In situ isolation of mRNA from individual plant cells: creation of cell-specific cDNA libraries.

A method for isolating and cloning mRNA populations from individual cells in living, intact plant tissues is described. The contents of individual cells were aspirated into micropipette tips filled with RNA extraction buffer. The mRNA from these cells was purified by binding to oligo(dT)-linked magnetic beads and amplified on the beads using reverse transcription and PCR. The cell-specific nature of the isolated mRNA was verified by creating cDNA libraries from individual tomato leaf epidermal and guard cell mRNA preparations. In testing the reproducibility of the method, we discovered an inhe

Metabolic and signalling processes in plant cell wall integrity maintenance.

BACKGROUND: Our understanding of plant cell walls has profoundly changed in recent decades. Originally seen as sturdy recalcitrant structures supporting plant growth, development, and interactions with the environment, we now appreciate the dynamic processes and plasticity modulating cell wall homeostasis, enabling cell walls to perform their respective biological functions in a highly adaptive manner. A key component underlying plasticity and responsible for adaptive changes is the plant cell wall integrity maintenance mechanism. It is responsible for monitoring and maintaining the functional

Mapping plant cell-type-specific responses to environmental stresses.

Plants constantly encounter diverse biotic and abiotic stresses that threaten their survival and productivity. With climate change intensifying the severity of these stressors, a deeper understanding of plant responses at the cellular level has become increasingly urgent. Traditional bulk RNA sequencing provides insights but masks critical cell-type-specific heterogeneity and fails to capture dynamic cellular responses. This review highlights recent advances in single-cell/nucleus RNA sequencing and spatial transcriptomics, enabling high-resolution mapping of transcriptional programs across di

Review on the impact of cell phone radiation effects on green plants.

The aim of this review is to assess the impact of cell phone radiation effects on green plants. Rapid progress in networking and communication systems has introduced frequency- and amplitude-modulated technologies to the world with higher allowed bands and greater speed by using high-powered radio generators, which facilitate high definition connectivity, rapid transfer of larger data files, and quick multiple accesses. These cause frequent exposure of cellular radiation to the biological world from a number of sources. Key factors like a range of frequencies, time durations, power densities,

A Review on the Impact of Oxidative Stress and Medicinal Plants on Leydig Cells.

Leydig cells are essential for steroidogenesis and spermatogenesis. An imbalance in the production of reactive oxygen species (ROS) and the cellular antioxidant level brings about oxidative stress. Oxidative stress (OS) results in the dysfunction of Leydig cells, thereby impairing steroidogenesis, spermatogenesis, and ultimately, male infertility. To prevent Leydig cells from oxidative insults, there needs to be a balance between the ROS production and the cellular protective capacity of antioxidants. Evidence indicates that medicinal plants could improve Leydig cell function at specific conce

A plant-derived natural photosynthetic system for improving cell anabolism.

Insufficient intracellular anabolism is a crucial factor involved in many pathological processes in the body1,2. The anabolism of intracellular substances requires the consumption of sufficient intracellular energy and the production of reducing equivalents. ATP acts as an 'energy currency' for biological processes in cells3,4, and the reduced form of NADPH is a key electron donor that provides reducing power for anabolism5. Under pathological conditions, it is difficult to correct impaired anabolism and to increase insufficient levels of ATP and NADPH to optimum concentrations1,4,6-8. Here we

Phytomedicines (medicines derived from plants) for sickle cell disease.

BACKGROUND: Sickle cell disease, a common recessively inherited haemoglobin disorder, affects people from sub-Saharan Africa, the Middle East, Mediterranean basin, Indian subcontinent, Caribbean and South America. It is associated with complications and a reduced life expectancy. Phytomedicines (medicine derived from plants in their original state) encompass many of the plant remedies from traditional healers which the populations most affected would encounter. Laboratory research and limited clinical trials have suggested positive effects of phytomedicines both in vivo and in vitro. However,

Induction of programmed cell death by Prangos uloptera, a medicinal plant.

Inhibition of the cell growth or induction of cell death is the most promising area in cancer therapy. The induction of apoptosis by dichloromethane extract of Prangos uloptera was evaluated on the McCoy cell line. This plant's roots, aerial parts and fruit have medicinal value. Cell growth inhibitory and cell cytotoxicity effects of the extract were assayed by MTT and Trypan-blue tests, respectively. Morphological changes and DNA fragmentation were also evaluated. The viability tests showed 0.49 and 0.3 mg/ml as 50% inhibition concentration and 50% cytotoxicity concentration after 24 hours of

Use of plant cell cultures to study the metabolism of environmental chemicals.

The metabolism of the following environmental chemicals has been studied in cell suspension cultures of wheat (Triticum aestivum L.) and soybean (Glycine max L.):2, 4-dichlorophenoxyacetic acid (2,4-D), 2,4,5-trichlorophenoxyacetic acid (2,4,5-T), hexachlorobenzene, pentachlorophenol, diethylhexylphthalate , benzo [alpha] pyrene, and DDT. All chemicals tested, including the persistent ones, were partially metabolized. Polar conjugates predominated in all cases. A covalent incorporation into lignin could be demonstrated for 2,4-D and pentachlorophenol. A specific deposition in the cellular vacu

Single-Cell and Spatial Perspectives on Source-Sink Dynamics in Plants.

Source-sink relationships describe how plants coordinate carbon production, transport, and utilization to support growth, reproduction, and stress responses. Classical physiological studies established a robust organ-level framework for carbon allocation, but these approaches largely treat tissues as homogeneous units. Recent advances in single-cell and spatial omics technologies now reveal that both sources and sinks consist of diverse cell populations with specialized metabolic and transport functions. In this review, we synthesize emerging insights from single-cell transcriptomics, spatial

Pathogen-Induced Cell Wall Remodeling and Immune Signaling in Plants.

The plant cell wall is a primary barrier to pathogen invasion and a dynamic extracellular interface involved in defense signaling. During infection, pathogens remodel the wall through cell wall-degrading enzymes and effectors, whereas plants reinforce the wall and perceive wall-derived damage-associated molecular patterns that activate immune responses. Recent studies have refined this view by challenging a simple wallassociated kinase-centered model of oligogalacturonide perception, linking MIK2-associated peptide-receptor pathways to pattern-triggered immunity, and showing that pathogens can

A plant vesicle-dendritic cell chimera for enhancing cancer immunotherapy.

The tumour microenvironment (TME) causes mitochondrial dysfunction in resident dendritic cells (DCs), resulting in inadequate antigen presentation and weak T cell priming. Herein, we identify hypoxia as a key factor for causing pathological mitochondrial fission in tumour-associated DCs, and develop a plant vesicle-DC chimera to relieve hypoxia-induced mitochondrial dysfunction for enhancing cancer immunotherapy. The biohybrid chimera is fabricated by loading algae-derived nanovesicles (ANVs) with genetically engineered CCR2 overexpressing-DCs. The CCR2-DC-ANVs target tumour by leveraging the

Plant Cellulose as a Substrate for 3D Neural Stem Cell Culture.

Neural stem cell (NSC)-based therapies are at the forefront of regenerative medicine strategies for various neural defects and injuries such as stroke, traumatic brain injury, and spinal cord injury. For several clinical applications, NSC therapies require biocompatible scaffolds to support cell survival and to direct differentiation. Here, we investigate decellularized plant tissue as a novel scaffold for three-dimensional (3D), in vitro culture of NSCs. Plant cellulose scaffolds were shown to support the attachment and proliferation of adult rat hippocampal neural stem cells (NSCs). Further,

Isolation of Plant Root Nuclei for Single Cell RNA Sequencing.

The characterization of the transcriptional similarities and differences existing between plant cells and cell types is important to better understand the biology of each cell composing the plant, to reveal new molecular mechanisms controlling gene activity, and to ultimately implement meaningful strategies to enhance plant cell biology. To gain a deeper understanding of the regulation of plant gene activity, the individual transcriptome of each plant cell needs to be established. Until recently, single cell approaches were mostly limited to bulk transcriptomic studies on selected cell types.

[The power plants of the cell: Treatment of psychiatric symptoms in patients with mitochondriopathy].

Introduction Mitochondriopathies are pathologies of cell organelles, which are essential for the formation of adenosine triphosphate (ATP), which is responsible for cellular energy stock. When mitochondrial mutations occur, symptoms arise frequently in those organs that rely on a continuous energy supply, such as the nervous system. Although psychiatric illness is increasingly prevalent in patients with mitochondrial disease, less attention has been paid to its psychiatric presentations. Case Report We describe a case of a 21-year-old woman who presented in our outpatient department with panic
