Inactivation of Mycobacterium avium subsp. paratuberculosis during cooking of hamburger patties.

The role of Mycobacterium avium subsp. paratuberculosis (MAP) in Crohn's disease in humans has been debated for many years. Milk and milk products have been suggested as possible vectors for transmission since the beginning of this debate, whereas recent publications show that slaughtered cattle and their carcasses, meat, and organs can also serve as reservoirs for MAP transmission. The objective of this study was to generate heat-inactivation data for MAP during the cooking of hamburger patties. Hamburger patties of lean ground beef weighing 70 and 50 g were cooked for 6, 5, 4, 3, and 2 min,

Mycobacterium avium sub. paratuberculosis in tissue samples of Crohn's disease patients.

Crohn's disease is a non-specific chronic transmural inflammatory disease. The disease was associated with a frameshit mutation in the NOD2 gene. Nevertheless, other researchers associated the presence of M. paratuberculosis within the intestinal tissues of patients with the disease. An adapted "in situ hybridization" technique was used to detect IS900 M. paratuberculosis DNA in paraffin embedded tissue from Crohns tissue disease samples. We were able to identify M. paratuberculosis DNA in around 69% of the paraffine embedded intestinal samples of Crohn's disease patients analysed. The presenc

East Asian Mycobacterium avium subsp. paratuberculosis: molecular typing and population structure.

IMPORTANCE: The global molecular epidemiology of Mycobacterium avium subsp. paratuberculosis (MAP) has been well documented in North America and Europe, but the population-genetic framework of the East Asian lineages is underrepresented, hindering a complete understanding of the global genotype distribution and transmission dynamics. OBJECTIVE: This study aimed to establish a regional molecular-typing framework for MAP in East Asia and investigate its genetic relatedness with global lineages to explore potential intercontinental connections associated with international livestock movements.

Mycobacterium avium subspecies paratuberculosis (MAP) and Crohn's disease: the debate continues.

Crohn's disease (CD) in humans and Johne's disease (JD) in ruminants share numerous clinical and pathologic similarities. As Mycobacteria avium subspecies paratuberculosis (MAP) is known to fulfill Koch's postulates as the cause of JD, there has been considerable debate over the past century about whether MAP also plays a role in CD. With recent advances in MAP identification techniques, we can now demonstrate a higher presence of MAP in CD patients compared to the general population. However, it remains unclear if MAP is playing a bystander role or is directly pathogenic in these patients. St

Macrophage polarization in cattle experimentally exposed to Mycobacterium avium subsp. paratuberculosis.

Mycobacterium avium subspecies paratuberculosis (MAP), the causative agent of Johne's disease (JD) in cattle, has significant impacts on the livestock industry and has been implicated in the etiology of Crohn's disease. Macrophages play a key role in JD pathogenesis, which is driven by the manipulation of host immune mechanisms by MAP. A change in the macrophage microenvironment due to pathogenic or host-derived stimuli can lead to classical (M1) or alternative (M2) polarization of macrophages. In addition, prior exposure to antigenic stimuli has been reported to alter the response of macropha

Mycobacterium avium ss paratuberculosis-associated diseases: piecing the Crohn's puzzle together.

The relation of Mycobacterium avium ss paratuberculosis (MAP) to Crohn's Disease (CD) and other MAP-associated conditions remains controversial. New data, coupled with the analogous Helicobacter pylori (H. pylori) story, has permitted us to piece together the MAP puzzle and move forward with a more scientific way of treating inflammatory bowel disease, particularly CD. As infection moves centre stage in inflammatory bowel disease, the dated "aberrant reaction" etiology has lost scientific credibility. Now, our growing understanding of MAP-associated diseases demands review and articulation. We

Prophylactic effect of monensin sodium against experimentally induced paratuberculosis in mice.

Monensin sodium (0, 15, or 30 mg/kg of complete feed) was fed ad libitum for 1 week to female mice (strain C57BL6/J) that were genetically susceptible to infection with Mycobacterium paratuberculosis. Ten mice in each of the 3 groups were inoculated intraperitoneally with M paratuberculosis (10(9) organisms). Sterile saline solution was injected intraperitoneally into 10 other mice in each group. Rations were continued for 50 days, then mice were euthanatized, and body weight, splenic weight, and hepatic weight for each mouse were recorded. Ratios of body weight to splenic weight and of body w

Identification of restriction fragment length polymorphisms in DNA from Mycobacterium paratuberculosis.

DNAs from 34 mycobactin-dependent isolates of Mycobacterium paratuberculosis and 1 isolate of M. paratuberculosis 18 were digested with four restriction endonucleases. Southern hybridization experiments were performed with a 32P-labeled oligonucleotide DNA probe derived from the sequence of IS900, an insertion sequence present in 15 to 20 copies per M. paratuberculosis chromosome. The probe hybridized with DNA from each of the mycobactin-dependent isolates, and restriction fragment length polymorphisms were detected among the isolates with each restriction endonuclease used. Restriction fragme

Paratuberculosis vaccination induces a phenotype consistent with trained immunity in bovine neutrophils.

Vaccination decreases bacterial shedding and reduces the incidence and severity of infection by Mycobacterium avium subsp. paratuberculosis (Map), the causative agent of paratuberculosis (PTB), a chronic granulomatous enteritis that economically impacts the dairy industry. An inactivated Map vaccine exhibits features suggestive of trained immunity in cattle, along with enhanced neutrophil (PMN) activation in rabbits. Although PMNs from healthy cattle exhibit antimicrobial effects against Map, the impact of vaccination on bovine PMN function remains undetermined. This study has further explored

Evidence of intrauterine transmission of Mycobacterium avium subspecies paratuberculosis in pregnant goats.

Paratuberculosis (PTBC) is a chronic granulomatous enteritis of ruminants caused by Mycobacterium avium subsp. paratuberculosis (Map). Although PTBC intrauterine transmission has been described in cattle, sheep, and deer, data in dairy goats are limited. The present study investigated intrauterine Map infection in 32 pregnant Saanen goats from a herd with high PTBC prevalence in Jujuy, Argentina. Maternal and fetal tissues were analyzed using a range of methods, including bacteriological culture, IS900 PCR, histopathology, and serology. All goats were positive to the intradermal test, and 75%

Mycobacterium avium Subspecies paratuberculosis and Colorectal Cancer: Putting MAP on the Map.

The rising incidence of early-onset colorectal cancer (CRC) remains incompletely explained despite extensive investigation into genetic, environmental and metabolic factors. Emerging evidence suggests that infectious agents may contribute to colorectal carcinogenesis. A recent study by Tehrani et al. demonstrated the presence of Mycobacterium avium subspecies paratuberculosis (MAP) in a majority of colorectal cancer lesions and in approximately half of precancerous lesions, providing a critical epidemiologic anchor. MAP, a zoonotic intracellular pathogen long associated with Crohn's disease, e

Exploring the role of circulating microRNAs as potential diagnostic markers in Mycobacterium paratuberculosis.

Extracellular vesicles (EVs) are cell-derived and play a notable role in the development of chronic diseases and can be used as biomarkers as they transport microRNAs (miRNA). Existing research has found that most miRNA functions are carried out via intercellular transmission of EVs, which can protect and sort miRNAs. Early detection of disease is crucial for controlling the spread of the disease and improving livestock prognosis. miRNAs play a promising role as circulating biomarkers for early identification of disease. miRNA expressed in paratuberculosis infection has been identified using a

Paratuberculosis in South American camelids: two independent cases in alpacas in Germany.

BACKGROUND: Paratuberculosis, caused by Mycobacterium avium subspecies paratuberculosis (MAP), is a chronic granulomatous enteritis that affects domestic and wild ruminants and camelids. The disease has rarely been reported in alpacas in Germany. This publication describes epidemiologically independent cases of paratuberculosis in two alpacas in Germany. CASE PRESENTATION: Two alpacas, a 26-year-old female zoo animal (case 1) and a 2.5-year-old breeding stallion from a private owner (case 2), presented with progressive emaciation, leading to death (case 2) or euthanasia (case 1) because of de

IP10 is a predictor of successful vaccine protection against paratuberculosis infection in sheep.

The cell mediated immune response and ability of immune cells to migrate to the site of infection are both key aspects of protection against many pathogens. Mycobacterium avium subsp. paratuberculosis (MAP) is an intracellular pathogen and the causative agent of paratuberculosis, a chronic wasting disease of ruminants. Current commercial vaccines for paratuberculosis reduce the occurrence of clinical disease but not all animals are protected from infection. Therefore, there is a need to understand the immune responses triggered by these vaccines at the site of infection, in circulating immune

On deaf ears, Mycobacterium avium paratuberculosis in pathogenesis Crohn's and other diseases.

The historic suggestion that Mycobacterium avium subsp. paratuberculosis (Map) might be a zoonotic pathogen was based on the apparent similarity of lesions in the intestine of patients with Crohn's disease (CD) with those present in cattle infected with Map, the etiological agent of Johne's disease. Reluctance to fully explore this possibility has been attributed to the difficulty in demonstrating the presence of Map in tissues from patients with CD. Advances in technology have resolved this problem and revealed the presence of Map in a significant proportion of patients with CD and other dise

Facts, myths and hypotheses on the zoonotic nature of Mycobacterium avium subspecies paratuberculosis.

Mycobacterium avium subspecies paratuberculosis (MAP) is the causative agent of paratuberculosis (Johne's disease [JD]), a chronic granulomatous enteritis in ruminants. JD is one of the most widespread bacterial diseases of domestic animals with significant economic impact. The histopathological picture of JD resembles that of Crohn's disease (CD), a human chronic inflammatory bowel disease of still unresolved aetiology. An aetiological relevance of MAP for CD has been proposed. This and the ambiguity of other published epidemiological findings raise the question whether MAP represents a zoono

Assessment of Dietzia subsp. C79793-74 for treatment of cattle with evidence of paratuberculosis.

The objective of the present investigation was to determine whether the bacterium Dietzia subsp. C79793-74, previously shown to inhibit growth of Mycobacterium subsp. paratuberculosis under in vitro culture conditions, has therapeutic value as a probiotic for adult cattle with paratuberculosis.  Animals were obtained from several herds with evidence of disease based on seropositivity and/or fecal shedding.  Sixty-eight cows with initial evidence of Stage II or III paratuberculosis and 2 with an initial Stage IV disease were evaluated longitudinally.  Animals were either treated daily with vari

Where are all the Mycobacterium avium subspecies paratuberculosis in patients with Crohn's disease?

Mycobacterium avium subspecies paratuberculosis (MAP) causes a chronic granulomatous inflammation of the intestines, Johne's disease, in dairy cows and every other species of mammal in which it has been identified. MAP has been identified in the mucosal layer and deeper bowel wall in patients with Crohn's disease by methods other than light microscopy, and by direct visualization in small numbers by light microscopy. MAP has not been accepted as the cause of Crohn's disease in part because it has not been seen under the microscope in large numbers in the intestines of patients with Crohn's dis

Use of the bead beater for preparation of Mycobacterium paratuberculosis template DNA in milk.

Mycobacterium paratuberculosis is a recognized chronic enteric pathogen that can affect many different species of animals, including primates. It has been suggested that this organism is associated with Crohn's disease in humans, and that milk is a potential source of human exposure to this organism. The limit of the detection of M. paratuberculosis in milk samples by direct PCR was 10(5) cfu/mL if the traditional boiling method was used for template DNA preparation. In this study, an improved method for template DNA preparation was examined. The method involves the use of a bead beater, which

Exploring the link between Mycobacterium avium subsp. Paratuberculosis and colorectal cancer development.

BACKGROUND: Colorectal cancer (CRC) ranks third in incidence and fourth in cancer-related mortality worldwide. It typically begins as benign polyps that may progress to malignancy through cumulative genetic alterations. Inflammatory bowel disease (IBD) is a major risk factor for CRC, and Mycobacterium avium subspecies paratuberculosis (MAP) has been implicated in its pathogenesis and may also contribute to CRC development, particularly in regions with MAP-infected livestock. METHODS: In this case-control study which included 147 participants, we analyzed 74 patients with colorectal conditions
