Preterm birth in relation to the bisphenol A replacement, bisphenol S, and other phenols and parabens.

INTRODUCTION: Preterm birth continues to be a significant public heath concern and is a leading cause of perinatal and infant mortality. Environmental exposures to phenols and parabens are suspected to potentially contribute to the pathology of preterm birth, yet limited human studies have characterized the extent to which these toxicants are associated with birth outcomes. METHODS: We examined the associations between phenols, parabens, and preterm birth, within pregnant women who were recruited early in gestation into the LIFECODES cohort at Brigham and Women's Hospital in Boston, Massachus

Parabens promotes invasive properties of multiple human cells: A potential cancer-associated adverse outcome pathway.

Parabens are esters of p-hydroxybenzoic acid, which have been used as preservatives and considered safe for nearly a century, until the last two decades when concerns began to be raised about their association with cancers. Knowledge of the mode of action of parabens on the metastatic properties of different cancer cells is still very limited. In the present study, we investigated the effects of methylparaben (MP) and propylparaben (PP) on cell invasion and/or migration in multiple human cancerous and noncancerous cells, including hepatocellular carcinoma cells (HepG2), cervical carcinoma cell

Synthesis of 13 C-labeled parabens from isotopically enriched phenols using the Houben-Hoesch reaction.

Parabens are antimicrobial additives found in a wide array of consumer products. However, the halogenated compounds formed from parabens during wastewater disinfection are a potential environmental concern. In order to identify these transformation products and investigate their mechanism of formation, a synthetic route to ethyl parabens labeled with the stable isotope carbon-13 at specific positions within the benzene ring was developed. This efficient two-step procedure starts from commercially available 13 C-labeled phenols and involves (1) initial acylation of the phenol via a Houben-Hoesc

Associations between mixed urinary phenols and parabens metabolites and bone mineral density: Four statistical models.

Phenols and parabens widely exist in personal care and consumer products and have been proved to be endocrine disrupting chemicals that could disturb bone metabolism. The current studies focusing on the associations between phenols and parabens with bone mineral density (BMD) drew contradictory conclusions. Considering the bias might be due to not considering the effects of mixed exposure, we conducted the first cross-sectional study to investigate the associations of both single and mixed metabolites of phenols and parabens with BMD in three populations by setting up four models: generalized

Toxicity of the cosmetic preservatives parabens, phenoxyethanol and chlorphenesin on human meibomian gland epithelial cells.

Recently, we discovered that the cosmetic preservatives, benzalkonium chloride and formaldehyde, are especially toxic to human meibomian gland epithelial cells (HMGECs). Exposure to these agents, at concentrations approved for human use, leads within hours to cellular atrophy and death. We hypothesize that these effects are not unique, and that other cosmetic preservatives also exert adverse effects on HMGECs. Such compounds include parabens, phenoxyethanol and chlorphenesin, which have been reported to be toxic to corneal and conjunctival epithelial cells, the liver and kidney, as well as to

Microbial inactivation properties of a new antimicrobial/antithrombotic catheter lock solution (citrate/methylene blue/parabens).

BACKGROUND: Microbial infections are the most serious complications associated with indwelling central venous catheters. A catheter lock solution that is both antibacterial and antithrombotic is needed. The goal of this study was to determine whether a new catheter lock solution containing citrate, methylene blue and parabens has antimicrobial properties against planktonic bacteria and against sessile bacteria within a biofilm. These effects were compared to the antimicrobial properties of heparin at 2500 units/ml. METHODS: The tested solution (C/MB/P comprising 7% sodium citrate, 0.05% methy

Next generation risk assessment for skin sensitisation: A case study with propyl paraben.

Skin sensitisation is a key adverse health effect to be addressed in the safety assessment of cosmetic ingredients. Regulatory demands have urged the development of Next Generation Risk Assessment (NGRA) using New Approach Methodologies (NAM) and Defined Approaches (DA) instead of animal models. An illustrative NGRA case study shall demonstrate if the use of propyl paraben at 0.2% in a face cream was safe for consumers. A sequential stacking tier testing DA based on NAM data predicted propyl paraben to be a non-sensitiser, while some NAM input data showed positive results. To increase confiden

Evaluation of EDCs (phthalates, bisphenols, parabens, and benzophenones) in coffee: cross-sectional study in Algiers.

Endocrine disrupting chemicals (EDCs) are emerging environmental pollutants. The present study focused on evaluating the concentrations of EDCs in packed coffee. Among 12 investigated EDCs, only DEP, DEHP, DnBP, BPA, and BPF were found in packed coffee (20 %, 40 %, 80 %, 20 %, and 20 %, respectively). The mean values ranged from 0.230 ng/g for BPA to 23.214 ng/g for DnBP. DnBP was the most frequently detected EDC in coffee powder (60 %), followed by DEP, DEHP, BPA, and BPF (20 % each). Migration assessment showed that DEHP, DnBP, and BPF amounts were significantly higher after coffee preparati

MIL-68 (Ga) for the extraction of derivatized and non-derivatized parabens from healthcare products.

This study was the first-ever attempt to apply MIL-68 (Ga) in developing an analytical method. The method extracts and preconcentrates some parabens from mouthwash and hydrating gel samples. The variable extraction parameters were optimized, and the figures of merit were documented. Avogadro software was used besides discussing intermolecular interactions to clarify the absorption process. ComplexGAPI software was also exploited to assess the greenness of the method. After the derivatization of the parabens using acetic anhydride in the presence of sodium carbonate, sodium chloride was added t

Phthalate esters, parabens and bisphenol-A exposure among mothers and their children in Greece (Rhea cohort).

Exposure to endocrine disruptors, used as additives, preservatives, plasticisers and solvents in numerous consumer products, might cause adverse health effects. Humans exposed to these chemicals, metabolise and excrete them mostly via urine. Urinary metabolite concentrations are used as biomarkers of exposure. We evaluated the exposure of 4-month pregnant women and their children at 2 years of age to phthalates, parabens and bisphenol-A. Concentrations of eight phthalate metabolites, six parabens and bisphenol-A were measured in 239 mother-child pairs of the "Rhea" cohort in Greece. Concentrat

A network toxicology approach to decipher paraben-induced molecular dysregulation in breast cancer pathogenesis.

Paraben, extensively utilized as preservatives in cosmetics, pharmaceuticals, industrial products, and food due to its antimicrobial properties, represent pervasive environmental contaminants capable of bioaccumulation through dietary, dermal, and respiratory exposure, potentially leading to diseases including endocrine disruption, skin allergies, and breast cancer. As the endocrine-disrupting chemical (EDC) with estrogenic activity, paraben bind estrogen receptors (ERs), potentially disrupting hormonal homeostasis and increasing breast cancer risk. However, the molecular mechanisms linking pa

Maternal paraben exposure and intra-pair thyroid-stimulating hormone difference in twin neonates.

Thyroid hormones are essential for fetal growth and neurodevelopment. The recent frequent use of parabens has raised concerns about their endocrine-disrupting potential. However, the effects of maternal paraben exposure on neonatal thyroid hormone levels are still largely unknown. In our study, a co-twin control design was employed to analyze the relationships between maternal paraben exposure and neonatal thyroid-stimulating hormone (TSH) difference. We collected information from 252 mother-twin pairs from a twin birth cohort in Wuhan, China. Concentrations of six parabens were measured in ma
