Alterations in prefrontal cortical serotonin and antidepressant-like behavior in a novel C3H/HeJxDBA/2J recombinant inbred mouse strain.

In the present study, two genetically related inbred mouse strains selectively bred for high and low fear-sensitized acoustic startle reflex (FSS) were assessed in the forced swim test model of anti-depressant action and central monoamine concentrations in several brain regions were investigated. These mice were generated through backcrossing C3H/HeJ mice on DBA/2J mice, followed by inbreeding for several generations. The high-FSS and low-FSS strains are known to differ in their acquisition and extinction of fear following auditory fear conditioning. Significantly increased concentrations of 5

Do malaria parasites mate non-randomly in the mosquito midgut?

Polymerase chain reaction (PCR)-based genotyping of oocysts dissected from mosquito midguts has previously been used to investigate overall levels of inbreeding within malaria parasite populations. We present a re-analysis of the population structure of Plasmodium falciparum malaria using diploid genotypes at three antigen-encoding loci in 118 oocysts dissected from 34 mosquitoes. We use these data to ask whether mating is occurring at random within the mosquito midgut, as is generally assumed. We observe a highly significant deficit of heterozygous oocysts within mosquitoes at all three loci,

[Long Sperm Storage in the Siberian Crane (Grus leucogeranus Pallas): Analysis of Paternity and Relatedness under Artificial Insemination].

Using ten microsatellite loci, paternity analysis has been conducted for 71 individuals of the Siberian crane (Grus leucogeranus Pallas) obtained under artificial insemination in Oka Crane Breeding Center in 2001–2014. The fathers of 39 chicks were the sires whose sperm was used for insemination directly before fertilized egg laying. Paternity of 23 fertilizations belonged to the sires whose sperm was used in the beginning or middle of insemination cycle. Nine cases of fertilization resulted from natural copulation of artificially inseminated females with their social partners. The terms of sp

Abundant genetic variation is retained in many laboratory schistosome populations.

Schistosomes are obligately sexual blood flukes that can be maintained in the laboratory using freshwater snails as intermediate and rodents as definitive hosts. The genetic composition of laboratory schistosome populations is poorly understood: whether genetic variation has been purged due to serial inbreeding or retained is unclear. We sequenced 19 - 24 parasites from each of five laboratory Schistosoma mansoni populations and compared their genomes with published exome data from four S. mansoni field populations. We found abundant genomic variation (0.897 - 1.22 million variants) within lab

Fertilization of Ascidians: Gamete Interaction, Self/Nonself Recognition and Sperm Penetration of Egg Coat.

Fertilization is one of the most important events in living organisms to generate a new life with a mixed genetic background. To achieve successful fertilization, sperm and eggs must undergo complex processes in a sequential order. Fertilization of marine invertebrate Ciona intestinalis type A (Ciona robusta) has been studied for more than a hundred years. Ascidian sperm are attracted by chemoattractants from eggs and bind to the vitelline coat. Subsequently, sperm penetrate through the vitelline coat proteolytically and finally fuse with the egg plasma membrane. Here, we summarize the fertili

Ancestral self-compatibility facilitates the establishment of allopolyploids in Brassicaceae.

Self-incompatibility systems based on self-recognition evolved in hermaphroditic plants to maintain genetic variation of offspring and mitigate inbreeding depression. Despite these benefits in diploid plants, for polyploids who often face a scarcity of mating partners, self-incompatibility can thwart reproduction. In contrast, self-compatibility provides an immediate advantage: a route to reproductive viability. Thus, diploid selfing lineages may facilitate the formation of new allopolyploid species. Here, we describe the mechanism of establishment of at least four allopolyploid species in Bra

Advanced Generation Seed Orchard of Abies alba Mill. in Romania Combining Genetic Gain and Diversity.

The genetic parameters at 6, 9 and 12 years were studied in two progeny trials (one half-sib and one full-sib) of silver fir (Abies alba Mill.) in Romania, in order to establish an appropriate breeding strategy for advancing second-generation seed orchards. The half-sib trial (HS) consists of 60 open-pollinated families of plus trees from four first-generation seed orchards, while the full-sib trial (FS) consists of 51 half-diallel crosses of 11 plus trees from one seed orchard. Tree height and diameter were found to be under moderate to strong genetic control at both the family and individual

Uncovering genetic population structure in the Endangered northern rockhopper penguin (Eudyptes moseleyi) across islands in the Southern Atlantic and Indian oceans.

BACKGROUND: The northern rockhopper penguin (Eudyptes moseleyi) is a threatened species, listed as Endangered globally by the IUCN owing to rapid population decreases in the past, combined with a limited distributional range. Their breeding is confined to five islands in the central South Atlantic Ocean (Gough, Nightingale, Inaccessible, Alex (= Middle), and Tristan da Cunha) and two islands in the southern Indian Ocean (Amsterdam and Saint Paul). Non-breeding birds forage widely in these oceans and vagrant individuals have been recorded in the Falkland Islands/Malvinas, South Africa, Kerguele

Genetic rescue by distant trees mitigates qualitative pollen limitation imposed by fine-scale spatial genetic structure.

Restricted seed dispersal frequently leads to fine-scale spatial genetic structure (i.e., FSGS) within plant populations. Depending on its spatial extent and the mobility of pollinators, this inflated kinship at the immediate neighbourhood can critically impoverish pollen quality. Despite the common occurrence of positive FSGS within plant populations, our knowledge regarding the role of long-distance pollination preventing reproductive failure is still limited. Using microsatellite markers, we examined the existence of positive FSGS in two low-density populations of the tree Pyrus bourgaeana.

Oocyte and embryo preservation in wild animals: an update.

The reduction in population genetic diversity due to inbreeding depression and the negative impact of human activity on habitats ultimately generates an extinction debt. Therefore, there is always a dire need to save wild population and to protect biodiversity. Preservation of wildlife female germplasm, i.e., oocytes and embryos, is a promising biotechnological tool to conserve species' biodiversity. Other applied tools of Assisted Reproductive Technology (ART) which assure conservation of endangered species include artificial insemination (AI), embryo transfer technology (ETT), and sperm cryo

Population Genetic Structure of the Endangered Long-Tailed Goral (Naemorhedus caudatus) in South Korea Revealed by Genome-Wide Markers from a 3-RADseq Approach.

The long-tailed goral (Naemorhedus caudatus) is facing severe habitat fragmentation along the Baekdudaegan Mountain Range, South Korea, due to expanding anthropogenic infrastructures and ASF fence installations. To evaluate the evolutionary impacts of these barriers, we assessed the genome-wide genetic diversity and structural connectivity of South Korean goral populations using a high-resolution triple-digest restriction site-associated DNA sequencing (3-RADseq) framework. Out of 36 biological samples collected across regional demes, 34 individuals were successfully genotyped, yielding an ini

Sex Allocation and Reproductive Success in a Perennial Hermaphroditic Herb: Both Size and Timing Matter.

AbstractIn simultaneous hermaphrodites, resource availability and the temporal distribution of mates determine male and female fitness and optimal sex allocation. In insect-pollinated plants, we expect individuals to allocate more to their female function when they are large and more to their male function when other individuals have many ovules available to be fertilized. Here, we studied the dependence of sex allocation and male and female components of reproductive success on both the size and the timing of reproduction in the plant Pulsatilla alpina (Ranunculaceae), accounting for inbreedi

Quantitative genetics of trauma induced mortality in Drosophila melanogaster.

Traumatic brain injury is a major cause of chronic neurological impairment worldwide, and there is evidence that both genetic and environmental variation contribute to the likelihood of recovery. Using an insect model of traumatic brain injury, we examined variation in the risk of mortality using quantitative genetic approaches applied previously for life history traits in Drosophila melanogaster. We quantified additive genetic variance for mortality risk using a controlled breeding design and found levels of variation consistent with existing data on major fitness components. We did not detec

Unraveling genetic diversity and population structure of pineapple germplasm using genome-wide SNP markers.

Pineapple (Ananas comosus (L.) Merr.) is one of the most important tropical fruits worldwide. It is primarily propagated clonally and exhibits high heterozygosity which can mask deleterious mutations, potentially leading to inbreeding depression and hindering breeding efforts. To address this, we conducted a comprehensive genome-wide analysis of 91 pineapple accessions and identified over 7.9 million high-quality SNPs. We utilized these SNPs to investigate the genetic structure and reproductive patterns of diverse cultivars and related varieties. Population structure analysis revealed a unique

Floral Biology, Breeding System and Conservation Implications for the Azorean Endemic Azorina vidalii (Campanulaceae).

This study investigates the seasonal and floral phenology, breeding strategies, and floral morphology of Azorina vidalii, an Azorean endemic Campanulaceae with hermaphroditic, protandrous flowers, dichogamy and secondary pollen presentation. Seasonal phenology was recorded in four field populations and floral phenology in a garden population. Reproductive strategies were assessed via controlled hand pollinations in one field population. Floral morphometrics were analysed using 23 floral and five pollen traits from 121 flowers across fourteen populations throughout the Azores archipelago. Non-p

Evolution from mixed to fixed handedness in mirror-image flowers: insights from adaptive dynamics.

Mirror-image flowers (enantiostyly) involve a form of sexual asymmetry in which a flower's style is deflected either to the left or right side, with a pollinating anther orientated in the opposite direction. This curious floral polymorphism, which was known but not studied by Charles Darwin, occurs in at least 11 unrelated angiosperm families and represents a striking example of adaptive convergence in form and function associated with cross-pollination by insects. In several lineages, dimorphic enantiostyly (one stylar orientation per plant, both forms occurring within populations) has evolve

Reproductive physiology and artificial insemination studies in wild and captive gerenuk ( Litocranius walleri walleri ).

Gerenuk antelope in North American zoos are descended from 28 founders imported from Kenya approximately 20 years ago. Intensive management is required to prevent inbreeding depression. Artificial insemination has potential for augmenting genetic management, but successful application requires a thorough understanding of species' reproductive norms. Semen collected from captive (n = 10) and wild (n = 6) gerenuk contained low numbers of morphologically normal spermatozoa (approximately 40%). Age, but not season, influenced (P < 0.05) the proportion of morphologically normal spermatozoa (mean +/

Genetics then and now: breeding the best and biotechnology.

In the past, domesticated animals were genetically improved by identifying meritorious individuals, mating animals displaying desired traits, continued breeding of related animals to perpetuate their superior traits and crossbreeding when inbreeding depression became evident. Today, assisted reproduction and biotechnology allow breeders to design and direct the reproductive course, disseminate desired traits and hasten genetic improvement. Generation interval can be greatly reduced by combining artificial insemination, which is the oldest and most widely used assisted reproductive technology,

The sexual reproductive strategy in clonal Scopolia carniolica is based on obligatory cross-pollination and self-incompatibility.

Understanding the reproductive biology and breeding system of rare species is crucial for effective conservation. We examined floral biology, assessed the true pollinators, and investigated the reproductive effect of six pollination treatments (spontaneous and induced autogamy, geitonogamy, spontaneous and supplemental cross-pollination, control - flowers exposed to natural pollinators) on the fruit and seed set and their quality (size, viability) in two populations (artificial and natural) of clonal Scopolia carniolica (Solanaceae), a rare species distributed across Central and South-Eastern

Allograft outcomes in outbred mice.

Inbreeding depression and lack of genetic diversity in inbred mice could mask unappreciated causes of graft failure or remove barriers to tolerance induction. To test these possibilities, we performed heart transplantation between outbred or inbred mice. Unlike untreated inbred mice in which all allografts were rejected acutely (6-16 days posttransplantation), untreated outbred mice had heterogeneous outcomes, with grafts failing early (<4 days posttransplantation), acutely (6-24 days) or undergoing chronic rejection (>75 days). Blocking T cell costimulation induced long-term graft acceptance
