Dynamical quorum sensing and synchronization in large populations of chemical oscillators.

Populations of certain unicellular organisms, such as suspensions of yeast in nutrient solutions, undergo transitions to coordinated activity with increasing cell density. The collective behavior is believed to arise through communication by chemical signaling via the extracellular solution. We studied large, heterogeneous populations of discrete chemical oscillators (approximately 100,000) with well-defined kinetics to characterize two different types of density-dependent transitions to synchronized oscillatory behavior. For different chemical exchange rates between the oscillators and the su

Pseudomonas aeruginosa from canine otitis externa exhibit a quorum sensing deficiency.

Pseudomonas aeruginosa LasB elastase gene (lasB) transcription depends on cell density-dependent quorum-sensing mechanisms of gene activation. Previously, we collected several non-mucoid P. aeruginosa veterinary isolates and showed that the total matrix protease phenotype was similar for isolates regardless of host and site of isolation. In contrast, isolates from chronic canine ear infections (otitis externa) were significantly more likely to exhibit less elastase activity as measured by elastin Congo red than from any other site [Clin. Diag. Lab. Immun. 8 (2001) 632]. In this study, we found

LuxS-mediated quorum sensing in Borrelia burgdorferi, the lyme disease spirochete.

The establishment of Borrelia burgdorferi infection involves numerous interactions between the bacteria and a variety of vertebrate host and arthropod vector tissues. This complex process requires regulated synthesis of many bacterial proteins. We now demonstrate that these spirochetes utilize a LuxS/autoinducer-2 (AI-2)-based quorum-sensing mechanism to regulate protein expression, the first system of cell-cell communication to be described in a spirochete. The luxS gene of B. burgdorferi was identified and demonstrated to encode a functional enzyme by complementation of an Escherichia coli l

Engineered Phage Modulates Quorum Sensing and Biofilm Formation in Pseudomonas aeruginosa.

Pseudomonas aeruginosa is an opportunistic Gram-negative pathogen frequently associated with chronic and biofilm-related infections, largely driven by quorum sensing (QS)-related genes/phenotypes. In this study, we investigated the antivirulence activity of an engineered M13-derived phage-display particle (P9b), selected for specific binding to P. aeruginosa, which acts as a non-lytic modulator of QS through specific binding to a bacterial surface target. P9b induced a transient delay in early planktonic growth, without affecting long-term proliferation. In contrast, P9b significantly reduced

Natural Lactone-Based Quorum Sensing Inhibitors Attenuate Virulence in Pseudomonas aeruginosa.

Natural lactones represent a promising anti-virulence strategy against Pseudomonas aeruginosa by targeting quorum-sensing (QS) pathways. In this work, we investigate γ-caprolactone (Capro), γ-octalactone (Octa), γ-decalactone (GDL), δ-decalactone (DDL), and a laboratory-generated lactone mixture (Lx) as modulators of QS-regulated virulence. Treatment with γ-decalactone and engineered lactone formulations significantly downregulated rhlR expression (∼twofold) and suppressed pyocyanin and rhamnolipid production. At 48 h, lactone exposure reduced extracellular virulence factors by up to 74.81% (p

A Critical Review of Quorum-Sensing Inhibition Strategies in Klebsiella Pneumoniae.

Klebsiella pneumoniae is an antibiotic-resistant microbe that causes pneumonia, urinary tract infections, bloodstream infections, and liver abscesses. Its pathogenicity is fueled by the polysaccharide capsule, lipopolysaccharides, and quorum-sensing-regulated biofilm formation, which enhance immune evasion and antibiotic resistance. The rise of multidrug-resistant and hypervirulent pathogens poses a significant challenge to treatment. Targeting quorum sensing with quorum quenching is a promising method for disrupting bacterial interaction and reducing virulence without increasing resistance. T

Multidrug-resistant bacteria quorum-sensing inhibitors: A particular focus on Pseudomonasaeruginosa.

Many widely used conventional antibiotics have failed to show clinical efficacy against Pseudomonas aeruginosa (P. aeruginosa) due to the strain's rising resistance to most clinically relevant antimicrobials. P. aeruginosa uses quorum sensing to regulate its virulence and biofilm development, which contributes to its pathogenicity and drug resistance. This mechanism is responsible for the resurgence and persistence of infections. Therefore, targeting the virulence and pathogenicity of P. aeruginosa through quorum sensing (QS) is regarded as a potential target for anti-infective therapy. Howeve

Combatting biofilm-mediated infections in clinical settings by targeting quorum sensing.

Biofilm-associated infections constitute a significant challenge in managing infectious diseases due to their high resistance to antibiotics and host immune responses. Biofilms are responsible for various infections, including urinary tract infections, cystic fibrosis, dental plaque, bone infections, and chronic wounds. Quorum sensing (QS) is a process of cell-to-cell communication that bacteria use to coordinate gene expression in response to cell density, which is crucial for biofilm formation and maintenance.. Its disruption has been proposed as a potential strategy to prevent or treat biof

Synthetic bacterial therapies for intestinal diseases based on quorum-sensing circuits.

Bacterial therapy has become a key strategy against intestinal infectious diseases in recent years. Moreover, regulating the gut microbiota through traditional fecal microbiota transplantation and supplementation of probiotics faces controllability, efficacy, and safety challenges. The infiltration and emergence of synthetic biology and microbiome provide an operational and safe treatment platform for live bacterial biotherapies. Synthetic bacterial therapy can artificially manipulate bacteria to produce and deliver therapeutic drug molecules. This method has the advantages of solid controllab

Metabolic reprogramming of Pseudomonas aeruginosa by phage-based quorum sensing modulation.

The Pseudomonas quinolone signal (PQS) is a multifunctional quorum sensing molecule of key importance to P. aeruginosa. Here, we report that the lytic Pseudomonas bacterial virus LUZ19 targets this population density-dependent signaling system by expressing quorum sensing targeting protein (Qst) early during infection. We demonstrate that Qst interacts with PqsD, a key host quinolone signal biosynthesis pathway enzyme, resulting in decreased levels of PQS and its precursor 2-heptyl-4(1H)-quinolone. The lack of a functional PqsD enzyme impairs LUZ19 infection but is restored by external supplem

Quorum Sensing Communication: Molecularly Connecting Cells, Their Neighbors, and Even Devices.

Quorum sensing (QS) is a molecular signaling modality that mediates molecular-based cell-cell communication. Prevalent in nature, QS networks provide bacteria with a method to gather information from the environment and make decisions based on the intel. With its ability to autonomously facilitate both inter- and intraspecies gene regulation, this process can be rewired to enable autonomously actuated, but molecularly programmed, genetic control. On the one hand, novel QS-based genetic circuits endow cells with smart functions that can be used in many fields of engineering, and on the other, r

A quorum sensing-independent path to stumpy development in Trypanosoma brucei.

For persistent infections of the mammalian host, African trypanosomes limit their population size by quorum sensing of the parasite-excreted stumpy induction factor (SIF), which induces development to the tsetse-infective stumpy stage. We found that besides this cell density-dependent mechanism, there exists a second path to the stumpy stage that is linked to antigenic variation, the main instrument of parasite virulence. The expression of a second variant surface glycoprotein (VSG) leads to transcriptional attenuation of the VSG expression site (ES) and immediate development to tsetse fly inf

Innovative application of ceftriaxone as a quorum sensing inhibitor in Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a Gram-negative bacterium that has the potential to induce various healthcare-related infections through its array of virulence factors. The control of virulence factor expression is mainly regulated by a communication process among cells called quorum sensing (QS). Blocking QS could be a viable tactic to suppress virulence factors and reduce pathogenicity without impacting bacterial growth. This approach has the potential to significantly decrease the multiple drug resistance emergence. In this study, we explored the impact of ceftriaxone (CRO), which is a commonly u

Yersiniabactin is a quorum-sensing autoinducer and siderophore in uropathogenic Escherichia coli.

Siderophores are secreted ferric ion chelators used to obtain iron in nutrient-limited environmental niches, including human hosts. While all Escherichia coli express the enterobactin (Ent) siderophore system, isolates from patients with urinary tract infections additionally express the genetically distinct yersiniabactin (Ybt) siderophore system. To determine whether the Ent and Ybt systems are functionally redundant for iron uptake, we compared the growth of different isogenic siderophore biosynthetic mutants in the presence of transferrin, a human iron-binding protein. We observed that Ybt

Attenuation of quorum sensing system and virulence in Vibrio cholerae by phytomolecules.

The Vibrio cholerae, a gram-negative bacterium, is the causative agent of cholera. Quorum sensing is a cell-to-cell communication that leads to gene expression, accumulation of signaling molecules, biofilm formation, and production of virulence factors. The quorum sensing pathway in V. cholerae is regulated by luxO, and biofilm formation and other virulence factors are positively controlled by aphA and negatively by hapR. Hence, targeting the global regulator luxO would be a promising approach to modulate the QS to curtail V. cholerae pathogenesis. The present study investigated the modulating

The role of Staphylococcus aureus quorum sensing in cutaneous and systemic infections.

BACKGROUND: Staphylococcus aureus is a leading cause of human bacterial infections worldwide. It is the most common causative agent of skin and soft tissue infections, and can also cause various other infections, including pneumonia, osteomyelitis, as well as life-threatening infections, such as sepsis and infective endocarditis. The pathogen can also asymptomatically colonize human skin, nasal cavity, and the intestine. S. aureus colonizes approximately 20-30% of human nostrils, being an opportunistic pathogen for subsequent infection. Its strong ability to silently spread via human contact m

Relay Architectures and Ecological Strategies of Quorum Sensing Effects on Microbial Interactions.

Microbial communities assemble in heterogeneous habitats, where quorum sensing (QS) influences interactions beyond signalling populations. Yet QS studies are commonly organized by signal type, circuit, or phenotype, obscuring the routes by which a QS event reaches neighbouring populations and why they prevail in particular settings. This Review synthesizes evidence from host-associated, terrestrial, marine, and aquatic systems to develop a two-dimensional framework for QS-mediated ecological effects. The framework distinguishes three relay architectures by which QS activity in one population a

[Research Progress on Quorum Sensing-Regulated Oral Microbial Interaction Networks in Periodontitis].

Quorum sensing (QS) is a mechanism by which bacteria communicate and coordinate group behaviors through the secretion of signaling molecules, playing a critical role in oral microbial interactions and the pathogenesis of periodontitis. This review summarizes recent advances in QS-mediated regulation of oral microbial interaction networks in periodontitis, focusing on the molecular mechanisms that modulate biofilm formation, virulence factor expression, and pathogen community dynamics. In addition, novel QS-targeted therapeutic strategies are discussed. Although periodontitis is fundamentally a

Visible-light-based modulation of bacterial quorum sensing with photoswitchable signal molecules.

Controlling bacterial collective behaviour through manipulation of the quorum sensing (QS) system presents applicability for various ends, in both the inhibition and induction of these bacterial regulatory circuits using external signals. In addition to native functions, the repurposing of natural control elements for biotechnological applications has also garnered interest. Homoserine lactone (HSL) mediated QS has previously been coupled to light signals by photoswitchable signal molecule mimics; however, existing compounds have made use of UV-responsive azobenzenes, which may limit their use
