Antidepressant-like effect of essential oil of Perilla frutescens in a chronic, unpredictable, mild stress-induced depression model mice.

Perilla frutescens (Perilla leaf), a garnishing vegetable in East Asian countries, as well as a plant-based medicine, has been used for centuries to treat various conditions, including depression. Several studies have demonstrated that the essential oil of P. frutescens (EOPF) attenuated the depressive-like behavior in mice. The present study was designed to test the anti-depressant effects of EOPF and the possible mechanisms in an chronic, unpredictable, mild stress (CUMS)-induced mouse model. With the exposure to stressor once daily for five consecutive weeks, EOPF (3, 6, and 9 mg·kg(-1)) an

Perilla-derived chalcone inhibits α-synuclein fibrillization and prevents aggregate-induced disruption of intracellular α-synuclein conformation in neuronal cells.

The pathological aggregation of α-synuclein (α-syn) is a hallmark of Parkinson's disease, where extracellular α-syn aggregates can disrupt intracellular α-synuclein conformation in neurons. In this study, we investigated the impact of 2',3'-dihydroxy-4',6'-dimethoxychalcone (DDC), isolated from green perilla, on α-syn fibrillization and subsequently on intracellular α-synuclein conformation. DDC concentration-dependently inhibited α-syn fibrillization in vitro, as determined by thioflavin S fluorescence and blue native polyacrylamide gel electrophoresis. DDC also disaggregated preformed α-syn

Antibacterial mechanism of action of perilla leaf essential oil on Shigella flexneri and its preservation effectiveness in fresh-cut lettuce.

Shigella flexneri is a major foodborne pathogen and a leading cause of bacillary dysentery in developing countries, typically transmitted via contaminated water and fresh produce. Although perilla leaves-used as both food and medicine-are rich in bioactive compounds, the antimicrobial and antibiofilm activities of perilla leaf essential oil (PLEO) against S. flexneri remain unclear. This study investigated the antibacterial mechanisms of PLEO against S. flexneri and evaluated its potential for use in preserving fresh-cut lettuce. Gas chromatography-mass spectrometry (GC-MS) analysis identified

Aqueous Extract of Perilla frutescens var. acuta Relaxes the Ciliary Smooth Muscle by Increasing NO/cGMP Content In Vitro and In Vivo.

The leaves of Perilla frutescens var. acuta (PFA) are commonly used as a traditional medicine in Korea, Japan, and China. We previously showed that PFA attenuates eye fatigue by improving visual accommodation through a clinical study. However, detailed mechanisms and chemical compounds have not been studied. In this study, we analyzed the active compounds in an aqueous extract of PFA involved in ciliary muscle relaxation in vitro and in vivo. NMR and MS analyses showed that the PFA extract contained mainly luteolin-7-O-diglucuronide and apigenin-7-O-diglucuronide. The composition after freeze-

From molecular interactions to functional enhancement: unraveling the non-covalent binding between Perilla seed protein fibrils and structurally diverse polyphenols.

Plant protein fibrils have gained increasing attention as promising delivery carriers for hydrophobic bioactive compounds. In this study, perilla seed protein fibrils (PSPF) prepared via acid-heat induced self-assembly were used to systematically investigate the non-covalent interactions with four structurally different polyphenols (quercetin, luteolin, naringenin, and curcumin) at pH 3.5. The functional properties and delivery potential of the non-covalent complexes were also evaluated. Thermodynamic analysis and molecular docking demonstrated that hydrogen bonds, van der Waals forces, and hy

Effects of 2'-3'-dihydroxy-4',6'-dimethoxychalcone derived from green perilla on auricle thickness in chronic contact dermatitis model mice.

Oxidative stress has been implicated in the pathogenesis of allergic contact dermatitis. The nuclear factor erythroid 2-related factor 2 (Nrf2)-antioxidant response element (ARE) pathway, an in vivo antioxidant system, induces antioxidant enzymes. In our previous studies, we isolated 2',3'-dihydroxy-4',6'-dimethoxychalcone (DDC) from green perilla and identified it as a novel activator of the Nrf2-ARE pathway. We also discovered that it exerted cytoprotective effects against oxidative stress in PC12 cells. However, its effects on skin disease model animals in vivo remain unclear. In the presen
