Identification of blood-feeding sources in Panstrongylus, Psammolestes, Rhodnius and Triatoma using amplicon-based next-generation sequencing.

BACKGROUND: Triatomines are hematophagous insects that play an important role as vectors of Trypanosoma cruzi, the causative agent of Chagas disease. These insects have adapted to multiple blood-feeding sources that can affect relevant aspects of their life-cycle and interactions, thereby influencing parasitic transmission dynamics. We conducted a characterization of the feeding sources of individuals from the primary circulating triatomine genera in Colombia using amplicon-based next-generation sequencing (NGS). METHODS: We used 42 triatomines collected in different departments of Colombia.

A phospholipase A2 gene is linked to Jack bean urease toxicity in the Chagas' disease vector Rhodnius prolixus.

BACKGROUND: Ureases are multifunctional enzymes that display biological activities independent of their enzymatic function, including exocytosis induction and insecticidal effects. The hemipteran Rhodnius prolixus is one of the known susceptible models for this toxicity. It has been shown that Jack bean urease (JBU) has deleterious effects on R. prolixus, and these effects are modulated by eicosanoids, which are synthesized in a cascade involving phospholipase A2 (PLA2) enzymes. METHODS: R. prolixus genome was screened for putative PLA2s and matching transcripts were cloned. Predicted amino a

Gut microbiota, blood feeding sources, and Trypanosoma cruzi infection in Rhodnius colombiensis from center-west of Colombia.

Chagas disease, caused by Trypanosoma cruzi, remains a major public health concern in Latin America. In Tolima, Colombia, the vector R. colombiensis has acquired epidemiological relevance due to its ability to colonize domiciliary environments and its high rates of T. cruzi infection, which increase transmission in areas where primary vectors such as R. prolixus have been controlled. In this study, we characterized the gut microbiome of R. colombiensis using culture-dependent and culture-independent approaches, including next-generation sequencing (NGS) of the 16S rRNA gene, specific detection

Jack bean (Canavalia ensiformis) urease induces eicosanoid-modulated hemocyte aggregation in the Chagas' disease vector Rhodnius prolixus.

Ureases are multifunctional proteins that display biological activities independently of their enzymatic function, such as induction of exocytosis and insecticidal effects. Rhodnius prolixus, a major vector of Chagas' disease, is a model for studies on the entomotoxicity of jack bean urease (JBU). We have previously shown that JBU induces the production of eicosanoids in isolated tissues of R. prolixus. In insects, the immune response comprises cellular and humoral reactions, and is centrally modulated by eicosanoids. Cyclooxygenase products signal immunity in insects, mainly cellular reaction

Rhodnius prolixus Colonization and Trypanosoma cruzi Transmission in Oil Palm (Elaeis guineensis) Plantations in the Orinoco Basin, Colombia.

Trypanosoma cruzi is the etiological agent of Chagas disease that infects more than seven million people in Latin America. The parasite is transmitted by triatomine insects, of which some species are often associated with palms. The establishment of oil palm plantations (Elaeis guineensis) in the Orinoco region (Colombia) has been rapidly growing, possibly constituting a new environment for the establishment and increase in triatomine populations. In this study, the potential of Rhodnius prolixus to colonize E. guineensis plantations and maintain T. cruzi transmission was assessed. Fieldwork w

Domiciliary biting frequency and blood ingestion of the Chagas's disease vector Rhodnius prolixus Ståhl (Hemiptera: Reduviidae), in Venezuela.

Demolition of a rural house in the State of Cojedes, Venezuela, provided a collection of 7.934 Rhodnius prolixus of which a random sample of 1,415 was weighed within 48 hours. The field weights, coupled with laboratory information of weight loss (in %) with time, average blood ingestion and meal size sufficient to promote moulting, were used to estimate biting rate under domiciliary conditions. The results show that in this particularly highly infested house, the R. prolixus population bites, on the average, at a rate of 58 times/person/day, draining blood at a rate of about 100 cm3/person/mon

Histology and histochemistry of the accessory gland of the female reproductive tract of Rhodnius neglectus Lent, 1954 (Hemiptera: Reduviidae).

Rhodnius neglectus is a wild triatomine, vector of the protozoan Trypanosoma cruzi, which causes Chagas' disease, and feeds on the blood of small mammals, being essential for its growth and reproduction. Accessory glands of the female reproductive tract are important in insect reproduction, but their anatomy and histology in R. neglectus are poorly studied. The aim of this work was to describe the histology and histochemistry of the accessory gland of the female reproductive tract of R. neglectus. The reproductive tract of five females of R. neglectus was dissected and the accessory glands tra

Genome of Rhodnius prolixus, an insect vector of Chagas disease, reveals unique adaptations to hematophagy and parasite infection.

Rhodnius prolixus not only has served as a model organism for the study of insect physiology, but also is a major vector of Chagas disease, an illness that affects approximately seven million people worldwide. We sequenced the genome of R. prolixus, generated assembled sequences covering 95% of the genome (∼ 702 Mb), including 15,456 putative protein-coding genes, and completed comprehensive genomic analyses of this obligate blood-feeding insect. Although immune-deficiency (IMD)-mediated immune responses were observed, R. prolixus putatively lacks key components of the IMD pathway, suggesting

Shelter selection in the kissing bugs Triatoma infestans and Rhodnius prolixus: Intra- and inter-specific interactions and quality evaluation.

Triatomines display most of their activities during the night. Before sunrise, they search, select and occupy adequate shelters to stay during the photophase, avoiding exposure to diurnal predators. In this work, we first explored the interactions between individuals of the same or different species in the shelter selection process of Triatoma infestans (Klug 1834) and Rhodnius prolixus (Stål 1859). When two groups of insects (either of different nutritional status, nymphal instar or species) were released together over an experimental arena containing two identical shelters, all nymphs were d

Could the Chagas disease elimination programme in Venezuela be compromised by reinvasion of houses by sylvatic Rhodnius prolixus bug populations?

The Andean Pact Initiative (1997) committed Andean countries to eliminate vectorial transmission of Chagas disease by 2010 via widespread residual insecticide spraying. In Venezuela, this aim could be compromised by reinvasion of houses by palm tree populations of the major vector Rhodnius prolixus. To test this hypothesis, a multivariate logistic regression was undertaken of risk factors for triatomine infestation and colonization in 552 houses and 1068 peri-domestic outbuildings in Barinas State. After adjusting for other risk factors, including palm roofs, R. prolixus infestation and coloni

Genotyping of Trypanosoma cruzi DTUs and Trypanosoma rangeli genetic groups in experimentally infected Rhodnius prolixus by PCR-RFLP.

The specific detection and genetic typing of trypanosomes that infect humans, mammalian reservoirs, and vectors is crucial for diagnosis and epidemiology. We utilized a PCR-RFLP assay that targeted subunit II of cytochrome oxidase and 24Sα-rDNA to simultaneously detect and discriminate six Trypanosoma cruzi discrete typing units (DTUs) and two genetic groups of Trypanosoma rangeli (KP1+/KP1-) in intestinal contents of experimentally infected Rhodnius prolixus. The PCR assays showed that in 23 of 29 (79.4%) mixed infections with the six T. cruzi DTUs and mixed infections with individual DTUs an

Functional characterization of vitellogenin unveils novel roles in RHBP uptake and lifespan regulation in the insect vector Rhodnius prolixus.

In insects, vitellogenesis plays a critical role in providing the energy reserves needed for embryonic development as it ensures the accumulation of yolk in the oocytes. Vitellogenin (Vg), the precursor to vitellin (Vt), is primarily synthesized in the fat body of females and transported to the oocytes via receptor-mediated endocytosis. In Rhodnius prolixus, a key vector of Chagas disease, two Vg genes, Vg1 and Vg2, were characterized. These genes share 65 % amino acid identity and present the conserved Vitellogenin_N, DUF1943, and VWD domains typical of Vg proteins across various species. We

Gonadulin: A newly discovered insulin-like peptide involved in ovulation and oviposition in Rhodnius prolixus, a vector of Chagas disease.

Insulin-like peptides (ILPs) are vital hormones involved in a wide range of physiological processes in all organisms. In insects, insulin signaling has a key role in detecting and interpreting nutrient levels for egg production. Based on publicly available transcriptomes, a new ILP named gonadulin has been reported and suggested to be expressed by the gonads (hence its name). Although the identification of gonadulin establishes its existence, its physiological relevance remains poorly understood. A rhamnose-binding lectin from Rhodnius prolixus and the impact of its silencing on gut bacterial microbiota and Trypanosoma cruzi.

Lectins are ubiquitous proteins involved in the immune defenses of different organisms and mainly responsible for non-self-recognition and agglutination reactions. This work describes molecular and biological characterization of a rhamnose-binding lectin (RBL) from Rhodnius prolixus, which possesses a 21 amino acid signal peptide and a mature protein of 34.6 kDa. The in-silico analysis of the primary and secondary structures of RpLec revealed a lectin domain fully conserved among previous insects studied. The three-dimensional homology model of RpLec was similar to other RBL-lectins. Docking p

Rhodnius prolixus smells repellents: Behavioural evidence and test of present and potential compounds inducing repellency in Chagas disease vectors.

Insect repellents are known since many decades ago and constitute a major tool for personal protection against the biting of mosquitoes. Despite their wide use, the understanding of why and how repellents repel is relatively recent. In particular, the question about to what extent insects other than mosquitoes are repulsed by repellents remains open. We developed a series of bioassays aimed to test the performance of well established as well as potential repellent molecules on the Chagas disease vector Rhodnius prolixus. Besides testing their ability to prevent biting, we tested the way in whi

Dual silencing of lipophagy and lipolysis in Rhodnius prolixus induces lipid droplet remodeling without TAG accumulation in the fat body.

Autophagy and lipolysis are key metabolic pathways involved in lipid mobilization and energy homeostasis. In Rhodnius prolixus, a major vector of Chagas disease, previous studies have shown that silencing autophagy-related genes (RpAtg8, RpAtg6) or the brummer lipase gene RpBmm alters lipid metabolism and impairs flight activity. Here, we investigate the combined roles of lipolysis and lipophagy by performing a dual RNAi-mediated silencing of RpAtg8 and RpBmm under both fed and starved conditions. In the fat body, dual silencing did not exacerbate triacylglycerol (TAG) accumulation beyond sing

Correlation between populations of Rhodnius and presence of palm trees as risk factors for the emergence of Chagas disease in Amazon region, Brazil.

Among the states that comprise the legally defined Amazon region of Brazil, Pará has presented the highest occurrences of acute cases of Chagas disease over the last two decades. These cases have been attributed to consumption of fruits from native palm trees. In surveys in rural and wild areas of the municipality of Oriximiná, Pará, triatomine fauna, their main ecotopes and the infection rate due to Trypanosoma cruzi were identified using active and passive search methods: manual capture and Noireau traps, respectively. A total of 582 ecotopes were surveyed using 1496 Noireau traps. Out of 44

Investigating the distribution of a rare Colombo-Venezuelan kissing bug, Rhodnius neivai, Lent, 1953, using geographical information system-based analyses.

BACKGROUND: Rhodnius neivai, a kissing bug found in the dry regions of Colombia and Venezuela, has limited documented occurrences. While it is not deemed a significant vector for Chagas disease, distributional and ecological studies are essential in monitoring species domiciliation and shedding light on the evolutionary aspects of the Rhodniini tribe. OBJECTIVES: The study aims to provide a detailed revision of R. neivai distribution and evaluate general spatial data quality for ecological niche modelling (ENM). It will also provide the first published ENM for the species, which may aid speci

Triatoma venosa and Panstrongylus geniculatus challenge the certification of interruption of vectorial Trypanosoma cruzi transmission by Rhodnius prolixus in eastern Colombia.

Reactivation of Trypanosoma cruzi transmission by native vectors with different domiciliation capabilities is a major concern for Chagas disease control programs. T. cruzi transmission via intra-domestic Rhodnius prolixus was certified as interrupted by the Pan American Health Organization in Miraflores municipality (Boyacá, Colombia) in 2019. However, Triatoma venosa, a native vector infected with T. cruzi has been increasingly found inside human dwellings across rural areas.
