Role of innate immunity in the development of cancer immunotherapy immune-mediated adverse events.

Immune checkpoint inhibitors (ICIs) have transformed modern cancer therapy by restoring antitumor T-cell responses through blockade of immune tolerance pathways such as CTLA-4 and PD-1/PD-L1. However, the same immune activation that underlies their clinical efficacy can also lead to immune-related adverse events (irAEs), a broad spectrum of inflammatory and autoimmune toxicities that may affect virtually any organ system. The incidence and severity of these events vary according to the specific agent, tumor type, and treatment strategy. While irAEs have traditionally been attributed to dysregu

Resistance to PD-1/PD-L1 blockade: Mechanism and pharmacological strategies.

Immune checkpoint inhibitors (ICIs) targeting the PD-1/PD-L1 axis have transformed the realm of cancer treatment. Nevertheless, fewer than one in five patients derive durable benefit, and a substantial proportion of initial responders eventually relapse, reflecting a heterogeneous milieu of intrinsic and acquired mechanisms of resistance. Resistance to ICIs can arise from impaired neoantigen generation and presentation (low tumor mutational burden, loss of functional HLA class I and β2-microglobulin), defective IFN-γ/JAK-STAT signaling, co-expression of alternative checkpoints (LAG-3, TIM-3, T

Immune-pressure redistribution in resistance to PD-1/PD-L1 blockade: mechanisms, biomarkers, and therapeutic design.

PD-1/PD-L1 blockade can produce durable tumor control, yet primary, adaptive, and acquired resistance remain common. Existing accounts often catalogue resistance by cellular compartment, obscuring the coordinated nature of tumor adaptation. Here, we introduce immune-pressure redistribution as a treatment-oriented framework that complements cancer immunoediting by asking where therapeutic immune pressure is diverted after checkpoint release. Resistance is organized into three coupled routes: transfer into tumor-intrinsic escape through antigen-presentation loss, interferon-response defects, onc

Multifocal ground-glass opacities in lung cancer: a narrative review of genomic evolution, immune ecology, and biology-informed clinical management.

BACKGROUND AND OBJECTIVE: Ground-glass opacity (GGO) is a computed tomography descriptor rather than a histologic diagnosis. Multifocal GGOs (mGGOs) are increasingly detected during lung cancer screening, yet their clonal origins, immune ecology, progression risk, and optimal management remain difficult to reconcile within conventional lung cancer frameworks. This narrative review evaluates genomic, immunologic, and clinical evidence relevant to mGGOs and examines how biological findings may inform surveillance and local treatment without encouraging overtreatment. METHODS: PubMed/MEDLINE, We

Tumor cell derived osteopontin and prostaglandin E2 synergistically promote the expansion of myeloid derived suppressor cells during the tumor immune escape phase.

The immune escape stage in cancer immunoediting is a pivotal feature, transitioning immune-controlled tumor dormancy to progression, and augmenting invasion and metastasis. Tumors employ diverse mechanisms for immune escape, with generating immunosuppressive cells from skewed hematopoiesis being a crucial mechanism. This led us to suggest that tumor cells with immune escape properties produce factors that induce dysregulations in hematopoiesis. In support of this suggestion, this study found that mice bearing advanced-stage tumors exhibited dysregulated hematopoiesis characterized by the devel

In Situ Cancer Vaccines: Redefining Immune Activation in the Tumor Microenvironment.

Cancer is one of the leading causes of mortality worldwide. Nanomedicines have significantly improved life expectancy and survival rates for cancer patients in current standard care. However, recurrence of cancer due to metastasis remains a significant challenge. Vaccines can provide long-term protection and are ideal for preventing bacterial and viral infections. Cancer vaccines, however, have shown limited therapeutic efficacy and raised safety concerns despite extensive research. Cancer vaccines target and stimulate responses against tumor-specific antigens and have demonstrated great poten

Targeting Arginine Metabolism to Reverse Immune Paralysis in Cancer.

The immune system plays an essential role in protecting the host from malignant transformation through immune surveillance, a concept that was later expanded by the cancer immunoediting framework. While immune responses eliminate emerging tumors, they can also impose selective pressure that promotes tumor adaptation and immune escape. Among the mechanisms involved in this process, metabolic immune suppression has gained increasing attention, as it weakens immune function without causing immune cell death. In this context, the semi-essential amino acid L-arginine has emerged as an important met

The Immune Landscape of Pheochromocytoma and Paraganglioma: Current Advances and Perspectives.

Pheochromocytomas and paragangliomas (PPGLs) are rare neuroendocrine tumors derived from neural crest cells from adrenal medullary chromaffin tissues and extra-adrenal paraganglia, respectively. Although the current treatment for PPGLs is surgery, optimal treatment options for advanced and metastatic cases have been limited. Hence, understanding the role of the immune system in PPGL tumorigenesis can provide essential knowledge for the development of better therapeutic and tumor management strategies, especially for those with advanced and metastatic PPGLs. The first part of this review outlin

The Immune Microenvironment in Human Papilloma Virus-Induced Cervical Lesions-Evidence for Estrogen as an Immunomodulator.

Globally, human papilloma virus (HPV) infection is a common sexually transmitted disease. However, most of the HPV infections eventually resolve aided by the body's efficient cell-mediated immune responses. In the vast majority of the small group of patients who develop overt disease too, it is the immune response that culminates in regression of lesions. It is therefore a rarity that persistent infection by high-risk genotypes of HPV compounded by other risk factors progresses through precancer (various grades of cervical intraepithelial neoplasia-CIN) to cervical cancer (CxCa). Hence, althou

Old and New Players of Inflammation and Their Relationship With Cancer Development.

Pathogens or genotoxic agents continuously affect the human body. Acute inflammatory reaction induced by a non-sterile or sterile environment is triggered for the efficient elimination of insults that caused the damage. According to the insult, pathogen-associated molecular patterns, damage-associated molecular patterns, and homeostasis-altering molecular processes are released to facilitate the arrival of tissue resident and circulating cells to the injured zone to promote harmful agent elimination and tissue regeneration. However, when inflammation is maintained, a chronic phenomenon is indu

Exosome RAB10 inhibits JAK1/STAT1 to hinder macrophage M1 polarization and promote tumor immune escape.

Exosomes are key mediators of communication between tumor cells and the tumor microenvironment(TME); however, the mechanisms underlying exosome-mediated crosstalk between tumor cells and macrophages remain largely unclear. This study investigated the effect of exosomal RAB10 on macrophage polarization and tumor growth. Mechanistically, RAB10 delivered by breast cancer cells binds to the interferon receptor IFNAR1 and inhibits JAK1/STAT1 pathway phosphorylation, thereby impeding M1 polarization and promoting M2 polarization. RAB10 expression was significantly upregulated in drug-resistant breas

CREBBP loss drives immune evasion by epigenetically silencing STING and MHC-I in small cell lung cancer.

Most small cell lung cancers (SCLCs) exhibit low immunogenicity and intrinsic resistance to immunotherapy, yet the underlying mechanisms remain poorly understood. Here, we identify CREBBP loss, a frequent genetic alteration in SCLC, as a key suppressor of tumor cell-intrinsic immune responses that drives an immune-cold tumor microenvironment in Rb1/Trp53-driven autochthonous mouse models. Consistently, Crebbp deletion promotes immune escape in syngeneic SCLC models. Mechanistically, Crebbp loss suppresses Sting and major histocompatibility complex class-I expression by reducing histone H3 acet

Peptide Loading Complex in Cancer: From Peptide Translocation, Editing, and Loading into MHC-I to a Potential Therapeutic Target.

The peptide-loading complex (PLC) is a pivotal endoplasmic reticulum (ER) protein machinery that facilitates peptide translocation, editing, and loading onto major histocompatibility complex class I (MHC-I) to allow recognition of infected or transformed cells by CD8⁺ cytotoxic T lymphocytes (CTLs). PLC comprises the peptide transporter complex formed of transporters associated with antigen processing (TAP1/2), the adapter protein tapasin, chaperones such as calreticulin (CRT) and ERp57, and β2-microglobulin (β2M), ensuring the proper assembly of MHC-I/peptide complexes for subsequent antigen

CCL21-DC in situ vaccination in murine NSCLC overcomes resistance to immunotherapy and generates systemic tumor-specific immunity.

BACKGROUND: Despite recent advances in immunotherapy, many patients with non-small cell lung cancer (NSCLC) do not respond to immune checkpoint inhibitors (ICI). Resistance to ICI may be driven by suboptimal priming of antitumor T lymphocytes due to poor antigen presentation as well as their exclusion and impairment by the immunosuppressive tumor microenvironment (TME). In a recent phase I trial in patients with NSCLC, in situ vaccination (ISV) with dendritic cells engineered to secrete CCL21 (CCL21-DC), a chemokine that facilitates the recruitment of T cells and DC, promoted T lymphocyte tumo

Accelerating tumor evolution and enhancing immunotherapy efficacy in lung adenocarcinoma based on EXO1 inhibition.

BACKGROUND: Emerging evidence highlights defects in DNA damage repair as critical modulators of tumor immunogenicity, yet the mechanistic interplay between repair pathway dynamics and immune checkpoint inhibitor efficacy remains elusive. This study aims to investigate the role of exonuclease 1 (EXO1), a dual-function nuclease involved in homologous recombination (HR) and mismatch repair (MMR), in immunogenomic regulation and immunotherapy response in lung adenocarcinoma (LUAD). METHODS: An integrated multi-omics analysis was performed using LUAD cohorts from The Cancer Genome Atlas (TCGA) to

E3 ubiquitin ligase NKLAM/RNF19b suppresses myc-driven B cell lymphomagenesis in Eμ-myc mice.

INTRODUCTION: Natural Killer Lytic-Associated Molecule (NKLAM) (also known as RNF19b) is a membrane-bound E3 ubiquitin ligase. Previous studies demonstrated a role of NKLAM in natural killer (NK) cell function and pro-inflammatory cytokine production by macrophages. Studies using lymphoma, melanoma and breast cancer cells found that tumor dissemination and metastasis are greater in NKLAM-deficient knockout (KO) mice than in wild type (WT) mice, indicating that NKLAM participates in controlling tumor development in vivo. METHODS: We employed the Eμ-myc mouse model to determine whether NKLAM in

Up-regulation of indoleamine 2,3-dioxygenase 1 (IDO1) expression and catalytic activity is associated with immunosuppression and poor prognosis in penile squamous cell carcinoma patients.

BACKGROUND: Indoleamine 2,3-dioxygenase 1 (IDO1) and tryptophan (Trp) catabolism have been demonstrated to play an important role in tumor immunosuppression. This study examined the expression and catalytic activity of IDO1 in penile squamous cell carcinoma (PSCC) and explored their clinical significance. METHODS: IDO1 expression level, serum concentrations of Trp and kynurenine (Kyn) were examined in 114 PSCC patients by immunohistonchemistry and solid-phase extraction-liquid chromatography-tandem mass spectrometry. The survival was analyzed using Kaplan-Meier method and the log-rank test. H

How does spaceflight affect the acquired immune system?

Prostate cancer (PCa) remains a leading cause of cancer‑related mortality in men despite advances in screening and localized treatment. The clinical heterogeneity of PCa, ranging from indolent disease to aggressive, lethal phenotypes, underscores the urgent need for reliable biomarkers that improve diagnosis, prognostication and therapeutic decision‑making. While prostate‑specific antigen testing has reduced mortality, its limited specificity has resulted in overdiagnosis and overtreatment. The present review provides a comprehensive overview of contemporary and emerging biomarkers that suppor
