Alfaxalone does not have long-term effects on goldfish pyramidal neuron action potential properties or GABAA receptor currents.

Anesthetics have varying physiological effects, but most notably alter ion channel kinetics. Alfaxalone is a rapid induction and washout neuroactive anesthetic, which potentiates γ-aminobutyric acid (GABA)-activated GABAA receptor (GABAA-R) currents. This study aims to identify any long-term effects of alfaxalone sedation on pyramidal neuron action potential and GABAA-R properties, to determine if its impact on neuronal function can be reversed in a sufficiently short timeframe to allow for same-day electrophysiological studies in goldfish brain. The goldfish (Carassius auratus) is an anoxia-t

Sedative effects of intramuscular alfaxalone-methadone-midazolam versus ketamine-methadone-midazolam in cats.

OBJECTIVE: To assess depth and quality of sedation and the ability to place an intravenous catheter in cats after intramuscular (IM) injection with alfaxalone-methadone-midazolam (AMM) or ketamine-methadone-midazolam (KMM). STUDY DESIGN: Blinded randomized clinical trial. ANIMALS: A group of 46 healthy Domestic Short Hair cats. METHODS: Cats were randomly assigned to be administered IM methadone 0.3 mg kg-1 and midazolam 0.3 mg kg-1 with either alfaxalone 2 mg kg-1 (AMM group, n = 23) or ketamine 3 mg kg-1 (KMM group, n = 23). Before IM injection, a temperament score was assigned. The react

Evaluation of intramuscular sodium nitroprusside injection to improve oxygenation in white-tailed deer (Odocoileus virginianus) anesthetized with medetomidine-alfaxalone-azaperone.

OBJECTIVE: In ungulates, α2-adrenergic agonists can decrease oxygenation possibly through alteration of pulmonary perfusion. Sodium nitroprusside can decrease pulmonary vascular resistance (PVR) and increase cardiac output (Q˙t) through vasodilation. The objective was to determine if sodium nitroprusside would improve pulmonary perfusion and attenuate the increased alveolar-arterial (a-a) gradient resulting from medetomidine-azaperone-alfaxalone (MAA) administration. STUDY DESIGN: Prospective, randomized, crossover study with a 2 week rest period. ANIMALS: A group of eight adult female capti

Effects of alfaxalone on cerebral blood flow and intrinsic neural activity of rhesus monkeys: A comparison study with ketamine.

OBJECTIVE: Alfaxalone has been used increasingly in biomedical research and veterinary medicine of large animals in recent years. However, its effects on the cerebral blood flow (CBF) physiology and intrinsic neuronal activity of anesthetized brains remain poorly understood. METHODS: Four healthy adult rhesus monkeys were anesthetized initially with alfaxalone (0.125 mg/kg/min) or ketamine (1.6 mg/kg/min) for 50 min, then administrated with 0.8% isoflurane for 60 min. Heart rates, breathing beats, and blood pressures were continuously monitored. CBF data were collected using pseudo-continuous

Challenges of bringing a new sedative to market!

PURPOSE OF REVIEW: The current review examines the success and failures of the development of new hypnotic compounds for the human market. One of the important aspects is that one of the key present agents, propofol, is considered by many anaesthesiologists as 'the ideal'. However, all drugs have adverse or side-effects. RECENT FINDINGS: The last 30 years since the introduction of propofol has seen many new compounds evaluated; but as at the present time, only three agents may achieve a pivotal position in the market - fospropofol (a sedative agent which may have a role in endoscopic surgery)

Synthesis and pharmacological evaluation of neurosteroid photoaffinity ligands.

Neuroactive steroids are potent positive allosteric modulators of GABAA receptors (GABAAR), but the locations of their GABAAR binding sites remain poorly defined. To discover these sites, we synthesized two photoreactive analogs of alphaxalone, an anesthetic neurosteroid targeting GABAAR, 11β-(4-azido-2,3,5,6-tetrafluorobenzoyloxy)allopregnanolone, (F4N3Bzoxy-AP) and 11-aziallopregnanolone (11-AziAP). Both photoprobes acted with equal or higher potency than alphaxalone as general anesthetics and potentiators of GABAAR responses, left-shifting the GABA concentration - response curve for human α

A comparison of cardiopulmonary and anesthetic effects of an induction dose of alfaxalone or propofol in dogs.

OBJECTIVE: To compare the physiological parameters, arterial blood gas values, induction quality, and recovery quality after IV injection of alfaxalone or propofol in dogs. STUDY DESIGN: Prospective, randomized, blinded crossover. ANIMALS: Eight random-source adult female mixed-breed dogs weighing 18.7 ± 4.5 kg. METHODS: Dogs were assigned to receive up to 8 mg kg(-1) propofol or 4 mg kg(-1) alfaxalone, administered to effect, at 10% of the calculated dose every 10 seconds. They then received the alternate drug after a 6-day washout. Temperature, pulse rate, respiratory rate, direct blood p

Effects of Saffan administered intravenously in the horse.

Saffan was injected intravenously on 41 occasions in 11 horses and ponies to investigate its possible use in clinical equine anaesthesia. The optimum dose for induction was 1-90 mg per kg. This dose was divided into two halves, the first half given in five seconds and the second half, containing suxamethonium chloride 0.1 mg per kg, in the next 10 seconds. Induction was associated with excitement for up to 30 secs after the assumption of recumbency. At this dose rate anaesthesia lasted five to eight minutes. Muscle relaxation was poor. Recovery was associated with marked tactile and audible hy

Retrospective evaluation of the induction of anaesthesia with alfaxalone or propofol in cats undergoing caesarean section.

OBJECTIVES: Several anaesthetic protocols for caesarean section (c-section) have been described in dogs; however, anaesthesiological studies in cats undergoing c-section are lacking. The aim of this study was to assess the viability and outcome of kittens born from queens undergoing emergency c-section and receiving propofol or alfaxalone for anaesthetic induction and isoflurane for maintenance of general anaesthesia. METHODS: All cats admitted to the Veterinary Teaching Hospital of the University of Bologna between January 2014 and December 2022 for dystocia that had undergone an emergency c
