Review of health consequences from high-, intermediate- and low-level exposure to organophosphorus nerve agents.

Short and long-term health effects from exposure to organophosphorus (OP) military and insecticidal nerve agents are evaluated based on the abundant scientific literature published over five decades on health effects in humans (from human experimentation and occupational exposures) and in laboratory animals. Four distinct health effects are identified: acute cholinergic toxicity; organophosphate-induced delayed neuropathy (OPIDN); subtle long-term neuropsychological and neurophysiological effects; and a reversible muscular weakness called 'intermediate syndrome'. Some effects are subtle and di

An investigation into the use of Caenorhabditis elegans as a model organism for nerve agent exposure research.

Nerve agents exert their toxicological effects via inhibition of acetylcholinesterase (AChE). This inhibition results in excessive cholinergic signalling, which can result in fatality through cessation of breathing. Current therapies for nerve agent poisoning antagonise the overstimulation of muscarinic acetylcholine receptors and use benzodiazepines to enhance GABAergic inhibition as a counter to increased excitability in the central nervous system. Caenorhabditis elegans (C. elegans) can be effectively used to assess the toxic effects of AChE inhibition by paraoxon-ethyl, which shares the sa

Enhancing oxime efficacy into brain using ultrasound to counteract nerve agent exposure.

Organophosphates (OP) found in pesticides and chemical weapons irreversibly inhibit acetylcholinesterases (AChE) and cause toxic accumulation of acetylcholine throughout the organism. Due to their lipophilicity, OP easily cross the blood-brain barrier (BBB) and affect the central nervous system (CNS), resulting in epileptic seizures and long-term cognitive impairment. The antidote includes oximes which reactivate inhibited AChE. Unfortunately, oximes have limited BBB penetration and therefore fail to prevent neurological damage. Improving the penetration of oximes through the CNS and their the

Design and development of a prototype for specific naked-eye detection of blister and nerve agents.

In view of the strong need to strengthen the national security arising from chemical terrorism, a rapid, specific, and onsite detection of chemical warfare agents (CWAs) employing a simple and easy-to-use kit is of utmost importance. Constant and sincere efforts are being carried out by the scientific community to find reliable techniques/methods for early warning detection. Herein, we designed a prototype technique in the form of a smart and portable chemical weapon detection kit (CWDK) to facilitate rapid and onsite detection. In this portable kit, a range of unique chemical probes were cond

Neuroprotection by delayed triple therapy following sarin nerve agent insult in the rat.

The development of refractory status epilepticus (SE) induced by sarin intoxication presents a therapeutic challenge. In our current research we evaluate the efficacy of a delayed combined triple treatment in ending the abnormal epileptiform seizure activity (ESA) and the ensuing of long-term neuronal insult. SE was induced in male Sprague-Dawley rats by exposure to 1.2LD50 sarin insufficiently treated by atropine and TMB4 (TA) 1 min later. Triple treatment of ketamine, midazolam and valproic acid was administered 30 min or 1 h post exposure and was compared to a delayed single treatment with

Effects of non-depolarizing neuromuscular blocking agents on the cardiac vagus nerve in the guineapig.

The ability of gallamine, metocurine, pancuronium, tubocurarine and atropine to block the response to postganglionic stimulation of the vagus nerve were measured in isolated, spontaneously beating guineapig atria. From analysis of the results in conjunction with those of earlier assays of concentrations that block effects of preganglionic vagal stimulation, it was concluded that only tubocurarine possessed ganglionic blocking activity. From calculation of the extent of block of muscarinic receptors at drug concentrations which blocked the response to postganglionic vagal stimulation, it was co

Assessing oxime reactivation efficacy using principal component analysis: Insights from nerve agents inhibited human butyrylcholinesterase.

BACKGROUND AND PURPOSE: The toxicity of organophosphorus compounds (OPs) and related nerve agents (NAs) impairs the cholinergic system via irreversible inhibition of acetylcholinesterase (AChE) activity by phosphylation of the catalytic serine. Reactivation of the enzyme activity largely depends on the structural compatibility between the enzyme, an oxime reactivator, and the specific OP compound. EXPERIMENTAL APPROACH: For this study, we used our recently published data on the reactivation of human butyrylcholinesterase inhibited by the NAs sarin, cyclosarin, tabun and VX, using a library of

Inhibition of tPA-NMDAR interaction prevents neurovascular and functional deficits induced by organophosphorus nerve agents.

Organophosphorus compounds (OP), including pesticides and nerve agents such as sarin, soman and Novichok, irreversibly inhibit cholinesterases (ChE), resulting in excessive cholinergic signaling, excitotoxicity, and neuroinflammation. Although current countermeasures provide partial protection, OP exposure remains associated with central nervous system (CNS) damage, including blood-brain barrier (BBB) disruption and cognitive impairments. In this study, we identify a critical role for tissue-type plasminogen activator (tPA)-a serine protease that modulates neuronal and endothelial N-methyl-D-

Acetylcholinesterase inhibitors (nerve agents) as weapons of mass destruction: History, mechanisms of action, and medical countermeasures.

Nerve agents are organophosphorus acetylcholinesterase inhibitors. Acute exposure to nerve agents can cause rapid death. In this review, we summarize the history of nerve agent development and use in warfare, the mechanisms by which these agents cause death or long-term brain damage, and the treatments for preventing death or long-term morbidity. The G-series nerve agents, tabun, sarin, soman, ethyl sarin, and cyclosarin, were developed by the Nazis. VX, the best-known of the V-series agents, was synthesized in the 1950's by a British scientist. Little is known about the development of the nov

A photoluminescent nanocrystal-based signaling protocol highly sensitive to nerve agents and highly toxic organophosphate pesticides.

A photoluminescent II-VI group semiconductor nanocrystal (NC)-based signaling platform composed of thioglycolic acid capped CdS NCs and acetylcholinesterase-acetylthiocholine enzyme catalytic reaction system was developed that was shown to be highly sensitive to nerve agents and toxic organophosphate pesticides with detection limits down to sub-nM levels. This new sensing protocol does not require troublesome conjugation of biomacromolecules onto the surface of NCs.

Effects of muscarinic agonists and depolarizing agents on inositol monophosphate accumulation in the rabbit vagus nerve.

The effects of muscarinic agonists and depolarizing agents on inositol phospholipid hydrolysis in the rabbit vagus nerve were assessed by the measurement of [3H]inositol monophosphate production in nerves that had been preincubated with [3H]inositol. After 1 h of drug action, carbachol, oxotremorine, and arecoline increased the inositol monophosphate accumulation, though the maximal increase induced by these agonists differed. Addition of the muscarinic antagonists atropine or pirenzepine shifted the carbachol dose-response curves to the right, without decreasing the carbachol maximal stimulat

Integrated Colorimetric/Fluorescence Nanozyme Dual-Signal Probe for Simultaneous Detection and Degradation of Organophosphorus Nerve Agents.

Organophosphorus nerve agents pose grave risks to human health and ecology, creating an urgent demand for high-performance detection and remediation techniques. This work fabricates an all-in-one sensing-degradation platform with UiO-67@AuNCs/MnO2 nanozyme and AChE biorecognition element, where MnO2 acts as a reversible switch to regulate fluorescence and oxidase-like activity. The system delivers inverse dual optical signals for self-verification, greatly boosting detection precision. Tested on real sarin and soman, its colorimetric and fluorescent LODs reach 10-3-10-4 ng·mL-1, 2-7 orders of

Contribution of inflammatory processes to nerve cell toxicity by bilirubin and efficacy of potential therapeutic agents.

Hyperbilirubinemia is a common condition in neonatal life, where elevated levels of unconjugated bilirubin (UCB) may lead to adverse neurologic outcomes, namely in the presence of inflammatory features. In this review, we summarize recent concepts on UCB damage to brain cells and associated neuroinflammation research. Exposure of astrocytes and microglia to UCB initiates an inflammatory response with the release of proinflammatory cytokines, such as TNF-alpha, IL-1beta and IL-6, accumulation of extracellular glutamate and a time-dependent cell death. Moreover, undifferentiated cells revealed t

Neurological manifestations of encephalitic alphaviruses, traumatic brain injuries, and organophosphorus nerve agent exposure.

Encephalitic alphaviruses (EEVs), Traumatic Brain Injuries (TBI), and organophosphorus nerve agents (NAs) are three diverse biological, physical, and chemical injuries that can lead to long-term neurological deficits in humans. EEVs include Venezuelan, eastern, and western equine encephalitis viruses. This review describes the current understanding of neurological pathology during these three conditions, provides a comparative review of case studies vs. animal models, and summarizes current therapeutics. While epidemiological data on clinical and pathological manifestations of these conditions

Amelioration of nerve demyelination by hydrogen-producing silicon-based agent in neuropathic pain rats.

Trigeminal neuralgia (TN) is a complex orofacial neuropathic pain. The crippling condition's underlying mechanism is still not completely understood. The main cause of lightning-like pain in patients with TN may be chronic inflammation that causes nerve demyelination. Nano-silicon (Si) can safely and continuously produce hydrogen in the alkaline environment of the intestine to exert systemic anti-inflammatory effects. Hydrogen has a promising anti-neuroinflammatory impact. The study aimed to determine how intra-intestinal application of a hydrogen-producing Si-based agent affected the demyelin

Aqueous-Phase Destruction of Nerve-Agent Simulants at Copper Single Atoms in UiO-66.

Metal-organic frameworks (MOFs) have shown great success in aqueous-phase hydrolysis of nerve agents, with some even showing promise in the gas phase. However, both aqueous-phase reactivity and gas-phase reactivity are hindered because of the binding of the hydrolyzed products to the MOF nodes in a stable, bridging configuration, which limits turnover. Single transition-metal atoms in MOFs have been a growing field of interest for catalytic applications, and single atoms have been proposed to prevent the unwanted bridged conformation and increase catalytic turnover. To date, there has been lit

4-Aminopyridine: A Single-Dose Diagnostic Agent to Differentiate Axonal Continuity in Nerve Injuries.

INTRODUCTION: Traumatic peripheral nerve injuries (TPNIs) are increasingly prevalent in battlefield trauma, and the functional recovery with TPNIs depends on axonal continuity. Although the physical examination is the main tool for clinical diagnosis with diagnostic work up, there is no diagnostic tool available to differentiate nerve injuries based on axonal continuity. Therefore, treatment often relies on "watchful waiting," and this leads to muscle weakness and further reduces the chances of functional recovery. 4-aminopyridine (4-AP) is clinically used in multiple sclerosis patients for wa
