2023 to August 2025) from four different species of food-producing animals - pigs, cows, sheep and goats. The presence of HEV RNA was confirmed by real-tim

Drosophila melanogaster: A Powerful Tiny Animal Model for the Study of Metabolic Hepatic Diseases.

Animal experimentation is limited by unethical procedures, time-consuming protocols, and high cost. Thus, the development of innovative approaches for disease treatment based on alternative models in a fast, safe, and economic manner is an important, yet challenging goal. In this paradigm, the fruit-fly Drosophila melanogaster has become a powerful model for biomedical research, considering its short life cycle and low-cost maintenance. In addition, biological processes are conserved and homologs of ∼75% of human disease-related genes are found in the fruit-fly. Therefore, this model has been

A modified animal model of hepatic regeneration induced by hilar bile duct ligation.

Mechanisms of the proliferation of liver are mainly studied in animal model of liver regeneration after partial hepatectomy (PH). However, the PH model involves complex regeneration mechanisms, including hemodynamic factors, cytokines, growth factors, and metabolites. Among liver metabolites, bile acid (BA) is a key signaling molecule that regulates liver regeneration. This study aimed to establish a new type of rapid liver hyperplasia model induced mainly by bile acid pathway through hepatoenteral circulation with hilar bile duct ligation (HBDL). We first established the HBDL model by ligatin

Liver cell transplantation -- novel animal model for human hepatic viral infections.

AIM: To generate a model of human hepatitis B infection (HBV) in immunocompetent rats with chimeric human liver. METHODS AND RESULTS: Normal rats were tolerized to human hepatocytes by exposure to human hepatocytes at day 17 of gestation. We transplanted human hepatocytes and inoculated HBV into the rats after birth. Mixed lymphocytes assay, a measure for tolerance, indicated that animals fetally exposed to human hepatocytes developed tolerance to human hepatocytes. Spleen lymphocytes from tolerized animals did not proliferate when challenged with donor human hepatocytes. In contrast, control

Gamma-aminobutyric acid and benzodiazepine receptors in an animal model of fulminant hepatic failure.

Hepatic encephalopathy (HE) is a syndrome of generalized neural inhibition, which complicates both acute and chronic liver failure. Since gamma-aminobutyric acid (GABA) is the principal inhibitory neurotransmitter of the brain and HE is associated with increased responsiveness to certain drugs (benzodiazepines and barbiturates) that mediate neural inhibition by binding to the GABA receptor complex on postsynaptic neural membranes, the status of this receptor complex in HE was investigated. Fulminant hepatic failure was induced in rabbits by the intravenous injection of galactosamine hydrochlor

Animal naming test at discharge is associated with hepatic encephalopathy after elective TIPS.

BACKGROUNDS & AIMS: Overt hepatic encephalopathy (OHE) is the most feared complication after transjugular intrahepatic portosystemic shunt (TIPS) placement. We aimed to evaluate the usefulness of the animal naming test (ANT) in predicting the occurrence of OHE after elective TIPS placement. METHODS: The ANT (1 min) was evaluated before TIPS, at discharge, and at 1, 3, and 6 months in all patients treated with elective TIPS between September 2019 and November 2024. RESULTS: We included 100 consecutive patients (80% men; median age 59 years; median MELD score 11). Indications for TIPS were asc

Safety and Effectiveness of Muse Cell Transplantation in a Large-Animal Model of Hepatic Fibrosis.

Background: In recent years, liver regeneration therapy using mesenchymal stem cells (MSC) has been investigated as an alternative therapy for end-stage liver diseases. Among these MSCs, multilineage-differentiating stress enduring (Muse) cells are reported to be effective in mouse models. The present study investigated the safety and effectiveness of Muse cell transplantation in large animal models of hepatic fibrosis. Methods: Muse cells and MSC were prepared from bone marrow cells of male mini pigs (Göttingen strain). Recipients mini pigs (female Göttingen strain) were repeatedly administer

Dietary restriction and hepatic cancer: Systematic review and meta-analysis of animal studies.

The effect of calorie restriction, fasting, and ketogenic diets on the treatment of liver cancer remains uncertain. Therefore, we conducted a systematic review to evaluate the effect of restrictive diets on the development and progression of liver cancer in animal models. We did a meta-analysis using the Cochrane Collaboration's Review Manager software, with the random effects model and the inverse variance technique. We examined 19 studies that were conducted between 1983 and 2020. Of these, 63.2% investigated calorie restriction, 21.0% experimented with a ketogenic diet, and 15.8% investigat

Hepatitis E virus and related viruses in wild, domestic and zoo animals: A review.

Hepatitis E is a human disease mainly characterized by acute liver illness, which is caused by infection with the hepatitis E virus (HEV). Large hepatitis E outbreaks have been described in developing countries; however, the disease is also increasingly recognized in industrialized countries. Mortality rates up to 25% have been described for pregnant women during outbreaks in developing countries. In addition, chronic disease courses could be observed in immunocompromised transplant patients. Whereas the HEV genotypes 1 and 2 are mainly confined to humans, genotypes 3 and 4 are also found in a

Role of hemostatic factors in hepatic injury and disease: animal models de-liver.

Chronic liver damage is associated with unique changes in the hemostatic system. Patients with liver disease often show a precariously rebalanced hemostatic system, which is easily tipped towards bleeding or thrombotic complications by otherwise benign stimuli. In addition, some clinical studies have shown that hemostatic system components contribute to the progression of liver disease. There is a strong basic science foundation for clinical studies with this particular focus. Chronic and acute liver disease can be modeled in rodents and large animals with a variety of approaches, which span c

An overview of animal models for investigating the pathogenesis and therapeutic strategies in acute hepatic failure.

Acute hepatic failure (AHF) is a severe liver injury accompanied by hepatic encephalopathy which causes multiorgan failure with an extremely high mortality rate, even if intensive care is provided. Management of severe AHF continues to be one of the most challenging problems in clinical medicine. Liver transplantation has been shown to be the most effective therapy, but the procedure is limited by shortage of donor organs. Although a number of clinical trials testing different liver assist devices are under way, these systems alone have no significant effect on patient survival and are only re

Troglitazone-induced hepatic necrosis in an animal model of silent genetic mitochondrial abnormalities.

Troglitazone, a first-generation thiazolidinedione antidiabetic drug, was withdrawn from the market due to an unacceptable risk of idiosyncratic hepatotoxicity. Troglitazone does not cause hepatotoxicity in normal healthy rodents, but it produces mitochondrial injury in vitro at high concentrations. The aim of this study was to explore whether genetic mitochondrial abnormalities might sensitize mice to hepatic adverse effects of troglitazone. We used heterozygous superoxide dismutase 2 (Sod2(+/-)) mice as a model of clinically silent mitochondrial stress. Troglitazone was daily administered fo

Animal models of fulminant hepatic failure: need to test liver support devices.

Fulminant hepatic failure is one of the most dramatic entities in clinical medicine, but experimental studies of its pathogenesis, evolution and treatment have, so far been limited by the lack of satisfactory animal models for testing new supportive treatment options. The variable aetiology, complex pathogenetic mechanisms and inconstant clinical evolution of human fulminant hepatic failure make it particularly difficult to establish an "ideal fulminant hepatic failure animal model" suitable for all studies: it is no longer mandatory to develop one single model serving all possible scientific

Animal models for the study of human hepatitis B and D virus infection: New insights and progress.

Hepatitis B virus (HBV) is a member of the Hepadnaviridae family and infects hepatocytes, leading to liver pathology in acutely and chronically infected individuals. Co-infection with Hepatitis D virus (HDV), which requires the surface proteins of HBV to replicate, can exacerbate this disease progression. Thus, the >250 million people living with chronic HBV infection, including 13 million co-infected with HDV, would significantly benefit from an effective and affordable curative treatment. Animal models are crucial to the development of innovative disease therapies, a paradigm repeated again

Broad antifibrotic activities of AK3280 in pulmonary, hepatic, cardiac, and skin fibrosis animal models.

Fibrosis is the pathological outcome of many chronic inflammatory diseases, affecting various human organs. It is a significant contributor to global morbidity and mortality that affects nearly half of the elderly population. Pirfenidone (PFD) and nintedanib are approved by the FDA for treating pulmonary fibrosis, but these treatments are associated with poor tolerability and limited efficacy. Moreover, no antifibrotic drugs are approved for other fibrosis-related diseases, highlighting an urgent unmet medical need for more effective therapies. Here we report the in vivo pharmacological activi
