Alpha-linolenic acid-enriched diet prevents myocardial damage and expands longevity in cardiomyopathic hamsters.

Randomized clinical trials have demonstrated that the increased intake of omega-3 polyunsaturated fatty acids significantly reduces the risk of ischemic cardiovascular disease, but no investigations have been performed in hereditary cardiomyopathies with diffusely damaged myocardium. In the present study, delta-sarcoglycan-null cardiomyopathic hamsters were fed from weaning to death with an alpha-linolenic acid (ALA)-enriched versus standard diet. Results demonstrated a great accumulation of ALA and eicosapentaenoic acid and an increased eicosapentaenoic/arachidonic acid ratio in cardiomyopath

Effects of metal ions on lipid peroxidation in cultured rat hepatocytes loaded with alpha-linolenic acid.

We investigated the ability of various redox-active metal ions to induce lipid peroxidation in normal and alpha-linolenic acid-loaded (LNA-loaded) cultured rat hepatocytes. Lipid peroxidation was estimated by the accumulation of malondialdehyde (MDA) in the culture medium. At low concentrations induction was highest with ferrous ions (Fe), whereas at high concentrations, vanadium (V) and copper ions (Cu) had the greatest effect on both groups of hepatocytes. With any one of the three metal ions, the extent of lipid peroxidation in LNA-loaded hepatocytes was several times greater compared to no

Eicosapentaenoic and Docosahexaenoic Acid Levels in Mouse Tissues After Intake of Echium and Ahiflower Oils Rich in Stearidonic and α-Linolenic Acids.

Omega-3 fatty acids (n-3 FA), particularly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), have been consumed aiming to reduce cardiovascular disease. Fish oil is the main dietary source of EPA and DHA but has limitations that have stimulated interest in sustainable alternatives such as Echium ( Echium plantagineum ) and Ahiflower ( Buglossoides arvensis ) oils, both rich in α-linolenic acid (ALA) and stearidonic acid (SDA), precursors of EPA. However, the ability of these two oils to increase EPA and DHA levels in different tissues remains unclear. Thus, this study aimed to invest

Lactic acid bacteria-derived γ-linolenic acid metabolites are PPARδ ligands that reduce lipid accumulation in human intestinal organoids.

Gut microbiota regulate physiological functions in various hosts, such as energy metabolism and immunity. Lactic acid bacteria, including Lactobacillus plantarum, have a specific polyunsaturated fatty acid saturation metabolism that generates multiple fatty acid species, such as hydroxy fatty acids, oxo fatty acids, conjugated fatty acids, and trans-fatty acids. How these bacterial metabolites impact host physiology is not fully understood. Here, we investigated the ligand activity of lactic acid bacteria-produced fatty acids in relation to nuclear hormone receptors expressed in the small inte

Murine hepatic proteome adaptation to high-fat diets with different contents of saturated fatty acids and linoleic acid : α-linolenic acid polyunsaturated fatty acid ratios.

INTRODUCTION: Some health disorders, such as obesity and type 2 diabetes, are associated with a poor diet and low quality of the fat in it. The type and duration of the diet have an impact on the liver. This investigation uses the proteomic approach to identify changes in the mouse liver protein profile in adaptation to high-fat diets with different saturated fatty acid contents and linoleic acid (18:2n-6) to α-linolenic acid (18:3n-3) fatty acid ratios. MATERIAL AND METHODS: Four groups of male mice were fed different diets: one standard diet and three high-fat diets were investigated. After

Uptake of α-linolenic acid and its conversion to long chain omega-3 fatty acids in rats fed microemulsions of linseed oil.

The present work was designed to prepare linseed oil (LSO) microemulsion and explore the possibility of enhancing the uptake and utilization of α-linolenic acid (ALA) present in LSO. The bioavailability of encapsulated LSO as against native oil was monitored in rats by measuring the uptake in vitro using the intestinal everted sac model and in-vivo administration of microemulsions of LSO to rats for a period of 30 days. Microemulsions were prepared by using different binding materials such as gum acacia, whey protein and lipoid. When LSO was encapsulated with gum acacia, whey protein and lipoi

Microbial cell factories for gamma-linolenic acid biosynthesis: metabolic engineering, downstream processing, and future perspectives.

Gamma-linolenic acid (GLA) is a vital anti-inflammatory polyunsaturated fatty acid, yet its traditional supply from plant seeds such as evening primrose and borage is constrained by low yields and climatic instability. Consequently, microbial cell factories present a sustainable and season-independent alternative for scalable GLA production. This review critically synthesizes recent biotechnological advancements in utilizing oleaginous microorganisms as production platforms. It systematically elucidates the microbial lipid biosynthesis pathways and evaluates strategies to enhance GLA accumulat

Lipid-mediated broad-spectrum antiviral mechanism of alpha-linolenic acid against respiratory enveloped viruses.

BACKGROUND: α-Linolenic acid (ALA), an essential plant-derived polyunsaturated fatty acid with high safety, has been preliminarily shown to possess antiviral activity in vitro. However, the lack of in vivo activity evaluation and mechanism research has limited the understanding of its precise antiviral mechanism and therapeutic potential. PURPOSE: This study elucidates the mechanism by which ALA disrupts viral lipid envelopes, conferring broad-spectrum antiviral activity, and evaluates its feasibility as a natural botanical agent against respiratory enveloped viruses. METHODS: ALA (100 μM) w

Multiomics analyses identify diet-derived creatine and α-linolenic acid linking with metastatic thyroid cancer.

BACKGROUND: Thyroid cancer (TC) has traditionally been regarded as an indolent malignancy. However, it retains the potential for metastasis and lethality, underscoring its status as a malignant tumor. To explore the interplay between serum metabolite profiles, gut microbial communities, and TC metastasis, we conducted a comprehensive study. METHODS: Serum metabolites were analyzed using untargeted metabolomics methods to characterize the metabolic changes. Two diet-derived metabolites, creatine (Cr) and α-linolenic acid (ALA), were selected for in vitro validation. Finally, 16S rRNA gene sequ

In Vitro Evidence of α-Linolenic Acid Ethyl Ester Hydrolysis and Transesterification in the Gastrointestinal Tract in the Presence of Acylglycerols.

Omega-3 polyunsaturated fatty acids (n-3 PUFA), such as α-linolenic acid (ALA), are crucial for human health. Although n-3 PUFAs in food are mainly in triacylglycerol (TAG) form, they are often provided as ethyl esters (EE) in supplements. Previous research has demonstrated improved postprandial bioavailability of n-3 PUFAs in the EE form when coingested with fat, potentially due to transesterification during digestion. To shed light on the bioavailability improvement, this study investigated the influence of triolein and 2-monoolein on the digestion of ALA-EE using an in vitro model, employin

Roseburia hominis enriched by baicalin reverses the non-response to metformin via upregulating linolenic acid metabolism.

Metformin is the most commonly used hypoglycemic drug for patients with type 2 diabetes (T2D), but about 30% of patients show non-response potentially linked to gut microbiota imbalance. Although baicalin exhibits potent gut microbiota-modulating activity, its role in reversing metformin non-response remains unclear. Here, we recruited patients with T2D who were non-responders to metformin treatment and collected their fecal samples to construct a humanized mouse model via fecal microbial transplantation. We found that baicalin combined with metformin improved the abnormal glucose tolerance in

Associations of serum gamma-linolenic acid levels with erythema severity and anxiety/depression status in patients with rosacea.

BACKGROUND: The development of rosacea is suggested to be closely associated with lipid metabolism, inflammation, and anxiety/depression. Gamma linolenic acid (GLA) is a key factor participating in lipid metabolism, which is also confirmed to regulate the inflammatory response. However, the associations of serum GLA levels with rosacea severity and psychological status still remain unclear. OBJECTIVE AND LIMITATIONS OF THE STUDY: The present study aimed to investigate the associations of gamma linolenic acid (GLA), a key factor participating in lipid metabolism and the inflammatory response,

The Activity of Liposomal Linolenic Acid Against Helicobacter pylori In Vitro and Its Impact on Human Fecal Bacteria.

Helicobacter pylori (H. pylori) infection is associated with a variety of gastrointestinal diseases. Here, we focused on the activity of a novel nanomedicine-liposomal linolenic acid (LipoLLA) against H. pylori and its impact on human fecal bacteria in vitro. The minimum inhibitory concentrations (MICs) of LipoLLA against 30 H. pylori clinical strains were determined in combination with amoxicillin (AMX), metronidazole (MTZ), levofloxacin (LVFX) and clarithromycin (CAM). Bactericidal activity was measured by generating concentration-bactericidal curves at different times and pH values. Leakage

Failure of short-term gamma-linolenic acid treatment to reduce urinary calcium loss of diabetic rats.

Calcium re-absorption in the kidney is impaired in streptozotocin (STZ) diabetic rats, thereby causing hypercalciuria. Increased calcium loss starts within 1-2 days after induction of diabetes and reaches a plateau after 2 weeks. The excessive calcium excretion was previously shown to be reduced by treatment with gamma-linolenic acid (GLA) or evening primrose oil rich in GLA. However, in these studies, the animals were pre-treated for several weeks before injection of STZ. In the present study we investigated whether GLA can reduce calcium excretion when treatment starts at the same time as in

Vitamin A deficiency reduces liver and colon docosahexaenoic acid levels in rats fed high linoleic and low alpha-linolenic acid diet.

Studies indicate that the transcription factor peroxisome proliferator-activated receptors (PPARs) regulate the activity of delta-6 and -5 desaturases and several key enzymes of peroxisomal beta-oxidation, including acyl-CoA oxidase. These enzymes are vital for the synthesis of docosahexaenoic (22:6 omega 3; DHA) and osbond (22:5 omega 6, OA) acids. An activated PPAR must form a hetrodimer with the obligate cofactor retinoid X receptor (RXR) to interact with a peroxisome proliferator responsive element (PPRE) of a target gene and to regulate transcriptional expression. The vitamin A metabolite

Alpha-Linolenic Acid Modulates T Cell Incorporation in a 3D Tissue-Engineered Psoriatic Skin Model.

Psoriasis is an autoimmune skin disease with an increased number of leukocytes infiltrating the dermal and epidermal compartments compared with normal skin. N-3 polyunsaturated fatty acids (n-3 PUFAs) are frequently used in the clinic in order to attenuate the symptoms of psoriasis. For psoriatic patients, a supplementation of the diet with alpha-linolenic acid (ALA) reduces the activation of T cell signaling pathways, leading to a significant reduction in inflammatory cytokine secretion. However, the precise mechanism of action of n-3 PUFAs in psoriasis is still not understood. In the present

Effect of alpha linolenic acid on membrane fluidity and respiration of liver mitochondria in normoglycemic and diabetic Wistar rats.

The omega 3 fatty acids (ω3FA) have been recommended for the treatment of Type 2 Diabetes Mellitus (T2DM) and its complications, but there are studies questioning those beneficial effects. In this research, we supplemented the short-chain ω3FA, alpha-linolenic acid (ALA), to a model of rats with T2DM and normoglycemic controls, for 5 months. We were mainly interested in studying the effects of diabetes and ALA on the physicochemical properties of mitochondrial membranes and the consequences on mitochondrial respiration. We found that the Respiratory Control (RC) of diabetic rats was 46% lower
