Crosstalk between bile acid-activated receptors and microbiome in entero-hepatic inflammation.

Bile acids are potent signaling molecules exerting diverse actions through bile acid-activated receptors. Among them, the Farnesoid X receptor (FXR) and the Takeda G protein-coupled receptor 5 (TGR5; GPBAR1), modulate the inflammation occurring in chronic/acute hepatitis, cholestasis, and inflammatory bowel disease. A role for other bile acid-responsive receptors in this context is emerging. This review aims to summarize recent advances on the immune-modulatory actions of the bile acid-responsive receptors Shingosine-1-phosphate receptor 2 (S1PR2), pregnane X receptor (PXR), constitutive andro

Intestinal Sirtuin 1 Controls Liver Regeneration via Regulating the Intestinal Farnesoid X Receptor/Fibroblast Growth Factor 15 Axis in Mice.

BACKGROUND & AIMS: The contribution of the gut-liver axis during liver regeneration remains poorly understood. Understanding the mechanisms underlying this process is critical for developing novel therapeutic strategies for liver diseases and transplantation. Here, we aimed at defining the role of intestinal sirtuin-1 during liver regeneration in mice. METHODS: We performed partial hepatectomy to intestinal-specific sirtuin-1 knockout mice (SIRTintKO) and wild-type littermates. Immunostaining and immunoblotting were performed on liver and intestinal tissues to assess proliferation and senesce

Work in Progress: Drugs in Development.

Primary biliary cholangitis is an archetypal autoimmune disease that causes cholestasis, fibrosis, and liver failure. Ursodeoxycholic acid and obeticholic acid are approved for its treatment. Not all patients respond, some are intolerant, many have ongoing symptoms, and new therapies are required. Herein we describe drugs in development and potential future biological targets. We consider compounds acting on the farnesoid X receptor/fibroblast growth factor 19 pathway, fibrates and other agonists of the peroxisome proliferator-activated receptor family, transmembrane-G-protein-receptor-5 agoni

Farnesoid X receptor agonists in biliary tract disease.

The farnesoid X receptor (FXR) is a member of ligand-activated nuclear receptor superfamily. FXR is a bile sensor and is part of a complex network of nuclear receptors that includes also the constitutive androstane receptor and the pregnane X receptor. These receptors act coordinately to regulate essential steps of bile acids and xenobiotics uptake, metabolism and excretion in hepatocytes, cholangiocytes and kidney cells. Preclinical models indicate that FXR agonists are effective in reducing liver injury in nonobstructive models of cholestasis. FXR ligands are currently under investigation fo

Nuclear Receptor Modulation for the Treatment of Nonalcoholic Fatty Liver Disease.

Nuclear receptors (NRs) are ligand-activated transcriptional regulators of several key metabolic processes including hepatic lipid and glucose metabolism, bile acid homeostasis, and energy expenditure as well as inflammation, fibrosis, and cellular proliferation in the liver. Dysregulation of these processes contributes to the pathogenesis and progression of nonalcoholic fatty liver disease (NAFLD). This places NRs at the forefront of novel therapeutic approaches for NAFLD. Some NRs are already pharmacologically targeted in metabolic disorders such as hyperlipidemia (peroxisomal proliferator-a

Pulsatile FXR activation for liver diseases.

Sustained receptor activation is often prioritized in drug development. Zang and colleagues challenge this paradigm with linafexor, a rapidly cleared farnesoid X receptor agonist that produces pulsatile activation, preserves receptor responsiveness, reduces broad transcriptional disruption, and improves efficacy and safety, highlighting temporal alignment as a design principle for metabolic therapeutics.

Lipidome remodeling in primary biliary cholangitis.

Primary biliary cholangitis (PBC) is a chronic, cholestatic liver disease characterized by progressive destruction of intrahepatic bile ducts, impaired bile flow, and complex disturbances in bile acid and lipid metabolism. PBC is associated with a distinctive dyslipidemic profile, marked by elevated total cholesterol, accumulation of lipoprotein X, and paradoxically high HDL levels. At the molecular level, dysregulation of bile acid-activated nuclear receptors - particularly farnesoid X receptor (FXR) and peroxisome proliferator-activated receptors (PPARs) - contributes to abnormalities in cho

Gut microbiota-derived deoxycholic acid shapes an immunosuppressive tumor microenvironment and promotes breast cancer progression.

The gut microbiota is increasingly recognized as a contributor to breast cancer progression. Here, we report that the gut bacterium Enterocloster bolteae, a member of the Lachnospiraceae family, is progressively enriched during tumor development and is associated with increased levels of the microbiota-derived metabolite deoxycholic acid. Deoxycholic acid accumulates in tumors and activates the farnesoid X receptor in tumor cells, inducing interleukin-6 production through nuclear factor κB signaling. Interleukin-6 promotes the recruitment of granulocytic myeloid-derived suppressor cells and T

Use of dominant negative nuclear receptors to study xenobiotic-inducible gene expression in primary cultured hepatocytes.

INTRODUCTION: To determine the feasibility of using dominant negative nuclear receptors to dissect the regulation of inducible gene expression in primary cultured hepatocytes, a series of dominant negative nuclear receptor expression plasmids were designed with truncated AF-2 subdomains. METHODS: Plasmids expressing dominant negative or wild-type constitutive androstane receptor (CAR), pregnane X receptor (PXR), farnesoid X receptor (FXR), liver X receptor (LXR), or peroxisome proliferator-activated receptor alpha (PPARalpha) were transiently cotransfected into primary cultured rat hepatocyte

Spatial metabolite profiling identifies taurocholic acid as a protective mediator in hepatic ischemia-reperfusion injury: A mass spectrometry imaging and pharmacological study.

Hepatic ischemia-reperfusion injury (IRI) remains a major clinical obstacle during liver surgery and liver transplantation, and effective targeted intervention strategies are still limited. In the present study, we combined mass spectrometry imaging, in vitro cellular experiments, and in vivo animal tests to explore key functional metabolites involved in the progression of hepatic IRI. Spatial metabolite profiling via mass spectrometry imaging systematically mapped metabolic disturbances in liver tissues during IRI progression and confirmed that taurocholic acid (TCA) undergoes distinct spatia

Environmental Chemical Contribution to the Modulation of Bile Acid Homeostasis and Farnesoid X Receptor Signaling.

Maintaining bile acid (BA) homeostasis is important and regulated by BA activated receptors and signaling pathways. Farnesoid X receptor (FXR) and its regulated target networks in both the liver and the intestines are critical in suppressing BA synthesis and promoting BA transport and enterohepatic circulation. In addition, FXR is critical in regulating lipid metabolism and reducing inflammation, processes critical in the development of cholestasis and fatty liver diseases. BAs are modulated by, but also control, gut microflora. Environmental chemical exposure could affect liver disease develo

Obeticholic acid prevents carbon tetrachloride-induced liver fibrosis through interaction between farnesoid X receptor and Smad3.

Liver fibrosis results from sustained liver injury and is characterized by inflammation, hepatic stellate cell (HSC) activation, extracellular matrix (ECM) accumulation and liver structure destruction. The Farnesoid-X receptor (FXR) antagonizes toxic liver injury and fibrosis, yet the mechanism in liver fibrosis remains unclear. We investigated the effects of FXR agonist obeticholic acid (OCA) on liver fibrosis in mice. Mice were injected with carbon tetrachloride (CCl4) for 3 weeks or 6 weeks to induce liver fibrosis. OCA (5 mg/kg) or PBS is administered daily during CCl4-treatment. At sacrif

Modulating bile acid pathways and TGR5 receptors for treating liver and GI diseases.

Bile acids are central signals in enterohepatic communication and also integrate microbiota-derived signals into this signaling axis. Discovery of the tissue distribution and signaling pathways activated by the natural receptors for bile acids, farnesoid X receptor and G protein-coupled bile acid receptor 1 (GPBAR1) also known as TGR5, and bile acid transporters has led to the development of therapeutic agents that target these molecules. Obeticholic acid, a selective FXR agonist, and NGM282, a non-mitogenic FGF-19 analog, are two of the agents in this pipeline. Obeticholic acid has been appro

Clinical Trials in PBC Going Forward.

New treatments for primary biliary cholangitis (PBC) are progressively emerging, including first and second generations of farnesoid X receptor and peroxisome proliferator-activated receptors agonists. Even though ursodeoxycholic acid monotherapy remains the standard of care treatment for PBC, these additional therapeutic options, already or soon to be available, lead us to revise our priorities and strategies with respect to future clinical trials. The present article is a personal view of where we currently stand in this field and where and how we should be going to achieve new progress.

The enabling power of chemical probes: examples from the nuclear hormone receptor field.

Nuclear receptors (NRs) are ligand-regulated transcription factors that control many physiological processes, from metabolism and inflammation to development and circadian rhythms. Beginning in the 1990s, systematic chemical probe development transformed many 'orphan' NRs with unknown endogenous ligands into chemically tractable, well-studied molecular targets, uncovering their physiological functions and potential for therapeutic development. The present review examines how small molecule ligands for peroxisome proliferator-activated receptors, liver X receptors, farnesoid X receptor, pregnan

Bile acids induce visceral hypersensitivity via mucosal mast cell-to-nociceptor signaling that involves the farnesoid X receptor/nerve growth factor/transient receptor potential vanilloid 1 axis.

Increased colonic bile acid (BA) exposure, frequent in diarrhea-predominant irritable bowel syndrome (IBS-D), can affect gut function. Nerve growth factor (NGF) is implicated in the development of visceral hypersensitivity (VH). In this study, we tested the hypothesis that BAs cause VH via mucosal mast cell (MMC)-to-nociceptor signaling, which involves the farnesoid X receptor (FXR)/NGF/transient receptor potential vanilloid (TRPV)1 axis. BAs were intracolonically administered to rats for 15 d. Visceral sensitivity to colorectal distention and colonic NGF expression were examined. BAs caused V

Beta-catenin/sirtuin 1/farnesoid X receptor pathway promotion of portal vein ligation and parenchymal transection-induced rapid liver regeneration.

BACKGROUND: By accelerating the regeneration of the future liver remnant, portal vein ligation and parenchymal transection allows for more extensive hepatectomy. Given that the mechanism remains poorly understood, the aim of this study was to investigate the mechanism of portal vein ligation and parenchymal transection-induced liver regeneration. METHODS: A portal vein ligation and parenchymal transection-induced liver regeneration mouse model was established, followed by RNA microarray analysis to identify candidate molecules. Liver-targeting drugs and their effect on blood glucose and hepatic lipids.

The global epidemic of non-alcoholic fatty liver disease (NAFLD) and steatohepatitis (NASH) and the high prevalence among individuals with type 2 diabetes has attracted the attention of clinicians specialising in liver disorders. Many drugs are in the pipeline for the treatment of NAFLD/NASH, and several glucose-lowering drugs are now being tested specifically for the treatment of liver disease. Among these are nuclear hormone receptor agonists (e.g. peroxisome proliferator-activated receptor agonists, farnesoid X receptor agonists and liver X receptor agonists), fibroblast growth factor-19 an

Linking liver metabolic and vascular disease via bile acid signaling.

Non-alcoholic fatty liver disease (NAFLD) is a metabolic disorder affecting over one quarter of the global population. Liver fat accumulation in NAFLD is promoted by increased de novo lipogenesis leading to the development of a proatherosclerotic lipid profile and atherosclerotic cardiovascular disease (CVD). The CVD component of NAFLD is the main determinant of patient outcome. The farnesoid X receptor (FXR) and the G protein bile acid-activated receptor 1 (GPBAR1) are bile acid-activated receptors that modulate inflammation and lipid and glucose metabolism in the liver and CV system, and are

Development of bile acid activated receptors hybrid molecules for the treatment of inflammatory and metabolic disorders.

The farnesoid-x-receptor (FXR) and the G protein bile acid activated receptor (GPBAR)1 are two bile acid activated receptors highly expressed in entero-hepatic, immune, adipose and cardiovascular tissues. FXR and GPBAR1 are clinically validated targets in the treatment of metabolic disorders and FXR agonists are currently trialled in patients with non-alcoholic steato-hepatitis (NASH). Results of these trials, however, have raised concerns over safety and efficacy of selective FXR ligands suggesting that the development of novel agent designed to impact on multiple targets might have utility i
