Cistanoside A decreases Tau hyperphosphorylation and neuronal apoptosis through the AKT/GSK3β pathway in okadaic acid-induced in vivo and in vitro models of Alzheimer's disease.

BACKGROUND: The multifactorial nature of Alzheimer's disease (AD) pathogenesis has driven the search for therapeutic agents with low toxicity that can act on multiple targets. Cistanoside A (Cis A), a bioactive compound derived from Cistanches Herba, exhibits anti-inflammatory, antioxidant, and antiapoptotic effects. Given that these processes are implicated in AD progression, we propose that Cis A may be a promising candidate for AD therapy. METHODS: We employed an okadaic acid (OA)-induced rat model in vivo and SH-SY5Y cells in vitro. Cognitive function was assessed using the Morris water m

Okadaic acid as a major problem for the seafood safety (Mytilus galloprovincialis) and the dynamics of toxic phytoplankton in the Slovenian coastal sea (Gulf of Trieste, Adriatic Sea).

This article presents the first results on shellfish toxicity in the Slovenian sea (Gulf of Trieste, Adriatic Sea) since the analytical methods for the detection of biotoxins (PSP, ASP, DSP and other lipophilic toxins) in bivalve molluscs were included in the national monitoring program in 2013. In addition to toxins, the composition and abundance of toxic phytoplankton and general environmental characteristics of the seawater (surface temperature and salinity) were also monitored. During the 2014-2019 study period, only lipophilic toxins were detected (78 positive tests out of 446 runs), of w

Galangin decreases p‑tau, Aβ42 and β‑secretase levels, and suppresses autophagy in okadaic acid‑induced PC12 cells via an Akt/GSK3β/mTOR signaling‑dependent mechanism.

Okadaic acid (OA)‑induced neurotoxicity may be considered a novel tool used to study Alzheimer's disease (AD) pathology, and may be helpful in the development of a novel therapeutic approach. It has been reported that galangin inhibits β‑site amyloid precursor protein‑cleaving enzyme 1 expression, which is a key enzyme for amyloid β (Aβ) generation and is a potential drug candidate for AD therapy. However, further studies are required to confirm its neuroprotective effects in other AD models. The present study aimed to explore the neuroprotective effects of galangin on OA‑induced neurotoxicity

Long term study (2012-2021) on Okadaic acid and Yessotoxins in Northwestern Adriatic Sea mussels (Italy). The role of inland rainfall in the dynamics of contamination events.

The Northwestern Adriatic Sea is highly exploited for mussel farming although it is periodically affected by Harmful Algal Bloom. In the last 30 years Okadaic acid (OA) and Yessotoxins (YTXs) predominated the mussels' toxin profile with serious threat to public health and damage to the shellfish industry. The Official Control Authorities address this issue by monitoring Marine Biotoxins in shellfish and toxic phytoplankton in seawater with good results in terms of safeguarding public health but with poor forecasting power. This work reports ten years (2012-2021) monitoring of OA and YTXs in mu

Immunosuppression in hamsters with progressive visceral leishmaniasis is associated with an impairment of protein kinase C activity in their lymphocytes that can be partially reversed by okadaic acid or anti-transforming growth factor beta antibody.

Progressive visceral infection of golden hamsters by Leishmania donovani amastigotes led to gradual impairment of the proliferative responses of their splenic or peripheral blood mononuclear cells (SPMC or PBMC, respectively) to in vitro stimulation with phorbol 12-myristate 13-acetate (PMA) and ionomycin (Io). Removal of macrophage-like adherent cells from SPMC or PBMC of infected animals (I-SPMC or I-PBMC) was earlier shown to restore almost completely their lymphoproliferative responses to PMA plus Io. The present study was directed to evaluate the status of protein kinase C (PKC), a molecu

Activation of p38 mitogen-activated protein kinase and mitochondrial Ca(2+)-mediated oxidative stress are essential for the enhanced expression of grp78 induced by the protein phosphatase inhibitors okadaic acid and calyculin A.

We have reported that treatment with okadaic acid, a potent protein phosphatase inhibitor, has the ability to enhance the synthesis of the 78-kDa glucose-regulated protein (GRP78). This article reports our investigation of another protein phosphatase inhibitor, calyculin A, demonstrating the signaling pathways elicited by the protein phosphatase inhibitors that lead to the induction of grp78. Our data showed that the induction process is abolished by SB203580, a specific inhibitor of p38 mitogen-activated protein kinase (p38(MAPK)). Phosphorylation-activation of p38(MAPK) in the treated cells

Role of phosphorylation sites and the C2 domain in regulation of cytosolic phospholipase A2.

Cytosolic phospholipase A2 (cPLA2) mediates agonist-induced arachidonic acid release, the first step in eicosanoid production. cPLA2 is regulated by phosphorylation and by calcium, which binds to a C2 domain and induces its translocation to membrane. The functional roles of phosphorylation sites and the C2 domain of cPLA2 were investigated. In Sf9 insect cells expressing cPLA2, okadaic acid, and the calcium-mobilizing agonists A23187 and CryIC toxin induce arachidonic acid release and translocation of green fluorescent protein (GFP)-cPLA2 to the nuclear envelope. cPLA2 is phosphorylated on mul

Protein phosphatases 1 and 2A in rabbit ciliary epithelium and iris-ciliary body.

A significant dephosphorylation of exogenous phosphorylase a by serine/threonine protein phosphatases in tissue extracts of rabbit ciliary epithelium and iris-ciliary body was observed. Okadaic acid caused a concentration-dependent inhibition of this dephosphorylation. In a series of diluted samples, selective inhibitions of protein phosphatase 2A by 1 nM okadaic acid and protein phosphatase 1 by 1 microM okadaic acid were determined. In the preparation of ciliary epithelium, the ratio of protein phosphatase 1 to protein phosphatase 2A activity was estimated to be 1:2. Dephosphorylation by oth

Effects of environmental variables on phycotoxin degradation and preliminary identification of transformation products.

Phycotoxins are released from phytoplankton into the water column during rapid cell division or during bloom termination. These processes introduce pulses of dissolved phycotoxins into coastal systems that can deleteriously impact animal, environmental, and human health. Despite evidence that they can persist months beyond their extracellular release, environmental factors controlling phycotoxin fate and persistence within aquatic environments have not been thoroughly studied. As such, we investigated the stability of four phycotoxins (pectenotoxin-2, yessotoxin, azaspiracid-2, and okadaic aci

Inhibitors of protein phosphatases 1 and 2A differentially prevent intrinsic and extrinsic apoptosis pathways.

Inhibitors of serine/threonine protein phosphatases can inhibit apoptosis. We investigated which protein phosphatases are critical for this protection using calyculin A, okadaic acid, and tautomycin. All three phosphatase inhibitors prevented anisomycin-induced apoptosis in leukemia cell models. In vitro, calyculin A does not discriminate between PP1 and PP2A, while okadaic acid and tautomycin are more selective for PP2A and PP1, respectively. Increased phosphorylation of endogenous marker proteins was used to define concentrations that inhibited each phosphatase in cells. Concentrations of ea

Serine/threonine phosphatase inhibitors decrease adrenergic arylalkylamine n-acetyltransferase induction in the rat pineal gland.

Adrenergic regulation of the pineal enzyme serotonin N-acetyltransferase [arylalkylamine N-acetyltransferase (AA-NAT); EC 2.3.1.87] accounts for the circadian rhythm in melatonin formation. In the present study, the role of protein phosphatases in the adrenergic regulation of rat pineal AA-NAT was investigated using specific inhibitors. In cultured pineals, the serine/threonine phosphatase type 1 and type 2A inhibitors okadaic acid and calyculin A significantly decreased adrenergically or cAMP-induced AA-NAT activity, whereas the serine/threonine phosphatase type 2B inhibitor cypermethrin and

Occurrence and profiles of lipophilic toxins in shellfish harvested from Argentina.

The presence of phytoplankton responsible for the production of lipophilic marine biotoxins is well recognised throughout parts of South America. To date, the quantitation of lipophilic toxins in Argentinean shellfish has been limited to select and highly focussed geographical studies. This work reports the analysis for lipophilic marine biotoxins in shellfish harvested across five regions of Argentina between 1992 and 2012. LC-MS/MS analysis was used for the quantitation of all regulated lipophilic toxins. High concentrations of okadaic acid group toxins were quantified, with a clear dominanc

Easy biodosimetry for high-dose radiation exposures using drug-induced, prematurely condensed chromosomes.

PURPOSE: To propose a practical method of biodosimetry for high-dose estimation. MATERIALS AND METHODS: Okadaic acid in the range 100 nM to 500 nM was given for 1-6 h to cultured lymphocytes that had been exposed in the range of 0 Gy to 40 Gy. The frequency of the cells with prematurely condensed chromosomes (PCC) was compared with that of metaphases accumulated by 0.05 microg/ml colcemid. The dose-response relationship of the yield of prematurely condensed ring chromosomes (PCC rings) in the irradiated lymphocytes was examined. RESULTS: The 0.05 microg/ml colcemid collected few metaphases w

Multidirectional effects of calmodulin kinase II on transmitter release in mature and newly formed mouse motor synapses.

Calmodulin inhibitor W-7 did not cause changes in the quantal content of postsynaptic end-plate potentials (EPP) in newly formed synapses, but prevented facilitation of acetylcholine secretion induced by L-type Ca(2+)channels blocker nitrendipine. CaMKII inhibitor KN-62 produced similar effect and suppressed the increase in EPP quantal content caused by blockade of L-type Ca(2+)channels. Phosphatase PP2A inhibitor okadaic acid significantly facilitated secretion in newly formed synapses; the effect was completely blocked by KN-62. In mature synapses, okadaic acid had no effect on transmitter s

Involvement of type 2C phosphatase in the dephosphorylation of 26 kDa phosphoprotein in rat parotid acinar cells.

Phosphorylation of three particulate proteins with molecular masses of 34, 26, and 22 kDa was stimulated in the presence of cyclic AMP/3-isobutyl-1-methylxanthine in saponin-permeabilized rat parotid acinar cells. When the particulate fraction isolated from the cells labeled with [gamma-32p]ATP was incubated at 30 degrees C, dephosphorylation of the 26 kDa phosphoprotein occurred in the presence of Mg2+ or Mn2+. Okadaic acid had no effect on the Mg(2+)-dependent dephosphorylation of the 26 kDa phosphoprotein. Addition of the recombinant type 2C phosphatase, Mg(2+)-dependent and okadaic acid-in

Na(+)-K(+)-2Cl- cotransport, Na+/H+ exchange, and cell volume in ferret erythrocytes.

Ferrets have high-Na+ and low-K+ erythrocytes (113 and 5.4 mmol/l cell water) due to the lack of Na(+)-K+ pumps. Because ferret erythrocytes have a high capacity for Na(+)-K(+)-2Cl- cotransport, the present study was undertaken to evaluate cell volume-related changes in cotransport activity and its role in volume regulation. With cell shrinkage, Na(+)-K(+)-2Cl- cotransport is activated about twofold. A large bumetanide-insensitive Na+ uptake component that has not yet been described is found in shrunken erythrocytes. Its inhibition by amiloride (concn inhibiting 50% of maximal response = 12 mi

Development of enzyme-linked immunosorbent assay with nanogold-labeled probes for detection of diarrhetic shellfish toxins.

Diarrheal shellfish toxins (DSTs) are a type of liposoluble toxins produced by toxic algae in the ocean, with main components of okadaic acid (OA), dinotricin-1 (DTX-1), and dinotricin-2 (DTX-2). They can accumulate in shellfish through food chain. Human consumption of toxic shellfish can have adverse effects on health. Therefore, conducting DSTS detection can effectively ensure food safety. Base on the principle of competitive enzyme-linked immunosorbent assay (ELISA), referring to the seed growth method, we prepared goldnano rods using gold nanoparticles (AuNPs), and conjugated them with oka

Phosphorylated alpha-synuclein at Ser-129 is targeted to the proteasome pathway in a ubiquitin-independent manner.

α-Synuclein (a-Syn) is a major component of fibrillar aggregates in Lewy bodies (LBs), a characteristic hallmark of Parkinson disease. Almost 90% of a-Syn deposited in LBs is phosphorylated at Ser-129. However, the role of Ser-129-phosphorylated a-Syn in the biogenesis of LBs remains unclear. Here, we investigated the metabolism of Ser-129-phosphorylated a-Syn. In SH-SY5Y cells, inhibition of protein phosphatase 2A/1 by okadaic acid, and inhibition of the proteasome pathway by MG132 or lactacystin accumulated Ser-129-phosphorylated a-Syn. However, these inhibitions did not alter the amounts of

Force-inhibiting effect of Ser/Thr protein phosphatase 2A inhibitors on bovine ciliary muscle.

Ciliary muscle is a smooth muscle characterized by a rapid response to muscarinic receptor stimulation and sustained contraction. Although it is evident that these contractions are Ca(2+)-dependent, detailed molecular mechanisms are still unknown. In order to elucidate the role of Ser/Thr protein phosphatase 2A (PP2A) in ciliary muscle contraction, we examined the effects of okadaic acid and other PP2A inhibitors on contractions induced by carbachol (CCh) and ionomycin in bovine ciliary muscle strips (BCM). Okadaic acid inhibited ionomycin-induced contraction, while it did not cause significan

Carcinogenic aspects of protein phosphatase 1 and 2A inhibitors.

Okadaic acid is functionally a potent tumor promoter working through inhibition of protein phosphatases 1 and 2A (PP1 and PP2A), resulting in sustained phosphorylation of proteins in cells. The mechanism of tumor promotion with okadaic acid is thus completely different from that of the classic tumor promoter phorbol ester. Other potent inhibitors of PP1 and PP2A - such as dinophysistoxin-1, calyculins A-H, microcystin-LR and its derivatives, and nodularin - were isolated from marine organisms, and their structural features including the crystal structure of the PP1-inhibitor complex, tumor pro
