Effects of partial liquid ventilation with FC-77 on acute lung injury in newborn piglets.

Partial liquid ventilation (PLV) with various types of perfluorochemicals (PFC) has been shown to be beneficial in treating acute lung injury. FC-77 is a type of PFC with relatively high vapor pressure and evaporative losses during PLV. This study tested the hypothesis that using FC-77 for PLV with hourly replacement is effective in treating acute lung injury. Fifteen neonatal piglets were randomly and evenly divided into 3 study groups: 1) lavage-induced lung injury followed by conventional mechanical ventilation (Lavage-CMV); 2) lavage-induced lung injury followed by PLV using FC-77 with hou

Partial liquid ventilation with perfluorocarbon in acute lung injury: light and transmission electron microscopy studies.

Liquid ventilation using perfluorocarbon has been shown to improve gas exchange in animal models of acute lung injury as well as in children with acute respiratory distress syndrome. This study was designed to define structural features of lung injury following partial liquid ventilation (PLV) using light and transmission electron microscopy in a rabbit model of acute respiratory distress. Animals were treated with either conventional mechanical ventilation (CMV-gas) (n = 6) or PLV (n = 5) for 4 h after the induction of acute lung injury with saline lavage. Control animals were killed after th

The hemodynamic effect of total liquid ventilation in piglets with reduced cardiac function after cardiopulmonary bypass.

Total liquid ventilation (TLV) is a promising treatment for severe lung injury and heart resuscitation. We aim to investigate the hemodynamic effect of TLV in piglets with preexisting cardiac dysfunction. After onset of cardiopulmonary dysfunction induced by cardiopulmonary bypass (CPB), piglets were randomly treated with TLV (n = 6) or conventional gas ventilation (n = 6) for 240 min. Hemodynamics was monitored before and at a 30-min interval after CPB. CPB with cardiac arrest resulted in significant decrease in cardiac index (CI) (96.19 ± 6.74 vs. 48.58 ± 3.96 mL/min/kg, P < 0.001) and incre

Computer tomographic assessment of perfluorocarbon and gas distribution during partial liquid ventilation for acute respiratory failure.

The average in vivo chest computed tomographic (CT) attenuation number (air = -1,000, soft tissue = 0, perflubron = +2,300 Hounsfield units [HU]) of 10 ventrodorsal-oriented lung segments was calculated to assess the distribution of gas and perflubron in 14 oleic acid lung-injured adult sheep during partial liquid ventilation (PLV, n = 7) or gas ventilation (GV, n = 7). Partial liquid ventilation was associated with a significant decrease in shunt fraction (PLV = 40 +/- 12%, GV = 76 +/- 12%, p = 0.004). Computed tomographic attenuation data during expiration (HUexp) demonstrated minimal gas ae

Alkaline Liquid Ventilation of the Membrane Lung for Extracorporeal Carbon Dioxide Removal (ECCO2R): In Vitro Study.

Extracorporeal carbon dioxide removal (ECCO2R) is a promising strategy to manage acute respiratory failure. We hypothesized that ECCO2R could be enhanced by ventilating the membrane lung with a sodium hydroxide (NaOH) solution with high CO2 absorbing capacity. A computed mathematical model was implemented to assess NaOH-CO2 interactions. Subsequently, we compared NaOH infusion, named "alkaline liquid ventilation", to conventional oxygen sweeping flows. We built an extracorporeal circuit with two polypropylene membrane lungs, one to remove CO2 and the other to maintain a constant PCO2 (60 ± 2 m

Liquid ventilation improves pulmonary function, gas exchange, and lung injury in a model of respiratory failure.

OBJECTIVE: The authors evaluated gas exchange, pulmonary function, and lung histology during perfluorocarbon liquid ventilation (LV) when compared with gas ventilation (GV) in the setting of severe respiratory failure. BACKGROUND: The efficacy of LV in the setting of respiratory failure has been evaluated in premature animals with surfactant deficiency. However, very little work has been performed in evaluating the efficacy of LV in older animal models of the adult respiratory distress syndrome (ARDS). METHODS: A stable model of lung injury was induced in 12 young sheep weighing 16.4 +/- 3.0

Liquid movements in ventilated and perfused isolated lungs of fetal sheep at 0.87, 0.90, and 0.95 of term.

To study lung liquid movements, an isolated lung model, adapted from adult physiology, was developed to measure pulmonary weight changes after the onset of artificial ventilation of lungs from 16 fetal sheep at 0.87 (n = 5), 0.90 (n = 6), and 0.95 (n = 5) of gestation. The fetuses were delivered by Caesarean section. After tracheotomy and thoracotomy under general anaesthesia, a blocked air-free tracheal cannula and a pulmonary arterial catheter were inserted and secured to ensure in situ perfusion of the pulmonary circulation (Krebs-Henseleit buffer) without ventilation before the lungs were

Tracheal gas insufflation with partial liquid ventilation to treat LPS-induced acute lung injury in juvenile piglets.

OBJECTIVES: Partial liquid ventilation (PLV) with perfluorocarbons (PFC) seems not superior to conventional ventilation clinically. We hypothesized that a combination of continuous tracheal gas insufflation (TGI) with protective strategy of PLV (low dose of PFC, low inflation pressure, moderate inhalation of oxygen and moderate anesthesia) would improve cardiopulmonary function in acute lung injury. METHODS: Twenty-four healthy juvenile piglets were anesthetized and mechanically ventilated at PEEP of 2 cmH(2)O with a peak inspiratory pressure of 10 cmH(2)O and FIO(2) of 0.4. The piglets were

Simultaneous identification of the three active constituents in Lung-Ventilating-Regulating Oral Liquid by RP-HPLC.

The study aimed to develop a HPLC method for ephedrine, hesperidin, and baicalin in Lung-Ventilating-Regulating Oral Liquid. The three active constituents were identified in an Agilent TC-C18 (2) chromatographic column (250mm × 4.6mm, 5μm), with 0.2% phosphoric acid solution - methyl cyanides as mobile phase, which was performed at a gradient elution column temperature of 25oC and a flow rate of 0.8 mL•min-1. Then the eluate was detected at detection wavelengths of 207 nm (for ephedrine) and 278 nm (for hesperidin and baicalin). Under the chromatographic conditions, ephedrine, hesperidin, and

Partial liquid ventilation combined with inhaled nitric oxide in acute respiratory failure with pulmonary hypertension in piglets.

This study was a prospective, randomized, controlled design to evaluate gas exchange, lung mechanics, and pulmonary hemodynamics during partial liquid ventilation (PLV) combined with inhaled nitric oxide (NO) in acute respiratory failure (ARF) with pulmonary hypertension (PH). ARF with PH was induced in 12 piglets weighing 9.7-13.7 kg by repeated lung lavages and the continuous infusion of the stable endoperoxane analog of thromboxane. Thereafter the animals were randomly assigned either for PLV or conventional mechanical ventilation (CMV) at a fractional concentration of inspired O2 (Fio2) of

Perfluorocarbon protects lung epithelial cells from neutrophil-mediated injury in an in vitro model of liquid ventilation therapy.

Liquid ventilation with perfluorocarbon has been effective in improving gas exchange and pulmonary function in the setting of acute respiratory failure. How improvement is brought about remains unknown. In the present study, we examined perfluorocarbon for effects on neutrophil function and for effects on neutrophil-epithelial cell interactions that could underlie its in vivo activity. Exposure of neutrophils in vitro to perfluorocarbon followed by washing did not interfere with their ability to generate oxidants or release proteolytic enzymes upon subsequent stimulation. Likewise, such treatm

Effects of perfluorochemicals for intrapulmonary vancomycin administration and partial liquid ventilation in an animal model of meconium-injured lungs.

BACKGROUND: Acute lung injury, such as meconium aspiration syndrome in neonates, may present with exacerbated ventilation and perfusion abnormalities. This can impair the efficacy of intravenous antibiotic therapy in treating pulmonary infection. Intrapulmonary administration via perfluorochemical as a vehicle may adequately deliver drugs, such as the poorly pulmonary-penetrative antibiotic vancomycin, to affect lung regions while maintaining gas exchange and non-toxic serum level. METHODS: Twelve newborn piglets were injured with intra-tracheally installed human meconium, and randomly groupe

Influence of partial liquid ventilation on bacterial growth and alveolar expansion in newborn rabbits with group B-streptococcal pneumonia.

Partial liquid ventilation (PLV) with perfluorocarbons has been considered as an alternative therapy for severe inflammatory lung disease. The present study was performed to test whether PLV influences bacterial growth and lung histology in a rabbit model of congenital pneumonia caused by group B streptococci. Near-term newborn rabbits were tracheotomized, inoculated via the airways with group B streptococci, and subsequently ventilated for 5 h with either PLV or conventional ventilation. At 30 min after group B streptococci administration, animals in the PLV group (n = 16) received 30 mL/kg b
